1Zika virus infection induces RNAi-mediated antiviral immunity in human neural progenitors and brain organoids显示文摘The re-emergence of Zika virus(ZIKV)in the Western Hemisphere has resulted in global public health crisis since 2015.ZIKV preferentially infects and targets human neural progenitor cells(hNPCs)and causes fetal microcephaly upon maternal infection.hNPCs not only play critical roles during fetal brain development,but also persist in adult brain throughout life.Yet the mechanism of innate antiviral immunity in hNPCs remains largely unknown.Here,we show that ZIKV infection triggers the abundant production of virus-derived small interfering RNAs in hNPCs,but not in the more differentiated progenies or somatic cells.Ablation of key RNAi machinery components significantly enhances ZIKV replication in hNPCs.Furthermore,enoxacin,a broad-spectrum antibiotic that is known as an RNAi enhancer,exerts potent anti-ZIKV activity in hNPCs and other RNAi-competent cells.Strikingly,enoxacin treatment completely prevents ZIKV infection and circumvents ZIKV-induced microcephalic phenotypes in brain organoid models that recapitulate human fetal brain development.Our findings highlight the physiological importance of RNAi-mediated antiviral immunity during the early stage of human brain development,uncovering a novel strategy to combat human congenital viral infections through enhancing RNAi.Yan-Peng Xu Yang Qiu Boya Zhang Guilai Chen Qi Chen Miao Wang Fan Mo Jiuyue Xu Jin Wu Rong-Rong Zhang Meng-Li Cheng Na-Na Zhang Bao Lyu Wen-Liang Zhu Meng-Hua Wu Qing Ye Da Zhang Jiang-Hong Man Xiao-Feng Li Jie Cui Zhiheng Xu Baoyang Hu Xi Zhou Cheng-Feng Qin 2019Cell Research2019,29,4:8
2Nanoporous titanium implant surface promotes osteogenesis by suppressing osteoclastogenesis via integrin β1/FAKpY397/MAPK pathway显示文摘Macrophages and osteoclasts are both derived from monocyte/macrophage lineage,which plays as the osteoclastic part of bone metabolism.Although they are regulated by bone implant surface nanoarchitecture and involved in osseointegration,the beneath mechanism has not been simultaneously analyzed in a given surface model and their communication with osteoblasts is also blurring.Here,the effect of implant surface topography on monocyte/macrophage lineage osteoclastogenesis and the subsequent effect on osteogenesis are systematically investigated.The nanoporous surface is fabricated on titanium implant by etching and anodizing to get the nanotubes structure.The early bone formation around implant is significantly accelerated by the nanoporous surface in vivo.Meanwhile,the macrophage recruitment and osteoclast formation are increased and decreased respectively.Mechanistically,the integrin mediated FAK phosphorylation and its downstream MAPK pathway(p-p38)are significantly downregulated by the nanoporous surface,which account for the inhibition of osteoclastogenesis.In addition,the nanoporous surface can alleviate the inhibition of osteoclasts on osteogenesis by changing the secretion of clastokines,and accelerate bone regeneration by macrophage cytokine profiles.In conclusion,these data indicate that physical topography of implant surface is a critical factor modulating monocyte/macrophage lineage commitment,which provides theoretical guidance and mechanism basis for promoting osseointegration by coupling the osteogenesis and osteoclastogenesis.Yide He Zhe Li Xin Ding Boya Xu Jinjin Wang Yi Li Fanghao Chen Fanhui Meng Wen Song Yumei Zhang 2022Bioactive Materials2022,7,2:4
3A Novel Multi-Modal OAM Vortex Electromagnetic Wave Microstrip Array Antenna显示文摘In this paper,we combine the circular polarization technology and orbital angular momentum(OAM)into the array antenna design for the first time,achieve free switch of the different topological charges in the array by using high-speed radio frequency(RF)switch technology.We arrange microstrip patch antenna elements equidistantly along the circumference to form eight elements multi-modal OAM vortex electromagnetic wave microstrip array antenna.It can generate elec-tromagnetic waves with dual characteristics of circular polarization and multi-modal vortex OAM(where OAM mode values are l=0,l=±1,l=±2,l=±3).Through simulation,we find mutual coupling between the radiating elements is small relatively,and increasing the number of array elements can not only improve the beam quality,but also generate electromagnetic waves with a higher order of OAM modes.Antenna model can meet the basic demands of ordinary array antenna,and also confirm the practicality of this circular polarized microstrip antenna array model.Xuehong Sun Jianfeng Shao Boya Dan Qiang Li 2019Journal of Communications and Information Networks2019,4,4:3
