1Add-on pegylated interferon augments hepatitis B surface antigen clearance vs continuous nucleos(t)ide analog monotherapy in Chinese patients with chronic hepatitis B and hepatitis B surface antigen≤1500 IU/mL:An observational study显示文摘BACKGROUND Nucleos(t)ide analog(NA)has shown limited effectiveness against hepatitis B surface antigen(HBsAg)clearance in chronic hepatitis B(CHB)patients.AIM To evaluate the efficacy and safety of add-on peginterferonα-2a(peg-IFNα-2a)to an ongoing NA regimen in CHB patients.METHODS In this observational study,195 CHB patients with HBsAg≤1500 IU/m L,hepatitis B e antigen(HBeAg)-negative(including HBeAg-negative patients or HBeAg-positive patients who achieved HBeAg-negative after antiviral treatment with NA)and hepatitis B virus-deoxyribonucleic acid<1.0×10^2 IU/mL after over 1 year of NA therapy were enrolled between November 2015 and December2018 at the Second Affiliated Hospital of Xi'an Jiaotong University,China.Patients were given the choice between receiving either peg-IFNα-2a add-on therapy to an ongoing NA regimen(add-on group,n=91)or continuous NA monotherapy(monotherapy group,n=104)after being informed of the benefits and risks of the peg-IFNα-2a therapy.Total therapy duration of peg-IFNα-2a was 48 wk.All patients were followed-up to week 72(24 wk after discontinuation of peg-IFNα-2a).The primary endpoint was the proportion of patients with HBsAg clearance at week 72.RESULTS Demographic and baseline characteristics were comparable between the two groups.Intention-to-treatment analysis showed that the HBsAg clearance rate in the add-on group and monotherapy group was 37.4%(34/91)and 1.9%(2/104)at week 72,respectively.The HBsAg seroconversion rate in the add-on group was 29.7%(27/91)at week 72,and no patient in the monotherapy group achieved HBsAg seroconversion at week 72.The HBsAg clearance and seroconversion rates in the add-on group were significantly higher than in the monotherapy group at week 72(P<0.001).Younger patients,lower baseline HBsAg concentration,lower HBsAg concentrations at weeks 12 and 24,greater HBsAg decline from baseline to weeks 12 and 24 and the alanine aminotransferase≥2×upper limit of normal during the first 12 wk of therapy were strong predictors of HBsAg clearance in patients with peg-IFNα-2a add-on treatment.Regarding the safety of the treatment,4.4%(4/91)of patients in the add-on group discontinued peg-IFNα-2a due to adverse events.No severe adverse events were noted.CONCLUSION Peg-IFNα-2a as an add-on therapy augments HBsAg clearance in HBeAg-negative CHB patients with HBsAg≤1500 IU/m L after over 1 year of NA therapy.Feng-Ping Wu Ying Yang Mei Li Yi-Xin Liu Ya-Ping Li Wen-Jun Wang Juan-Juan Shi Xin Zhang Xiao-Li Jia Shuang-Suo Dang 2020World Journal of Gastroenterology2020,26,13:27
2Deep soil water infiltration and its dynamic variation in the shifting sandy land of typical deserts in China显示文摘Soil moisture is the key resource constraint in arid ecosystems, and has been a focus of research on restoration. However, quantitative studies on the contribution of rainfall to deep soil rainfall infiltration are lacking. In this study, we used the YWB-01 Deep Soil Infiltration Water Recorder which had been invented by ourselves to measure the quantity of rain infiltration into deep soil, 150 cm below ground, in four locations in China: Mu Us Sandy Land and Ulan Buh, Tengger, and Badan Jilin deserts over a 2-year period. We found:(1) Deep soil rainfall infiltration decreased progressively from east to west and from semiarid to arid areas, with two locations completely lacking rainfall infiltration. Heavy rain was important to deep soil infiltration in shifting sandy land of arid and semiarid areas.(2) Seasonal variation of infiltration was correlated with rainfall, with a time lag that was less apparent in areas with more rainfall.(3) For single intense rainfall events, infiltration maximums occurred 40–55 h after the rainfall, during which the infiltration rates increased rapidly before reaching a peak, and then decreased slowly. Continuous infiltration could last about 150 h. Rainfall infiltration was determined by the combined action of intensity, quantity and duration. Rainfall with low intensity, long duration, and large quantity was most favorable for deep soil infiltration. 