| 2 | Fabrication of a hydroxyapatite-PDMS microfluidic chip for bone-related cell culture and drug screening显示文摘Bone is an important part of the human body structure and plays a vital role in human health.A microfluidic chip that can simulate the structure and function of bone will provide a platform for bone-related biomedical research.Hydroxyapatite(HA),a bioactive ceramic material,has a similar structure and composition to bone mineralization products.In this study,we used HA as a microfluidic chip component to provide a highly bionic bone environment.HA substrates with different microchannel structures were printed by using ceramic stereolithography(SLA)technology,and the minimum trench width was 50μm.The HA substrate with microchannels was sealed by a thin polydimethylsiloxane(PDMS)layer to make a HA-PDMS microfluidic chip.Cell culture experiments demonstrated that compared with PDMS,HA was more conducive to the proliferation and osteogenic differentiation of the human foetal osteoblast cell line(hFOB).In addition,the concentration gradient of the model drug doxorubicin hydrochloride(DOX)was successfully generated on a Christmas tree structure HA-PDMS chip,and the half maximal inhibitory concentration(IC50)of DOX was determined.The findings of this study indicate that the HA-PDMS microfluidic chip has great potential in the field of high-throughput bonerelated drug screening and bone-related research. | Qiangqiang Tang Xiaoyu Li Chen Lai Lei Li Hongkai Wu Yingjun Wang Xuetao Shi | 2021 | Bioactive Materials2021,6,1: | 5 |
| 3 | Analysis of severe human adenovirus infection outbreak in Guangdong Province,southern China in 2019显示文摘During 2018–2019,a severe human adenovirus(HAdV)infection outbreak occurred in southern China.Here,we screened 18 respiratory pathogens in 1704 children(≤14 years old)hospitalized with acute respiratory illness in Guangzhou,China,in 2019.In total,151 patients had positive HAdV test results;34.4%(52/151)of them exhibited severe illness.HAdV infection occurred throughout the year,with a peak in summer.The median patient age was 3.0(interquartile range:1.1–5.0)years.Patients with severe HAdV infection exhibited increases in12 clinical indexes(P≤0.019)and decreases in four indexes(P≤0.007),compared with patients exhibiting nonsevere infection.No significant differences were found in age or sex distribution according to HAdV infection severity(P>0.05);however,the distributions of comorbid disease and HAdV co-infection differed according to HAdV infection severity(P<0.05).The main epidemic types were HAdV-3(47.0%,71/151)and HAdV-7(46.4%,70/151).However,the severe illness rate was significantly higher in patients with HAdV-7(51.4%)than in patients with HAdV-3(19.7%)and other types of HAdV(20%)(P<0.001).Sequencing analysis of genomes/capsid genes of 13 HAdV-7 isolates revealed high similarity to previous Chinese isolates.A representative HAdV-7isolate exhibited a similar proliferation curve to the curve described for the epidemic HAdV-3 strain Guangzhou01(accession no.DQ099432)(P>0.05);the HAdV-7 isolate exhibited stronger virulence and infectivity,compared with HAdV-3(P<0.001).Overall,comorbid disease,HAdV co-infection,and high virulence and infectivity of HAdV-7 were critical risk factors for severe HAdV infection;these data can facilitate treatment,control,and prevention of HAdV infection. | Wenkuan Liu Shuyan Qiu Li Zhang Hongkai Wu Xingui Tian Xiao Li Duo Xu Jing Dai Shujun Gu Qian Liu Dehui Chen Rong Zhou | 2022 | Virologica Sinica2022,37,3: | 5 |

| 16 | The impacts of climate change on hydrology in a typical glacier region A case study in Hailuo Creek watershed of Mt.Gongga in China显示文摘The glaciers of the Hengduan Mountains play an important role in the hydrology processes of this region. In this study, the HBV Light model, which relies on a degree-day model to simulate glacier melting, was employed to simulate both glacier runoff and total runoff. The daily temperature and precipitation at the Hailuo Creek No. 1 Glacier from 1952 to 2009 were obtained from daily meteorological observed data at the glacier and from six national meteorological stations near the Hailuo Creek Basin. The daily air temperature, precipitation, runoff depth, and monthly potential evaporation in 1995, 1996, and 2002 were used to obtain a set of optimal parameters, and the annual total runoff and glacier runoff of the Hailuo Creek Glacier(1952–2009) were calculated using the HBV Light model. Results showed the average annual runoff in the Hailuo Creek Basin was 2,114 mm from 1952 to 2009, of which glacial melting accounted for about 1,078 mm. The river runoff in the Hailuo Creek catchment increased as a result of increased glacier runoff. Glacier runoff accounted for 51.1% of the Hailuo Creek stream flow in 1994 and increased to 72.6% in 2006. | 