| 2 | Therapeutic imaging window of cerebral infarction revealed by multisequence magnetic resonance imaging An animal and clinical study显示文摘In this study, we established a Wistar rat model of right middle cerebral artery occlusion and observed pathological imaging changes (T2-weighted imaging [T2WI], T2FLAIR, and diffusion-weighted imaging [DWI]) following cerebral infarction. The pathological changes were divided into three phases: early cerebral infarction, middle cerebral infarction, and late cerebral infarction. In the early cerebral infarction phase (less than 2 hours post-infarction), there was evidence of intracellular edema, which improved after reperfusion. This improvement was defined as the ischemic penumbra. In this phase, a high DWI signal and a low apparent diffusion coefficient were observed in the right basal ganglia region. By contrast, there were no abnormal T2WI and T2FLAIR signals. For the middle cerebral infarction phase (2-4 hours post-infarction), a mixed edema was observed. After reperfusion, there was a mild improvement in cell edema, while the angioedema became more serious. A high DWI signal and a low apparent diffusion coefficient signal were observed, and some rats showed high T2WI and T2FLAIR signals. For the late cerebral infarction phase (4-6 hours post-infarction), significant angioedema was visible in the infarction site. After reperfusion, there was a significant increase in angioedema, while there was evidence of hemorrhage and necrosis. A mixed signal was observed on DWI, while a high apparent diffusion coefficient signal, a high T2WI signal, and a high T2FLAIR signal were also observed. All 86 cerebral infarction patients were subjected to T2WI, T2FLAIR, and DWI. MRI results of clinic data similar to the early infarction phase of animal experiments were found in 51 patients, for which 10 patients (10/51) had an onset time greater than 6 hours. A total of 35 patients had MRI results similar to the middle and late infarction phase of animal experiments, of which eight patients (8/35) had an onset time less than 6 hours. These data suggest that defining the 'therapeutic time window' as the time 6 hours after infarction may not be suitable for all patients. | 3 | Gene interference regulates aquaporin-4 expression in swollen tissue of rats with cerebral ischemic edema Correlation with variation in apparent diffusion coefficient显示文摘To investigate the effects of mRNA interference on aquaporin-4 expression in swollen tissue of rats with ischemic cerebral edema, and diagnose the significance of diffusion-weighted MRI, we injected 5 μL shRNA- aquaporin-4 (control group) or siRNA- aquaporin-4 solution (1:800) (RNA interference group) into the rat right basal ganglia immediately before occlusion of the middle cerebral artery. At 0.25 hours after occlusion of the middle cerebral artery, diffusion-weighted MRI displayed a high signal; within 2 hours, the relative apparent diffusion coefficient decreased markedly, aquaporin-4 expression increased rapidly, and intracellular edema was obviously aggravated; at 4 and 6 hours, the relative apparent diffusion coefficient slowly returned to control levels, aquaporin-4 expression slightly increased, and angioedema was observed. In the RNA interference group, during 0.25- 6 hours after injection of siRNA- aquaporin-4 solution, the relative apparent diffusion coefficient slightly fluctuated and aquaporin-4 expression was upregulated; during 0.5-4 hours, the relative apparent diffusion coefficient was significantly higher, while aquaporin-4 expression was significantly lower when compared with the control group, and intracellular edema was markedly reduced; at 0.25 and 6 hours, the relative apparent diffusion coefficient and aquaporin-4 expression were similar when compared with the control group; obvious angioedema remained at 6 hours. Pearson's correlation test results showed that aquaporin-4 expression was negatively correlated with the apparent diffusion coefficient (r = -0.806, P < 0.01). These findings suggest that upregulated aquaporin-4 expression is likely to be the main molecular mechanism of intracellular edema and may be the molecular basis for decreased relative apparent diffusion coefficient. Aquaporin-4 gene interference can effectively inhibit the upregulation of aquaporin-4 expression during the stage of intracellular edema with time-effectiveness. Energy dispersive X-ray spectroscopy(EDX), X-ray diffraction(XRD), X-Ray photoelectron spectroscopy(XPS),high resolution transmission electron microscopy(HRTEM) detection show that high quality AIN nanowires were prepared. Nanostructures including nanorings, nanosprings, nanohelices, chainlike nanowires, six-fold symmetric nanostructure and rod-like structure were successfully obtained by controlling the growth duration and