3Surveillance-based evidence:elimination of schistosomiasis as a public health problem in the Peoples’Republic of China显示文摘Background:A steady progress on schistosomiasis control in the Peoples’Republic of China(P.R.China)was achieved and broadened into the twelve-year medium and long term national plan(MLNP)which marled the implementation of an integrated control strategy across all endemic areas in P.R.China in 2004.To understand the endemic trends of schistosomiasis to assess the effectiveness of an integrated strategy,we conducted an analysis of schistosomiasis surveillance data spanned from 2005 to 2015.Methods:The schistosomiasis sentinel surveillance data from sentinel sites were collected and analyzed from 2005 to 2015.In these sentinel sites,residents aged 6 years or above were screened annually by indirect hemagglutination assay(IHA),while only antibody positives were followed by stool examination either Kato-katz method(KK)and/or hatching technique(HT).Domestic animals raised in sentinel sites were examined by HT for confirming the infection of schistosomes.Snail investigation was conducted each year through systematic sampling method combined with environmental sampling method.The snails collected from field were tested by microscopic dissection method.The infection rates of schistosomes in residents,domestic animals and snails,as well as the indicators reflecting the snails’distribution were calculated and analyzed.ANOVA analysis was used to examine the changes of the number of eggs per gram feces in population and Chi-square test was used to examine any change in proportions among groups.Results:A total of 148902 residents from sentinel sites attended this study and 631676 blood samples were examined by IHA test during the 11 covered years.The annual average antibody positive rates presented a significant decrease trends,from 17.48%(95%CI:17.20–17.75%)in 2005 to 5.93%(95%CI:5.71–6.15%)(χ2=8890.47,P<0.001)in 2015.During 2005–2015,the average infection rate of schistosomes in residents declined from 2.07%(95%CI:1.96–2.17%)to 0.13%(95%CI:0.09–0.16%),accompanied by significant decrease of infection intensity in population.In 2015,the stool positives were only found in farmers,fishermen and boatmen with infection rate of 0.16%(95%CI:0.11–0.20%),0.17%(95%CI:0–0.50%)respectively.The infection rate of schistosomes in domestic animals dropped from 9.42%(538/5711,95%CI:8.66–10.18%)to 0.08%(2/2360,95%CI:0–0.20%)from 2005 to 2015.Infections were found in eight species of domestic animals at the beginning of surveillance while only two cattle were infected in 2015.Totally 98 ha of new snail habitats were found,while 94.90%(93/98)distributed in lake and marshland regions.The percentage of frames with snails decreased from 16.96%(56884/335391,95%CI:16.83–17.09%)in 2005 to 4.28%(18121/423755,95%CI:4.22–4.34%)in 2014,with a slightly increase in 2015.Meanwhile,the infection rate of schistosomes in snails was decreased from 0.26%(663/256531,95%CI:0.24–0.28%)to zero during 2005–2015.Conclusions:The infection rate of schistosomes declined significantly,providing evidence that the goal of the MLNP was achieved.Elimination of schistosomiasis as a public health problem defined as WHO was also reached in P.R.China nationwide.Surveillance-response system should be improved and strengthened to realize the final goal of schistosomiasis elimination.Jing Xu Shi-Zhu Li Li-Juan Zhang Robert Bergquist Hui Dang Qiang Wang Shan Lv Tian-Ping Wang Dan-Dan Lin Jian-Bing Liu Guang-Hui Ren Kun Yang Yang Liu Yi Dong Shi-Qing Zhang Xiao-Nong Zhou 2020Infectious Diseases of Poverty2020,9,3:20
