1OsGRF4 controls grain shape, panicle length and seed shattering in rice显示文摘Traits such as grain shape, panicle length and seed shattering, play important roles in grain yield and harvest. In this study, the cloning and functional analysis of PANICLE TRAITS 2(PT2), a novel gene from the Indica rice Chuandali(CDL), is reported. PT2 is synonymous with Growth-Regulating Factor 4(OsGRF4), which encodes a growth-regulating factor that positively regulates grain shape and panicle length and negatively regulates seed shattering. Higher expression of OsGRF4 is correlated with larger grain, longer panicle and lower seed shattering. A unique OsGRF4 mutation, which occurs at the Osmi RNA396 target site of OsGRF4, seems to be associated with high levels of OsGRF4 expression, and results in phenotypic difference. Further research showed that OsGRF4 regulated two cytokinin dehydrogenase precursor genes(CKX5 and CKX1) resulting in increased cytokinin levels,which might affect the panicle traits. High storage capacity and moderate seed shattering of OsGRF4 may be useful in high-yield breeding and mechanized harvesting of rice. 2Fine mapping of GS2, a dominant gene for big grain rice显示文摘Grain shape as a major determinant of rice yield and quality is widely believed to be controlled by quantitative trait loci(QTL). We have identified a novel gene 'GS2' to largely regulate grain length and width in rice. The GS2 allele in the big-grain rice line ‘CDL’functioned in a dominant manner. In the present study, we employed a chromosome walking strategy in the residual heterozygous lines from recombinant inbred population between cultivar 'R1126' and CDL, and located the GS2 gene in an interval of ~33.2 kb flanked by marker GL2-35-1 and GL2-12 in the long arm of rice chromosome 2. According to genome annotations, three putative gene loci, LOC_Os02g47280, LOC_Os02g47290 and LOC_Os02g47300, exist in this candidate region. In addition, allelic analysis with previously reported genes demonstrated that GS2 was novel for regulating rice grain shape. 3Rapid drift of the Tethyan Himalaya terrane before two-stage India-Asia collision显示文摘The India-Asia collision is an outstanding smoking gun in the study of continental collision dynamics.How and when the continental collision occurred remains a long-standing controversy.Here we present two new paleomagnetic data sets from rocks deposited on the distal part of the Indian passive margin,which indicate that the Tethyan Himalaya terrane was situated at a paleolatitude of∼19.4°S at∼75 Ma and moved rapidly northward to reach a paleolatitude of∼13.7°N at∼61 Ma.This implies that the Tethyan Himalaya terrane rifted from India after∼75 Ma,generating the North India Sea.We document a new two-stage continental collision,first at∼61 Ma between the Lhasa and Tethyan Himalaya terranes,and subsequently at∼53−48 Ma between the Tethyan Himalaya terrane and India,diachronously closing the North India Sea from west to east.Our scenario matches the history of India-Asia convergence rates and reconciles multiple lines of geologic evidence for the collision.Jie Yuan Zhenyu Yang Chenglong Deng Wout Krijgsman Xiumian Hu Shihu Li Zhongshan Shen Huafeng Qin Wei An Huaiyu He Lin Ding Zhengtang Guo Rixiang Zhu 2021National Science Review2021,8,7:7
4Genome-wide CRISPR screen identifies synthetic lethality between DOCK1 inhibition and metformin in liver cancer显示文摘Metformin is currently a strong candidate anti-tumor agent in multiple cancers.However,its anti-tumor effectiveness varies among different cancers or sub-populations,potentially due to tumor heterogeneity.It thus remains unclear which hepatocellular carcinoma(HCC)patient subpopulation(s)can benefit from met-formin treatment.Here,through a genome-wide CRISPR-Cas9-based knockout screen,we find that DOCK1 levels determine the anti-tumor effects of met-formin and that DOCK1 is a synthetic lethal target of metformin in HCC.Mechanistically,metformin promotes DOCK1 phosphorylation,which activates RAC1 to facilitate cell survival,leading to metformin resistance.The DOCK1-selective inhibitor,TBOPP,potentiates anti-tumor activity by metformin in vitro in liver cancer cell lines and patient-derived HCC organoids,and in vivo in xenografted liver cancer cells and immunocompetent mouse liver cancer models.Notably,metformin improves overall survival of HCC patients with low DOCK1 levels but not among patients with high DOCK1 expression.This study shows that metformin effective-ness depends on DOCK1 levels and that combining metformin with DOCK1 inhibition may provide a promising personalized therapeutic strategy for met-formin-resistant HCC patients.Junru Feng Hui Lu Wenhao Ma Wenjing Tian Zhuan Lu Hongying Yang Yongping Cai Pengfei Cai Yuchen Sun Zilong Zhou Jiaqian Feng Jiazhong Deng Ying Shu Kun Qu Weidong Jia Ping Gao Huafeng Zhang 2022Protein & Cell2022,13,11:5
