1Brain-derived neurotrophic factors increase the proliferation and differentiation of endogenous neural stem cells in mouse models of cerebral infarction显示文摘BACKGROUND: It has been confirmed that brain-derived neurotrophic factor (BDNF) can promote the proliferation of neural stem cells (NSCs) and protect neuron-like cells in vitro. However, its effect on endogenous NSCs in vivo is still unclear. OBJECTIVE: To evaluate whether BDNF can induce the endogenous NSCs to proliferate and differentiate into the neurons in the mice model of cerebral infarction. DESIGN: A synchronal controlled observation. SETTINGS: Department of Neurology, Microbiology Division of the Department of Laboratory, Tianjin First Central Hospital; Howard Florey Institute, Medical College, the University of Melbourne. MATERIALS: Twenty-four pure breed C57BL/6J mice at the age of 10 weeks old (12 males and 12 females) were divided into saline control group and BDNF-treated group, 6 males and 6 females in each group. METHODS: The experiments were performed at the University of Melbourne from July 2004 to February 2005. ① The left middle cerebral artery (MCA) was ligated in both groups to establish models of cerebral infarction and the Matsushita measuring method was used to monitor the blood flow of the lesioned region supplied by MCA. 75% reduction of blood flow should be reached in the lesioned region. ② At 24 hours after infarction, mice in the BDNF-treated group were administrated with BDNF, which was slowly delivered using an ALZET osmium pump design. BDNF was dissolved in saline at the dosage of 500 mg/kg and injected into the pump, which could release the solution consistently in the following 28 days. The mice in the saline control group accepted the same volume of saline at 24 hours after infarction. ③ The Rotarod function test began at 1 week preoperatively, the time stayed on Rotarod was recorded. The mice were tested once a day till the end of the experiment. At 4 weeks post cerebral infarction, double labeling of Nestin and GFAP, BIII tubulin and CNPase immunostaining was performed to observe the differentiation directions of the re-expressed endogenous NSCs, and the percentages of the cells differentiated into astrocytes, neurons and oligodendrocytes were calculated. MAIN OUTCOME MEASURES: ① The differentiation directions of the re-expressed endogenous NSCs, and the percentage of the cells differentiated into astrocytes, neurons and oligodendrocytes. ② Comparison of motor function between the two groups. RESULTS: All the 24 pure C57BL/6J mice were involved in the analysis of results. ① Positively expressed endogenous NSCs appeared in the mice of both groups, and they mainly distributed around the focus of lesion, as well as the contralateral side. The expressed cells in the BDNF-treated group were obviously more than those in the saline control group. ②Activations of endogenous NSCs: At 4 weeks after infarction, re-expressions of endogenous NSCs appeared in both groups. The number of the re-expressed cells in the BDNF-treated group was about 4.2 times higher than that in the saline control group. The percentage of the cells differentiated into neurons in the BDNF-treated group was significantly higher than that in the saline control group (36%, 15%), the percentage of the cells differentiated into astrocytes was lower than that in the saline control group (54%, 77%), whereas the percentage of the cells differentiated into oligodendrocytes was similar to that in the saline control group (10%, 8%). ③ Results of motor functional test: Compared with before cerebral infarction, the mice in both groups manifested as obvious decrease in motor function at 1week after infarction, whereas the recovery of motor function in the BDNF-treated group was significantly superior to that in the saline control group at 2, 3 and 4 weeks (P < 0.01). CONCLUSION: BDNF can promote the proliferation of endogenous NSCs in the brain of mice with cerebral infarction, it can decrease the differentiation rate of astrocytes, and increase the differentiation rate of neurons. BDNF has small influence on the differentiation of endogenous NSCs into oligodendrocytes, which was not benefit for the recovery of neural axon. 2Density estimated physicochemical properties of alanine-based ionic liquid [C_7mim][Ala] and its application in selective transesterification of soybean oil显示文摘Amino acid ionic liquid (AAIL) based on alanine, 1-n-heptyl-3-methylimidazolium alanine ([C7mim][Ala]), has been prepared and characterized. Since the IL can form strong hydrogen bonds with water, trace amount of water is a problematic impurity in the IL. Using the standard addition method (SAM), the densities were measured in the temperature range from 293.15 to 343.15±0.05 K. On the basis of the experimental data of density, the surface tension , molar volume, and the molecular volume Vm, molar enthalpy of vaporization lgHm0, and the thermal expansion coefficients , for [C7mim][Ala] were estimated using semi-empirical methods. Also the interstice model has been verified with good accordance. Then the [C7mim][Ala] was taken as catalyst in the transesterification reaction with fatty acid, where soybean oil was used as raw material and methanol as reactant. The amount of catalyst, alcohol consumption, reaction time, temperature and other factors of the reaction were investigated. Using GC determination, the conversions were calculated by the internal standard method. 