4Isolation and Characterization of a Cinnamoyl CoA Reductase Gene(CCR) in Pear(Pyrus pyrifolia)显示文摘Pear is a popular and commercially important fresh fruit, and its texture is related to the presence of sclereid formatted by parenchyma cell with lignification in vascular plants. Previous studies have demonstrated that content of lignin may be regulated by cinnamoyl CoA reductase(CCR) in various plants. However, the function of CCR in pears remains very limited. In the present study, we isolated a cDNA encoding CCR(PpCCR, GenBank accession No. KF999958) and its promoter(proPpCCR) from Whangkeumbae pear to investigate the function of CCR in lignin biosynthesis. PpCCR-GFP expressed in rice mesophyll protoplast demonstrated that PpCCR-GFP was localized in the cytoplasm, indicating that CCR may function in cytoplasm without localization signals. In transgenic plants carrying PpCCR, we observed higher lignin content compared with that in wild type plants, further suggesting that PpCCR can affect the lignin contents through regulating lignin biosynthesis in Arabidopsis thaliana. 6MOF-derived Co_(9)S_(8) nano-flower cluster array modified separator towards superior lithium sulfur battery显示文摘Lithium sulfur batteries with high energy density are thought to be the most potential energy storage technology that can be commercialized.However,the shuttle effect of polysulfides deteriorates its electrochemical performance.Herein,a novel Co_(9)S_(8)nanostructure derived from metal organic framework material(MOF)was explored by simple liquid phase reaction and heat vulcanization of2-methylimidazole and Co(NO_(3))_(2)·6 H_(2)O on the surface of the original PP separator.The Co_(9)S_(8)nano-flower cluster array wall was vertically and closely arranged with the thickness of 200 nm,and the polysulfide can be adsorbed by its physical and chemical action to slow down the'shuttle effect'.It is found that the cell with the modified separator can achieve an ideal discharge capacity of about 600 mAh/g at 1 C.The specific capacity is maintained at 500 mAh/g after 200 cycles,with only 0.11%of capacity decay per cycle.It provides a new way for the utilization of MOF material derivatives to modify the separator in order to improve the electrochemical performance of lithium-sulfur batteries.Qi Wang Hanqing Zhao Boya Li Chenhui Yang Mingyang Li Yanan Li Peng Han Miaomiao Wu Tengyu Li Ruiping Liu 2021Chinese Chemical Letters2021,32,3:2

9Incidence and Clinical Risk Factors for Preeclampsia and Its Subtypes:A Population-Based Study in Beijing,China显示文摘Objective:This study aimed to evaluate the incidence and associated clinical risk factors for preeclampsia(PE)and its subtypes in a large multicentre retrospective study of Beijing,China.Methods:This study was conducted as a secondary analysis from the Gestational diabetes mellitus Prevalence Survey(GPS),a multicentre retrospective cohort study,which included 15 hospitals in Beijing,China.This analysis included 15,003 pregnant women who delivered in Beijing from June 20th to November 30th,2013.The incidence of PE was calculated.Risk factors for PE,including maternal age,pre-gestational body mass index(BMI),parity,chronic hypertension,pre-existing diabetes,and gestational diabetes mellitus,were assessed.PE was defined as early-or late-onset PE based on clinical manifestations during the week of delivery,and mild or severe PE based on the severity of the disease.Logistic regression analysis was used to quantify the association with the risk factors,and data were displayed as odds risks(OR)and 95%confidence interval(CI).Results:The overall incidence of PE was 2.65%(397/15,003).The prevalence of early-onset and late-onset PE was 0.36%(54/15,003)and 2.29%(343/15,003),respectively.The prevalence of mild and severe PE was 0.91%(137/15,003)and 1.73%(260/15,003),respectively.Risk factors including high BMI considered overweight(adjusted odds risk(aOR):1.48;95%CI:1.06-2.05;P=0.02)and obesity(aOR:2.15;95%CI:1.50-3.08;P<0.001),nulliparity(aOR:1.73;95%CI:1.32-2.25;P<0.001),multiple gestation(aOR:4.58;95%CI:2.86-7.32;P<0.001),and chronic hypertension(aOR:34.95;95%CI:26.60-45.93;P<0.001),were associated with increased risk for PE.Only chronic hypertension(aOR:13.75;95%CI:4.78-39.58;P<0.001)was a significant risk factors for early-onset PE,whereas high BMI considered both overweight(aOR:1.54;95%CI:1.09-2.18;P=0.01)and obesity(aOR:2.23;95%CI:1.53-3.27;P<0.001),nulliparity(aOR:2.00;95%CI:1.49-2.68;P<0.001),multiple gestation(aOR:4.11;95%CI:2.40-7.05;P<0.001),and chronic hypertension(aOR:35.57;95%CI:26.66-47.47;P<0.001)were more relevant risk factors for