| 1 | Systematic Characterization of Long Non-Coding RNAs and Their Responses to Drought Stress in Dongxiang Wild Rice显示文摘Long non-coding RNAs(lncRNAs) play important roles in response to various biotic and abiotic stresses. So far, systematic identification and characterization of lncRNAs have been reported in a few model plant species and major crops, but their roles in abiotic stress response have not yet been reported in common wild rice(Oryza rufipogon). Dongxiang wild rice(DXWR) possesses a high degree of drought resistance and has been well recognized as a precious genetic resource for drought resistant rice breeding. We presented the reference catalog of 1 655 novel lncRNA transcripts in DXWR using strand-specific RNA sequencing and bioinformatics approaches. Meanwhile, a total of 1092 lncRNAs were determined as differentially expressed lncRNAs under drought stress. Quantitative real-time PCR results exhibited a high concordance with RNA sequencing data, which confirmed that the expression patterns of lncRNAs based on RNA sequencing were highly reliable. Furthermore, 8 711 transcripts were predicted as target genes of the differentially expressed lncRNAs. Functional annotation analysis based on Gene Ontology and Kyoto Encyclopedia of Genes and Genomes databases showed that the target genes were significantly enriched in cellular and metabolic processes, cell part, binding and plant hormone signal transduction, as well as many other terms related to abiotic stress resistance. | 2 | Identification and Characterization of Drought Stress-Responsive Novel microRNAs in Dongxiang Wild Rice显示文摘MicroRNAs(miRNAs) are non-coding small RNAs, which play important regulatory roles in response to biotic and abiotic stresses. Dongxiang wild rice(Oryza rufipogon, DXWR) can survive in extreme drought environment, but its molecular mechanism of drought resistance is still largely unknown. To further explore miRNA regulatory mechanisms involved in drought resistance, we identified 138 novel miRNAs in DXWR using small RNA sequencing and bioinformatics approaches, and found that the expression levels of 67 novel miRNAs were significantly affected by drought stress. In total, 200 candidate target genes were predicted and annotated for the drought stress-responsive novel miRNAs. Gene Ontology(GO) analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathways suggested that most of the target genes were related to metabolism. Stem-loop quantitative real-time PCR(qRT-PCR) results exhibited high concordance with sequencing data, which confirmed that miRNA expression patterns based on small RNA sequencing in the present study were reliable. Meanwhile, qRT-PCR validated the inverse expression patterns between several miRNAs and their target genes. | 8 | B-Site Nanoscale-Ordered Structure Enables Ultra-High Tunable Performance显示文摘Tunable devices constructed by ferroelectric thin flms are often desired to possess a low dielectric loss while maintainging a high dielectric tunability over a broad operating temperature range in applications,for example,resonators,filters,or phase shifters.However,it is dificult to simultaneously achieve these characteristics by traditional strategies,such as doping and strain modifying.Here,we demonstrate that the dielectric tunability of the sol-gel-prepared Pb(Sc_(1/2)Nb_(1/2))_(0.9)(Mg_(1/3)Nb_(2/3))_(0.O3)(PSNMN)thin film can be almost doubled from~47%to~80.0%(at 10kHz)at a low electric field(~530kV/cm),and the dielectric loss can be sharply reduced by more than an order of magnitude,from~0.50 to~0.037(at 1 kHz)when the thin flm was annealed in air at 650℃ for 15h under the help of an atmosphere-compensating-block(ACB)made from the proto-PSNMN gel.Moreover,the PSNMN thin flm annealed with ACB also exhibited an extremely high thermally-stable dielectric tunability in an ultrabroad temperature range(>130 K),which could be attributed to the Maxwell-Wagner(MW)effect generated by the interface between the PSNMN disordered matrix and the B-site nanoscale-ordered structure formed during the long-term annealing process.The reduced dielectric loss is mainly benefited from the reduced concentration of oxygen vacancy and the possibie MW efects,and the enhanced dielectric tunability could be ascribed to the weaker domain-pinning effect by oxygen vacancy.The breakthrough provides a new universal strategy to achieve utrahigh tunable performance in A(B'_(1/2)B'_(1/2))O3 ferroelectric thin films with a B-site nanoscale-ordered structure,meanwhile it paves the way for ultraintergrated tunable thin-flm-devices with great phase shifter performance in practical applications. | Biaolin Peng Qiuping Lu Yi-Chi Wang Jing-Feng Li Qi Zhang Haitao Huang Laijun Liu Chao Li Limei Zheng Zhong Lin Wang | 2023 | Research2023,,1: | 0 |