4Long noncoding RNAs in hepatocellular carcinoma: Novel insights into their mechanism显示文摘Hepatocellular carcinoma(HCC) is the predominant subject of liver malignancies which arouse global concern. Advanced studies have found that long noncoding RNAs(lnc RNAs) are differentially expressed in HCC and implicate they may play distinct roles in the pathogenesis and metastasis of HCC. However, the underlying mechanisms remain largely unclear. In this review, we summarized the functions and mechanisms of those known aberrantly expressed lncR NAs identified in human HCC tissues. We hope to enlighten more comprehensive researches on the detailed mechanisms of lncR NAs and their application in clinic, such as being used as diagnostic and prognostic biomarkers and the targets for potential therapy. 5Prognosis of HER2 over-expressing gastric cancer patients with liver metastasis显示文摘AIM:To study the risk factors for liver metastasis and the prognosis in patients with human epidermal growth factor receptor 2(HER2) over-expressing gastric cancer(GC).METHODS:A total of 84 GC patients recruited from the General Hospital of the People's Liberation Army(PLA) between 2003 and 2010 were randomly enrolled in this study.HER2 expression was detected by immunohistochemistry in 84 GC patients with liver metastases.The study group consisted of 66 men and 18 women,with an average age of 54 years(range:19-74 years).Liver metastasis was diagnosed by magnetic resonance imaging or computed tomography.Patients were followed-up and predictive factors of liver metastasis were evaluated.RESULTS:The median follow-up period was 47 mo(range:6-85 mo).The characteristics of 35(25.7%) patients with HER2 over-expression of liver metastatic GC are presented.HER2 over-expression was detected in 23 out of 49(46.9%) patients with intestinal GC,and 9 out of 35(25.7%) patients with diffuse GC.29 out of 59(49.2%) patients aged < 60 years were HER2positive,while 8 out of 25(32%) patients aged ≥ 60 were HER2-positive;a significant difference(P < 0.05).Univariate analysis(log-rank test) showed that HER2 over-expression,sex,Lauren classification,differentiation and disease-free interval were correlated with poor survival(P < 0.05).Survival analysis with a survival curve showed that HER2 over-expression was significantly relevant,with a reduced survival time in GC patients with liver metastases(P < 0.01).2-year survival was not associated with the patient's age.A diseasefree survival longer than 12 mo has a significant association with extended overall survival(OS) in GC patients with liver metastases.The median survival time after the diagnosis of liver metastases was 18 mo [95% confidence interval(CI):9.07-26.94] among HER2 positive GC patients with liver metastases.In comparison,for 49(69.4%) out of 84 HER2 negative patients with liver metastatic GC,the median survival time was 47 mo(95% CI:19.37-74.63).In patients with HER2 positive liver metastatic GC,the median OS was significantly shorter than in HER2 negative patients(median,20.32 mo;95% CI:16.51-24.13 vs median,50.14 mo;95% CI:37.83-62.45;P < 0.01).CONCLUSION:HER2 over-expressing GC patients with liver metastases have a poor prognosis.Overall survival was significantly lower in HER2 positive patients.HER2overexpression is correlated with a lower survival rate.Hai-Zhen Dang Yang Yu Shun-Chang Jiao 2012World Journal of Gastroenterology2012,18,19:17
6Effects of sodium ferulate on preventing steroid-induced femoral head osteonecrosis in rabbits显示文摘The aim of this study is to investigate the effects and possible mechanisms of sodium ferulate (SF) on anti-apoptosis in steroid-induced femoral head osteonecrosis in rabbits. Japanese white rabbits were randomly divided into three groups (control group, treatment group, and model group), each with 24 rabbits. The model and treatment groups were first injected with an intravenous dose of horse serum, 10 ml/kg, three weeks later with an intravenous dose of 7.5 ml/kg, and two weeks later with an intramuscular dose of methylprednisolone, 45 mg/kg, three times in order to establish rabbit models of osteonecrosis. Concurrently, the treatment group was injected with intravenous doses of SF 20 mg/kg for two weeks, once per day. Three time points, Weeks 2, 4, and 8, were selected after modeling was completed. Osteonecrosis was verified by histopathology with haematoxylin-eosin (HE) staining. The apoptosis rate of osteonecrosis was observed by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay. The apoptosis expressions of caspase-3 and Bcl-2 were analyzed by immunohistochemistry and Western blot. The rabbit models of osteonecrosis were successfully established and observed by HE staining. SF was effective in intervening in apoptosis and decreasing the apoptosis rate in femoral head necrosis by the immunohistochemistry and TUNEL assay (P<0.01). Western blot analysis indicated that there were statistical significances in the protein levels of caspase-3 and Bcl-2 (P<0.01). 