| 3 | A high-strength mineralized collagen bone scaffold for large-sized cranial bone defect repair in sheep显示文摘Large-sized cranial bone defect repair presents a great challenge in the clinic.The ideal cranioplasty materials to realize the functional and cosmetic recovery of the defect must have sufficient mechanical support,excellent biocompatibility,good osseointegration and biodegradability as well.In this study,a high-strength mineralized collagen(MC)bone scaffold was developed with biomimetic composition,microstructure and mechanical properties for the repair of sheep largesized cranial bone defects in comparison with two traditional cranioplasty materials,polymethyl methacrylate and titanium mesh.The compact MC scaffold showed no distinct pore structure and therefore possessed good mechanical properties.The strength and elastic modulus of the scaffold were much higher than those of natural cancellous bone and slightly lower than those of natural compact bone.In vitro cytocompatibility evaluation revealed that the human bone marrow mesenchymal stem cells(hBMSC)had good viability,attachment and proliferation on the compact MC scaffold indicating its excellent biocompatibility.An adult sheep cranial bone defect model was constructed to evaluate the performances of these cranioplasty materials in repairing the cranial bone defects.The results were investigated by gross observation,computed tomography scanning as well as histological assessments.The in vivo evaluations indicated that compact MC scaffold showed notable osteoconductivity and osseointegration with surrounding cranial bone tissues by promoting bone regeneration.Our results suggested that the compact MC scaffold has a promising potential for large-sized cranial bone defect repair. | Shuo Wang Zhijun Zhao Yongdong Yang Antonios G.Mikos Zhiye Qiu Tianxi Song Fuzhai Cui XiumeiWang Chunyang Zhang | 2018 | Regenerative Biomaterials2018,5,5: | 9 |
| 4 | Potassium 2-(l-hydroxypentyl)-benzoate attenuates neuroinflammatory responses and upregulates heme oxygenase-1 in systemic lipopolysaccharide-induced inflammation in mice显示文摘A neuroinflammatory response is commonly involved in the progression of many neurodegenerative diseases. Potassium 2-(1-hydroxypentyl)-benzoate(PHPB), a novel neuroprotective compound, has shown promising effects in the treatment of ischemic stroke and Alzheimer's disease(AD). In the present study, the anti-inflammatory effects of PHPB were investigated in the plasma and brain of C57BL/6 mice administered a single intraperitoneal(i.p.) injection of lipopolysaccharide(LPS). Levels of i NOS and the cytokines TNFα, IL-1β and IL-10 were elevated in plasma, cerebral cortex and hippocampus after LPS injection and the number of microglia and astrocytes in cortex and hippocampus were increased. LPS also upregulated the expression of heme oxygenase-1(HO-1) in the cortex and hippocampus. PHPB reduced the levels of i NOS and cytokines in the plasma and brain, decreased the number of microglia and astrocytes and further enhanced the upregulation of HO-1. In addition, PHPB inhibited the LPS-induced phosphorylation of ERK, P38 and JNK. | 5 | Causality Diagram-based Scheduling Approach for Blast Furnace Gas System显示文摘Rational use of blast furnace gas(BFG) in steel industry can raise economic profit, save fossil energy resources and alleviate the environment pollution. In this paper, a causality diagram is established to describe the causal relationships among the decision objective and the variables of the scheduling process for the industrial system, based on which the total scheduling amount of the BFG system can be computed by using a causal fuzzy C-means(CFCM) clustering algorithm. In this algorithm,not only the distances among the historical samples but also the effects of different solutions on the gas tank level are considered.The scheduling solution can be determined based on the proposed causal probability of the causality diagram calculated by the total amount and the conditions of the adjustable units. The causal probability quantifies the impact of different allocation schemes of the total scheduling amount on the BFG system. An evaluation method is then proposed to evaluate the effectiveness of the scheduling solutions. The route combined high-temperature solid phase reaction with chemical vapor deposition (CVD) using Fe2O3 and LiH2PO4 as the precursors for LiFePO4 growth and acetylene (C2H2) as the carbon source for CNFs growth. The composites were characterized by X-ray diffraction (XRD), Brunauer-Emmett-Teller (BET) specific surface area, field emission scanning electron microscopy (FE-SEM), and transmission electron microscopy (TEM). The electrochemical performance of the composites was studied by galvanostatic cycling and cyclic voltammetry (CV). The results showed that the in-situ CNFs growth could be realized by the catalytic effect of the Fe2O3 precursor. | 10 | Hepatitis B virus evades immune recognition via RNA adenosine deaminase ADAR1-mediated viral RNA editing in hepatocytes显示文摘HBV is considered as a“stealth”virus that does not invoke interferon(IFN)responses;however,the mechanisms by which HBV bypasses innate immune recognition are