1Effectiveness of probiotics in irritable bowel syndrome:Updated systematic review with meta-analysis显示文摘AIM:To investigate the efficacy of probiotics in irritable bowel syndrome(IBS) patients.METHODS:Pub Med,Cochrane library,Scopus,Google Scholar,and Clinicaltrial.gov databases were searched for literature published between September 2007 and December 2013.The applied Mesh terms were 'probiotics,' 'irritable bowel syndrome,' and 'irritable bowel syndrome treatment.' The collected data contained24 clinical trials,of which 15 were eligible for meta-analysis and nine were reviewed systematically.All studies were randomized placebo-controlled trials in patients with IBS that investigated the efficacy of probiotics in IBS improvement.The Jadad score was used to assess the methodological quality of trials.The quality scale ranges from 0 to 5 points,with a score ≤ 2 indicating a low quality report,and a score of ≥3 indicating a high quality report.Relative risk(RR),standardized effect size,and 95%CI were calculated using the Der Simonian-Laird method.The Cochran Q test was used to test heterogeneity with P < 0.05.Funnel plots were constructed and Egger's and BeggMazumdar tests were performed to assess publication bias.RESULTS:A total of 1793 patients were included in the meta-analysis.The RR of responders to therapies based on abdominal pain score in IBS patients for two included trials comparing probiotics to placebo was 1.96(95%CI:1.14-3.36;P = 0.01).RR of responders to therapies based on a global symptom score in IBS patients for two included trials comparing probiotics with placebo was 2.43(95%CI:1.13-5.21;P = 0.02).For adequate improvement of general symptoms in IBS patients,the RR of seven included trials(six studies) comparing probiotics with placebo was 2.14(95%CI:1.08-4.26;P = 0.03).Distension,bloating,and flatulence were evaluated using an IBS severity scoring system in three trials(two studies) to compare the effect of probiotic therapy in IBS patients with placebo,the standardized effect size of mean differences for probiotics therapy was-2.57(95%CI:-13.05--7.92).CONCLUSION:Probiotics reduce pain and symptom severity scores.The results demonstrate the beneficial effects of probiotics in IBS patients in comparison with placebo.Tina Didari Shilan Mozaffari Shekoufeh Nikfar Mohammad Abdollahi 2015World Journal of Gastroenterology2015,21,10:51

3Intestinal microbiota, probiotics and prebiotics in inflammatory bowel disease显示文摘It has been presumed that aberrant immune response to intestinal microorganisms in genetically predisposed individuals may play a major role in the pathogenesis of the inflammatory bowel disease, and there is a good deal of evidence supporting this hypothesis. Commensal enteric bacteria probably play a central role in pathogenesis, providing continuous antigenic stimulation that causes chronic intestinal injury. A strong biologic rationale supports the use of probiotics and prebiotics for inflammatory bowel disease therapy. Many probiotic strains exhibit anti-inflammatory properties through their effects on different immune cells, proinflammatory cytokine secretion depression, and the induction of anti-inflammatory cytokines. There is very strong evidence supporting the use of multispecies probiotic VSL#3 for the prevention or recurrence of postoperative pouchitis in patients. For treatment of active ulcerative colitis, as well as for maintenance therapy, the clinical evidence of efficacy is strongest for VSL#3 and Escherichia coli Nissle 1917. Moreover, some prebiotics, such as germinated barley foodstuff, Psyllium or oligofructose-enriched inulin, might provide some benefit in patients with active ulcerative colitis or ulcerativecolitis in remission. The results of clinical trials in the treatment of active Crohn's disease or the maintenance of its remission with probiotics and prebiotics are disappointing and do not support their use in this disease. 