7Experimental exploration and research prospect of physical bases and functional characteristics of meridians显示文摘The physical basis of meridians and acupoints (acupuncture points) is in a complex system mainly of connective tissue and interwoven with the blood capillaries, nerves, lymph vessels,etc. Elements of Ca, P, K, Fe, Zn, Mn, etc are found concentrated in the deep connective tissue structures in locations corresponding to acupoints. On this physical basis, the specific waveband of high efficiency for transmission of infrared rays exists in the collagenous fibre in a liquid crystal state. 9Changes in count and function of splenic lymphocytes from patients with portal hypertension显示文摘AIM: To investigate changes in numbers and proliferative function of splenic lymphocytes in patients with hypersplenism due to portal hypertension (PH), to provide evidence for further study of immune status of the spleen during PH. METHODS: Twelve spleens from patients with hypersplenism due to PH served as the PH group, and four spleens from cases of traumatic spleen rupture were regarded as the control group. After weighing the spleen, lymphocytes were separated and counted using a cell counting plate to calculate the lymphocyte count per gram of spleen tissue (relative quantity) and total lymphocyte count in whole spleen (absolute quantity). The immunohistochemical SP method was used to observe the density and distribution of lymphocytes in the spleen. The MTT method was used to observe changes in lymphocyte proliferative function. RESULTS: As compared to the control group, the splenic lymphocytes in the PH group showed that: (1) There was no difference in distribution but a significant decreasein density; (2) the number of lymphocytes per gram of spleen (relative quantity) decreased significantly (0.822 ± 0.157) × 108 vs (1.174 ± 0.254) × 108, P < 0.01]; (3) with the significant increase in the weight of the PH spleen (832.6 ± 278.2 g vs 211.7 ± 85.6 g, P < 0.01), the total quantity of lymphocytes (absolute quantity) increased significantly (0.685 ± 0.072) × 1011 vs (0.366 ± 0.057) × 1011, P < 0.01]; and (4) the proliferative function of lymphocytes was enhanced: T lymphocytes, (0.022 ± 0.005 vs 0.015 ± 0.003, P < 0.05), and B lymphocytes (0.034 ± 0.006 vs 0.023 ± 0.001, P < 0.01). CONCLUSION: Although lymphocyte density in the spleen decreased in patients with PH, the total quantity of lymphocytes increased because spleen weight increased greatly, along with the proliferating function. With respect to changes in lymphocytes, PH spleens may still have immune function, although it may be disordered. 10Morphological and cytological changes of meibomian glands in patients with type 2 diabetes mellitus显示文摘AIM: To observe the changes of microcellular structure of meibomian glands(MGs) in type 2 diabetes mellitus(DM), and to explore its correlation with the duration of diabetes.METHODS: The study assessed 132 eyes of 132 patients with type 2 diabetes mellitus(DM group) and 100 eyes of 100 non-diabetic participants(NDM group). All patients underwent the examination of the Keratograph 5 M system to obtain the meibography which were used to evaluate the structure dropout of the MGs. And then laser scanning confocal microscopy(LSCM) was performed for observing the acinar cells and ducts of the MGs to obtain the following parameters: the MG acinar unit density(MGAUD), MG acinar longest diameter(MGALD) and MG acinar shortest diameter(MGASD). The examination results of the right eye were selected for analysis.RESULTS: