1The second natural gas hydrate production test in the South China Sea显示文摘Clayey silt reservoirs bearing natural gas hydrates(NGH)are considered to be the hydrate-bearing reservoirs that boast the highest reserves but tend to be the most difficult to exploit.They are proved to be exploitable by the first NGH production test conducted in the South China Sea in 2017.Based on the understanding of the first production test,the China Geological Survey determined the optimal target NGH reservoirs for production test and conducted a detailed assessment,numerical and experimental simulation,and onshore testing of the reservoirs.After that,it conducted the second offshore NGH production test in 1225 m deep Shenhu Area,South China Sea(also referred to as the second production test)from October 2019 to April 2020.During the second production test,a series of technical challenges of drilling horizontal wells in shallow soft strata in deep sea were met,including wellhead stability,directional drilling of a horizontal well,reservoir stimulation and sand control,and accurate depressurization.As a result,30 days of continuous gas production was achieved,with a cumulative gas production of 86.14×104 m3.Thus,the average daily gas production is 2.87×10^4 m^3,which is 5.57 times as much as that obtained in the first production test.Therefore,both the cumulative gas production and the daily gas production were highly improved compared to the first production test.As indicated by the monitoring results of the second production test,there was no anomaly in methane content in the seafloor,seawater,and atmosphere throughout the whole production test.This successful production test further indicates that safe and effective NGH exploitation is feasible in clayey silt NGH reservoirs.The industrialization of hydrates consists of five stages in general,namely theoretical research and simulation experiments,exploratory production test,experimental production test,productive production test,and commercial production.The second production test serves as an important step from the exploratory production test to experimental production test.Jian-liang Ye Xu-wen Qin Wen-wei Xie Hai-long Lu Bao-jin Ma Hai-jun Qiu Jin-qiang Liang Jing-an Lu Zeng-gui Kuang Cheng Lu Qian-yong Liang Shi-peng Wei Yan-jiang Yu Chunsheng Liu Bin Li Kai-xiang Shen Hao-xian Shi Qiu-ping Lu Jing Li Bei-bei Kou Gang Song Bo Li He-en Zhang Hong-feng Lu Chao Ma Yi-fei Dong Hang Bian 2020China Geology2020,3,2:22

3Chronic intermittent hypoxia accelerates liver fibrosis in rats with combined hypoxia and nonalcoholic steatohepatitis via angiogenesis rather than endoplasmic reticulum stress显示文摘In the present study,we aimed to investigate the role of endoplasmic reticulum stress(ERS)and its related inflammation and angiogenesis in liver fibrosis in a rat model of combined hypoxia and nonalcoholic steatohepatitis(NASH)and to confirm whether the intervention of hypoxia-inducible factor 1α(HIF1α)can improve fibrosis.Liver histological changes and biochemical indices,HIF1α,inflammatory factors,ERS-related parameters(GRP78,CHOP,caspase-3,and caspase-12),and angiogenesis indices(VEGFA,VEGFR2,and CD34)were evaluated.Compared with the control rats,the liver tissue of rats with hypoxia and NASH had obvious NASH characteristics and hepatic fibrosis was significantly aggravated,including bridging fibrosis in some rats.The mRNA expression levels of HIF1α,VEGFA,and VEGFR2 and total immunohistochemical staining scores of VEGFR2 and CD34 were significantly increased.In addition,HIF1α silencing significantly decreased HIF1α,biochemical indices(ALT,AST,and TG),inflammatory factors(TNFα,IL6,and IL1β),and angiogenesis indices(CD34 and VEGFR2),consequently,improved the hepatic fibrosis score in the rat model of combined hypoxia and NASH.Taken together,chronic intermittent hypoxia accelerates liver fibrosis in rats with combined hypoxia and NASH via angiogenesis rather than ERS and HIF1α intervention can improve liver fibrosis,angiogenesis,inflammatory factors,and biochemical indices.Therefore,HIF1α is a key regulatory factor of liver fibrosis in rats with combined hypoxia and NASH.Wei Wu Weiping Li Jiaojiao Wei Chunsheng Wang Yunliang Yao Weihua Zhu Weimei He Weimei Zhou Jiang Liu 2019Acta Biochimica et Biophysica Sinica2019,51,2:8
4Impending HRT wave precursors to the Wenchuan M_s8.0 earthquake and methods of earthquake impending prediction by using HRT wave显示文摘We deployed four geo-electric monitoring stations in Sichuan and Yunnan provinces from 2004, using the new generation of equipment (PS-100) and technologies to capture the HRT wave earthquake precursor. Before the Wenchuan Ms8.0 earthquake, we recorded the HRT wave precursor at the only operating station in Hongge (HG, Δ=465 km) and found that significant impending signal had been recorded at the station in the early morning ( 0―5 am) of 12th of May, 2008. The precursor for this earthquake is consistent with precursors recorded for other strong earthquakes. The measured physical properties (geo-resistivity and telluric-current) show tidal wave period oscillations from several days to several months before the earthquakes and the amplitude of such HT oscillation increases significantly towards the occurrence of an earthquake. These HT and RT waves from the epicenter have a causal relationship with the earthquakes that happened several days