4Increased susceptibility for intrahepatic cholestasis of pregnancy and contraceptive-induced cholestasis in carriers of the 1331T>C polymorphism in the bile salt export pump显示文摘AIM: To study the association of three common ABCB11 and ABCC2 polymorphisms (ABCB11: 1331T>C V444A; ABCC2: 3563T>A V1188E and 4544G>A C1515Y) with intrahepatic cholestasis of pregnancy (ICP) and contraceptive-induced cholestasis (CIC). METHODS: ABCB11 and ABCC2 genotyping data were available from four CIC patients and from 42 and 33 ICP patients, respectively. Allele-frequencies of the studied polymorphisms were compared with those in healthy pregnant controls and Caucasian individuals. Furthermore, serum bile acid levels were correlated with the presence or absence of the 1331 C allele.RESULTS: The ABCB11 1331T>C polymorphism was significantly more frequent in cholestatic patients than in pregnant controls: C allele 76.2% (CI, 58.0-94.4) vs 51.3% (CI 35.8-66.7), respectively (P = 0.0007); and CC allele 57.1% (CI 36.0-78.3) vs 20% (CI 7.6-32.4), respectively (P = 0.0065). All four CIC patients were homozygous carriers of the C allele. In contrast, none of the studied ABCC2 polymorphism was overrepresented in ICP or CIC patients. Higher serum bile acid levels were found in carriers of the 1331CC genotype compared to carriers of the TT genotype.CONCLUSION: Our data support a role for the ABCB11 1331T>C polymorphism as a susceptibility factor for the ←←← development of estrogen-induced cholestasis, whereas no such association was found for ABCC2. 7乌苏酸联合吉西他滨对胰腺癌PANC-1细胞增殖和凋亡的影响显示文摘目的:探讨乌苏酸联合吉西他滨对胰腺癌PANC-1细胞增殖和凋亡的影响。方法:以乌苏酸、吉西他滨单独用药作用于体外培养的胰腺癌细胞PANC-1细胞,采用细MTT法检测细胞半抑制浓度(IC_(50)),确定药物给药浓度;以乌苏酸(2μmol/L)、吉西他滨(0.2μmol/L)单独及联合作用于PANC-1细胞,采用锥虫蓝染色法检测PANC-1细胞存活率,PI染色后PANC-1细胞中检测细胞凋亡,采用细胞划痕实验检测PANC-1细胞迁移和伤口愈合情况,采用Western blotting检测对照组、乌苏酸组与吉西他滨组单独及联合用药组细胞中P-JNK、Bcl-2、IL-6、P-Stat 3、NF-κB和Cox-2蛋白的表达。结果:乌苏酸和吉西他滨对胰腺癌PANC-1细胞均有较强的抑制作用,其IC_(50)分别是13.67和2.78μmol/L,据此选择它们的实验浓度分别为2和0.2μmol/L。两者联合用药比分别单独用药能更显著抑制癌细胞的增殖活性[(46.47±5.07)%vs(78.38±8.65)%、(76.12±3.23)%,均P<0.05],能更明显诱导癌细胞凋亡[(39.78±7.01)%vs(20.35±8.51)%、(20.35±8.51)%,均P<0.01]和抑制细胞迁移能力(P<0.01),同时更显著抑制凋亡相关蛋白Bcl-2、IL-6的表达及促进P-JNK的表达。结论:乌苏酸和吉西他滨协同增强抑制胰腺癌PANC-1细胞的增殖和迁移以及促凋亡作用,其机制可能与抑制Bcl-2、IL-6、P-Stat 3和促进P-JNK蛋白表达有关。江森 郑希 何燕 Tina LIAO 张焜 2018中国肿瘤生物治疗杂志2018,25,3:11

10Microscopic colitis: A review of etiology, treatment and refractory disease显示文摘Microscopic colitis is a common cause of chronic,nonbloody diarrhea. Microscopic colitis is more common in women than men and usually affects patients in their sixth and seventh decade. This article reviews the etiology and medical management of microscopic colitis. The etiology of microscopic colitis is unknown, but it is associated with autoimmune disorders, such as celiac disease, polyarthritis, and thyroid disorders. Smoking has been identified as a risk factor of mi-croscopic colitis. Exposure to medications, such as non-steroidal anti-inflammatory drugs, proton pump inhibitors, and selective serotonin reuptake inhibitors, is suspected to play a role in microscopic colitis, although their direct causal relationship has not been proven. Multiple medications, including corticosteroids, anti-diarrheals, cholestyramine, bismuth, 5-aminosalicylates, and immunomodulators, have been used to treat microscopic colitis with variable response rates. Budesonide is effective in inducing and maintaining clinical remission but relapse rate is as high as 82% when budesonide is discontinued. There is limited data on management of steroid-dependent microscopic colitis or refractory microscopic colitis. Immunomodulators seem to have low response rate 0%-56% for patients with refractory microscopic colitis. Response rate 66%-100% was observed for use of anti-tumor necrosis factor(TNF) therapy for refractory microscopic colitis. 11靶区和剂量控制下125Ⅰ粒子治疗颅内恶性肿瘤的再评价显示文摘目的再次评价靶区和剂量控制下125I粒子治疗颅内恶性肿瘤的疗效和并发症。方法回顾分析2003年11月—2016年5月青岛大学附属医院、山东省滕州市中心人民医院、山东省邹平中医院等12家医院310例颅内恶性肿瘤患者412次手术的临床资料,分析靶区和剂量控制下125I粒子治疗颅内恶性肿瘤的技术,评价疗效和并发症。对颅内恶性肿瘤进行分层研究,行靶区和剂量控制下125I粒子治疗。结果单发脑转移瘤、低级别胶质瘤、高级别胶质瘤、手术后复发、放疗后复发、其他(脑膜瘤、垂体瘤等)中位生存期分别是19、67、41、26、23、46个月;中位无进展生存时间分别是19、42、6、9、11、12个月。并发症种类多,发生率低,总并发症发生率10.4%;术后7~45 d死亡3例。结论靶区和剂量控制下125I粒子治疗颅内恶性肿瘤可以取得较好的疗效,而并发症较低,与外科切除、外放射治疗各具特点,应该进一步研究,并形成临床路径。胡效坤 彭丽静 刘士锋 张开贤 刘院生 张福君 朱旭东 巩秀珍 高阳 王德峰 Tina Kapur 李子祥 2017中华医学杂志2017,97,19:9