18 | Venovenous extracorporeal membrane oxygenation for COVID-19 and influenza H1N1 associated acute respiratory distress syndrome:A comparative cohort study in China显示文摘Background Venovenous extracorporeal membrane oxygenation(VV-ECMO)has been demonstrated to be effective in treating patients with virus-induced acute respiratory distress syndrome(ARDS).However,whether the management of ECMO is different in treating H1N1 influenza and coronavirus disease 2019(COVID-19)-associated ARDS patients remains unknown.Methods This is a retrospective cohort study.We included 12 VV-ECMO-supported COVID-19 patients admitted to The First Affiliated Hospital of Guangzhou Medical University,Guangzhou Eighth People's Hospital,and Wuhan Union Hospital West Campus between January 23 and March 31,2020.We retrospectively included VV-ECMO-supported patients with COVID-19 and H1N1 influenza-associated ARDS.Clinical characteristics,respiratory mechanics including plateau pressure,driving pressure,mechanical power,ventilatory ratio(VR)and lung compliance,and outcomes were compared.Results Data from 25 patients with COVID-19(n=12)and H1N1(n=13)associated ARDS who had received ECMO support were analyzed.COVID-19 patients were older than H1N1 influenza patients(P=0.004).The partial pressure of arterial carbon dioxide(PaCO_(2))and VR before ECMO initiation were significantly higher in COVID-19 patients than in H1N1 influenza patients(P<0.001 and P=0.004,respectively).COVID-19 patients showed increased plateau and driving pressure compared with H1N1 subjects(P=0.013 and P=0.018,respectively).Patients with COVID-19 remained longer on ECMO support than did H1N1 influenza patients(P=0.015).COVID-19 patients who required ECMO support also had fewer intensive care unit and ventilator-free days than H1N1.Conclusions Compared with H1N1 influenza patients,COVID-19 patients were older and presented with increased PaCO_(2) and VR values before ECMO initiation.The differences between ARDS patients with COVID-19 and influenza on VV-ECMO detailed herein could be helpful for obtaining a better understanding of COVID-19 and for better clinical management. | Yonghao Xu Yin Xi Shuijiang Cai Yuheng Yu Sibei Chen Weijie Guan Weibo Liang Hongkai Wu Weiqun He Xilong Deng Yuanda Xu Rong Zhang Manshu Li Jieyi Pan Zhenting Liang Ya Wang Shaofeng Kong Xiaoqing Liu Zheng Lv Yimin Li | 2023 | Journal of Intensive Medicine2023,3,4: | 0 |

| 20 | Schiff-base silver nanocomplexes formation on natural biopolymer coated mesoporous silica contributed to the improved curative effect on infectious microbes显示文摘Infectious microbes that spread easily in healthcare facilities remain as the severe threat for the public health,especially among immunocompromised populations.Given the intricate problem of dramatic increase in resistance to common biocides,the development of safe and efficient biocide formulated agents to alleviate drug resistance is highly demanding.In this study,Schiff-base ligands were successfully formed on natural biopolymer of epsilon-poly-L-lysine(ε-PL)decorated aldehyde functionalized mesoporous silica SBA-15(CHO-SBA-15)for the selective coordination of silver ions,which was affirmed by various physicochemical methods.Besides the identified broad-spectrum antibacterial activities,the as-prepared Schiff-base silver nanocomplex(CHO-SBA-15/ε-PL/Ag,CLA-1)exhibited an improved inhibitory effect on infectious pathogen growth typified by Escherichia coli and Staphylococcus aureus in comparison with two control silver complexes without Schiff-base conjugates,SBA-15/ε-PL/Ag and CHO-SBA-15/Ag,respectively.In addition,CLA-1 remarkably inhibited the growth of Mycobacterium tuberculosis due to the excellent antimicrobial activity of silver species.Significantly,CLA-1 kills Candida albicans cells,inhibits biofilm formation,and eliminates preformed biofilms,with no development of resistance during continuous serial passaging.The antifungal activity is connected to disruption of bacterial cell membranes and increased levels of intracellular reactive oxygen species.In mouse models of multidrug-resistant C.albicans infection,CLA-1 exhibited efficient in vivo fungicidal efficacy superior to two antifungal drugs,amphotericin B and fluconazole.Moreover,CLA-1 treatment induces negligible toxicity against normal tissues with safety.Therefore,this study reveals the pivotal role of the molecular design of Schiff-base silver nanocomplex formation on biopolymer surface-functionalized silica mesopores as a green and efficient nanoplatform to tackle infectious microbes. | Ling Cai Yanqiang Huang Yuanyuan Duan Qiao Liu Qilan Xu Jia Jia Jianming Wang Qian Tong Peipei Luo Yujie Wen Luming Peng Qian Wu Xudong Hang Huijun Jiang Ping Zhu Yanmei Yang Boshen Zhou Liping Zeng Hongkai Bi Jin Chen | 2021 | Nano Research2021,14,8: | 0 |
