The second natural gas hydrate production test in the South China Sea显示文摘Clayey silt reservoirs bearing natural gas hydrates(NGH)are considered to be the hydrate-bearing reservoirs that boast the highest reserves but tend to be the most difficult to exploit. They are proved to be exploitable by the first NGH production test conducted in the South China Sea in 2017.Based on the understanding of the first production test, the China Geological Survey determined the optimal target NGH reservoirs for production test and conducted a detailed assessment, numerical and experimental simulation, and onshore testing of the reservoirs. After that,it conducted the second offshore NGH production test in 1225 m deep Shenhu Area,South China Sea(also referred to as the second production test)from October 2019 to April 2020.During the second production test,a series of technical challenges of drilling horizontal wells in shallow soft strata in deep sea were met, including wellhead stability, directional drilling of a horizontal well, reservoir stimulation and sand control, and accurate depressurization. As a result,30 days of continuous gas production was achieved, with a cumulative gas production of 86.14×104 m3.Thus,the average daily gas production is 2.87×10^4 m^3,which is 5.57 times as much as that obtained in the first production test. Therefore, both the cumulative gas production and the daily gas production were highly improved compared to the first production test.As indicated by the monitoring results of the second production test, there was no anomaly in methane content in the seafloor, seawater, and atmosphere throughout the whole production test. This successful production test further indicates that safe and effective NGH exploitation is feasible in clayey silt NGH reservoirs. The industrialization of hydrates consists of five stages in general, namely theoretical research and simulation experiments, exploratory production test, experimental production test, productive production test, and commercial production. The second production test serves as an important step from the exploratory production test to experimental production test. | Jian-liang Ye Xu-wen Qin Wen-wei Xie Hai-long Lu Bao-jin Ma Hai-jun Qiu Jin-qiang Liang Jing-an Lu Zeng-gui Kuang Cheng Lu Qian-yong Liang Shi-peng Wei Yan-jiang Yu Chunsheng Liu Bin Li Kai-xiang Shen Hao-xian Shi Qiu-ping Lu Jing Li Bei-bei Kou Gang Song Bo Li He-en Zhang Hong-feng Lu Chao Ma Yi-fei Dong Hang Bian | 2020 | China Geology2020,3,2: | 22

Selective modification of two-dimensional MoS_2 nanosheets by polymer grafting显示文摘As an emerging two-dimensional (2D) nanomaterial, single layer ora few layers of molybdenum disulfide (MoS_2) has drawn significant attention in the past decade. In the present work, we report a strategy in direct creating polymer brushes on MoS_2 surfaces via S—C bond in the presence of UV light. The modification of MoS_2 nanosheets can be achieved bilaterally or homolaterally by performing the UVgrafting polymerization on exfoliated MoS_2 nanosheets or a single layer of MoS_2 deposited on a silicon substrate. A series of vinyl monomers including methyl methacrylate (MMA), styrene (St) and 2-isopropenyl-2-oxazoline (IPOx) could be applied to this approach, leading to the formation of poly (methyl methacrylate) (PMMA), polystyrene (PS) and poly (2-isopropenyl-2-oxazoline) (PIPOx) brushes.AFM, IR, and XPS characterizations indicate the successful formation of homogeneous brush layers on MoS_2 surfaces. The polymer brushes modified MoS_2 may found potential applications in photo dynamic therapy and sensing technologies. | Hang Bian Xuejian Zhang Dexin Huang Ning Zhang | 2019 | Chinese Chemical Letters2019,30,2: | 1
Polymeric nanoparticles of poly(2-oxazoline),tannic acid and doxorubicin for controlled release and cancer treatment显示文摘A straightforward coassembly strategy was developed for the preparation of polymeric nanoparticles driving by the intermolecular hydrogen bond between neutral poly(2-methyl-2-oxaozline)(PMeOx),tannic acid(TA) and doxorubicin hydrochloride(Dox). The occurrence of the hydrogen-bonding amongst the different functionalities within the formed nanoparticles was verified by infrared(IR) spectroscopy.Scanning electron microscopy(SEM),dynamic light scattering(DLS),UV-vis absorption and photoluminescent measurements indicated the rapid formation of uniform and water dispersible/stable nanoparticles. The relative poor stability of PMeOx-TA-Dox in fetal bovine serum(FBS) solution enabled the rapid separation of Dox and PMeOx-TA,facilitating the release of Dox and its entrance into cellular nuclei as revealed by confocal laser scanning microscopy(CLSM).The presented strategy may provide an efficient alternative for the construction of multifunctional nanomedicines. | Lin Liu Liying Yin Hang Bian Ning Zhang | 2020 | Chinese Chemical Letters2020,31,2: | 1