later. The arrival time of two RT waves is proportional to the distance from the station to the epicenter. The estimated natural decay of the amplitude is correlated with the natural period (T0) of the earthquake fault, which is proportional to the fault length. From this relationship, we can predict the earthquake magnitude. For magnitude 6―9 earthquakes, the natural period is about 1―6 hours. Such oscillation comes from the epicenter area and they can propagate several thousand kilometers in the Earth's crust. Before a strong earthquake in the shallow crust, the conductive pore fluid will experience major changes before the fault rapture. Such fluid change will emit an oscillation in the pore fluid pressure. This is the mechanism for the HRT wave generation. Since the China Earthquake Administration funded the HRT wave short-term earth-quake prediction project in 2003, the first record of HRT precursor wave has been recorded from the 2004-12-26 Sumatra Mw9.0 earthquake with the largest epicentre distance Δ=2900 km. Thereafter, we have captured HRT waves from more than twenty strong earthquakes, which are well-matched and show repeatability, consistency and regularity. All our observation with the HRT waves demonstrate that HRT wave precursors to earthquakes indeed exist. Strong earthquakes can be predicted and short-term and impending earthquake prediction is achievable in the very near future. 5Oxygen isotope evidence for two-stage water-rock interactions of the Nianzishan A-type granite in NE China显示文摘The oxygen isotope ratios of whole-rock, common rock-forming minerals and zircon from Mesozoic A-type granitic pluton at Nianzishan in northeastern China were analyzed by the conventional BrF5 method and the laser-probe technique, respectively. Both whole-rock and rock-forming minerals show large δ18O variations up to 5.5‰ with significant oxygen isotope disequilibrium between zircon and the other minerals, whereas the δ18O values of zircon are tightly clustered between 3.12‰ and 4.19‰ and thus lower than the normal-mantle δ18O values. These results indicate that the Nianzishan A-type granite experienced two-stage water-rock interactions subsequentially. The remarkably low zircon δ18O values are genetically due to sea-water exchange with granite protolith in the first stage, and the oxygen isotope disequilibrium fractionations between zircon and rock-forming minerals are caused by mete-oric-hydrothermal alteration in the second stage. 8The Escherichia coli O157:H7 DNA detection on a gold nanoparticle-enhanced piezoelectric biosensor显示文摘This paper presents development of a quartz crystal microbalance (QCM) biosensor for real-time de- tection of E. coli O157:H7 DNA based on nanogold particles amplification. Many inner Au nanoparticles were immobilized onto the thioled surface of the Au electrode, then more specific thiolated sin- gle-stranded DNA (ssDNA) probes could be fixed through Au-SH bonding. The hybridization was in- duced by exposing the ssDNA probe to the complementary target DNA of E. coli O157:H7 gene eaeA, then resulted in a mass change and corresponding frequency shifts ( △f ) of the QCM. The outer avidin-coated Au nanoparticles could combine with the target DNA to increase the mass. The electro- chemical techniques, cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) were adopted to manifest and character each step. The target DNA corresponding to 2.0×103 colony forming unit (CFU)/mL E. coli O157:H7 cells can be detected by this biosensor, so it is practical to develop a sensitive and effective QCM biosensor for pathogenic bacteria detection based on specific DNA analy- sis. 9A sulfur dioxide polymer prodrug showing combined effect with doxorubicin in combating subcutaneous and metastatic melanoma显示文摘Melanoma,as the most aggressive and treatment-resistant skin malignancy,is responsible for about 80%of all skin cancer mortalities.Prone to invade into the dermis and form distant metastases significantly reduce the patient survival rate.Therefore,early treatment of the melanoma in situ or timely blocking the deterioration of metastases is critical.In this study,a sulfur dioxide(SO_(2))polymer prodrug was designed as both an intracellular glutathione(GSH)-responsive SO_(2) generator and a carrier of doxorubicin(DOX),and used for the treatment of subcutaneous and metastatic melanoma.Firstly,chemical conjugation of 4-N-(2,4-dinitrobenzenesulfonyl)-imino-1-butyric acid(DIBA)onto the side chains of methoxy poly(ethylene glycol)grafted dextran(mPEG-g-Dex)resulted in the synthesis of the amphiphilic polymer prodrug of SO_(2),mPEG-g-Dex(DIBA).The obtained mPEG-g-Dex(DIBA)could self-assemble into stable micellar nanoparticles and exhibited a glutathione-responsive SO_(2) release behavior.Subsequently,DOX was encapsulated into the core of mPEG-g-Dex(DIBA)micelles to form DOX-loaded nanoparticles(PDDN-DOX).The formed PDDN-DOX could be internalized by B16F10 cells and synchronously release DOX and SO_(2) into the tumor cells.As a result,PDDN-DOX exerted synergistic anti-tumor effects in B16F10 melanoma cells because of the oxidative damage properties of SO_(2) and toxic effects of DOX.Furthermore,in vivo experiments verified that PDDN-DOX had great potential for the treatment of subcutaneous and metastasis melanoma.Collectively,our present work demonstrates that the combination of SO_(2)-based gas therapy and chemotherapeutics offers a new avenue for inhibiting melanoma progression and metastases.Lin An Peng Zhang Wei Shen Xuan Yi Weitian Yin Rihua Jiang Chunsheng Xiao 2021Bioactive Materials2021,6,5:3