Compared with that in NDM group, the meiboscore was significantly higher(Z=-4.057, P<0.001), and there were more MGs dropout in DM group. With the prolongation of the course of diabetes, the absence of MGs aggravated and the MGs dropout score increased(r=0.596;P<0.001). LSCM showed that there were various cytological alterations in acinar cells of MGs with the progress of diabetes duration, such as expansion, atrophy or fibrosis of MG acinar units, decreased density of MG acinar units, deposition of lipid substances, infiltration of inflammatory cells, proliferation of fibrous tissues, etc. And the opening of the glandular duct changed from smooth at the beginning to narrow, blocked, fibrotic and so on. Compared with that in NDM group, the MGAUD in DM group was significantly lower(Z=-9.713;P<0.001), the MGALD and MGASD were significantly larger(Z=-9.751,-6.416;P<0.001). With the duration of diabetes, the MGAUD reduced, the MGASD increased(r=0.860, 0.364, P<0.001);but the MGALD had no correlation with diabetic duration(r=0.133, P=0.151).CONCLUSION: With the progress of diabetes, the meibomian glandular acinar cells of diabetic patients show various manifestations. 11Caffeic acid phenethyl ester up-regulates antioxidant levels in hepatic stellate cell line T6 via an Nrf2-mediated mitogen activated protein kinases pathway显示文摘AIM To investigate the antioxidant effect of caffeic acid phenethyl ester(CAPE) in hepatic stellate cell-T6(HSC-T6) cells cultured in vitro and the potential mechanisms.METHODS HSC-T6 cells were cultured in vitro and treated with various concentrations of CAPE for 24, 48 and 72 h, respectively. Cell proliferation was investigated using the MTT assay, and cell ultrastructural alterations were observed by transmission electron microscopy. Flow cytometry was employed to investigate the effects of CAPE on apoptosis and the levels of reactive oxygen species in HSC-T6 cells cultured in vitro. An enzyme immunoassay instrument was used to evaluate antioxidant enzyme expression. The effect on α-smooth muscle actin was shown using immunofluorescence. Gene and protein levels of Nrf2, related factors, and mitogen activated protein kinases(MAPKs), in HSC-T6 cells were detected using RT-PCR and Western blot, respectively.RESULTS CAPE inhibited the proliferation and activation of HSC-T6 cells cultured in vitro. CAPE increased the antioxidant levels and the translocation of Nrf2 from the cytoplasm to the nucleus in HSC-T6 cells. Moreover,the phosphorylation of MAPKs in cells decreased in response to CAPE. 12Response of sediment yield to vegetation restoration at a large spatial scale in the Loess Plateau显示文摘The impact of vegetation coverage on erosion and sediment yield in the Loess Plateau has been extensively studied,but the research has been primarily based on observations from slope runoff plots or secondary forest regions;the scaling method remains unresolved when it is applied at a large spatial scale,and it is difficult to apply to regions with severe soil and water loss given the predominance of herbs and shrubs.To date,there is little data on the quantitative impact of changes to vegetation on sediment concentration at a large spatial scale.This paper is based on vegetation information from remote sensing images,measured rainfall and sediment data over nearly 60 years,and results from previous runoff and sediment variation research on the Yellow River.We introduce the concepts of a sediment yield coefficient and the percentage of effective vegetation and erodible area,analyze the impact of different vegetation conditions on the flood sediment concentration and sediment yield,and evaluate the effect of rainfall intensity on sediment yield under different vegetation conditions at the watershed scale.We propose models to evaluate the impact of vegetation on sediment yield in the loess gully hilly region,which are based on remote sensing data and support an application at a large spatial scale.The models can be used to assess sediment reduction that results from the current significant improvement of vegetation in the Loess Plateau.LIU XiaoYan YANG ShengTian DANG SuZhen LUO Ya LI XiaoYu ZHOU Xu 2014Science China(Technological Sciences)2014,57,8:11