Synthesis, supramolecular structures and luminescent properties of quinacridone derivatives bearing carbazole groups显示文摘The syntheses of three carbazyl-containing quinacridone derivatives, N,N’-di((N-carbazyl)-n-butyl) quinacridone (DCBQA), N,N’-di((N-carbazyl)-n-hexyl)quinacridone (DCHQA) and N,N’-di((N-carbazyl)-n-octyl)quinacridone (DCOQA), are reported, and the photoluminescent (PL) characteristics are pre- sented. The single crystal X-ray structures of DCBQA, DCHQA and DCOQA are investigated. The crystal of DCBQA is characterized by intermolecular π…π interactions between quinacridone cores and carbazole moieties resulting in the formation of DCBQA molecule layer, in which every quinacridone core is surrounded by four carbazole groups. In DCHQA crystal, molecules assemble into two kinds of oriented columns based on intermolecular π…π interactions between quinacridone cores. The DCOQA crystal displays intermolecular CH…π and hydrogen bonding interactions feature. In DCOQA solid, every quinacridone core is sandwiched by two alkyl chains from two adjacent DCOQA molecules and simultaneously linked together with two other quinacridone cores by hydrogen bonding interactions. The PL spectra of the three compounds in solution exhibit concentration-dependent properties and their PL quantum causes decrease with the increasing concentration. | ZHAO GuoLi FAN Yan HUO Cheng BIAN Hang SONG WeiFeng ZHANG JingYing WANG Yue | 2009 | Chinese Science Bulletin2009,54,10: | 0
Certified defense against patch attacks via mask-guided randomized smoothing显示文摘The adversarial patch is a practical and effective method that modifies a small region on an image, making DNNs fail to classify. Existing empirical defenses against adversarial patch attacks lack theoretical analysis and are vulnerable to adaptive attacks. To overcome such shortcomings, certified defenses that provide a guaranteed classification performance in the face of strong unknown adversarial attacks are proposed. However, on the one hand, existing certified defenses either have low clean accuracy or need specified architecture, which is not robust enough. On the other hand, they can only provide provable accuracy but ignore the relationship to the number of perturbations. In this paper, we propose a certified defense against patch attacks that provides both the provable radius and high classification accuracy. By adding Gaussian noises only on the patch region with a mask, we prove that a stronger certificate with high confidence can be achieved by randomized smoothing. Furthermore, we design a practical scheme based on joint voting to find the patch with a high probability and certify it effectively. Our defense achieves 86.4%clean accuracy and 71.8% certified accuracy on CIFAR-10 exceeding the maximum 60% certified accuracy of existing methods. The clean accuracy of 67.8% and the certified accuracy of 53.6% on ImageNet are better than the state-of-the-art method, whose certified accuracy is 26%. | Kui ZHANG Hang ZHOU Huanyu BIAN Weiming ZHANG Nenghai YU | 2022 | Science China(Information Sciences)2022,65,7: | 0
Controlled synthesis of silver nanoparticles from polyoxometalates-immobilized poly(4-vinylpyridine) brushes显示文摘This contribution reports the immobilization of polyoxometalate(POM)into poly(4-vinyl pyridine)(P4 VP)brushes and the controlled reduction of silver ions,in-situ generating metal nanoparticles in the brushes.P4 VP brushes were straightforwardly created by UV-assisted photopolymerization of 4-vinyl pyridine(4VP)on silicon or glass substrates.Phosphotungstic acid(H3 PW12 O40),one of the most widely used Keggin-type POM was anchored onto these pyridine moieties through electrostatic interaction, leading to the P4 VP/POM hybrid brushes. The immobilized POM was further reduced to heteropolyblue, which could be used to generate silver nanoparticles in a controlled fashion.AFM,UV-vis and IR characterization indicate that P4 VP brushes not only provide an efficient platform in the controlled preparation of Ag nanoparticles, but also efficiently disperse and stabilize POM,thus preventing the aggregation of the generated Ag nanoparticles. | Hang Bian Xuejian Zhang Hongkai Zhao Ning Zhang | 2019 | Chinese Chemical Letters2019,30,5: | 0
