1mTOR/miR-145-regulated exosomal GOLM1 promotes hepatocellular carcinoma through augmented GSK-3b/MMPs显示文摘Golgi membrane protein 1(GOLM1/GP73)is a serum marker of hepatocellular carcinoma(HCC).We have previously shown that mTOR promoted tumorigenesis of HCC through stimulating GOLM1 expression.In this study,we demonstrated that the mammalian target of rapamycin(mTOR)was a negative regulator of microRNA-145(miR-145)expression.miR-145 inhibited GOLM1 expression by targeting a coding sequence of GOLM1 gene.GOLM1 and miR-145 were inversely correlated in human HCC tissues.GOLM1-enriched exosomes activated the glycogen synthase kinase-3β/matrix metalloproteinases(GSK-3β/MMPs)signaling axis of recipient cells and accelerated cell proliferation and migration.In contrast,miR-145 suppressed tumorigenesis and metastasis.We suggest that mTOR/miR-145/GOLM1 signaling pathway should be targeted for HCC treatment.Xiaochen Gai Bufu Tang Fangming Liu Yuting Wu Fang Wang Yanling Jing Fuqiang Huang Di Jin Ling Wang Hongbing Zhang 2019Journal of Genetics and Genomics2019,46,5:13
2Surface ozone monitoring and background characteristics at Zhongshan Station over Antarctica显示文摘Seasonal variation in surface ozone and the relationship between the background ozone concentration and wind were evaluated at Zhongshan Station,Antarctica using 2008 data.The wind frequency from the station area was only 2%,while the prevailing wind frequency was much larger (79.2%).This indicates that the surface ozone observations were not affected by the human activities at the station,and therefore could be counted as background concentrations of surface ozone along Antarctic coast.The concentration of surface ozone shows a distinct annual variation with the yearly mean of 25.0 nmol mol-1 and the maximum in winter,the minimum in summer.The surface ozone concentration had a strong negative correlation with ultraviolet radiation,and the mean values during polar night were one to two times higher than those in summer.These results imply that photochemical destruction of ozone dominates over Antarctica.The ozone depletion events at Zhongshan Station were obviously related to lower temperatures and higher BrO concentrations.Backward trajectory analysis reveals that the ozone depletion events are predominately caused by the high BrO concentrations.WANG YuTing BIAN LinGen MA YongFeng TANG Jie ZHANG DongQi ZHENG XiangDong 2011Chinese Science Bulletin2011,56,10:7
3Confined van der Waals Epitaxial Growth of Two-Dimensional Large Single-Crystal In_(2)Se_(3) for Flexible Broadband Photodetectors显示文摘The controllable growth of two-dimensional(2D)semiconductors with large domain sizes and high quality is much needed in order to reduce the detrimental efect of grain boundaries on device performance but has proven to be challenging.Here,we analyze the precursor concentration on the substrate surface which signifcantly infuences nucleation density in a vapor deposition growth process and design a confned micro-reactor to grow 2D In_(2)Se_(3) with large domain sizes and high quality.Te uniqueness of this confned micro-reactor is that its size is∼102-103 times smaller than that of a conventional reactor.Such a remarkably small reactor causes a very low precursor concentration on the substrate surface,which reduces nucleation density and leads to the growth of 2D In_(2)Se_(3) grains with sizes larger than 200�m.Our experimental results show large domain sizes of the 2D In_(2)Se_(3) with high crystallinity.Te fexible broadband photodetectors based on the as-grown In_(2)Se_(3) show rise and decay times of 140 ms and 25 ms,efcient response(5.6 A/W),excellent detectivity(7×10^(10) Jones),high external quantum efciency(251%),good fexibility,and high stability.Tis study,in principle,provides an efective strategy for the controllable growth of high quality 2D materials with few grain boundaries.Lei Tang Changjiu Teng Yuting Luo Usman Khan Haiyang Pan Zhengyang Cai Yue Zhao Bilu Liu Hui-Ming Cheng 2019Research2019,,1:7