13Spinal Cord Stimulation Frequency Influences the Hemodynamic Response in Patients with Disorders of Consciousness显示文摘Spinal cord stimulation(SCS) is a promising technique for treating disorders of consciousness(DOCs).However, differences in the spatio-temporal responsiveness of the brain under varied SCS parameters remain unclear. In this pilot study, functional near-infrared spectroscopy was used to measure the hemodynamic responses of 10 DOC patients to different SCS frequencies(5 Hz, 10 Hz, 50 Hz, 70 Hz, and 100 Hz). In the prefrontal cortex, a key area in consciousness circuits, we found significantly increased hemodynamic responses at 70 Hz and 100 Hz, and significantly different hemodynamic responses between 50 Hz and 70 Hz/100 Hz. In addition, the functional connectivity between prefrontal and occipital areas was significantly improved with SCS at 70 Hz. 14Current Status of Neuromodulatory Therapies for Disorders of Consciousness显示文摘Treatment for disorders of consciousness(DOCs) is still a Gordian knot. Evidence-based guidelines on the treatment of DOC patients are not currently available, while neuromodulation techniques are seen as a potential treatment. Multiple neuromodulation therapies have been applied. This article reviews the most relevant studies in the literature in order to describe a clear picture of the current state of neuromodulation therapies that could be used to treat DOC patients. Both invasive and noninvasive brain stimulation is discussed. Significant behavioral improvements in prolonged DOCs under neuromodulation therapies are rare. The efficacy of various such therapies remains a matter of debate. 15Combustion characteristics of semicokes derived from pyrolysis of low rank bituminous coal显示文摘Various semicokes were obtained from medium-low temperature pyrolysis of Dongrong long flame coal.The proximate analysis,calorific value and Hardgrove grindability index(HGI) of semicokes were determined,and the ignition temperature,burnout temperature,ignition index,burnout index,burnout ratio,combustion characteristic index of semicokes were measured and analyzed using thermogravimetry analysis(TGA).The effects of pyrolysis temperature,heating rate,and pyrolysis time on yield,composition and calorific value of long flame coal derived semicokes were investigated,especially the influence of pyrolysis temperature on combustion characteristics and grindability of the semicokes was studied combined with X-ray diffraction(XRD) analysis of semicokes.The results show that the volatile content,ash content and calorific value of semicokes pyrolyzed at all process parameters studied meet the technical specifications of the pulverized coal-fired furnaces(PCFF) referring to China Standards GB/T 7562-1998.The pyrolysis temperature is the most influential factor among pyrolysis process parameters.As pyrolysis temperature increases,the yield,ignition index,combustion reactivity and burnout index of semicokes show a decreasing tend,but the ash content increases.In the range of 400 and 450 °C,the grindability of semicokes is rational,especially the grindability of semicokes pyrolyzed at 450 °C is suitable.Except for the decrease of volatile content and increase of ash content,the decrease of combustion performance of semicokes pyrolyzed at higher temperature should be attributed to the improvement of the degree of structural ordering and the increase of aromaticity and average crystallite size of char.It is concluded that the semicokes pyrolyzed at the temperature of 450 °C is the proper fuel for PCFF.Qian wei Xie Qiang Huang Yuyi Dang Jiatao Sun Kaidi Yang Qian Wang Jincao 2012International Journal of Mining Science and Technology2012,22,5:10
