| 1 | Clinical features and mismatch repair gene mutation screening in Chinese patients with hereditary nonpolyposis colorectal carcinoma显示文摘AIM: Hereditary nonpolyposis colorectal cancer (HNPCC) is an autosomal dominantly- inherited cancer-susceptibility syndrome that confers an increased risk for colorectal cancer and a variety of other tumors at a young age. It has been associated with germline mutations in five mismatch repair (MMR) genes (hMSH2, hMLH1, hPMS1, hPMS2, and hMSH6/GTBP). The great majority of germline mutations were found in hMSH2 and hMLH1. The purpose of this study was to analyze the clinical features of Chinese HNPCC patients and to screen hMSH2 and hMLH1 gene mutations.METHODS: Twenty-eight independent Chinese families were collected, of which 15 met Amsterdam criteria I and 13 met the Japanese clinical diagnosis criteria. The data were recorded including sex, site of colorectal cancer (CRC),age of diagnosis, history of synchronous and/or metachronous CRC, instance of extracolonic cancers, and histopathology of tumors. Peripheral blood samples were collected from all pedigrees after formal written consents were signed. PCR and denaturing high-performance liquid chromatography (DHPLC) were used to screen the coding regions of hMSH2 and hMLH1 genes. The samples showing abnormal DHPLC profiles were sequenced by a 377 DNA sequencer.RESULTS: One hundred and seventy malignant neoplasms were found in one hundred and twenty-six patients (multiple cancer in twenty-three), including one hundred and twentyseven CRCs, fifteen gastric, seven endometrial, and five esophageal cancers. Seventy-seven point eight percent of the patients had CRCs, sharing the features of early occurrence (average age of onset, 45.9 years) and of the right-sided predominance reported in the literature. In Chinese HNPCC patients, gastric cancer occurred more frequently, accounting for 11.9% of all cancers patients and ranking second in the spectrum of HNPCC predisposing cancers. Synchronous CRCs occurred less frequently, only accounting for 3.1% of the total CRCs. Twenty percent of the colorectal patients had metachronous CRCs within 10 years after operation. Eight hMSH2 or hMLH1 gene sequence variations were found in twelve families, including the first Mongolian kindred with a hMSH2 gene mutation.CONCLUSION: HNPCC is characterized by an early-age onset, proximal predominance of CRC, multiple metachronous CRCs, and an excess of extra-colonic cancers. Frequent gastric cancer occurrence and less synchronous CRCs are the remarkable features in Chinese HNPCC patients. DHPLC is a powerful tool in hMSH2 and hMLH1 gene mutation screening, hMLH1 gene mutations, especially of the first nine exons, have been found more common than hMSH2 gene mutations in Chinese patients. | 2 | Improved catalytic performance of Ni catalysts for steam methane reforming in a micro-channel reactor显示文摘Milliseconds process to produce hydrogen by steam methane reforming(SMR) reaction, based on Ni catalyst rather than noble catalyst such as Pd, Rh or Ru, in micro-channel reactors has been paid more and more attentions in recent years. This work aimed to further improve the catalytic performance of nickel-based catalyst by the introduction of additives, i.e., MgO and FeO, prepared by impregnation method on the micro-channels made of metal-ceramic complex substrate. The prepared catalysts were tested in the same micro-channel reactor by switching the catalyst plates. The results showed that among the tested catalysts Ni-Mg catalyst had the highest activity, especially under harsh conditions,i.e., at high space velocity and/or low reaction temperature. Moreover, the catalyst activity and selectivity were stable during the 12 h on stream test even when the ratio of steam to carbon(S/C) was as low as 1.0. | 5 | Fabrication of high temperature grating on thermal barrier coatings based on solute-solvent separation soft lithography显示文摘In this study, an etched-SiO-film grating fabrication technique based on solute-solvent separation soft lithography is developed. By using this new technique, high temperature grating with frequency of 600 lines/mm is successfully fabricated on the surface of thermal barrier coatings(TBCs). During the fabrication process, the microstructure and chemical compositions of ceramic coating before and after polishing is analyzed with a digital microscope and the energy dispersive X-ray spectroscope(EDS) in scanning electron microscope(SEM). And then the grating on TBCs is heated at the temperature ranging from 300℃ to 1000℃ for examining the high temperature resistance. In the practical application, the displacement and strain field around the crack on the ceramic surface are investigated with geometry phase analysis(GPA). | 11 | Local chemical inhomogeneities in TiZrNb-based refractory high-entropy alloys显示文摘Multi-principal element solid solutions are prone to develop local chemical inhomogeneities,i.e.,chemi-cal order/clustering and/or compositional undulation.However,these structural details from short-range(first couple of nearest-neighbor atomic shells)to nanometer length scale are very challenging to re-solve in both experimental characterization and computer simulations.For instance,Monte Carlo model-ing based on density-functional-theory calculations is severely limited by the sample size and the sim-ulation steps practical in the simulations.Adopting the cluster expansion approach,here we systemati-cally reveal the local chemical inhomogeneity,including chemical order and compositional fluctuation,in three representative equiatomic TiZrNb-based body-centered cubic refractory high-entropy alloys(HEAs):TiZrNb,TiZrHfNb and TiZrHfNbTa.Ti-Zr pairs are found to exhibit the highest degree of chemical pref-erence among all atomic pairs.Such chemical short-range order(CSRO)induces an accompanying com-positional undulation,both extending to characteristic dimensions of the order of one nanometer.The chemical inhomogeneity trend uncovered for this series of TiZrNb-based HEAs is expected to impact their mechanical properties;e.g.,incorporating the CSRO effects in a current model significantly improves its agreement with experimental measured yield strength. | Kaihui Xun Bozhao Zhang Qi Wang Zhen Zhang Jun Ding En Ma | 2023 | Journal of Materials Science & Technology2023,,4: | 0 |
