1High-Resolution Genome-wide Association Study Identifies Genomic Regions and Candidate Genes for Important Agronomic Traits in Wheat显示文摘Wheat(Triticum aestivum)is a major staple food crop worldwide.Genetic dissection of important agronomic traits is essential for continuous improvement of wheat yield to meet the demand of the world's growing population.We conducted a large-scale genome-wide association study(GWAS)using a panel of 768 wheat cultivars that were genotyped with 327609 single-nucleotide polymorphisms generated by genotyping-by-sequencing and detected 395 quantitative trait loci(QTLs)for 12 traits under 7 environments.Among them,273 QTLs were delimited to≤1.0-Mb intervals and 7 of them are either known genes(Rht-D,Vrn-B1,and Vrn-D1)that have been cloned or known QTLs(TaGA2ox8,APO1,TaSus1-7B,and Rht12)that were previously mapped.Eight putative candidate genes were identified for three QTLs that enhance spike seed setting and grain size using gene expression data and were validated in three bi-parental populations.Protein sequence analysis identified 33 putative wheat orthologs that have high identity with rice genes in QTLs affecting similar traits.Large r^2 values for additive effects observed among the QTLs for most traits indicated that the phenotypes of these identified QTLs were highly predictable.Results from this study demonstrated that significantly increasing GWAS population size and marker density greatly improves detection and identification of candidate genes underlying a QTL,solidifying the foundation for large-scale QTL fine mapping,candidate gene validation,and developing functional markers for genomics-based breeding in wheat.Yunlong Pang Chunxia Liu Danfeng Wang Paul St.Amand Amy Bernardo Wenhui Li Fang He Linzhi Li Liming Wang Xiufang Yuan Lei Dong Yu Su Huirui Zhang Meng Zhao Yunlong Liangi Hongze Jia Xitong Shen Yue Lu Hongming Jiang Yuye Wu Anfei Li Honggang Wang Lingrang Kong Guihua Bai Shubing Liu 2020Molecular Plant2020,13,9:8
2Epidemiology of urticaria in China:a population-based study显示文摘Background:Urticaria is a common skin disease characterized by episodes of wheals,and it has a negative effect on patients’quality of life.Large-scale population-based epidemiological studies of urticaria are scarce in China.The aim of this survey was to determine the prevalence,clinical forms,and risk factors of urticaria in the Chinese population.Methods:This survey was conducted in 35 cities from 31 provinces,autonomous regions,and municipalities of China.Two to three communities in each city were selected in this investigation.Participants completed questionnaires and received dermatological examinations.We analyzed the prevalence,clinical forms,and risk factors of urticaria.Results:In total,44,875 questionnaires were distributed and 41,041 valid questionnaires were collected(17,563 male and 23,478 female participants).The lifetime prevalence of urticaria was 7.30%,with 8.26%in female and 6.34%in male individuals(P<0.05).The point prevalence of urticaria was 0.75%,with 0.79%in female and 0.71%in male individuals(P<0.05).Concomitant angioedema was found in 6.16%of patients.Adults had a higher prevalence of urticaria than adolescents and children.Living in urban areas,exposure to pollutants,an anxious or depressed psychological status,a personal and family history of allergy,thyroid diseases,and Helicobacter pylori infection were associated with a higher prevalence of urticaria.Smoking was correlated with a reduced risk of urticaria.Conclusion:This study demonstrated that the lifetime prevalence of urticaria was 7.30%and the point prevalence was 0.75%in the Chinese population;women had a higher prevalence of urticaria than men.Various factors were correlated with urticaria.Jiaqing Li Dandan Mao Shuoshuo Liu Ping Liu Jing Tian Chenhong Xue Xiaojing Liu Ruiqun Qi Bingxue Bai Jianjun Nie Siqi Ye Yu Wang Yuye Li Qing Sun Juan Tao Shuping Guo Hong Fang Jianqin Wang Qiri Mu Quanzhong Liu Yan Ding Jianzhong Zhang 2022Chinese Medical Journal2022,,11:6
3The number and cytotoxicity and the expression of cytotoxicity-related molecules in peripheral natural killer(NK)cells do not predict the repeated implantation failure(RIF)for the in vitro fertilization patients显示文摘Natural killer(NK)cells are thought to play a key role in the successful establishment of a pregnancy by facilitating immunological adaptation of the semi-allogeneic developing embryo.The aim of this study was to explore the cell number,immunophenotypic characteristics,and activities of peripheral blood NK cells in women with repeated implantation failure(RIF).Peripheral blood was obtained from 27 women with RIF and 11 healthy,fertile controls during the middle luteal phase of the menstrual cycle.CD3^-CD56^+NK cells were quantified and analyzed by flow cytometry for the expression of cytolytic molecules(granzyme B,granulysin,and perforin)as well as cell surface receptors responsible for NK cell activation or inhibition(NKG2D,NKp30,NKp46,CD158a,CD158b).NK