16Sorption of tylosin and sulfamethazine on solid humic acid显示文摘Tylosin(TYL) and sulfamethazine(SMT) are ionizable and polar antimicrobial compounds,which have seeped into the environment in substantial amounts via fertilizing land with manure or sewage. Sorption of TYL and SMT onto humic acid(HA) may affect their environmental fate. In this study, the sorption of TYL and SMT on HA at different conditions(pH, ionic strength) was investigated. All sorption isotherms fitted well to the Henry and Freundlich models and they were highly nonlinear with values of n between 0.5 and 0.8, which suggested that the HA had high heterogeneity. The sorption of TYL and SMT on HA decreased with increasing p H(2.0–7.5), implying that the primary sorption mechanism could be due to cation exchange interactions between TYL^+/SMT^+ species and the functional groups of HA.Increasing ionic strength resulted in a considerable reduction in the K_d values of TYL and SMT,hinting that interactions between H bonds and π–π EDA might be an important factor in the sorption of TYL and SMT on HA. Results of Fourier transform infrared(FT-IR) and ~13C-nuclear magnetic resonance(NMR) analysis further demonstrated that carboxyl groups and O-alkyl structures in the HA could interact with TYL and SMT via ionic interactions and H bonds,respectively. 17Environmental impacts on the distribution of microbial tetraether lipids in Chinese lakes with contrasting pH: Implications for lacustrine paleoenvironmental reconstructions显示文摘Glycerol dialkyl glycerol tetraethers(GDGTs) in lake sediments are useful biomarkers for the continental paleoclimatic reconstruction. However, the environmental controls on the distribution of these compounds, in particular the 6-methyl isomers of bacterial branched GDGTs(bGDGTs), in the lakes with contrasting pH, are still unknown, hindering their application for paleo-reconstructions. Here, we investigated the environmental impacts on the distribution of GDGTs in 17 alkaline lakes and 1 acid lake in China. It was found that the dissolved oxygen content in water column may have an impact on the distribution of archaeal isoprenoid GDGTs(iGDGTs) by causing the change in archaeal communities. The ratio of GDGT-0/crenarchaeol increases with decreasing oxygen content, indicating that the relative abundance of anaerobic methanogenic archaea or Miscellaneous Crenarchaeotic Group(MCG) vs. aerobic Thaumarchaeota is controlled by the oxygen content dissolved in water of these lakes. Thaumarchaeota are likely to contribute only a small proportion of iGDGTs in the relatively oxygen-depleted lakes, and thus TEX_(86) is not suitable for the reconstruction of the surface temperature of these lakes. The abundance ratio of iGDGTs to bGDGTs(R_(i/b)) appears to show no relationship with water pH in all the lakes, but exhibits a significant positive correlation with the water depth of the acid Lake Qinghai in Tengchong. As expected, the methylation degree of bGDGTs(MBT′) was found to correlate with both mean annual air temperature(MAT) and water pH, and the cyclization degree of bGDGTs(CBT) correlates only with water pH in these lakes. However, the MBT′_(5ME), an index to measure the methylation degree of 5-methyl bGDGTs, exhibits no relationship with MAT, whereas MBT′_(6ME), the methylation degree of 6-methyl bGDGTs, was found to correlate significantly with MAT. This is opposite to the situation observed in the global soils, pointing to a different adaptation of b GDGT-producing bacteria to environmental variables or different microbial sources of bGDGTs in these lakes. The relative abundance of 6- vs. 5-methyl bGDGTs is controlled by pH in these lakes, similar to that observed in worldwide soils. 19Clinical characteristics of coronavirus disease 2019 in patients aged 80 years and older显示文摘Objective: Coronavirus disease 2019(COVID-19) has raised concern around the world as an epidemic or pandemic. As data on COVID-19 has grown, it has become clear that older adults have a disproportionately high rate of death from COVID-19. This study describes the early clinical characteristics of COVID-19 in patients with more than 80 years of age.Methods: Epidemiological, clinical, laboratory, radiological, and treatment data from 17 patients diagnosed with COVID-19 between January 20 and February 20, 2020 were collected and analyzed retrospectively. Treatment outcomes among subgroups of patients with