4Genome-and transcriptome-wide association studies provide insights into the genetic basis of natural variation of seed oil content in Brassica napus显示文摘Seed oil content(SOC)is a highly important and complex trait in oil crops.Here,we decipher the genetic basis of natural variation in SOC of Brassica napus by genome-and transcriptome-wide association studies using 505 inbred lines.We mapped reliable quantitative trait loci(QTLs)that control SOC in eight environments,evaluated the effect of each QTL on SOC,and analyzed selection in QTL regions during breeding.Six-hundred and ninety-two genes and four gene modules significantly associated with SOC were identified by analyzing population transcriptomes from seeds.A gene prioritization framework,POCKET(prioritizing the candidate genes by incorporating information on knowledge-based gene sets,effects of variants,genome-wide association studies,and transcriptome-wide association studies),was implemented to determine the causal genes in the QTL regions based on multi-omic datasets.A pair of homologous genes,BnPMT6s,in two QTLs were identified and experimentally demonstrated to negatively regulate SOC.This study provides rich genetic resources for improving SOC and valuable insights toward understanding the complex machinery that directs oil accumulation in the seeds of B.napus and other oil crops.Shan Tang Hu Zhao Shaoping Lu Liangqian Yu Guofang Zhang Yuting Zhang Qing-Yong Yang Yongming Zhou Xuemin Wang Wei Ma Weibo Xie Liang Guo 2021Molecular Plant2021,14,3:6

6Purely organic optoelectronic materials with ultralong-lived excited states under ambient conditions显示文摘The exponential growth of utilizing synthetic organic molecules in optoelectronic applications poses strong demands for rational control over the excited states of the materials. The manipulation of excited states through molecular design has led to the development of high-performance optoelectronic devices with tunable emission colors, high quantum efficiencies and efficient energy/charge transfer processes. Recently, a significant breakthrough in lifetime tuning of excited states has been made;the purely organic molecules were found to have ultralonglived excited state under ambient conditions with luminescence lifetimes up to 1.35 s, which are several orders of magnitude longer than those of conventional organic fluorophores. Given the conceptual advance in understanding the fundamental behavior of excited state tuning in organic luminescent materials, the investigations of organic ultralong room-temperature phosphorescence(OURTP) should provide new directions for researches and have profound impacts on many different disciplines. 7Accelerated evolution of an Lhx2 enhancer shapes mammalian social hierarchies显示文摘Social hierarchies emerged during evolution,and social rank influences behavior and health of individuals.However,the evolutionary mechanisms of social hierarchy are still unknown in amniotes.Here we developed a new method and performed a genome-wide screening for identifying regions with accelerated evolution in the ancestral lineage of placental mammals,where mammalian social hierarchies might have initially evolved.Then functional analyses were conducted for the most accelerated region designated as placental-accelerated sequence 1(PAS1,P=3.15 × 10^-18).Multiple pieces of evidence show that PAS1 is an enhancer of the transcription factor gene Lhx2 involved in brain development.PAS1s isolated from various amniotes showed different c/s-regulatory activity in vitro,and affected the expression of Lhx2 differently in the nervous system of mouse embryos.PAS1 knock-out mice lack social stratification.PAS1 knock-in mouse models demonstrate that PAS1s determine the social dominance and subordinate of adult mice,and that social ranks could even be turned over by mutated PAS1.All homozygous mutant mice had normal huddled sleeping behavior,motor coordination and strength.Therefore,PAS1-Lhx2 modulates social hierarchies and is essential for establishing social stratification in amniotes,and positive Darwinian selection on PAS1 plays pivotal roles in the occurrence of mammalian social hierarchies.Yuting Wang Guangyi Dai Zhili Gu Guopeng Liu Ke Tang Yi-Hsuan Pan Yujie Chen Xin Lin Nan Wu Haoshan Chen Su Feng Shou Qiu Hongduo Sun Qian Li Chuan Xu Yanan Mao Yong Edward Zhang Philipp Khaitovich Yan-Ling Wang Qunxiu Liu Jing-Dong Jackie Han Zhen Shao Gang Wei Chun Xu Naihe Jing Haipeng Li 2020Cell Research2020,30,5:4

