1长江三角洲地区大气O3和PM10的区域污染特征模拟显示文摘以TRACE-P污染源资料及上海市地方排放清单为基础,采用Models-3/CMAQ环境空气质量模型和中尺度气象模式MM5,模拟研究了2001-01和2001-07长三角近地面二次污染物O3及PM10的浓度分布及输送状况,并以上海市国控点2001年冬、夏季各10 d的小时监测数据对模型进行了验证.验证结果显示,Models-3/CMAQ对O3和PM10的模拟结果与监测值的相关系数分别为0.77和0.52;一致性指数分别达到0.81和0.99.模型对O3小时最高浓度的估算偏低27%,标准偏差为-3.1%;对PM10小时平均浓度的估算偏低10%,标准偏差为46%.模型已具备再现和模拟长三角大气污染输送过程的能力,且误差落在可接受的范围之内.模拟结果显示,2001-07长三角区域16个主要城市中,有14个城市O3小时最大浓度超过国家二级标准,高浓度O3可覆盖苏南和浙北广大区域.2001-01泰州、扬州、南京、镇江、常州等城市受本地排放源和北部大气污染输送的影响显著,大气PM10日均浓度超过PM10国家二级标准.长三角地区环境空气质量与污染类型受大气污染传输与化学转化的影响十分明显.夏季太阳辐射较强时,南部城市排放的污染物常以二次污染物的形式影响下风向城市;太阳辐射较弱的情况下,则以一次污染物输送为主的形式影响周边地区.冬季长三角区域颗粒物污染总体水平较高,这与我国北方地区排放的颗粒物在西北风作用下向长三角输送造成的影响密切相关.长三角地区的大气污染已逐渐从局地转为区域问题.李莉 陈长虹 黄成 黄海英 李作攀 Joshua S. 2应用Models-3/CMAQ研究长三角区域大气污染及输送显示文摘选用美国国家环保局第3代空气质量模式(Models-3,/CMAQ),配合中尺度气象模式(MM5)进行研究,模拟了2001年1、7月份长江三角洲区域冬夏季典型天气条件下大气层二次污染物臭氧及颗粒物的浓度分布及输送状况。采用2001年冬.夏季各10d的小时监测数据对模式验证。结果表明,Models-3/CMAQ对O3、PM10的模拟相关系数分别为0.77和052;一致性指数分别达到081和0.99。模型对于O3略微低估,标准偏差为-31%,而对于PM10则有所高估,标准偏差为46%。对长三角7月份O3浓度及1月份PM10的模拟结果显示,7月份长三角区域16个主要城市O3日均浓度集中在0.043~0.086mg/m^3之间,其中,泰州、扬州.湖州、镇江O3月日均浓度相对较高,均超过0.064mg/m3o模拟时段内O3最高小时浓度达0.276mg/m^3。1月份整个模拟区域PM10月日均浓度为O056mg/m^3,其中,南京市PM10日均浓度最高,达0.080mg/m^3模拟时段内PM10最高小时浓度达0.432mg/m^3研究表明,长三角地区存在明显的污染物输送现象,大气污染已经从局地污染转化为区域污染。李莉 陈长虹 黄海英 黄成 李作攀 Joshua S Fu Carey J Jang David G Streets 2007上海环境科学2007,26,4:11
3A 2-kb Mycovirus Converts a Pathogenic Fungus into a Beneficial Endophyte for Brassica Protection and Yield Enhancement显示文摘Mycoviruses are viruses that infect fungi,and hypovirulence-associated mycoviruses have the potential to control fungal diseases.However,it is unclear how mycovirus-mediated hypovirulent strains live and survive in the field,and no mycovirus has been applied for field crop protection.In this study,we found that a previously identified small DNA mycovirus(SsHADV-1)can convert its host,Sclerotinia sclerotiorum,from a typical necrotrophic pathogen to a beneficial endophytic fungus.SsHADV-1 downregulates the expression of key pathogenicity factor genes in S.sclerotiorum during infection.When growing in rapeseed,the SsHADV-1-infected strain DT-8 significantly regulates the expression of rapeseed genes involved in defense,hormone signaling,and circadian rhythm pathways.As a result,plant growth is promoted and disease resistance is enhanced.Field experiments showed that spraying DT-8 at the early flowering stage can reduce the disease severity of rapeseed stem rot by 67.6%and improve yield by 14.9%.Moreover,we discovered that SsHADV-1 could also infect other S.sclerotiorum strains on DT-8-inoculated plants and that DT-8 could be recovered from dead plants.These findings suggest that the mycoviruses may have the ability to shape the origin of endophytism.Our discoveries suggest that mycoviruses may influence the origin of endophytism and may also offer a novel strategy for disease control in which mycovirus-infected strains are used to improve crop health and release mycoviruses into the field.Hongxiang Zhang Jiatao Xie Yanping Fu Jiasen Cheng Zheng Qu Zhenzhen Zhao Shufen Cheng Tao Chen Bo Li Qianqian Wang Xinqiang Liu Binnian Tian David B.Collinge Daohong Jiang 2020Molecular Plant2020,13,10:9

5Inactivation of SACE_3446, a TetR family transcriptional regulator, stimulates erythromycin production in Saccharopolyspora erythraea显示文摘Erythromycin A is a widely used antibiotic produced by Saccharopolyspora erythraea;however,its biosynthetic cluster lacks a regulatory gene,limiting the yield enhancement via regulation engineering of S.erythraea.Herein,six TetR family transcriptional regulators(TFRs)belonging to three genomic context types were individually inactivated in S.erythraea A226,and one of them,SACE_3446,was proved to play a negative role in regulating erythromycin biosynthesis.EMSA and qRT-PCR analysis revealed that SACE_3446 covering intact N-terminal DNA binding domain specifically bound to the promoter regions of erythromycin biosynthetic gene eryAI,the resistant gene ermE and the adjacent gene SACE_3447(encoding a longchain fatty-acid CoA ligase),and repressed their transcription.Furthermore,we explored the interaction relationships of SACE_3446 and previously identified TFRs(SACE_3986 and SACE_7301)associated with erythromycin production.Given demonstrated relatively independent regulation mode of SACE_3446 and