1Recent advances in cell sheet technology for bone and cartilage regeneration: from preparation to application显示文摘Bone defects caused by trauma,tumour resection,infection and congenital deformities,together with articular cartilage defects and cartilage–subchondral bone complex defects caused by trauma and degenerative diseases,remain great challenges for clinicians.Novel strategies utilising cell sheet technology to enhance bone and cartilage regeneration are being developed.The cell sheet technology has shown great clinical potential in regenerative medicine due to its effective preservation of cell–cell connections and extracellular matrix and its scaffold-free nature.This review will first introduce several widely used cell sheet preparation systems,including traditional approaches and recent improvements,as well as their advantages and shortcomings.Recent advances in utilising cell sheet technology to regenerate bone or cartilage defects and bone–cartilage complex defects will be reviewed.The key challenges and future research directions for the application of cell sheet technology in bone and cartilage regeneration will also be discussed.Yuezhi Lu Wenjie Zhang Jie Wang Guangzheng Yang Shi Yin Tingting Tang Chunhua Yu Xinquan Jiang 2019International Journal of Oral Science2019,11,2:14
2Evaluation of the Trend Uncertainty in Summer Ozone Valley over the Tibetan Plateau in Three Reanalysis Datasets显示文摘Trend uncertainty in the ozone valley over the Tibetan Plateau(OVTP)and the South Asian high(SAH)during1979–2009 in ERA-Interim(interim reanalysis data from the ECMWF),JRA-55(55-yr reanalysis data from the Japan Meteorological Agency),and NCEP-CFSR(Climate Forecast System Reanalysis)datasets was evaluated.The results showed that the NCEP-CFSR OVTP became strong in the summers of 1979–2009,whereas it became weak according to ERA-Interim and JRA-55.Satellite data merged with TOMS(Total Ozone Mapping Spectrometer)and OMI(Ozone Monitoring Instrument)agreed with the OVTP trend of NCEP-CFSR.The OVTP strengthening in NCEP-CFSR may have been caused by SAH intensification,a rising tropopause,and increasing ozone over non-TP(non-Tibetan Plateau)areas(27°–37°N,<75°E and>105°E).Analogously,the OVTP weakening in ERA-Interim and JRA-55 may have been affected by weakening SAH,descending tropopause,and decreasing non-TP ozone.Dong GUO Yucheng SU Xiuji ZHOU Jianjun XU Chunhua SHI Yu LIU Weiliang LI Zhenkun LI 2017Journal of Meteorological Research2017,31,2:13
3Different Expression Analysis in Fruit Softening and Ethylene Biosynthetic Pathways in Peaches of Different Flesh Textures显示文摘The aim of our study was to assess differences in the expression of genes involved in fruit softening and ethylene biosynthetic pathways under different temperature storage conditions. Different peach cultivars of ‘Xiacui' and ‘Yumyeong', which are stonyhard, ‘Yinhualu', which is softmelting, ‘Hujing Milu', which is hard-melting, and ‘Baby Gold 6', which is non-melting at 80% ripening, were collected as test materials. The results showed that only slight ethylene production was detected after harvesting of ‘Yumyeong' and ‘Xiacui' under either a room temperature(25 °C) or low temperature(4 °C). The fruit firmness of stonyhard cultivars was retained at a high level under room temperature over time, whereas a low temperature induced ‘Yumyeong' fruit to soften. Quantitative real-time PCR results indicated that the PpACS1 gene was highly expressed in soft-melting, hard-melting and non-melting cultivars; however, expression was extremely low in stonyhard peaches. PpACS2 or PpACS3, however,was not detected in all five cultivars. Interestingly, cold treatment significantly decreased firmness along with endo-PG expression obviously upregulated in ‘Yumyeong', but not in ‘Xiacui' peaches. 4The root structures of 21 aquatic plants in a macrophyte-dominated lake in China显示文摘Aims Aquatic plants play an important role in freshwater ecosystems.Previous works have largely focused on the functional significance of plant above ground parts,with much less attention on the root structures of aquatic plants.In this study,we divided 21 aquatic plants(including five introduced plants)into multiple plant groups(different life forms,monocot/eudicot and introduced/native)with the goal of addressing two questions:(i)what root structures do aquatic plants exhibit,and(ii)are there differences among these plant groups?Methods Twenty-one aquatic plants belonging to four life forms(free-float-ing,emergent,floating-leaved and submerged)were collected at the near flowering stage from a typical macrophyte-dominated lake in the yangtze River Basin,China.The following root topological parameters were quantified:altitude(a),path length(pe),magnitude(M),mean topological length(b),topological index(TI)and normed indices qa and qb.Important Findings The root topological indices