1miR-137 inhibits melanoma cell proliferation through downregulation of GLO1显示文摘Late-stage melanoma is refractory to current therapies. MicroRNAs (miRNAs) can modulate many physiological and pathological processes of melanoma. Studies have demonstrated that miR-137 acts as a tumor suppressor by inhibiting the proliferation of melanoma cells through targeting multiple mRNAs. The glyoxalase system member glyoxalase 1 (GLO1) is the principal scavenging enzyme of methylglyoxal (MG), a toxic byproduct of glycolysis. Using 35S in vivo/vitro labelling analysis for dynamic proteomics (SiLAD), we found that miR-137 downregulated the expression of GLO1 in melanoma cells.Bioinformatics analysis predicted that GLO1 is a direct target of miR-137. This was validated by dual luciferase reporter assay.Quantitative RT-PCR (qRT-PCR) and western blot analysis indicated that miR-137 could decrease endogenous GLO1 expression.Furthermore, siRNA targeting of GLO1 mimicked inhibition of melanoma cell proliferation caused by miR-137 overexpression.Re-expression of GLO1 was able to restore miR-137-mediated suppression of melanoma cell proliferation. 3Residual current compensation for single-phase grounding fault in coal mine power network显示文摘The way of neutral point to earth via full compensation arc suppression coil can solve the problem of residual current compensation in coal mine power network effectively. Based on the analysis on the grounding current detection results of Xieqiao coal mine, the conclusion that harmonic component of grounding current is dominated by higher harmonics with complex harmonic sources in coal mine power network system was obtained. The influences of harmonic source type and fault point position on harmonic voltage and harmonic current were analyzed theoretically. The influences of earthed fault feeder detection result and the estimation errors of parameters to earth on residual current compensation were analyzed. A new thought of residual current prediction and the selections of model method and control method were proposed on this basis. The simulation results prove that harmonic amplitudes of zero sequence voltage and zero sequence current are determined by harmonic source type as well as fault point position in coal mine power network, and also prove that zero sequence voltage detection can avoid the unstable problem of coal mine power network system caused by undercompensation of capacitive current. 4High-throughput screening for amyloid-β binding natural small-molecules based on the combinational use of biolayer interferometry and UHPLC−DAD-Q/TOF-MS/MS显示文摘Discovery of drugs rapidly and effectively is an important aspect for Alzheimer's disease(AD).In this study,a novel high-throughput screening(HTS)method aims at screening the small-molecules with amyloid-β(Aβ)binding affinity from natural medicines,based on the combinational use of biolayer interferometry(BLI)and ultra-high-performance liquid chromatography coupled with diode-array detector and quadrupole/time-of-flight tandem mass spectrometry(UHPLC−DAD-Q/TOF-MS/MS)has been firstly developed.Briefly,the components in natural medicines disassociated from biotinylated Aβwere collected to analyze their potential Aβbinding affinity by UHPLC−DAD-Q/TOF-MS/MS.Here,baicalein was confirmed to exhibit the highest binding affinity with Aβin Scutellaria baicalensis.Moreover,polyporenic acid C(PPAC),dehydrotumulosic acid(DTA),and tumulosic acid(TA)in Kai-Xin-San(KXS)were also identified as potent Aβinhibitors.Further bioactivity validations indicated that these compounds could inhibit Aβfibrillation,improve the viability in Aβ-induced PC-12 cells,and decrease the Aβcontent and improve the behavioral ability in Caenorhabditis elegans.The molecular docking results confirmed that PPAC,DTA,and TA possessed good binding properties with Aβ.Collectively,the present study has provided a novel and effective HTS method for the identification of natural inhibitors on Aβfibrillation,which may accelerate the process on anti-AD drugs discovery and development.Minsong Guo Fengdan Zhu Wenqiao Qiu Gan Qiao Betty Yuen-Kwan Law Lu Yu Jianming Wu Yong Tang Chonglin Yu Dalian Qin Xiaogang Zhou Anguo Wu 2022Acta Pharmaceutica Sinica B2022,12,4:3
5C.elegans-based screen identifies lysosome-damaging alkaloids that induce STAT3-dependent lysosomal cell death显示文摘Lysosomes are degradation and signaling centers within the cell,and their dysfunction impairs a wide variety of cellular processes.To understand the cellular effect of lysosome damage,we screened natural smallmolecule compounds that induce iysosomal abnormality using Caenorhabditis elegans (C.elegans)as a model system.A group of vobasinyl-ibogan type bisindole alkaloids (ervachinines A-D)were identified that caused lysosome enlargement in C.elegans macrophage-like cells.Intriguingly,these compounds triggered cell death in the germ line independently of the canonical apoptosis pathway.In mammalian cells, ervachinines A-D induced lysosomal enlargement and damage,leading to leakage of cathepsin proteases, inhibition of autophagosome degradation and necrotic cell death.Further analysis revealed that this ervachinine-induced lysosome damage and lysosomal cell death depended on STAT3 signaling,but not RIP1 or RIP3 signaling.These findings suggest that lysosomedamaging compounds are promising reagents for dissecting signaling mechanisms underlying lysosome homeostasis and lysosome-related human disorders.Yang Li Yu Zhang Qiwen Gan Meng Xu Xiao Ding Guihua Tang Jingjing Liang Kai Liu Xuezhao Liu Xin Wang Lingli Guo Zhiyang Gao Xiaojiang Hao Chonglin Yang 2018Protein & Cell2018,9,12:2
6Defective arginine metabolism impairs mitochondrial homeostasis in Caenorhabditis elegans显示文摘Arginine catabolism involves enzyme-dependent reactions in both mitochondria and the cytosol,defects in which may lead to hyperargininemia,a devastating developmental disorder.It is largely unknown if defective arginine catabolism has any effects on mitochondria.Here we report that normal arginine catabolism is essential for mitochondrial homeostasis in Caenorhabditis elegans.Mutations of the arginase gene argn-1 lead to abnormal mitochondrial enlargement and reduced adenosine triphosphate(ATP)production in C elegans hypodermal cells.ARGN-1 localizes to mitochondria and its loss causes arginine accumulation,which disrupts mitochondrial dynamics.Heterologous expression of human ARGl or ARG2 rescued the mitochondrial defects of argn-1 mutants.Importantly,genetic inactivation of the mitochondrial basic amino acid transporter SLC-25A29 or the mitochondrial glutamate transporter SLC-25A18.1 fully suppressed the mitochondrial defects caused by argn-1 mutations.These findings suggest that mitochondrial damage probably contributes to the pathogenesis of hyperargininemia and provide clues for developing therapeutic treatments for hyperargininemia.Ruofeng Tang Xin Wang Junxiang Zhou Fengxia Zhang Shan Zhao Qiwen Gan Liyuan Zhao Fengyang Wang Qian Zhang Jie Zhang Guodong Wang Chonglin Yang 2020Journal of Genetics and Genomics2020,47,3:2
