1Isolation and identification of CD4^+CD25^+ regulatory T cells in rat显示文摘Objective:To establish a stable and high efficient method for collection of CD4+CD25+ regulatory T cells from rats in vitro.Methods:CD4+CD25+ regulatory T cells were isolated from the rat splenic cells through two steps by magic cell sorting(MACS)system.The first step was negative selection of CD4+T cells by cocktail antibodies and anti-IgG magic microbeads,and the second step was positive selection of CD25+T cells by anti-CD25 PE and anti-PE magic microbeads.The purity and viability of separated cells were measured by flow cytometry(FACS)and Trypan blue staining.The suppressive ability of seperated cells on the proliferation of CD4+CD25-T cells was assessed by cell proliferation assay.Results:The purity of negatively enriched CD4+ T cells was 79%-87%(83.6% ± 2.5%),and the purity of positively enriched CD4+CD25+ T cells was 86%-93%(90.2 ± 1.8%)with the viability of 92%-95%(92.8% ± 3.4%).The enriched cells significantly suppressed the proliferation of CD4+CD25-T cells in mixed lymphocyte culture(P < 0.05).Conclusion:An effective method can be established for enrichment of CD4+CD25+ regulatory T cells in two steps by MACS,with satisfied cell purity,viability and function.Ling Lü Feng Zhang Liyong Pu Chao Jiang 2006Journal of Nanjing Medical University2006,20,4:1
2U-Pb zircon geochronology, geochemistry and kinetics of the Huaniushan A-type granite in Northwest China显示文摘The Huaniushan granite is located at the Beishan orogenic belt, northwestern China. At the contact zone between the granite and marble, a hydrothermal Pb-Zn and skarn Au deposit is formed. LA-ICP-MS zircon U-Pb dating yielded a weighted mean 206Pb/238U age of 229.5±2.6 Ma (MSDW=0.93) for the Huaniushan granite, imply-ing its Late Triassic intrusion. Geochemistry analyses show that the Huaniushan granite is enriched in Si, K, Na, and REE, and depleted in Mg and Ca, with contents of SiO2 (70.8% to 74.4%), Na2O+K2O (8.8% to 10.2%), CaO (0.93% to 1.44%), and MgO (0.14% to 0.48%). REE is characterized by obvious negative Eu anomaly. Rb, Th, U, K, Pb, Nb, Zr and Hf elements are rich in the granite while Ba, Sr, P, Ti and Eu are deplete. The granite has a high (Zr+Nb+Ce+Y) abundance and 104 Ga/Al ratios. Petrology, major and trace elements data all indicate that the Hua-niushan granite is A-type granite which intruded in a post-collisional extensional tectonic setting. The magma was dominantly sourced from partial melting of crustal intermediate-felsic igneous rocks. 3A novel method for improving cerussite sulfidization显示文摘Evaluation of flotation behavior, solution measurements, and surface analyses were performed to investigate the effects of chloride ion addition on the sulfidization of cerussite in this study. Micro-flotation tests indicate that the addition of chloride ions prior to sulfidization can significantly increase the flotation recovery of cerussite, which is attributed to the formation of more lead sulfide species on the mineral surface. Solution measurement results suggest that the addition of chloride ions prior to sulfidization induces the transformation of more sulfide ions from pulp solution onto the mineral surface by the formation of more lead sulfide species. X-ray diffraction and energy-dispersive spectroscopy indicate that more lead sulfide species form on the mineral surface when chloride ions are added prior to sulfidization. 4Enhanced Degradation of Methyl Parathion in the Ligand Stabilized Soluble Mn(Ⅲ)-Sulfite System显示文摘The ligand-stabilized soluble Mn(Ⅲ) recognized as active intermediate can potentially mediate the attenuation of contaminants. In this study,the abiotic degradation behaviors of methyl parathion in the ligand stabilized Mn(Ⅲ)-sulfite system were investigated. The results showed that the yield of soluble Mn(Ⅲ) produced from the redox reaction of MnO2 and oxalic acid was dependent linearly on the dosage of Mn O2 and caused the decomposition of methyl parathion up to 50.1% in Mn(Ⅲ)-sulfite system after 30 minutes. The fitted pseudo-first-order reaction constants of methyl parathion degradation increased with the increasing of the amount of produced Mn(Ⅲ) but was not effected linearly by the addition of sulfite. Other ligands,including pyrophosphate and oxalic acid,acted as effective complexing agents to stabilize soluble Mn(Ⅲ),and exhibited competitive effect on methyl parathion degradation with sulfite. The formation of Mn(Ⅲ)-sulfite complexes is the critical step in the system to produce abundant reactive oxygen species identified as SO3·-to facilitate methyl parathion degradation. The hydrolysis and oxidation of methyl parathion were acknowledged as two primary transformation mechanisms in Mn(Ⅲ)-sulfite system. 