late-onset PE.Risk factors including obesity(aOR:2.20;95%CI:1.28-3.76;P<0.01 and aOR:1.80;95%CI:1.16-2.80;P=0.01),nulliparity(aOR:2.28;95%CI:1.44-3.60;P<0.001 and aOR:1.48;95%CI:1.09-2.02;P=0.01),multiple gestation(aOR:5.50;95%CI:2.87-10.67;P<0.001 and aOR:3.51;95%CI:1.93-6.41;P<0.001),and chronic hypertension(aOR:33.98;95%CI:22.20-52.01;P<0.001 and aOR:35.03;95%CI:25.40-48.31;P<0.001)were associated with mild and severe PE.Moreover,we found that women with an increasing number of these risk factors had a higher risk of developing PE than pregnant women without any identified risk factors.Conclusion:The incidence of PE in this study is consistent with previous reported studies.Our findings indicate chronic hypertension and multiple gestation are the most important risk factors for PE in Chinese pregnant women.The risk for developing PE is associated with both the type and abundance of risk factors.These factors are valuable when monitoring patients at risk for PE,as this can help ensure an earlier diagnosis and prediction in women who are more likely to develop PE.Li Lin Jing Huai Rina Su Chen Wang Boya Li Huixia Yang 2021Maternal-Fetal Medicine2021,3,2:1
10Identification and characterization of Letm1 gene in Toxoplasma gondii显示文摘Toxoplasma gondii is an obligate intracellular protozoan that causes toxoplasmosis.Previous studies have shown that the perturbation of mitochondrial metabolism in T.gondii results in growth deficiency in host cells and lack of virulence in animals.Members of this Letm1 protein family are inner mitochondrial membrane proteins which play a role in potassium and hydrogen ion exchange.Letm1 has not been characterized in T.gondii.In this study,a potential TgLetm1 gene (TgGT1_288400) with Letm1-like protein domain coding sequence was identified in T.gondii.Indirect immunofluorescence assays suggested that TgLetm1 localized to the mitochondria in tachyzoites,as indicated by the colocalization with mitochondrial marker Mitotracker.TgLetm1 was found in the membrane fraction by western blot analysis.To investigate the role of TgLetm1 in T.gondii,we generated a tetracycline-inducible TgLetm1-knock-down mutant.The conditional deletion of TgLetm1 resulted in mitochondrial swelling.Functional studies showed that the conditional deletion of TgLetm1 resulted in growth inhibition,deficiency in invasion and replication,and lack of virulence in mice.Le Chang Xichen Zhang Pengtao Gong Yuru Wang Boya Du Jianhua Li 2019Acta Biochimica et Biophysica Sinica2019,51,1:1

13Variability of fuel consumption and CO_(2) emissions of a gasoline passenger car under multiple in-laboratory and on-road testing conditions显示文摘An increasing divergence regarding fuel consumption(and/or CO_(2)emissions) between realworld and type-approval values for light-duty gasoline vehicles(LDGVs) has posed severe challenges to mitigating greenhouse gases(GHGs) and achieving carbon emissions peak and neutrality. To address this divergence issue, laboratory test cycles with more real-featured and transient traffic patterns have been developed recently, for example, the China Lightduty Vehicle Test Cycle for Passenger cars(CLTC-P). We collected fuel consumption and CO_(2)emissions data of a LDGV under various conditions based on laboratory chassis dynamometer and on-road tests. Laboratory results showed that both standard test cycles and setting methods of road load affected fuel consumption slightly, with variations of less than 4%. Compared to the type-approval value, laboratory and on-road fuel consumption of the tested LDGV over the CLTC-P increased by 9% and 34% under the reference condition(i.e., air conditioning off, automatic stop and start(STT) on and two passengers). On-road measurement results indicated that fuel consumption under the low-speed phase of the CLTC-P increased by 12% due to the STT off, although only a 4% increase on average over the entire cycle. More fuel consumption increases(52%) were attributed to air conditioning usage and full passenger capacity. Strong correlations(R2> 0.9) between relative fuel consumption and average speed were also identified. Under traffic congestion(average speed below 25 km/hr), fuel consumption was highly sensitive to changes in vehicle speed. 