TI,qa and qb for the 21 aquatic plants were 0.724±0.013,0.290±0.031 and 0.152±0.024(means±S.E.),respectively,revealing a general pattern of dichotomous branching,except for the aquatic root of Myriophyllum aquati-cum(Vell.)Verdc.,which displays herringbone branching.All three topological indices were significantly lower for monocots(TI=0.700±0.130,qa=0.191±0.149 and qb=0.086±0.236)than eudicots(TI=0.752±0.206,qa=0.405±0.569 and qb=0.229±0.393),indicating that the roots of monocots are typi-cally more dichotomous-like than those of eudicots.Among the four life forms,the three topological indices for emergent plants(TI=0.832±0.006,qa=0.616±0.018 and qb=0.381±0.014)were significantly higher than those of the other three life forms.Overall,there was no difference between the topological indices of introduced and native aquatic plants,but the introduced species M.aquaticum and Alternanthera philoxeroides(Mart.)Griseb.had both aquatic and edaphic roots as well as unusual functions,which may help explain their strong viability.Xiaolong Huang Ligong Wang Xin Guan Yuanyuan Gao Chunhua Liu Dan Yu 2018Journal of Plant Ecology2018,11,1:6
5Mesenchymal stem cell-derived small extracellular vesicles mitigate oxidative stress-induced senescence in endothelial cells via regulation of miR-146a/Src显示文摘Senescent endothelial cells (ECs) could impair the integrity of the blood vessel endothelium, leading to vascular aging and a series of diseases, such as atherosclerosis, diabetes. Preventing or mitigating EC senescence might serve as a promising therapeutic paradigm for these diseases. Recent studies showed that small extracellular vesicles (sEV) have the potential to transfer bioactive molecules into recipient cells and induce phenotypic changes. Since mesenchymal stem cells (MSCs) have long been postulated as an important source cell in regenerative medicine, herein we investigated the role and mechanism of MSC-derived sEV (MSC-sEV) on EC senescence. In vitro results showed that MSC-sEV reduced senescent biomarkers, decreased senescence-associated secretory phenotype (SASP), rescued angiogenesis, migration and other dysfunctions in senescent EC induced by oxidative stress. In the In vivo natural aging and type-2 diabetes mouse wound-healing models (both of which have senescent ECs), MSC-sEV promoted wound closure and new blood vessel formation. Mechanically, miRNA microarray showed that miR-146a was highly expressed in MSC-sEV and also upregulated in EC after MSC-sEV treatment. miR-146a inhibitors abolished the stimulatory effects of MSC-sEV on senescence. Moreover, we found miR-146a could suppress Src phosphorylation and downstream targets VE-cadherin and Caveolin-1. 6Physiological characteristics and commercial application of edible mushroom dietary fiber显示文摘Edible mushrooms are considered as healthy food because they are low in calories and fat but rich in proteins minerals and dietary fiber(DF). Edible mushrooms are recognized as new potential resource of DF since the components of edible mushroom dietary fiber(EMDF) have shown special physiological and pharmacological effects on human and animals. In this article,the soluble and insoluble fractions of DF in different edible mushroom species have been evaluated. Biological effects of EMDF are related to promoting desired responses,for example,reducing blood cholesterol,protecting cells from free radicals attack by antioxidative effects,attenuating levels and fluctuations of blood glucose and selectively supporting the growth of beneficial gut bacteria. The EMDF plays an important role in reducing risk of cardiovascular diseases,diabetes mellitus and intestinal diseases. The non-starch polysaccharides(NSP),a kind of EMDF,is the best known and most potent mushroom-derived substances with antitumor and immunomodulatory properties. EMDF has also been reported to take part in the control of body weight,lipid homeostasis and insulin sensitivity due to its effect on specific chemical structures and physical properties. Many pharmaceutical substances with potent and unique health- enhancing properties were isolated recently from edible mushrooms and distributed worldwide. Mushroom-based dietary supplements(DSs) with potential therapeutic effects are produced from the mycelia or the fruiting bodies of mushrooms,and are consumed in the forms of capsules,tablets,or extracts. 