7Amygdala-Inspired Affective Computing: to Realize Personalized Intracranial Emotions with Accurately Observed External Emotions显示文摘Artificial intelligence technology has revolutionized every industry and trade in recent years. However, its own development is encountering bottlenecks that it is unable to implement empathy with human emotions. So affective computing is getting more attention from researchers. In this paper, we propose an amygdala-inspired affective computing framework to realize the recognition of all kinds of human personalized emotions. Similar to the amygdala, the instantaneous emergency emotion is first computed more quickly in a low-redundancy convolutional neural network compressed by pruning and weight sharing with hashing trick. Then, the real-time process emotion is identified more accurately by the memory level neural networks, which is good at handling time-related signals. Finally, the intracranial emotion is recognized in personalized hidden Markov models. We demonstrate on Facial Expression of Emotion Dataset and the recognition accuracy of external emotions(including the emergency emotion and the process emotion) reached 85.72%. 9Numerical Simulation of Enhanced Oil-Water Separation in a Three-Stage Double-Stirring Extraction Tank显示文摘Numerical simulation of enhanced fluid flow characteristics in a three-stage double-stirring extraction tank was conducted with the coupling of an Eulerian multiphase flow model and a Morsi-Alexander interphase drag force model. Results show that the addition of a stirring device into the settler can efficiently reduce the volume fraction of out-of-phase impurity in the outlet, and accelerate the settling separation of oil-water mixture. Such addition can also effectively break down the oil-water-wrapped liquid droplets coming from the mixer, inhibit reflux from the outlet, and improve the oil-water separation. 10Characteristics of the turbidite fan in the Wenchang Formation of the Enping Sag, Pearl River Mouth Basin, China and its hydrocarbon significance显示文摘A turbidite fan in the Eocene upper Wenchang Formation in the Enping Sag, Pearl River Mouth Basin (PRMB) has been studied using seismic, logging and borehole data. The fan is characterized by parallel progradation on the dip seismic profile and is mound-shaped or lenticular-shaped on the strike seismic profile. The study of the core and logging data from well EP17-3-1, which is located in the front side of the turbidite fan, shows that this fan is a set of normal grading sand beds, interbedded within thick dark grey mudstones of semi-deep to deep lake deposits in the Wenchang Formation. The fan is interpreted as a sand/mud-rich turbidite fan that has an area of over 140 km2 and a maximum thickness of over 340 m. Combined with a study of the regional geological background and previous provenance analysis of the Eocene Wenchang Formation, the main potential provenances for the turbidite fan are considered to be the Panyu low-uplift and northern fault terrace zone. The Enping Sag is considered to be a half graben-like basin whose north side is faulted and whose south side is overlapped. Basement subsidence in the Eocene was mainly controlled by boundary faults which dip relatively steeply on the north side, causing the subsidence center of the Enping Sag in this stage to be close to the north boundary faults. Sustained faults developed in the Enping Sag during the Eocene caused an increase of the relative height difference between the north and the south uplift zone in the Enping Sag. Affected by the second episode of the Zhuqiong movement (39-36 Ma) in late Eocene, sediments which had accumulated on the Panyu low-uplift zone were triggered and moved toward the subsidence center of the Enping Sag and formed the turbidite fan. The second episode of the Zhuqiong movement is the most important triggering factor for the formation of the turbidite fan in the Wenchang Formation. Seismic attribute characterization shows that the low frequency energy is enhanced and high frequency energy is weakened when seismic waves propagate through the oil-bearing zone in this fan. Amplitude versus offset (AVO) anomalies are observed in the seismic data and abnormally high pressure is encountered. 11Effect of Stirring on Oil-Water Separation in Rare Earth Mixer-Settler显示文摘Oil-water separation is critical to solvent extraction process of rare earth, which can directly affect the yield and quality of the product. The experiments measure the two-phase separation time in a beaker, mixing uniformity of two phases in the mixer and the oil phase entrainment at oil exit by the Karl Fischer method and numerical simulation for the mixersettler to study the combined effect of gravity and stirring. Experimental results show that relative to the static situation, the separation efficiency resulted from low-speed stirring is increased by 25%. The water content in the oil is a minimum at an offset distance L of 10 cm and the clearance off the tank bottom z of 10 cm is as low as 0.49%. 