poorly understood.In this study,we identified adenosine deaminases acting on RNA 1(ADAR1),which is a key factor in HBV evasion from IFN responses in hepatocytes.Mechanically,ADAR1 interacted with HBV RNAs and deaminated adenosine(A)to generate inosine(I),which disrupted host immune recognition and thus promoted HBV replication.Loss of ADAR1 or its deficient deaminase activity promoted IFN responses and inhibited HBV replication in hepatocytes,and blocking the IFN signaling pathways released the inhibition of HBV replication caused by ADAR1 deficiency.Notably,the HBV X protein(HBx)transcriptionally promoted ADAR1 expression to increase the threshold required to trigger intrinsic immune activation,which in turn enhanced HBV escape from immune recognition,leading to persistent infection.Supplementation with 8-azaadenosine,an ADAR1 inhibitor,efficiently enhanced liver immune activation to promote HBV clearance in vivo and in vitro.Taken together,our results delineate a molecular mechanism by which HBx promotes ADAR1-derived HBV immune escape and suggest a targeted therapeutic intervention for HBV infection. | Liyuan Wang Yang Sun Xiaojia Song Zehua Wang Yankun Zhang Ying Zhao Xueqi Peng Xiaodong Zhang Chunyang Li Chengjiang Gao Nailin Li Lifen Gao Xiaohong Liang Zhuanchang Wu Chunhong Ma | 2021 | Cellular & Molecular Immunology2021,18,8: | 2 |

The materials were characterized by X-ray diffraction (XRD), field emission scanning electron microscopy (FESEM) and transmission electron microscopy (TEM). The electrochemical properties of the materials were investigated by galvanostatic cycling, cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). It was found that the materials consisted of nanorods with a diameter of 50 nm and a length of 500 nm. Galvanostatic cycling showed that the capacity of LiMnPO 4 could be largely increased by Fe2+ substitution. At a current rate of C/20, the capacity of the three samples (x=0,0.2,0.4) were 47, 107 and 150 mA·h·g-1 , respectively. CV result showed that the Fe2+ substitution could decrease the polarization during charging/discharging, accelerating the electrochemical process. | 14 | Glycosylation and an amino acid insertion in the head of hemagglutinin independently affect the antigenic properties of H5N1 avian influenza viruses显示文摘Antigenic drift forces us to frequently update influenza vaccines; however, the genetic basis for antigenic variation remains largely unknown. In this study, we used clade 7.2 H5 viruses as models to explore the molecular determinants of influenza virus antigenic variation. We generated eight monoclonal antibodies(MAbs) targeted to the hemagglutinin(HA) protein of the index virus A/chicken/Shanxi/2/2006 and found that two representative antigenically drifted clade 7.2 viruses did not react with six of the eight MAbs. The E131 N mutation and insertion of leucine at position 134 in the HA protein of the antigenically drifted strains eliminated the reactivity of the virus with the MAbs. We also found that the amino acid N131 in the H5 HA protein is glycosylated. | 17 | Safety and immunogenicity of a modified COVID-19 mRNA vaccine,SYS6006,as a fourth-dose booster following three doses of inactivated vaccines in healthy adults:an open-labeled Phase 1 trial显示文摘The continuous emergence of the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)variants led to a rapid decline in protection efficacy and neutralizing titers even after three doses of COVID-19 vaccines.Here,we report an open-labeled Phase I clinical trial of a modified mRNA vaccine(SYS6006)as a fourth-dose booster in healthy adults.Eighteen eligible participants,who had completed three doses of inactivated COVID-19 vaccines,received a fourth boosting dose of SYS6006-20μg.Eighteen convalescent COVID-19 patients were enrolled for the collection of serum samples as a comparator of immunogenicity.The primary endpoint of this trial was titers of anti-receptor binding domain of spike glycoprotein(RBD)antibodies of the Omicron strain(BA.2 and BA.4/5)in serum;titers of neutralizing antibodies against pseudovirus of the Omicron strain(BA.2 and BA.4/5).The secondary endpoint was the incidence of adverse events within 30 days after the boosting.The exploratory endpoint was the cellular immune responses(interferon gamma,IFN-γ).This trial was registered with the Chinese Clinical Trial Registry website.No serious adverse events were reported within 30 days after vaccination.No Grade 3 fever or serious adverse event was reported in the SYS6006 group.Notably,SYS6006 elicited higher titers and longer increases in anti-RBD antibodies and neutralizing antibodies(>90 days)compared with the convalescent group(P<0.0001)against Omicron strain(BA.2 and BA.4/5).Besides,higher positive spots of T-cell-secreting IFN-γwere observed in the SYS6006 group than those in the convalescent group(P<0.05).These data demonstrated that SYS6006 was well tolerated and highly immunogenic,generating a stronger and more durable immune response against different variants of SARS-CoV-2. | Yuzhou Gui Ye Cao Jiajin He Chunyang Zhao Wei Zheng Ling Qian Jie Cheng Chengyin Yu Chen Yu Kun Lou Gangyi Liu Jingying Jia | 2023 | Life Metabolism2023,2,3: | 1 |
