| 1 | Shoshonitic intrusive suite in SE Guangxi: Petrology and geochemistry显示文摘A NE-direction Mesozoic shoshonitic intrusive s黫te in SE Guangxi has been identified in terms of geological, petrological and geochemical investigations. The shoshonitic intrusives are characterized by enrichment of LILE, HFSE and LREE and no obvious Nb-Ta depletion, similar to those potassic rocks formed in within-plate and rifting environments. | 2 | CD24 aggravates acute liver injury in autoimmune hepatitis by promoting IFN-γ production by CD4^(+) T cells显示文摘The T-cell-mediated immune response is implicated in many clinical hepatic injuries, such as autoimmune hepatitis and acute virus hepatitis. CD24 is widely expressed by different immune cells and plays an important role in the pathogenesis of many autoimmune diseases. However, the role of CD24 in T-cell-mediated liver injury has not been elucidated until now. Here we showed that CD24 deficiency protects mice from concanavalin A (ConA)-induced fulminant liver injury by reducing serum interferon-γ (IFN-γ) levels. CD24 expression by hepatic T cells was markedly increased following ConA challenge. Moreover, decreased IFN-γ production by hepatic CD4^(+) T cells in CD24-deficient mice was detected, which was correlated with downregulated phosphorylation of STAT1 in hepatic tissue. In vitro experiments also supported the conclusion that CD24 deficiency impaired IFN-γ production by CD4^(+) T cells following ConA, CD3/CD28 and phorbol myristate acetate/ionomycin stimulation. | 3 | The transcription factor OpWRKY2 positively regulates the biosynthesis of the anticancer drug camptothecin in Ophiorrhiza pumila显示文摘The limited bioavailability of plant-derived natural products with anticancer activity poses major challenges to the pharmaceutical industry.An example of this is camptothecin,a monoterpene indole alkaloid with potent anticancer activity that is extracted at very low concentrations from woody plants.Recently,camptothecin biosynthesis has been shown to become biotechnologically amenable in hairy-root systems of the natural producer Ophiorrhiza pumila.Here,time-course expression and metabolite analyses were performed to identify novel transcriptional regulators of camptothecin biosynthesis in O.pumila.It is shown here that camptothecin production increased over cultivation time and that the expression pattern of the WRKY transcription factor encoding gene OpWRKY2 is closely correlated with camptothecin accumulation.Overexpression of OpWRKY2 led to a more than three-fold increase in camptothecin levels.Accordingly,silencing of OpWRKY2 correlated with decreased camptothecin levels in the plant.Further detailed molecular characterization by electrophoretic mobility shift,yeast one-hybrid and dual-luciferase assays showed that OpWRKY2 directly binds and activates the central camptothecin pathway gene OpTDC.Taken together,the results of this study demonstrate that OpWRKY2 acts as a direct positive regulator of camptothecin biosynthesis.As such,a feasible strategy for the over-accumulation of camptothecin in a biotechnologically amenable system is presented. | Xiaolong Hao Chenhong Xie Qingyan Ruan Xichen Zhang Chao Wu Bing Han Jun Qian Wei Zhou Hans-Wilhelm Nutzmann Guoyin Kai | 2021 | Horticulture Research2021,8,1: | 7 |
| 4 | Initial gut microbiota structure affects sensitivity to DSS-induced colitis in a mouse model显示文摘The dextran sulfate sodium (DSS)-induced colitis model is a widely applied mouse model, but controversial results have been obtained from experiments using the same mouse strain under the same conditions. Because the gut microbiota play an important role in DSS-induced colitis, it is essential to evaluate the influence of the initial gut microbiota in this model. Here, we identified significant variations in the initial gut microbiota of different batches of mice and found that the initial intestinal microbiota had a profound influence on DSS-induced colitis. We performed three independent trials using the same C57BL/6J mouse model with DSS treatment and used high-throughput 16S rRNA gene sequencing to analyze the gut microbiota. We found that the structure and composition of the gut microbiota in mice with severe colitis, as compared with mice with milder colon damage, had unique features, such as an increase in Akkermansia bacteria and a decrease in Barnesiella spp. Moreover, these varied gut bacteria in the different trials also showed different responses to DSS treatment. | 5 | Cloning and Characterization of an Abalone(Haliotis discus hannai)Actin Gene显示文摘An actin encoding gene was cloned by using RT-PCR, 3’ RACE and 5’ RACE from abalone Haliotis discus hannai. The full length of the gene is 1532 base pairs, which contains a long 3’ untranslated region of 307 base pairs and 79 base pairs of 5’ untranslated sequence. The open reading frame encodes 376 amino acid residues. Sequence comparison with those of human and other mollusks showed high conservation among species at amino acid level. The identities was 96%, 97% and 96% respectively compared with Aplysia californica, Biomphalaria glabrata and Homo sapience β-actin. | 6 | Epidemiology of urticaria in China:a population-based study显示文摘Background:Urticaria is a common skin disease characterized by episodes of wheals,and it has a negative