7Effects of Acanthopanax senticosus on learning and memory in a mouse model of Alzheimer's disease and protection against free radical injury to brain tissue显示文摘BACKGROUND: Acanthopanax senticosus,a plant of the Araliaceae family,is used in traditional Chinese medicine. It can be used to replenish Qi,strengthen the spleen,tonify the kidney,and relieve mental strain. OBJECTIVE: To observe effects of Acanthopanax senticosus on learning and memory in a mouse model of Alzheimer's disease and abnormal biochemical changes in the brain tissue. DESIGN: A completely randomized grouping,controlled animal experiment. SETTING: Department of Preventive Medicine,School of Basic Medical Sciences,Yanbian University. MATERIALS: A total of 50 Kunming mice,aged 1-1.5 months,equal numbers of males and females,were provided by the Laboratory Animal Center,Yanbian University Medical College. The study was performed in accordance with ethical guidelines for the use and care of animals. Acanthopanax was provided by Yanbian Chengda Pharmaceutical Co.,Ltd. Acanthopanax senticosus (0.5 kg) was soaked in water for 1 hour and transferred to 1.5 kg distilled water for extraction. It was boiled for 1 hour and extracted after 1 hour of boiling. The procedure was repeated 3 times. The extract was condensed to 500 mL and then adjusted to 500 and 1 000 g/L with water. Piracetam tablets were produced by Shandong Luoxin Pharmaceutical Corporation,China. Malonaldehyde (MDA),superoxide dismutase (SOD),and acetylcholinesterase (AChE) kits were purchased from Nanjing Jiancheng Bioengineering Co.,Ltd.,China. METHODS: This study was performed at the Department of Preventive Medicine,School of Basic Medical Sciences,Yanbian University from January to June 2007. All mice were randomly divided into 5 groups with 10 mice in each: control group,model group,low-,and high-dose Acanthopanax senticosus-treated groups,and piracetam-treated group. All groups were administered 0.1 mL/10 g. In the control and model groups,mice were intragastrically administered saline each morning for 5 days prior to experimentation. Five days later,they were intraperitoneally perfused with saline and aluminum trichloride (100 mg/kg),respectively,every other afternoon. In the low-and high-dose Acanthopanax senticosus-treated groups,as well as the piracetam-treated groups,mice were intragastrically administered 500 and 1 000 g/L Acanthopanax senticosus and 23 g/L piracetam suspension,respectively,every morning. Five days later,they were intraperitoneally perfused with aluminum trichloride (100 mg/kg) every other afternoon,20 successive times. MAIN OUTCOME MEASURES: On days 20,23,26,and 29 after treatment,time-to-platform,number of errors,and accuracy were measured by water maze. After anesthesia,mice were euthanized and whole brain tissues were immediately resected and homogenized. MDA levels,and SOD and AChE activities,were measured using the corresponding kits. RESULTS: Fifty mice were included in the final analysis. In the model group,accuracy increased,and time-to-platform was prolonged,compared to control group (P < 0.05-0.01). In the model group,MDA levels significantly increased (P < 0.05),while SOD activity significantly decreased (P < 0.01),compared to control group. MDA levels were significantly lower and SOD activity was significantly higher in the low-and high-dose Acanthopanax senticosus-treated groups,compared to the model group (P < 0.05-0.01). In the model group,AChE activity significantly increased,compared to the control group (P < 0.05). AChE activity was significantly lower in the high-dose Acanthopanax senticosus-treated and the piracetam-treated groups than in the model group (P < 0.05). 8Evaluating the Brewer–Dobson circulation and its responses to ENSO,QBO,and the solar cycle in different reanalyses显示文摘This study compares the climatology and long-term trend of northern winter stratospheric residual mean meridional circulation(RMMC), as well as its responses to El Ni?o-Southern Oscillation(ENSO), stratospheric Quasi Biennial Oscillation(QBO), and solar cycle in ten reanalyses and a stratosphere-resolving model, CESM1-WACCM. The RMMC is a large-scale meridional circulation cell in the stratosphere, usually referred to as the estimate of the Brewer Dobson circulation(BDC). The distribution of the BDC is generally consistent among multiple reanalyses except that the NOAA twentieth century reanalysis(20RC) largely underestimates it. Most reanalyses(except ERA40 and ERA-Interim) show a strengthening trend for the BDC during 1979–2010. All reanalyses and CESM1-WACCM consistently reveal that the deep branch of the BDC is significantly enhanced in El Ni?o winters as more waves from the troposphere dissipate in the stratospheric polar vortex region. A secondary circulation cell is coupled to the temperature anomalies below the QBO easterly center at 50 hPa with tropical upwelling/cooling and midlatitude downwelling/warming, and similar secondary circulation cells also appear between 50–10 hPa and above 10 hPa to balance the temperature anomalies. The direct BDC response to QBO in the upper stratosphere creates a barrier near 30°N to prevent waves from propagating to midlatitudes, contributing to the weakening of the polar vortex. The shallow branch of the BDC in the lower stratosphere is intensified during solar minima, and the downwelling warms the Arctic lower stratosphere. 