13Double role of CoOeCo_(4)N hetero-nanocages as sulfur host for lithiumsulfur batteries显示文摘Lithium-sulfur batteries(LiSBs)are widely deemed as the most promising energy storage devices to substitute for traditional Li-ion batteries.However,its wide application is impeded by the soluble lithium polysulfides(LiPSs),which is called the shuttle effect,and the irregular distribution of final product Li_(2)S.Herein,based on the interfaces engineering,CoO-Co_(4)N hetero-nanocages are used as the sulfur host for LiSBs.Taking advantage of the polarity of CoO with the conductivity of Co_(4)N,CoO-Co_(4)N nanocages not only can provide large void space for sulfur volume fluctuation,but also can adsorb polysulfides and simultaneously regulate the nucleation of solid Li_(2)S by the‘trapping-diffusion-conversion’mechanism,which significantly enhances the redox kinetic of LiSBs and the utilization of active materials.Eventually,LiSBs with CoO-Co_(4)N nanocages host exhibit higher rate capacity(737 mAh·g^(-1) at 2 C)and cycling stability(662 mAh·g^(-1) at^(-1) C after 350 cycles).Even when the areal sulfur loading is as high as 3.0 mg cm^(-2),a high capacity of 713 mAh·g^(-1) can still be achieved after 100 cycles at 0.2 C.This host with sufficient polar-conductive interfaces expands‘trapping-diffusion-conversion’concept for the design of fast kinetic and high performance LSBs.Yi-Ming Lu Jin-Lin Yang Shi-Xi Zhao Xiang-Tian Zeng Lü-Qiang Yu Chao Huang 2021Journal of Materiomics2021,7,6:0
14Detection of captopril based on its enhanced resonance light scattering signals of fluorosurfactant-capped gold nanoparticles显示文摘In this study,based on its enhancement effect on resonance light scattering (RLS) of fluorosurfactant (FSN)-capped gold nanoparticles (GNPs),we reported a simple approach for the rapid sensing of captopril. Under optimum conditions,the lowest detectable concentration of captopril through this approach (S/N=3) was 0.01μg/mL. The calibration curve was linear over the range of 0.08-4.0μg/mL for the detection of captopril. The recoveries of captopril were found to fall in the range between 99% and 100%. We have validated the applicability of our method through the analyses of captopril in pharmaceutical formulations. 15Variations in leaf carbon isotope composition along an arid and semi-arid grassland transect in northern China显示文摘Aims We aimed to quantify the variation of leafδ^(13)C along an arid and semi-arid grassland transect in northern China.We also evaluated the effects of environmental factors(i.e.precipitation,temperature and altitude)on the spatial variation of leafδ^(13)C in northern grasslands and Tibetan Plateau,China.Method We sampled leaves of plant species belonging to three herb genera(Stipa spp.,Leymus spp.and Cleistogenes spp.)and three shrub genera(Caragana spp.,Reaumuria spp.and Nitraria spp.)for carbon isotope analysis from 50 locations along a 3200-km arid and semiarid grassland transect in northern China.Leafδ^(13)C data in Tibetan Plateau and northern grasslands in China were also compiled from studies in literature.Important Findings Along the transect,leafδ^(13)C for C_(3)plants ranged from−28.0‰to−23.3‰,and from−16.3‰to−13.8‰for C_(4)plant Cleistogenes spp..The change in leafδ^(13)C ranged from−0.26‰to−3.51‰with every 100 mm increase of annual precipitation,and leafδ^(13)C of shrubs(Nitraria spp.,Reaumuria spp.and Caragana spp.)responded more markedly to climatic factors(precipitation and temperature)than that of herbs(Stipa spp.,Leymus spp.and Cleistogenes spp.),indicating higher sensitivity of shrubδ^(13)C to climatic changes.The most important factor regulating spatial variations of leafδ^(13)C in Tibetan Plateau was altitude,while it was precipitation in northern grasslands.Our results suggested that shrubs are more adapted to increasing drought in arid and semi-arid grassland.Controls of environmental factors on leafδ^(13)C depended on the most limiting factors in arid grassland(precipitation)and Tibetan grasslands(atmospheric CO_(2)concentration).Chao Wang Dongwei Liu Wentao Luo Yunting Fang Xiaobo Wang Xiaotao Lü Yong Jiang Xingguo Han Edith Bai 2016Journal of Plant Ecology2016,9,5:3