cytotoxicity was measured at three effector-to-target cell ratios.Women with RIF and fertile controls did not differ significantly in the percentage of circulating CD3CD56t NK cells,or in the proportions of these cells that expressed granzyme B,granulysin,or perforin.The two groups also did not differ significantly in the proportions of NK cells expressing the receptors NKG2D,NKp30,NKp46,CD158a or CD158b.General linear model analysis showed that NK cytotoxicity increased with effector-to-target cell ratio.However,NK cytotoxicity did not differ significantly between patients with RIF and fertile controls.These results suggest that RIF is not associated with significant alterations in the number or function of peripheral blood NK cells.Hongzhan Zhang Chunyu Huang Xian Chen Longfei Li Su Liu Yuye Li Yongnu Zhang Yong Zeng Lina HuDepartment of Obstetrics and Gynecology The Second Affiliated Hospital of Chongqing Medical 2020Genes & Diseases2020,7,2:2
4Transformation mechanism and fate of dissolved organic nitrogen(DON) in a full-scale drinking water treatment显示文摘Dissolved organic nitrogen(DON) has attracted much attention in drinking water treatment due to its potential to produce nitrogenous disinfection by-products(N-DBPs). This work was designed to explore the transformation and fate of DON and dissolved inorganic nitrogen(DIN) in drinking water treatment. The changes of DON and formation of N-DBPs were evaluated along the water treatment route(i.e., pre-ozonation and biologicalcontact oxidation, delivery pipes’ transportation, coagulation-sedimentation, sand filtration, post-ozonation, biological activated carbon, ultrafiltration and disinfection) of drinking water treatment plant(DWTP). The transformation mechanism of DON was comprehensively investigated by molecular weight fractionation, three-dimensional fluorescence, LCOCD(Liquid Chromatography-Organic Carbon Detection), total free amino acids. A detailed comparison was made between concentrations and variations of DON and DIN affected by seasons in the drinking water treatment. Regardless of seasonal variation in raw water concentration, the DON removal trends between different treatment processes remain constant in the present study. Compared to other treatment processes, pre-ozonation and coagulation-sedimentation exhibited the dominant DON removal in different seasons, i.e.,11.13%-14.45% and 14.98%-22.49%, respectively. Contrary, biological-contact oxidation and biological activated carbon negatively impacted the DON removal, in which DON increased by 1.76%-6.49% in biological activated carbon. 5The nonlinear mechanism for the same responses of neuronal bursting to opposite self-feedback modulations of autapse显示文摘In the traditional viewpoint,inhibitory and excitatory effects always induce opposite responses.In the present study,the enhanced bursting activities induced by excitatory autapses,which are consistent with the recent experimental observations,and those induced by inhibitory autapses,which is a paradoxical phenomenon,were simulated using the Chay model.The same bifurcations and different ionic currents for the same responses were acquired with fast-slow variable dissection and current decomposition,respectively.As the inhibitory or excitatory autaptic conductance increased,the ending phase of the burst related to a homoclinic bifurcation of the fast subsystem changed to widen the burst duration to contain more spikes,which was induced by an elevated minimal potential(V_(min)) of spiking of the fast subsystem.Larger inhibitory and excitatory autaptic conductances induced smaller and larger maximal potentials(V_(max)) of spiking,respectively.During the downstroke,a weaker potassium current induced by the smaller V_(max) played a dominant role for the inhibitory autapse,and the stronger potassium current induced by the larger V_(max) became weaker due to the opposite autaptic current of the excitatory autapse,which induced the V_(min) elevated.The results present the nonlinear and biophysical mechanisms of the same responses to opposite effects,which extends nonlinear dynamics knowledge and provides potential modulation measures for the nervous system.LI YuYe GU HuaGuang JIA Bing DING XueLi 2021Science China(Technological Sciences)2021,64,7:1
6Mechanical behaviors of extruded Mg alloys with high Gd and Nd content显示文摘The influence of alloying elements and heat treatment on the microstructure and mechanical behaviors of extruded Mg–Gd–Nd ternary alloys was investigated in this study.The grain sizes dramatically decreased after extrusion,and the particles which distributed in Mg matrix had great effect on the grain size.The grain sizes of extruded alloys decreased from 26 to 5μm with the alloying content increasing.The mechanical test results show that both Gd and Nd had positive effect on the hardness,yield strength and Young’s modulus.The ultimate tensile strength(UTS)was enhanced by Gd content,decreased with Nd content.The elongation of alloys was lower with higher alloying elements.Those extruded alloys were aged for 200 h in 200℃.The Young’s moduli were decreased by ageing treatment.Combined with microstructure study,the part of the reinforcement which identified as Mg5(Gd/Nd)was dissolved in Mg matrix.Nd element obviously has influence on the solubility of Gd in Mg alloys.Yuling Xu Shiwei Wang Yuye Wang Li Chen Lixiang Yang Lu Xiao Li Yang Norbert Hort 2021Progress in Natural Science:Materials International2021,31,4:1