9Hydrophilic bi-functional B-doped g-C_(3)N_(4) hierarchical architecture for excellent photocatalytic H_(2)O_(2) production and photoelectrochemical water splitting显示文摘Graphitic carbon nitride(g-C_(3)N_(4))has attracted great interest in photocatalysis and photoelectrocatalysis.However,their poor hydrophilicity poses a great challenge for their applications in aqueous environment.Here,we demonstrate synthesis of a hydrophilic bi-functional hierarchical architecture by the assembly of B-doped g-C_(3)N_(4)nanoplatelets.Such hierarchical B-doped g-C_(3)N_(4)material enables full utilization of their highly enhanced visible light absorption and photogenerated carrier separation in aqueous medium,leading to an excellent photocatalytic H_(2)O_(2)production rate of 4240.3μM g^(-1)h^(-1),2.84,2.64 and 2.13 times higher than that of the bulk g-C_(3)N_(4),g-C_(3)N_(4)nanoplatelets and bulk B doped g-C_(3)N_(4),respectively.Photoanodes based on these hierarchical architectures can generate an unprecedented photocurrent density of 1.72 m A cm^(-2)at 1.23 V under AM 1.5 G illumination for photoelectrochemical water splitting.This work makes a fundamental improvement towards large-scale exploitation of highly active,hydrophilic and stable metal-free g-C_(3)N_(4)photocatalysts for various practical applications.Yang Ding Soumyajit Maitra Chunhua Wang Runtian Zheng Meiyu Zhang Tarek Barakat Subhasis Roy Jing Liu Yu Li Tawfique Hasan Bao-Lian Su 2022Journal of Energy Chemistry2022,31,7:3
10Spatial structure of turbulent mixing of an anticyclonic mesoscale eddy in the northern South China Sea显示文摘Upper turbulent mixing in the interior and surrounding areas of an anticyclonic eddy in the northern South China Sea(SCS)was estimated from underwater glider data(May 2015)in the present study,using the Gregg-HenyeyPolzin parameterization and the Thorpe-scale method.The observations revealed a clear asymmetrical spatial pattern of turbulent mixing in the anticyclonic eddy area.Enhanced diffusivity(in the order of 10–3 m2/s)was found at the posterior edge of the anticyclonic mesoscale eddy;on the anterior side,diffusivity was one order of magnitude lower on average.This asymmetrical pattern was highly correlated with the eddy kinetic energy.Higher shear variance on the posterior side,which is conducive to the triggering of shear instability,may be the main mechanism for the elevated diffusivity.In addition,the generation and growth of sub-mesoscale motions that are fed by mesoscale eddies on their posterior side may also promote the occurrence of strong mixing in the studied region.The results of this study help improve our knowledge regarding turbulent mixing in the northern SCS.Yongfeng Qi Chenjing Shang Huabin Mao Chunhua Qiu Changrong Liang Linghui Yu Jiancheng Yu Xiaodong Shang 2020Acta Oceanologica Sinica2020,39,11:2
11Comparison of general practice residents’ attitudes and perceptions about training in two programmes in China: a mixed methods survey显示文摘Objective To understand general practice(GP)residents’attitudes about their residency training in China.Design Mixed methods survey administered cross-sectionally.Setting Two GP training programmes similar in most regards according to current GP training policy of 5 years’undergraduate degree in medicine and 3 years of postgraduate GP residency training-but differing as the Beijing programme has adopted educational innovations beyond the nationally prescribed standard curriculum used by the second Shenzhen programme.Participants 105(85%)of eligible GP trainees,35(90%)in the innovative Beijing programme and 70(83%)in the standard training Shenzhen programme.Results Overall,residents felt discrimination because of specialty choice,and that they lacked competency as a general practitioner.Many residents commented faculty had negative teaching attitudes.Beijing residents were more satisfied than Shenzhen residents with their training(p=0.001),and felt teaching faculty had sufficient knowledge(p<0.001),and appropriate attitudes towards teaching(p=0.004).Beijing residents more strongly agreed on five items about good future job prospects(all p<0.05).Conclusion These Chinese GP residents identify areas for improvement in their training as well as strengths.Higher satisfaction with faculty teaching and job optimism in Beijing where GP residents receive training from specifically qualified faculty,and can earn special certification,suggest that the educational innovations enhance training and promote positivity about job prospects.These findings imply that GP residents in China face many training challenges that are similar to other international reports,while also implicating benefits of using an innovative curricular approach.Siqing Lian Yu Xia Jinzhi Zhang Xiaoning Han Chunhua Chi Michael D Fetters 2019Family Medicine and Community Health2019,7,4:2