5北斗导航系统卫星激光测距综述显示文摘Satellite laser ranging(SLR)is an unambiguous measurement technique and generates high accuracy satellite orbit data.All satellites in the BeiDou navigation satellite system(BDS)carried laser retro-reflector arrays(LRAs),so they can be tracked by ground SLR stations in order to provide the accurate observation data.The Shanghai astronomical observatory(SHAO)designed the LRAs,and also developed the dedicated SLR systems using a 1 m-aperture telescope and a transportable cabin-based SLR system with a telescopes of 60 cm aperture.These enable BDS satellite ranging during daytime and nighttime with centimeter-level precision,allowing highly accurate estimations of satellite orbits.Moreover,some of the BDS satellites are also equipped with laser time transfer(LTT)payloads,which were developed by the SHAO and China Academy of Space Technology(CAST),providing a highly accurate time comparison between the satellites and ground clocks.This paper describes the dedicated SLR system and the design of the LRAs for BDS satellites,as well as global SLR measurements.The SLR tracking data is used for evaluating the orbit accuracy of BDS satellites and broadcast ephemeris,with an accuracy of less than 1 m.The LTT measurements to BDS satellites for a single shot have a precision of approximately 300 picoseconds,with a time stability of 20 picoseconds in 500 s.ZHANG Haifeng LONG Mingliang YANG Huafeng MENG Wendong CHENG Zhi’en DENG Huarong ZHANG Zhongping ZHANG A’li 2020Aerospace China2020,21,4:4
6A comparative study of magnetic properties between whole cells and isolated magnetosomes of Magnetospirillum magneticum AMB-1显示文摘The magnetic properties of magnetosome magnetite are of interdisciplinary interest because magnetosomes are potential carriers of natural remanent magnetization and paleoenvironment, as well as novel nano-biomaterials in biotechnological and biomedical applications. We carried out magnetic and electron transmission microscopy analyses of fresh Magnetospirillum magneticum AMB-1 whole cells and isolated magnetosomes. Results revealed that AMB-1 synthesized single-domain magnetite magneto-somes, which are arranged in the form of linear fragmental chain. The distinct differences of magnetic properties between these two samples can be faithfully interpreted in terms of spatial arrangement of magnetosomes and magnetostatic interaction. For the whole cells, the strong intra-chain interactions and weak inter-chain interactions generate behaviors of non-interacting uniaxial single-domain particles. Its δ-ratio is 3.0 and passes the Moskowitz test. In contrast, the isolated magnetosome sample has reduced values of coercivity and δ-ratio (1.5), due to increasing three-dimensional magnetostatic interactions and collapse of magneto-some chains. 8Analysis on the Breeding and Major Traits of Wide-adaptability Indica CMS Line Yungu 3A显示文摘Breeding sterile lines is the key to the development of hybrid rice with wide adaptability. However,at present,few wide-adaptability sterile lines have been applied in production practice. In this study,Yungu 3A,a new WA-type Indica CMS line with good grain quality,was developed by hybridization between Yunfeng 66 A with wide adaptability as female parent and F1 plants of Yunfeng 66 B / D62 B as male parent and continuous backcrossing. According to the results,the finally obtained Indica CMS line Yungu 3A has good plant type and leaf shape,strong tillering ability,excellent flowering habit,stable and complete male sterility with the sterile plant rate and pollen sterility rate of 100%,strong combining ability,high outcrossing rate,low sensitivity to light and temperature,and strong disease resistance,which was approved by Yunnan Provincial Crop Variety Appraisal Committee in 2013.
