| 1 | Biocompatibility of bio-Mg-Zn alloy within bone with heart, liver, kidney and spleen显示文摘A magnesium-zinc alloy rod was implanted into the marrow cavity of the distal femur in New Zealand rabbits. The femur with the implanted alloy was compared with the contralateral femur in which a bone tunnel without implant was formed as a control. Degradation of the magnesium-zinc alloy was analyzed via X-ray, scanning electron microscopy, and element energy spectrum analysis. Serum magnesium, liver and kidney function tests, and myocardial enzymes were measured. Heart, liver, kidney and spleen were sectioned for pathological analysis, and the effects of the implanted material on the histology and function of important organs were analyzed. Magnesium-zinc alloy was resorbed from the bone marrow cavity of the femur; 87% of the alloy was degraded within 14 weeks after the surgery. There were no significant differences in serum magnesium, liver or kidney function tests, or myocardial enzymes be- fore the surgery and after degradation of the magnesium-zinc alloy. Histology of the heart, liver, kidney, and spleen did not change. | 2 | Improved dissolution and bioavailability of silymarin delivered by a solid dispersion prepared using supercritical fluids显示文摘The objective of this study was to improve the dissolution and bioavailability of silymarin(SM).Solid dispersions(SDs)were prepared using solution-enhanced dispersion by supercritical fluids(SEDS)and evaluated in vitro and in vivo,compared with pure SM powder.The particle sizes,stability,and contents of residual solvent of the prepared SM-SDs with SEDS and solvent evaporation(SE)were investigated.Four polymer matrix materials were evaluated for the preparation of SM-SD-SEDS,and the hydrophilic polymer,polyvinyl pyrrolidone K17,was selected with a ratio of 1:5 between SM and the polymer.Physicochemical analyses using X-ray diffraction and differential scanning calorimetry indicated that SM was dispersed in SD in an amorphous state.The optimized SM-SD-SEDS showed no loss of SM after storage for 6 months and negligible residual solvent(ethanol)was detected using gas chromatography.In vitro drug release was increased from the SM-SDSEDS,as compared with pure SM powder or SM-SD-SE.In vivo,the area under the rat plasma SM concentration-time curve and the maximum plasma SM concentration were 2.4-fold and 1.9-fold higher,respectively,after oral administration of SM-SD-SEDS as compared with an aqueous SM suspension.These results illustrated the potential of using SEDS to prepare SM-SD,further improving the biopharmaceutical properties of this compound. | Gang Yang Yaping Zhao Nianping Feng Yongtai Zhang Ying Liu Beilei Dang | 2015 | Asian Journal of Pharmaceutical Sciences2015,10,3: | 6 |

| 7 | Cancer cell reprogramming:a promising therapy converting malignancy to benignity显示文摘In the past decade, remarkable progress has been made in reprogramming terminally differentiated somatic cells and cancer cells into induced pluripotent cells and cancer cells with benign phenotypes. Recent studies have explored various approaches to induce reprogramming from one cell type to another, including lineage-specific transcription factors-, combinatorial small molecules-, microRNAs- and embryonic microenvironment-derived exosome-mediated reprogramming. These reprogramming approaches have been proven to be technically feasible and versatile to enable re-activation of sequestered epigenetic regions, thus driving fate decisions of differentiated cells. One of the significant utilities of cancer cell reprogramming is the therapeutic potential of retrieving normal cell functions from various malignancies. However, there are several major obstacles to overcome in cancer cell reprogramming before clinical translation, including characterization of reprogramming mechanisms, improvement of reprogramming efficiency and safety, and development of delivery methods. Recently, several insights in reprogramming mecha-nism have been proposed, and determining progress has been achieved to promote reprogramming efficiency and feasibility, allowing it to emerge as a promising therapy against cancer in the near future. | 8 | Phytolith carbon sequestration in bamboos of different ecotypes:a case study in China显示文摘Occlusion of carbon(C) within phytoliths(Phyt OC) is becoming one of the most promising terrestrial C sequestration mechanisms. This study explored the production of Phyt OC within 35 bamboo species belonging to three ecotypes using methods of microwave digestion. The aim of this study is to explore the present and potential C sequestration rate within phytoliths of bamboo species from three ecotypes. Phyt OC content in bamboos of three ecotypes ranges from 0.07 % to 0.42 %. The mean Phyt OC production flux decreases as: clustered bamboo(0.050 ±0.016 t CO2ha-1a-1) & mixed bamboo(0.049 ± 0.016 t CO2ha-1a-1) [ scattered bamboo(0.038 ± 0.020 t CO2ha-1a-1). The phytolith carbon sequestration in Chinese bamboo is estimated to be 0.293 ± 0.127 Tg(1 Tg =1012g) CO2a-1; approximately 75 %, 3 %, and 22 % of which is contributed from scattered, mixed and clustered bamboo, respectively. Taking the Phyt OC production flux of 0.18 ± 0.12 t CO2ha-1a-1and current annual area increasing rate of 3 %, global bamboo phytoliths wouldsequester 11.9 ± 7.9 Tg CO2a-1by 2050. | 12 | Different patterns of agonist-induced modulation of α_(1B)-adrenoceptor density at different initial expression levels显示文摘We compared the norepinephrine (NE) induced α1B-adrenoceptor (α1B-AR) expression modulation between two transfected human embryonic kidney (HEK) 293 cell lines in which α1B-AR densities were (6 336 ±913) and (773±164) fmol · mg -1, respectively. Treatment of cells with NE (10 μmol · L1) for 48 h decreased high-level expressed α1B-AR density, but increased low-level expressed α1B-AR density. The protein kinase C inhibitor Calphostin C or Ro-31-8220 reversed, and its activator PMA mimicked the NE-induced down-regulation of high-level expressed α1B-AR. Moreover, PMA induced a down-regulation of low-level expressed α1B-AR. The endoplasmic reticulum Ca2+-ATPase inhibitor cyclopiazonic acid (CPA) and the calcium chelator BAPTA/AM did not affect the down-regulation of high-level expressed α1B-AR, but inhibited the up-regulation of low-level expression α1B-AR induced by NE. | 19 | Preparation and investigation of a novel iodine-based visible polyvinyl alcohol embolization material显示文摘Polyvinyl alcohol(PVA) embolization particles, currently used in clinical practice, have good expansibility and are capable of permanent embolization. However, the lack of adhesion of embolization particles contributes to facilitated recanalization after embolization, while the lack of visualization facilitates misembolization. At present, embolization materials with good expansion, adhesion, and visualization potential are urgently required in clinical practice. Here, we report the development of PVA/gelatin/iohexol(I) fiber blocks as a novel embolization material for liver embolization in rats. In our work, electrospun PVA/gelatin/I nanofibrous mats were first prepared, homogenized, centrifuged in a gradient manner, and freeze-dried to obtain fiber blocks(fiber diameter =296.2 ± 74.23 nm, length 99.6 ± 17.0 μm × width 46.9 ± 13.3 μm). The fiber blocks exhibited excellent cytocompatibility and hemocompatibility. Fiber blocks with a PVA/gelatin/I mass ratio of 8:2:10 were selected due to their excellent expansibility and adhesive properties. The PVA/gelatin/I fiber blocks display excellent liver embolic effects and computed tomography(CT) imaging potential due to a combination of the following characteristics: expansibility of PVA and gelatin, adhesive property of gelatin, and CT imaging potential of I.