12LncRNAs-directed PTEN enzymatic switch governs epithelial–mesenchymal transition显示文摘Despite the structural conservation of PTEN with dual-specificity phosphatases,there have been no reports regarding the regulatory mechanisms that underlie this potential dual-phosphatase activity.Here,we report that K27-linked polyubiquitination of PTEN at lysines 66 and 80 switches its phosphoinositide/protein tyrosine phosphatase activity to protein serine/threonine phosphatase activity.Mechanistically,high glucose,TGF-β,CTGF,SHH,and IL-6 induce the expression of a long non-coding RNA,GAEA(Glucose Aroused for EMT Activation),which associates with an RNA-binding E3 ligase,MEX3C,and enhances its enzymatic activity,leading to the K27-linked polyubiquitination of PTEN.The MEX3C-catalyzed PTEN^K27-polyUb activates its protein serine/threonine phosphatase activity and inhibits its phosphatidylinositol/protein tyrosine phosphatase activity.With this altered enzymatic activity,PTEN^K27-polyUb dephosphorylates the phosphoserine/threonine residues of TWIST1,SNAI1,and YAP1,leading to accumulation of these master regulators of EMT.Animals with genetic inhibition of PTEN^K27-polyUb,by a single nucleotide mutation generated using CRISPR/Cas9(Pten^K80R/K80R),exhibit inhibition of EMT markers during mammary gland morphogenesis in pregnancy/lactation and during cutaneous wound healing processes.Our findings illustrate an unexpected paradigm in which the lncRNA-dependent switch in PTEN protein serine/threonine phosphatase activity is important for physiological homeostasis and disease development.Qingsong Hu Chunlai Li Shouyu Wang Yajuan Li Bo Wen Yanyan Zhang Ke Liang Jun Yao Youqiong Ye Heidi Hsiao Tina K.Nguyen Peter K.Park Sergey D.Egranov David H.Hawke Jeffrey R.Marks Leng Han Mien-Chie Hung Bing Zhang Chunru Lin Liuqing Yang 2019Cell Research2019,29,4:8
13Data Management Planning:How Requirements and Solutions are Beginning to Converge显示文摘Effective stewardship of data is a critical precursor to making data FAIR.The goal of this paper is to bring an overview of current state of the art of data management and data stewardship planning solutions(DMP).We begin by arguing why data management is an important vehicle supporting adoption and implementation of the FAIR principles,we describe the background,context and historical development,as well as major driving forces,being research initiatives and funders.Then we provide an overview of the current leading DMP tools in the form of a table presenting the key characteristics.Next,we elaborate on emerging common standards for DMPs,especially the topic of machine-actionable DMPs.As sound DMP is not only a precursor of FAIR data stewardship,but also an integral part of it,we discuss its positioning in the emerging FAIR tools ecosystem.Capacity building and training activities are an important ingredient in the whole effort.Although not being the primary goal of this paper,we touch also the topic of research workforce support,as tools can be just as much effective as their users are competent to use them properly.We conclude by discussing the relations of DMP to FAIR principles,as there are other important connections than just being a precursor.Sarah Jones Robert Pergl Rob Hooft Tomasz Miksa Robert Samors Judit Ungvari Rowena I.Davis Tina Lee 2020Data Intelligence2020,2,1:7

19Garcinol sensitizes human head and neck carcinoma to cisplatin in a xenograft mouse model despite downregulation of proliferative biomarkers显示文摘OBJECTIVE Platinum compounds such as cisplatin and carboplatin are frequently used as the first-line chemotherapy for the treatment of the head and neck squamous cell carcinoma(HNSCC).In the present study,we investigated whether garcinol,apolyisoprenylated benzophenone can chemosensitize HNSCC to cisplatin.METHODS The effect of garcinol and cisplatin on HNSCC was assessed by MTT,Western blotting,real time PCR,FACS,immunohistochemistry,DNA binding assay and xenograft mouse model.RESULTS We found that garcinol inhibited the viability of a panel of diverse HNSCC cell lines,enhanced the apoptotic effect of cisplatin,suppressed constitutive as well as cisplatin-induced NF-κB activation,and downregulated the expression of various oncogenic gene products(cyclin D1,Bcl-2,survivin and VEGF).In vivo study showed that administration of garcinol alone(0.5 mg·kg-1,ip five times/week)significantly suppressed the growth of the tumor,and this effect was further increased by cisplatin.Both the markers of proliferation index(Ki-67)and microvessel density(CD31)were downregulated in tumor tissues by the combination of cisplatin and garcinol.The pharmacokinetic results of garcinol indicated that good systemic exposure was achievable after ip administration of garcinol at 0.5and 2mg·kg-1 with mean peak concentration(cmax)of 1825.4 and 6635.7nmol·L-1 in the mouse serum,respectively.CONCLUSION Overall,our results suggest that garcinol can indeed potentiate the effects of cisplatin by negative regulation of various inflammatory and proliferative biomarkers.Feng Li Muthu K.
