6A field investigation of wind erosion in the farming–pastoral ecotone of northern China using a portable wind tunnel: a case study in Yanchi County显示文摘The farming–pastoral ecotone in northern China is an extremely fragile ecological zone where wind erosion of cropland and rangeland is easy to occur. In this study, using a portable wind tunnel as a wind simulator, we conducted field simulated wind erosion experiments combined with laboratory analysis to investigate wind erosion of soils in trampled rangeland, non-tilled cropland and tilled cropland in Yanchi County, China. The results showed that compared with rangeland, the cropland had a higher soil water holding capacity and lower soil bulk density. The wind erosion rate of trampled rangeland was much higher than those of non-tilled cropland and tilled cropland. For cropland, the wind erosion rate of the soil after tilling was surprisingly less than that of the soil before tilling. With increasing of wind speed, the volume mean diameter of the eroded sediment collected by the trough in the wind tunnel generally increased while the clay and silt content decreased for all soils. 7Characterization of putative mannoprotein in Kluyveromyces lactis for lactase production显示文摘Lactase is a member of theβ-galactosidase family of enzymes that can hydrolyze lactose into galactose and glucose.However,extracellular lactase production was still restricted to the process of cell lysis.In this study,lactase-producing Kluyveromyces lactis JNXR-2101 was obtained using a rapid and sensitive method based on the fluorescent substrate 4-methylumbelliferyl-β-D-galactopyranoside.The purified enzyme was identified as a neutral lactase with an optimum pH of 9.To facilitate extracellular production of lactase,a putative mannoprotein KLLA0_E01057g of K.lactis was knocked out.It could effectively promote cell wall degradation and lactase production after lyticase treatment,which showed potential on other extracellular enzyme preparation.After optimizing the fermentation conditions,the lactase yield from mannoprotein-deficient K.lactis JNXR-2101ΔE01057g reached 159.62 U/mL in a 5-L fed-batch bioreactor.Xiuru Shen Lingtong Liao Guoqiang Zhang Jingwen Zhou Jianghua Li Guocheng Du 2023Synthetic and Systems Biotechnology2023,8,1:0