10Carrier-free highly drug-loaded biomimetic nanosuspensions encapsulated by cancer cell membrane based on homology and active targeting for the treatment of glioma显示文摘Nanosuspensions,as a new drug delivery system for insoluble drugs,are only composed of a drug and a small amount of stabilizer,which is dispersed in an aqueous solution with high drug-loading,small particle size,high dispersion,and large specific surface area.It can significantly improve the dissolution,bioavailability,and efficacy of insoluble drugs.In this study,paclitaxel nanosuspensions((PTX)NS)were prepared by an ultrasonic precipitation method,with the characteristics of simple preparation and easy repetition.With the help of a homologous targeting mechanism,a kind of glioma C6 cancer cell membrane(CCM)-coated(PTX)NS was developed and modified with DWSW peptide to obtain DWSW-CCM-(PTX)NS with the functions of BBB penetration and tumor targeting.The results showed that the cancer cell membrane could effectively camouflage the nanosuspensions so that it was not cleared by the immune system and could cross the blood-brain-barrier(BBB)and selectively target tumor tissues.Cell uptake experiments and in vivo imaging confirmed that the uptake of DWSW-CCM-(PTX)NS by tumor cells and the distribution in intracranial gliomas increased.Cytotoxicity test and in vivo anti-glioma studies showed that DWSW-CCM-(PTX)NS could significantly inhibit the growth of glioma cells and significantly prolong the survival time of glioma-bearing mice.Finally,the cancer cell membrane coating endowed the nanosuspensions with the biological properties of homologous adhesion and immune escape.This study provides an integrated solution for improving the targeting of nanosuspensions and demonstrates the encouraging potential of biomimetic nanosuspensions applicable to tumor therapy.Yueyue Fan Yuexin Cui Wenyan Hao Mengyu Chen Qianqian Liu Yuli Wang Meiyan Yang Zhiping Li Wei Gong Shiyong Song Yang Yang Chunsheng Gao 2021Bioactive Materials2021,6,12:3

13Effect of Sodium Chloride and Cadmium on the Growth, Oxidative Stress and Antioxidant Enzyme Activities of Zygosaccharomyces rouxii显示文摘Zygosaccharomyces rouxii is a salt-tolerant yeast species capable of removing cadmium(Cd) pollutant from aqueous solution. Presently, the physiological characteristics of Z. rouxii under the stress of sodium chloride(NaCl) and Cd are poorly understood. This study investigated the effects of NaCl and Cd on the growth, oxidative stress and antioxidant enzyme activities of Z. rouxii after stress treatment for 24 h. Results showed that NaCl or Cd alone negatively affected the growth of Z. rouxii, but the growth-inhibiting effect of Cd on Z. rouxii was reduced in the presence of NaCl. Flow cytometry assay showed that under Cd stress, NaCl significantly reduced the production of reactive oxygen species(ROS) and cell death of Z. rouxii compared with those in the absence of NaCl. The activities of superoxide dismutase(SOD), catalase(CAT), and peroxidase(POD) of Z. rouxii were significantly enhanced by 2%–6% NaCl, which likely contributed to the high salt tolerance of Z. rouxii. The POD activity was inhibited by 20 mg L-1Cd while the SOD and CAT activities were enhanced by 8 mg L-1 Cd and inhibited by 20 mg L-1 or 50 mg L-1 Cd. The inhibitory effect of high-level Cd on the antioxidant enzyme activities of Z. rouxii was counteracted by the combined use of NaCl, especially at 6%. This probably accounted for the decrease in Cd-induced ROS production and cell death of Z. rouxii after incubation with NaCl and Cd. 19A light-addressable microfluidic device for label-free functional assays of bioengineered taste receptor cells via extracellular recording显示文摘The functional assay of chemical sensitive cells is of great importance for the preparation of sensitive elements towards the development of cell-based biosensors for chemical sensing. In this study, a novel light-addressable microfluidic device was developed by the integration of a light-addressable potentiometric sensor (LAPS) with a microfluidic chip for label-free functional assays of bioengineered taste receptor cells via extracellular recording. Extracellular potential changes of single bioengineered cells were recorded by LAPS. Microfluidic chip was capable of providing stable microenvironments for cell measurements w让h a well-defined concentration stimulus. Bioengineered taste receptor cells were utilized as a model of chemical sensitive cells and functional assayed by this light-addressable microfluidic device using bitter stimulation. The results indicate that this microfluidic device can efficiently monitor the membrane potential changes originated from bioengineered taste receptor cells in response to specific bitter stimulation. The bioengineered cells with responsive functions can be easily identified via the analysis on the firing rates of extracellular recording data. The expression of bitter receptors in the bioengineered taste receptor cells was further validated by the immunofluorescent staining results, which proved that the expression of specific bitter receptor was the main contribution to the responsive function of bioengineered cells.