temperature. | 6 | Sensitivity analysis of longitudinal cracking on asphalt pavement using MEPDG in permafrost region显示文摘Longitudinal cracking is one of the most important distresses of asphalt pavement in permafrost regions. The sensitivity analysis of design parameters for asphalt pavement can be used to study the influence of every parameter on longitudinal cracking, which can help optimizing the design of the pavement structure. In this study, 20 test sections of Qinghai e Tibet Highway were selected to conduct the sensitivity analysis of longitudinal cracking on material parameter based on Mechanistic-Empirical Pavement Design Guide(MEPDG) and single factorial sensitivity analysis method. Some computer aided engineering(CAE) simulation techniques, such as the Latin hypercube sampling(LHS) technique and the multiple regression analysis are used as auxiliary means.Finally, the sensitivity spectrum of material parameter on longitudinal cracking was established. The result shows the multiple regression analysis can be used to determine the remarkable influence factor more efficiently and to process the qualitative analysis when applying the MEPDG software in sensitivity analysis of longitudinal cracking in permafrost regions. The effect weights of the three parameters on longitudinal cracking in descending order are air void, effective binder content and PG grade. The influence of air void on top layer is bigger than that on middle layer and bottom layer. | 7 | A review on the best practices in concrete pavement design and materials in wet-freeze climates similar to Michigan显示文摘The research presented in this paper aims to identify best practices of design and materials for concrete pavements in wet-freeze climates similar to the Michigan State. For the purposes of this paper, a best practice is a procedure that has been shown by field-validated research or experience to produce improved results and that is established or proposed as a standard suitable for widespread implementation. The local wet-freeze climate makes the requirements for Michigan's pavement system different from many other regions. Wetfreeze climates can result in various concrete pavement distress mechanisms such as thermally-induced cracking, freeze-thaw deterioration, accelerated cracking due to loss of support, frost heave, and material degradation. Therefore, appropriate procedures for design and material selection need to be selected to withstand high precipitation and freezing winter temperatures. Failure to take into account the climatic conditions may lead to inadequate or reduced pavement performance. However, utilizing appropriate techniques and materials could potentially improve the quality and increase the service life of the concrete pavement. Three design methods and five materials have been identified, and examples of their successful performance in wet-freeze climates are provided. In addition, the reasons that give them the superior performance in wet-freeze climates are discussed in detail. | Naser P. | 10 | Sleep architecture, periodic breathing and mood disturbance of expeditioners at Kunlun Station(4087 m) in Antarctica显示文摘Several studies have reported the detrimental impacts of hypoxia exposure on sleep.Chinese Kunlun Station(altitude 4087 m)is located at Dome A,the highest point on the Antarctic ice sheet and one of the most extreme environments on Earth.This study investigated alteration of sleep,breathing and mood status in healthy expeditioners at Kunlun Station at Dome A.The study examined 10 male volunteers of the inland transverse party to Kunlun Station during the 31st Chinese National Antarctic Research Expedition,and valid data from eight volunteers were analyzed.Sleep structure,breathing pattern and mood were monitored using portable polysomnography(PSG)and profile of mood state(POMS)at two time points:(1)at Zhongshan Station(10 m)before departure to Kunlun Station;(2)on nights 12–13 of residence at Kunlun Station.Slow-wave sleep(Stage 3 non-rapid eye movement)was markedly reduced at Kunlun Station(P<0.01).Total sleep time,sleep efficiency and sleep latency showed no significant changes.Total respiratory events(P<0.05),apnea/hypopnea index(AHI)(P<0.05)and hypopnea index(P<0.01)substantially increased at Kunlun Station.The most common respiratory disorder was periodic breathing,occurring almost exclusively during non-rapid eye movement sleep.The oxygen desaturation index increased markedly(P<0.05),while nocturnal oxygen saturation dramatically fell at Kunlun Station(P<0.05).Vigor scores decreased at Kunlun Station(P<0.05).Expeditioners exhibited reduced slow wave sleep,induced periodic breathing,decreased oxygen saturation and decreased vigor at Kunlun Station. | XU Chengli LIU Shiying KONG Zhanping CHEN Nan | 2020 | Advances in Polar Science2020,31,3: | 0 |