effect on patients’quality of life.Large-scale population-based epidemiological studies of urticaria are scarce in China.The aim of this survey was to determine the prevalence,clinical forms,and risk factors of urticaria in the Chinese population.Methods:This survey was conducted in 35 cities from 31 provinces,autonomous regions,and municipalities of China.Two to three communities in each city were selected in this investigation.Participants completed questionnaires and received dermatological examinations.We analyzed the prevalence,clinical forms,and risk factors of urticaria.Results:In total,44,875 questionnaires were distributed and 41,041 valid questionnaires were collected(17,563 male and 23,478 female participants).The lifetime prevalence of urticaria was 7.30%,with 8.26%in female and 6.34%in male individuals(P<0.05).The point prevalence of urticaria was 0.75%,with 0.79%in female and 0.71%in male individuals(P<0.05).Concomitant angioedema was found in 6.16%of patients.Adults had a higher prevalence of urticaria than adolescents and children.Living in urban areas,exposure to pollutants,an anxious or depressed psychological status,a personal and family history of allergy,thyroid diseases,and Helicobacter pylori infection were associated with a higher prevalence of urticaria.Smoking was correlated with a reduced risk of urticaria.Conclusion:This study demonstrated that the lifetime prevalence of urticaria was 7.30%and the point prevalence was 0.75%in the Chinese population;women had a higher prevalence of urticaria than men.Various factors were correlated with urticaria. | Jiaqing Li Dandan Mao Shuoshuo Liu Ping Liu Jing Tian Chenhong Xue Xiaojing Liu Ruiqun Qi Bingxue Bai Jianjun Nie Siqi Ye Yu Wang Yuye Li Qing Sun Juan Tao Shuping Guo Hong Fang Jianqin Wang Qiri Mu Quanzhong Liu Yan Ding Jianzhong Zhang | 2022 | Chinese Medical Journal2022,,11: | 6 |
| 7 | Application of digital pattern-less molding technology to produce art casting显示文摘Compared with the conventional casting process, digital pattern-less casting technology has many advantages such as good machining accuracy, a short processing cycle, and low production cost. It is a new rapid manufacturing technology for castings, integrated with CAD/CAM, casting, CNC machining and many other advanced technologies. With this digital casting technology, no pattern is needed for making molds; it is precise, f lexible, and green. Usually, art castings have complex structures and are made in small batches or even made in a single-piece, especially for large-sized art castings. So it has the shortcomings of high cost, low eff iciency and long time for making a pattern to produce art castings with the conventional casting processes. However, the digital pattern-less casting technology can be applied to fabricate art castings, since it can greatly shorten the manufacturing cycle and lower the production cost, thus having a very good prospect. | 8 | Measurement of the current and spectral analysis on the continental shelf in the East China Sea显示文摘Direct measurements of current velocity and water temperature were undertaken at the mooring Sta. M (125°29.38′ N,31°49.70′ E) on the continental shelf area in the East China Sea in June 1999 by R/V Xiangyanghong 14. The relationship between various time series of oceanic fluctuations is calculated by spectral analysis. The major results are as follows: (1) an average (u,v) of (6.9,-3.0 cm/s) at the 30 m depth is obtained during the 9-day observation, and that at the 45 m depth is (3.7,-1.1 cm/s), i. e., the mean flows are southeastward at both the 30 and 45 m depths;the currents become stronger gradually during the observation period; this may be mainly attributed to the transition of the tidal currents from neap to spring; (2)semidiurnal fluctuation is the most dominant in the current fluctuations, and rotates mainly clockwise; in the next place, there is also diurnal fluctuation;the local inertial period is close to the period of diurnal fluctuation, and an inertial motion is clockwise; thus, local inertial motion combines with diurnal fluctuation, and makes the spectral peaks in clockwise components much higher than those in counterclockwise ones;except for the fluctuations of above main periods, there is also the peak at 3 d period for counterclockwise components in the upper and lower layers; (3) the calculation of cross spectra between two time series of current velocities at the 30 and 45 m depths shows that both the current fluctuations at the 30 and 45 m depths are much alike, i. e.,they are synchro; this shows that the flow field here is rather vertically homogeneous; (4)power spectra of temperature time series at both the 30 and 45 m depths show that the semidiurnal peak is the most predominant, and the second highest peak is the diurnal period; besides spectral peaks at above periods, there are also obvious spectral peaks at 6.8 h and 2 d; (5)plots of temperature time series at 16,30,35,45 and 50 m depths show that the temporal variations of temperature at these depths are synchro, which are like those in the velocity field; temperature records also show a gradual rise in temperature, which are also like those in the velocity field. | LIUYonggang YUANYaochu LIUCho-Teng CHENHong | 2004 | Acta Oceanologica Sinica2004,23,2: | 4 |