Inhibitory effects of Pseudomonas aeruginosa mannose-sensitive hemagglutinin on rat C6 glioma cell proliferation显示文摘BACKGROUND: Pseudomonas aeruginosa mannose-sensitive hemagglutinin (PA-MSHA) parenteral injection is used as a broad-spectrum immunomodulator. It remains unclear whether PA-MSHA exhibits inhibitory effects on tumor cell growth. OBJECTIVE: To investigate inhibitory mechanisms of PA-MSHA-induced proliferation in rat C6 glioma cells in vitro. DESIGN, TIME AND SETTING: Comparative observation and in vitro experiments were performed at the Key Laboratory of Natural Medicine, Kunming Medical College, China from July 2008 to April 2009. MATERIALS: Rat C6 glioma cell line (Shanghai Institute of Cell Biology, Chinese Academy of Sciences, China) and PA-MSHA parenteral injection (Beijing Wanteer Bio-Pharmaceutical, China) were used in the present study. METHODS: Rat C6 glioma cells in logarithmic growth phase were harvested in vitro. Adherent monolayer cells were respectively treated with PA-MSHA at final colony-forming units (cfu) of 1 × 108 cfu/mL, 2 × 108 cfu/mL, 4 × 108 cfu/mL, 6 × 108 cfu/mL, and 8 × 108 cfu/mL following 24 hours of conventional culture. MAIN OUTCOME MEASURES: MTT colorimetric assay was utilized to determine the inhibitory rate of C6 glioma cells following treatment with various concentrations of PA-MSHA at different times. Cell apoptosis was detected by fluorescent microscopy following Hoechst 33258 staining. Flow cytometry was used to measure PA-MSHA effects on C6 cell cycle. RESULTS: Inhibitory rate of C6 glioma cells increased with prolonged time and increased dose. Hoechst 33258 staining revealed obvious morphological changes in apoptotic C6 glioma cells. Flow cytometry revealed hypodiploid peaks, i.e., apoptotic peak, and the apoptotic rate in cells during S-phase significantly increased with increased concentrations in the experimental groups. CONCLUSION: With in vitro experiments, PA-MSHA preparations inhibited C6 glioma cell proliferation in a time- and dose-dependent manner. These mechanisms are likely associated with cell apoptosis induction and inhibition of the S phase. | Jie Sun Jianchang Cen Qian Chang Ping Su Zhiyong Yang Jinkun Wang Peng Ding Hang Yin Zhiqiang Shen Peng Chen Dianhua Wang Ligong Bian Xiaobin Song Jun Liu | 2010 | Neural Regeneration Research2010,5,11: | 0
A microenvironment sensitive pillar[5]arene-based fluorescent probe for cell imaging and drug delivery显示文摘Dansylamide(DNSA) is a typical ICT probe that has a favorable serum albumin sensitivity. Inspired by this, we designed a microenvironment sensitive fluorescent probe 4 C-G through introducing DNSA into pillar[5]arene. Unlike DNSA, 4 C-G displayed differentiated sensitivity to multiple proteins, which was benefit from pillar[5]arene assisted the probe to form complexes with proteins. 4 C-G could not only be applied in imaging of Hep G2, but also act as a favorable drug carrier for regorafenib(REG) encapsulation. The 4 C-G-REG complex would aggregate into high drug-loading fluorescent nanoparticles in a physiological environment(pH 7.4). Such nanoparticles exhibited p H-triggered enrichment ability, which rapidly enriched REG in the acidic environment(pH 6.0). Furthermore, the complexation between 4 C-G and REG maintained the imaging property of the probe and the excellent anticancer activity of the drug on Hep G2. | Qing Wang Jingwen Fan Xiaoyan Bian Hang Yao Xiaohui Yuan Ying Han Chaoguo Yan | 2022 | Chinese Chemical Letters2022,33,4: | 0
Polymer brushes on hydrogen-terminated silicon substrates via stable Si--C bond显示文摘We demonstrate a straightforward and efficient method for the creation of polymer brushes on hydrogen-terminated silicon substrates through the UV-induced photopolymerization. The surface grafting polymerization is applicable to a series of monomers, allowing the direct formation of homogeneous polymer coatings ranging from hydrophilic poly(2-isopropenyl-2-oxazoline)(PIPOx),amphiphilic poly(N-isopropyl acrylamide)(PNIPAM), to hydrophobic polystyrene(PS) and poly(4-(1H,1H,2H,2H-perfluorohexyl)oxymethylstyrene)(PPHMS) on Si(100) and Si(111) surfaces via stable Si—C bonds. Polymerization kinetic investigation indicates a linear increase of polymer layer thickness with the polymerization time. Moreover, the as-prepared polymer brushes exhibit superior stability against basic conditions in contrast to those that were formed on silicon substrates via conventional Si—O—C bond. | Hang Bian Xiaozhe Dong Shanshan Chen Dewen Dong Ning Zhang | 2018 | Chinese Chemical Letters2018,29,1: | 0