9Effects of Angelica sinensis extracts on lipid oxidation in fish feeds and growth performance of juvenile Jian carp(Cyprinus carpio var. Jian)显示文摘The study was to explore the effect of the extracts of Angelica sinensis(EAs) on lipid oxidation in fish feeds compared with ethoxyquin(EQ) and the effect of dietary EAs on growth performance of carp(Cyprinus carpio var. Jian). Firstly, fish feeds were respectively added with EQ, and ethyl ether extract ethyl acetate extract(EAE), acetone extract, ethanol extract(EE) and aqueous extract(AQE) of Angelica sinensis, except for the control. The results showed that EAs and EQ inhibited lipid oxidation in fish feed(P < 0.05). Of all of the examined EAs, EAE showed the strongest protective effects against lipid oxidation(P < 0.05). Moreover, EAE at high concentrations showed a stronger effect on lipid oxidation compared with EQ(P < 0.05). Then, 7 experimental diets respectively supplemented with 0.0, 1.0, 2.0, 3.0, 4.0, 5.0 and 6.0 g/kg of EAE were fed to the respective treatment groups for 30 d. Four replicates were performed for each treatment group; 20 carp(mean weight: 12.10 ± 0.13 g) were in each replicate. The result indicated that dietary EAE improved the growth performance in carp(P < 0.05). The appropriate con centration of EAE for carp growth was estimated to be 3.643 g/kg diet. 10Model updating for real time dynamic substructures based on UKF algorithm显示文摘Combining the advantages of numerical simulation with experimental testing,real-time dynamic substructure(RTDS)testing provides a new experimental method for the investigation of engineered structures.However,not all unmodeled parts can be physically tested,as testing is often limited by the capacity of the test facility.Model updating is a good option to improve the modeling accuracy for numerical substructures in RTDS.In this study,a model updating method is introduced,which has great performance in describing this nonlinearity.In order to determine the optimal parameters in this model,an Unscented Kalman Filter(UKF)-based algorithm was applied to extract the knowledge contained in the sensors data.All the parameters that need to be identified are listed as the extended state variables,and the identification was achieved via the step-by-step state prediction and state update process.Effectiveness of the proposed method was verified through a group of experimental data,and results showed good agreement.Furthermore,the proposed method was compared with the Extended Kalman Filter(EKF)-based method,and better accuracy was easily found.The proposed parameter identification method has great applicability for structural objects with nonlinear behaviors and could be extended to research in other engineering fields.Su Tingli Tang Zhenyun Peng Lingyun Bai Yuting Jin Xuebo Kong Jianlei 2020Earthquake Engineering and Engineering Vibration2020,19,2:2

12Artemisinin analogue SM934 protects against lupus-associated antiphospholipid syndrome via activation of Nrf2 and its targets显示文摘Kidney is a major target organ in both antiphospholipid syndrome(APS)and systemic lupus erythematosus(SLE).The etiology of antiphospholipid syndrome nephropathy associated lupus nephritis(APSN-LN)is intricate and remains largely unrevealed.We proposed in present work,that generation of antiphospholipid antibodies(aPLs),especially those directed towards the oxidized neoepitopes,are largely linked with the redox status along with disease progression.Moreover,we observed that compromised antioxidative capacity coincided with turbulence of inflammatory cytokine profile in the kidney of male NZW×BXSB F1 mice suffered from APSN-LN.SM934 is an artemisinin derivative that has been proved to have potent immunosuppressive properties.In current study,we elaborated the therapeutic benefits of SM934 in male NZW×BXSB F1 mice,a murine model develops syndrome resembled human APS associated with SLE,for the first time.SM934 treatment comprehensively impeded autoantibodies production,inflammatory cytokine accumulation and excessive oxidative stress in kidney.Among others,we interpreted in present work that both anti-inflammatory and antioxidative effects of SM934 is closely correlated with the enhancement of Nrf2 signaling and expression of its targets.Collectively,our finding confirmed that therapeutic strategy simultaneously exerting antioxidant and anti-inflammatory efficacy provide a novel feasible remedy for treating APSN-LN.Zemin Lin Yuting Liu Li Chen Shiqi Cao Yueteng Huang Xiaoqian Yang Fenghua Zhu Wei Tang Shijun He Jianping Zuo 2021Science China(Life Sciences)2021,64,10:2