SACE_3986 in erythromycin biosynthesis,we individually and concomitantly inactivated them in an industrial S.erythraea WB.Compared with WB,the WBΔ3446 and WBΔ3446Δ3986 mutants respectively displayed 36%and 65%yield enhancement of erythromycin A,following significantly elevated transcription of eryAI and ermE.When cultured in a 5 L fermentor,erythromycin A ofWBΔ3446 and WBΔ3446Δ3986 successively reached 4095 mg/L and 4670 mg/L with 23%and 41%production improvement relative to WB.The strategy reported here will be useful to improve antibiotics production in other industrial actinomycete.Hang Wu Yansheng Wang Li Yuan Yongrong Mao Weiwei Wang Lin Zhu Panpan Wu Chengzhang Fu Rolf Muller David T.Weaver Lixin Zhang Buchang Zhang 2016Synthetic and Systems Biotechnology2016,1,1:7

7Surface effects on in-shoe plantar pressure and tibial impact during running显示文摘目的:探讨在不同表面上跑步对足底压力和胫骨加速度的影响。方法:13位男性跑步者被要求分别在水泥地、塑胶跑道、天然草地、跑步机以及配备EVA缓冲衬垫物的跑步机上以12 km/h的速度进行慢跑。用足底压力测试系统和安放在胫骨粗隆的加速度计,记录和分析足底压力与胫骨冲击的特点。结果:第一和第二足底压力峰值(发生时间)、压力时间积分、峰值压力分布在混凝土、塑胶、草地和正常的跑步机的表面间没有明显差异。在5种不同测试表面下,峰值加速度正值也无显著差异。但相比于混凝土表面,在配备EVA缓冲衬垫物的跑步机上跑步能明显减小在冲击阶段的足底压力第一峰值和压力积分(p<0.05)。上述结果可进一步归因为足后跟区域峰值压力下降(P<0.05)。结论:运动表面和下肢冲击之间也许并无必然关系。然而值得注意的是,在解释足底压力的表现及将其运用到陆地表面的跑步运动中时应考虑不同跑步机表面的影响。Weijie Fu Ying Fang David Ming Shuo Liu Lin Wang Sicong Ren Yu Liu 2015Journal of Sport and Health Science2015,4,4:4
8Herpetological phylogeographic analyses support a Miocene focal point of Himalayan uplift and biological diversification显示文摘The Himalaya are among the youngest and highest mountains in the world,but the exact timing of their uplift and origins of their biodiversity are still in debate.The Himalayan region is a relatively small area but with exceptional diversity and endemism.One common hypothesis to explain the rich montane diversity is uplift-driven diversification—that orogeny creates conditions favoring rapid in situ speciation of resident lineages.We test this hypothesis in the Himalayan region using amphibians and reptiles,two environmentally sensitive vertebrate groups.In addition,analysis of diversification of the herpetofauna provides an independent source of information to test competing geological hypotheses of Himalayan orogenesis.We conclude that the origins of the Himalayan herpetofauna date to the early Paleocene,but that diversification of most groups was concentrated in the Miocene.There was an increase in both rates and modes of diversification during the early to middle Miocene,together with regional interchange(dispersal)between the Himalaya and adjacent regions.Our analyses support a recently proposed stepwise geological model of Himalayan uplift beginning in the Paleocene,with a subsequent rapid increase of uplifting during the Miocene,finally giving rise to the intensification of the modern South Asian Monsoon.Wei Xu Wen-Jie Dong Ting-Ting Fu Wei Gao Chen-Qi Lu Fang Yan Yun-He Wu Ke Jiang Jie-Qiong Jin Hong-Man Chen Ya-Ping Zhang David M.Hillis Jing Che 2021National Science Review2021,8,9:3
9HDAC inhibitors improve CRISPR-mediated HDR editing efficiency in iPSCs显示文摘Genome-edited human induced pluripotent stem cells(iPSCs)hold great promise for therapeutic applications.However,low editing efficiency has hampered the applications of CRISPR-Cas9 technology in creating knockout and homology-directed repair(HDR)-edited iPSC lines,particularly for silent genes.This is partially due to chromatin compaction,inevitably limiting Cas9 access to the target DNA.Among the six HDAC inhibitors we examined,vorinostat,or suberoylanilide hydroxamic acid(SAHA),led to the highest HDR efficiency at both open and closed loci,with acceptable toxicity.HDAC inhibitors equally increased non-homologous end joining(NHEJ)editing efficiencies(~50%)at both open and closed loci,due to the considerable HDAC inhibitor-mediated increase in Cas9 and sgRNA expression.However,we observed more substantial HDR efficiency improvement at closed loci relative to open chromatin(2.8 vs.1.7-fold change).These studies provide a new strategy for HDRediting of silent genes in iPSCs.Jian-Ping Zhang Zhi-Xue Yang Feng Zhang Ya-Wen Fu Xin-Yue Dai Wei Wen Beldon Zhang Hannah Choi Wanqiu Chen Meredith Brown David Baylink Lei Zhang Hongyu Qiu Charles Wang Tao Cheng Xiao-Bing Zhang 2021Science China(Life Sciences)2021,64,9:3