Long-term passaging of pseudo-typed SARS-CoV-2 reveals the breadth of monoclonal and bispecific antibody cocktails显示文摘The continuous emergence of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)variants poses challenges to the effectiveness of neutralizing antibodies.Rational design of antibody cocktails is a realizable approach addressing viral immune evasion. However, evaluating the breadth of antibody cocktails is essential for understanding the development potential. Here, based on a replication competent vesicular stomatitis virus model that incorporates the spike of SARS-CoV-2(VSV-SARS-CoV-2),we evaluated the breadth of a number of antibody cocktails consisting of monoclonal antibodies and bispecific antibodies by long-term passaging the virus in the presence of the cocktails. Results from over two-month passaging of the virus showed that 9E12+10D4+2G1 and 7B9-9D11+2G1 from these cocktails were highly resistant to random mutation, and there was no breakthrough after 30 rounds of passaging. As a control, antibody REGN10933 was broken through in the third passage.Next generation sequencing was performed and several critical mutations related to viral evasion were identified. These mutations caused a decrease in neutralization efficiency, but the reduced replication rate and ACE2 susceptibility of the mutant virus suggested that they might not have the potential to become epidemic strains. The 9E12+10D4+2G1 and 7B9-9D11+2G1 cocktails that picked from the VSV-SARS-CoV-2 system efficiently neutralized all current variants of concern and variants of interest including the most recent variants Delta and Omicron,as well as SARS-CoV-1.Our results highlight the feasibility of using the VSV-SARS-CoV-2 system to develop SARS-CoV-2 antibody cocktails and provide a reference for the clinical selection of therapeutic strategies to address the mutational escape of SARS-CoV-2. | Hang Ma Hui-fang Zong Jun-jun Liu Ya-li Yue Yong Ke Yun-ji Liao Hao-neng Tang Lei Wang Shu-sheng Wang Yun-sheng Yuan Ming-yuan Wu Yan-lin Bian Bao-hong Zhang Hai-yang Yin Hua Jiang Tao Sun Lei Han Yue-qing Xie Jian-wei Zhu | 2023 | Acta Pharmacologica Sinica2023,44,7: | 0
The influence of heterogeneity on the analysis of sleep-wake architecture in the single-prolonged stress rats显示文摘Objective: To observe the influence of heterogeneity on sleep-wake architecture in single-prolonged stress(SPS) animal model. Methods: SPS rats were subdivided into low responders(LR) and high responders(HR) based on their freezing responses to a novel environment. Sleeping time(ST), awakening numbers(AN), brief awakening numbers(b AN) and frequency distribution of sleep bouts were used as observing indicators, single factor variance analysis combined with Dunnett t test were used to compare the differences between control, exposure, LR and HR groups. Results: We found sleeping time was increased only in HR group. Moreover, awakening numbers and brief awakening number increased in exposure group and HR group during the light phase, but not in LR group. The number of sleep bouts for the ranges of 40-80 s increased obviously in HR group, but not in exposure and LR group. In addition, there were significant correlation between sleep-related parameters and freezing in HR group, but not in LR group. Conclusion: Heterogeneity existed in SPS model in view of different sleep-wake architectures of SPS rats. Rats in HR group exactly mimicked the freezing response and sleep disorders of PTSD. So HR rats were more appropriate to be used as PTSD-like models, especially when studying sleep disorder in PTSD. | Ying WANG Hong-sheng BIAN Hang ZHAO Ting-li LI | 2016 | 中国应用生理学杂志2016,32,6: | 0
Global profiling of O-GlcNAcylated and/or phosphorylated proteins in hepatoblastoma显示文摘O-linked-β-N-acetylglucosamine(O-GlcNAc)glycosylation(O-GlcNAcylation)and phosphorylation are critical posttranslational modifications that are involved in regulating the functions of proteins involved in tumorigenesis and the development of various solid tumors. However,a detailed characterization of the patterns of these modifications at the peptide or protein level in hepatoblastoma(HB),a highly malignant primary hepatic tumor with an extremely low incidence in children, has not been performed. Here,we examined O-GlcNAc-modified or phospho-modified peptides and proteins in HB through quantitative proteomic analysis of HB tissues and paired normal liver tissues. Our results identified 114 O-GlcNAcylated peptides belonging to 78 proteins and 3494 phosphorylated peptides in 2088 proteins.Interestingly,41 proteins were modified by both O-GlcNAcylation and phosphorylation. These proteins are involved in multiple molecular and cellular processes, including chromatin remodeling, transcription, translation, transportation, and organelle organization. In addition,we verified the accuracy of the proteomics results and found a competitive inhibitory effect between O-GlcNAcylation and phosphorylation of HSPB1.Further,O-GlcNAcylation modification of HSPB1 promoted proliferation and enhanced the chemotherapeutic resistance of HB cell lines in vitro.Collectively, our research suggests that O-GlcNAc-modified and/or phospho-modified proteins may play a crucial role in the pathogenesis of HB. | Hang Song Ji Ma Zhixuan Bian Shuhua Chen Jiabei Zhu Jing Wang Nan Huang Minzhi Yin Fenyong Sun Min Xu Qiuhui Pan | 2019 | Signal Transduction and Targeted Therapy2019,4,1: | 0