11Programmable Analytical Feature of Ratiometric Electrochemical Biosensor by Alternating the Binding Site of Ferrocene to DNA Duplex for the Detection of Aflatoxin B1显示文摘Ratiometric biosensor is among the promising strategies for the advanced analysis;its applicability in multiple scenarios,however,is seriously limited for the fixed analytical feature.Here,we reported ratiometric electrochemical biosensors with programmable analytical feature by alternating the binding site of ferrocene(Fc)to DNA duplex for the detection of aflatoxin B1(AFB1).DNA duplex was synthesized with AFB1 aptamer and its complementary DNA(cDNA)with Fc molecular tagged on aptamer or cDNA.For analysis,aptamer-AFB1 complex stripped from the electrode,and free cDNA triggered hybridization chain reaction to produce double-stranded DNA for methylene blue(MB)absorption.In this way,redox currents of Fc(I_(Fc))and absorbed MB(I_(MB))at the electrode were collected to produce the ratiometric signal of I_(MB)/I_(Fc)as a yardstick for AFB1.When Fc was tagged on aptamer,IFc decreased with higher concentration of AFB1(C_(AFB1)),the resultant biosensor allowed detection with an On/Off(I_(MB)/I_(Fc))mode;when Fc was tagged on cDNA,I_(Fc) kept constant as C_(AFB1)varying,offering an On/Constant(I_(MB)/I_(Fc))mode for the detection.Consequently,biosensor with On/Off mode showed a linear range of 0.1—100 pg/mL with a limit of detection(LOD)of 0.0650 pg/mL,while biosensor with On/Constant mode offered a wider linear rang of 1—10000 pg/mL but a relatively poor LOD of 0.330 pg/mL.Our work revealed the vital importance of binding site of probes to DNA,and this principle can endow ratiometric biosensor with programmable analytical feature(e.g.,linear range,LOD),allowing detections in various scenarios.Dong Liu Fan Jia Ya Wei Yuye Li Shuyun Meng Tianyan You 2022Chinese Journal of Chemistry2022,40,18:0
12L-Selenocystine induce HepG2 cells apoptosis through ROS-mediated signaling pathways显示文摘At present,Hepatocarcinoma is one of the main causes of tumor related death all over the world.However,there are still many clinical restrictions on the treatment of liver cancer.Recently,L-Selenocystine has been shown to be a novel treatment for tumors,especially human glioma cells.But,the mechanism of L-Selenocystine against hepatocellular carcinoma remains unclear.Therefore,the main objective of this study was to investigate the effects of L-Selenocystine on HepG2 cell proliferation and activation of reactive oxygen species(ROS)mediated signaling pathway.L-Selenocystine can significantly inhibit HepG2 cell proliferation by activating caspase-3 and cleaving PARP to induce apoptosis.Moreover,the excessive production of ROS and the influence of Bax signaling pathway which can promote cell apoptosis are key factors for L-Selenocystine to induce HepG2 cell apoptosis.Therefore,the date of this study suggest that ROS mediated signal transduction mechanism may provide certain reference significance for L-Selenocystine induced HepG2 cell apoptosis.HAIYANG CHEN JINGYAO SU DANYANG CHEN YUYE DU RUILIN ZHENG QINGLIN DENG QIANQIAN DU BING ZHU YINGHUA LI 2022BIOCELL2022,46,10:0

14A Novel 2-dimensional Multiplex qPCR Assay for Single-Tube Detection of Nine Human Herpesviruses显示文摘Human herpesviruses are double-stranded DNA viruses that are classified into nine species.More than 90%of adults are ever infected with one or more herpesviruses.The symptoms of infection with different herpesviruses are diverse ranging from mild or asymptomatic infections to deadly diseases such as aggressive lymphomas and sarcomas.Timely and accurate detection of herpesvirus infection is critical for clinical management and treatment.In this study,we established a single-tube nonuple qPCR assay for detection of all nine herpesviruses using a 2-D multiplex qPCR method with a house-keeping gene as the internal control.The novel assay can detect and distinguish different herpesviruses with 30 to 300 copies per 25µL single-tube reaction,and does not cross-react with 20 other human viruses,including DNA and RNA viruses.The robustness of the novel assay was evaluated using 170 clinical samples.The novel assay showed a high consistency(100%)with the single qPCR assay for HHVs detection.The features of simple,rapid,high sensitivity,specificity,and low cost make this assay a high potential to be widely used in clinical diagnosis and patient treatment.Yingxue Li Zhenzhou Wan Lulu Zuo Shenwei Li Honglian Liu Yingying Ma Lianqun Zhou Xia Jin Yuye Li Chiyu Zhang 2021Virologica Sinica2021,36,4:0