16Establishment and validation of a model for brain injury state evaluation and prognosis prediction显示文摘Purpose:Traumatic brain injury(TBI)is one of the leading causes of disability and death in modern times,whose evaluation and prognosis prediction have been one of the most critical issues in TBI management.However,the existed models for the abovementioned purposes were defective to varying degrees.This study aims to establish an ideal brain injury state clinical prediction model(BISCPM).Methods:This study was a retrospective design.The six-month outcomes of patients were selected as the end point event.BISCPM was established by using the split-sample technology,and externally validated via different tests of comparison between the observed and predicted six-month mortality in validating group.TBI patients admitted from July 2006 to June 2012 were recruited and randomly divided into establishing model group and validating model group.Twenty-one scoring indicators were included in BISCPM and divided into three parts,A,B,and C.Part A included movement,pupillary reflex and diameter,CT parameters,and secondary brain insult factors,etc.Part B was age and part C was medical history of the patients.The total score of part A,B and C was final score of BISCPM.Results:Altogether 1156 TBI patients were included with 578 cases in each group.The score of BISCPM from validating group ranged from 2.75 to 31.94,averaging 13.64±5.59.There was not statistical difference between observed and predicted mortality for validating group.The discrimination validation showed that the BISCPM is superior to international mission for prognosis and analysis of clinical trials(IMPACT)lab model.Conclusion:BISCPM is an effective model for state evaluation and prognosis prediction of TBI patients.The use of BISCPM could be of great significance for decision-making in management of TBI.Xia Li Chao Lü Jun Wang Yi Wan Shu-Hui Dai Lei Zhang Xue-An Hu Xiao-Fan Jiang Zhou Fei 2020Chinese Journal of Traumatology2020,23,5:3
17Molecular behavior and interactions with microbes during anaerobic degradation of bio-derived DOM in waste leachate显示文摘It is the key to control bio-derived dissolved organic matters (DOM) in order to reduce the effluent concentration of wastewater treatment, especially for waste leachate with high organic contaminants. In the present study, the anaerobic degradation of aerobically stabilized DOM was investigated with DOM substrate isolated through electrodialysis. The degradation of bio-derived DOM was confirmed by reduction of 15%of total organic carbon in 100 days.We characterized the molecular behavior of bio-derived DOM by coupling molecular and biological information analysis. Venn based Sankey diagram of mass features showed the transformation of bio-derived DOM mass features. Occurrence frequency analysis divided mass features into six categories so as to distinguish the fates of intermediate metabolites and persistent compounds. Reactivity continuum model and machine learning technologies realized the semi-quantitative determination on the kinetics of DOM mass features in the form of pseudo-first order, and confirmed the reduction of inert mass features. Furthermore, network analysis statistically establish relationship between DOM mass features and microbes to identify the active microbes that are able to utilize bio-derived DOM. 19FBG Temperature and Pressure Sensing System for Hot Water Pipeline of Petrochemical Factory显示文摘A spatial and wavelength division multi- plexing fiber Bragg grating (FBG) sensing system is reported for monitoring the temperature and the pressure (T-P) of hot water pipeline in petrochemical factory. The FBG sensing system has 72 channels independently, and it provides the capability to monitor large number sensors at same time. 20Is the dinoflagellate Takayama xiamenensis a synonym of Takayama acrotrocha (Kareniaceae, Dinophyceae)?显示文摘The naked dinoflagellate Takayama acrotrocha was identified as responsible for a bloom in Shenzhen Bay,Guangdong,China,in early spring 2021.The identification was confirmed by light,scanning,and transmission electron microscopy and molecular data based on the LSU ribosomal DNA(rDNA)and ITS rDNA sequences.This is the first record of T.acrotrocha bloom in the South China Sea.The sulcus of T.acrotrocha was wide and extended onto the epicone as a short intrusion in general,sometime the intrusion was not apparent and some were finger-like.The apical groove was deeply sigmoid.The nucleus was large,ovoid to cup-shaped and occupied most of the epicone.A large,rounded pyrenoid surrounded by a starch sheath was located at the left side to the centre of the hypocone.Under epifluorescence illumination,a row of large vesicular knobs was observed on the upper border of the cingulum.The intraspecific morphological variabilities in the clonal cultures of T.acrotrocha were investigated carefully.Cells that share the same diagnostic characters used for the description of Takayama xiamenensis such as the finger-like sulcus,a large nucleus located in the epicone and the similar pyrenoid type were observed.The LSU rDNA sequences of T.acrotrocha and T.xiamenensis only differed in 3 base pairs(bp)for a sequence length of 673 bp(with a similarity of 99.55%).For these reasons,we propose T.xiamenensis as a junior synonym of T.acrotrocha.Songhui LÜ Aimin CHAO Qianyan LIANG Jingyi CEN Jianyan WANG Tao JIANG Si LI 2022Journal of Oceanology and Limnology2022,40,6:0