14RAD6B participates in neuronal DNA damage response to resist neurodegeneration显示文摘Protein ubiquitination plays an important role in DNA damage repair.To clarify the role of RAD6B,identified as an important ubiquitin conjugating enzyme,in neuronal genome integrity and survival,we first silenced RAD6B gene in RPE1 and found that RAD6B deficiency resulted in increased genomic instability.While in brains of RAD6B knockout mouse generated by homologous recom bination,we found that RAD6B deficiency caused a significant reduction of H2B ubiquitination and impaird DNA damage repair compared with wild type mouse brains.The accumulation of unrepaired DNA damage often leads to apoptosis or senescence,even tumor formation.Furthermore,we evaluated the effect of RAD6B deficiency on the survival of neurons,and noticed that RAD6B-deficient mice exhibited marked neuronal senescence,neurodegeneration and reduced neuronal numbers.We also observed learning and memory impairment in neuron-specific RAD6B-deficient mice.In conclusion,RAD6B is involved in DNA damage repair of neurons,and is essential to maintain neuronal genome stability.Guo Zhao Tian Yingxia Guo Yingli Li Boya Liu Xiangwen Xie Kun Song Yanfeng Wang Degui 2021解剖学杂志2021,44,S01:0
15Polystyrene nanoplastics induce glycolipid metabolism disorder via NF-κB and MAPK signaling pathway in mice显示文摘Nanoplastics-induced developmental and reproductive toxicity,neurotoxicity and immunotoxicity are a focus of widespread attention.However,the effects of nanoplastics(NPs)on glycolipid metabolism and the precise underlying mechanisms are unclear at present.Here,we showed that oral administration of polystyrene nanoparticles(PS-NPs)disrupts glycolipid metabolism,with reactive oxygen species(ROS)identified as a potential key signaling molecule.After PS-NPs treatment,excessive production of ROS induced the infammatory response and activated the antioxidant pathway through nuclear factor-erythroid factor 2-related factor 2.The activation of nuclear factor-κB(NFκB)signaling pathway induced the phosphorylation of the mitogen-activated protein kinases(MAPK)signaling pathway,which induced the activation of extracellular regulated kinases(ERK)and p38.Constitutive activation of the MAPK signaling proteins induced high continued phosphorylation of insulin receptor substrate-1,in turn,leading to decreased protein kinase B(Akt)activity,which weakened the sensitivity of liver cells to insulin signals and induced insulin resistance.In parallel,phosphorylation of Akt led to loss of control of Fo XO1,a key gene of gluconeogenesis,activating transcription of glucose-6-phosphatase(G6PC)and phosphoenolpyruvate carboxykinase(PEPCK)in a manner dependent on PGC1α.Moreover,the activated ERK promoted lipid accumulation through ERK-PPARγcascades.Therefore,sterol regulatory element-binding protein-1 and levels of its downstream lipogenic enzymes,ACC-1,were up-regulated.Upon treatment with the antioxidant resveratrol,PS-NPs-induced glucose and lipid metabolic disorders were improved by inhibiting ROS-induced activation of NFκB and MAPK signaling pathway in mice.Based on above,PS-NPs exposure disrupts glycolipid metabolism in mice,with ROS identified as a potential key signaling molecule.Xingpei Fan Xiaoyan Li Jiaxin Li Yuxia Zhang Xiangjuan Wei Hailong Hu Boya Zhang Haining Du Meimei Zhao Ruijiao Zhu Daqian Yang Yuri Oh Ning Gu 2024Journal of Environmental Sciences2024,,3:0

20Dispersed distribution derived integrated anode for lithium ion battery显示文摘With the development of portable communication devices and electric vehicles, there is a great need for energy storage devices with lighter weight and higher energy density. In this paper, a new method by combining waster-paper-synthesized conductive paper(CP) and active material Mn O2 together is developed to obtain a new type of anode without any binder for lithium ion batteries. By this way, we can obtain low density anode with active material in CP, instead of the commonly-used heavy metal current collector. Also, binder has been abandoned, which are usually used to combine active material into anode, to further decrease weight. The multi walled carbon nanotube(MWCNT) was added in serves as a component of CP and the conductive agent for active material. Compared to traditional anode coated on Cu current collector, the CP-combined anode can greatly improve the electrochemical performance of active material Mn O2. It can let more particles to fully participate in the reaction and therefore boost the specific capacity to a great extent(about 3 times higher). It delivered an initial specific capacity of 1629.9 m A h g^-1 at a current density of 100 m A g^-1 and maintained about 67% even after 100 cycles.What’s more, it shows reversible capacity of about 260 m A h g^-1 at high current density of 1000 m A h g^-1.Our original synthesis method of anode, which shows far-reaching referential value and environmental significance, can be generalized to other electrodes and other battery systems.Boya Zhang Dongxiang Li Jiaqi Wan 2019Journal of Materials Science & Technology2019,35,10:0