non-severe and severe symptoms of COVID-19 were compared.Results: Of the 17 hospitalized patients with COVID-19, the median age was 88.0 years(interquartile range, 86.6–90.0 years;range, 80.0–100.0 years) and 12(70.6%) were men. The age distribution of patients was not significantly different between non-severe group and severe group. All patients had chronic pre-existing conditions. Hypertension and cardiovascular diseases were the most common chronic conditions in both subgroups. The most common symptoms at the onset of COVID-19 were fever(n = 13;76.5%), fatigue(n = 11;64.7%), and cough(n = 5;29.4%). Lymphopenia was observed in all patients, and lymphopenia was significantly more severe in the severe group than that in non-severe group(0.4×10~9/L vs 1.2×10~9/L, P = 0.014). The level of serum creatinine was higher in the severe group than in the non-severe group(99.0 lmol/L vs 62.5 lmol/L, P = 0.038). The most common features of chest computed tomography images were nodular foci in 10(58.8%) patients and pleural thickening in 7(41.2%) patients. All patients received antiviral therapy, while some patients also received intravenous antibiotics therapy(76.5%), Chinese medicinal preparation therapy(Lianhuaqingwen capsule, 64.7%), corticosteroids(35.3%) or immunoglobin(29.4%). Eight patients(47.1%) were transferred to the intensive care unit because of complications. Ten patients(58.8%) received intranasal oxygen, while 3(17.6%)received non-invasive mechanical ventilation, and 4(23.5%) received high-flow oxygen. As of June 20,7(41.2%) patients had been discharged and 10(58.8% of this cohort, 77.8% of severe patients) had died.Conclusion: The mortality of patients aged 80 years and older with severe COVID-19 symptoms was high.Lymphopenia was a characteristic laboratory result in these patients, and the severity of lymphopenia was indicative of the severity of COVID-19. 20Comparison of paleotemperature reconstructions using microbial tetraether thermometers of the Chinese loess-paleosol sequence for the past 350000 years显示文摘The recently proposed global and local calibrations for the mean annual air temperature(MAT) reconstruction on the basis of 5-and 6-methyl brGDGTs have rarely been applied to the Chinese LPS yet, leaving the applicability of these calibrations unclear. Here, we used the improved chromatography method to analyze 198 loess-paleosol samples from the Weinan section in the southern CLP for the past 350 kyr. The 6-methyl brGDGTs comprise a major proportion of total brGDGTs, pointing to alkaline conditions for most soil samples from the Weinan LPS. The decoupled profile variation of MBT′6ME and MBT′_(5ME) suggests their response to different enviromental factors, possibly soil pH(or soil moisture) and temperature, respectively. This discrimination further corraborates that temperature and monsoonal precipitation were not in phase during the last four deglaciations on the CLP. Temperature estimates for the six calibrations tested show similar trends but remarkably differ in amplitudes. The soil moisture appears to affect the global MBT′/CBT calibration, the global MATmr calibration(a calibration based on the multiple linear regression) and the Chinese local SSM(Stepwise Selection Method) calibration, resulting in a significant underestimation of late Holocene temperature. In contrast, the dry climate has no effect on the global MBT′5ME calibration. Of the six calibrations,only the Chinese local SSM calibration and global MATmr calibration produce temperature variation amplitude over the past 350 ka that is consistent with other independent proxy data. The recently proposed local calibrations based on 5-and 6-methyl brGDGTs for North China yield the amplitude of temperature changes that is much larger than the results determined by other approaches.All the six calibrations have their own weakness in the MAT reconstruction, due partly to the inconsistency between the brGDGT distribution in the Weinan LPS and the modern soils used to establish these calibrations.TANG ChangYan YANG Huan DANG XinYue XIE ShuCheng 2017Science China Earth Sciences2017,60,6:7