3Effects of leukemia inhibitory factor on endogenous neural stem cell proliferation and glycoprotein-130 expression in a mouse model of cerebral infarction显示文摘Leukemia inhibitory factor (LIF) has been shown to promote proliferation of endogenous neural stem cells. In this study, we treated mice with cerebral infarction using LIF to investigate whether the LIF receptor subunit glycoprotein (gp)130 is involved in neuroprotection. After LIF treatment, the motor function of model mice was significantly improved. Immunofluorescence histochemistry showed increased numbers of endogenous neural stem cells surrounding the infarct foci. Western blot analysis revealed that gp130 expression was significantly decreased surrounding the infarcted foci. 4Development of HEP-XXMF Series Hall Thrusters in BICE显示文摘Hall thruster is a type of electric propulsion thruster which is often regarded as a moderate specific impulse space propulsion technology and is used primarily for station keeping and orbit maintenance tasks. Magnetic field is the most important aspect in Hall thruster's design. Each time the performance improvement of Hall thrusters is accompanied with the regeneration of magnetic field design. Now, all the major space nations treat the magnetic field design as the key technology for Hall thrusters, and many Hall thrusters have emerged in recent years, for example, BPT-4000 and NASA-173 M. In China, based on magnetic focus technology, the Beijing Institute of Control Engineering(BICE) has been developing Hall thrusters ranged from hundreds of watts to 5 kilowatts, including the 1500 W HEP-100 MF and the 5 k W HEP-140 MF Hall thrusters. This paper briefly reviews the development of the HEP-XXMF series Hall thruster. 5Effects of leukemia inhibitory factor and basic fibroblast growth factor on free radicals and endogenous stem cell proliferation in a mouse model of cerebral infarction显示文摘The present study established a mouse model of cerebral infarction by middle cerebral artery occlusion,and monitored the effect of 25 μg/kg leukemia inhibitory factor and(or) basic fibroblast growth factor administration 2 hours after model establishment.Results showed that following administration,the number of endogenous neural stem cells in the infarct area significantly increased,malondialdehyde content in brain tissue homogenates significantly decreased,nitric oxide content,glutathione peroxidase and superoxide dismutase activity significantly elevated,and mouse motor function significantly improved as confirmed by the rotarod and bar grab tests.In particular,the effect of leukemia inhibitory factor in combination with basic fibroblast growth factor was the most significant.Results indicate that leukemia inhibitory factor and basic fibroblast growth factor can improve the microenvironment after cerebral infarction by altering free radical levels,improving the quantity of endogenous neural stem cells,and promoting neurological function of mice with cerebral infarction.Weihui Huang Yadan Li Yufeng Lin Xue Ye Dawei Zang 2012Neural Regeneration Research2012,7,19:2
62-9 Half-life Measurement of 94mRu44+ at CSRe显示文摘A radioactive nucleus is characterized with an intrinsic half-life. However, for a nuclear species, the half-lives inneutral atoms could be very different from that in highly charged ions. The half-lives of some highly charged ionshave been directly measured at GSI for multiple motivations[1]. In the same case, the nuclear state(i:e the isomer)may be in the range of several tens of microseconds and their half-live can be measured using isochronous massspectrometry. The J = 8+ isomeric state in 94Ru was chosen to test this method. The half-life of this isomer is71 s [2] in neutral atoms, and the excitation energy is 2.64 MeV. The internal conversion coefficient of this decayin neutral atom is 0.335. 7Basic fibroblast growth factor increases the numbe of endogenous neural stem cells and inhibits the expression of amino methyl isoxazole propionic acid receptors in amyotrophic lateral sclerosis mice显示文摘This study aimed to investigate the number of amino methyl isoxazole propionic acid (AMPA) receptors and production of endogenous neural stem cells in the SOD1 G93AG1H transgenic mouse model of amyotrophic lateral sclerosis, at postnatal day 60 following administration of basic fibroblast growth factor (FGF-2). A radioligand binding assay and immunohistochemistry were used to estimate the number of AMPA receptors and endogenous neural stem cells respectively. Results showed that the number of AMPA receptors and endogenous neural stem cells in the brain stem and sensorimotor cortex were significantly increased, while motor function was significantly decreased at postnatal days 90 and 120. After administration of FGF-2 into mice, numbers of endogenous neural stem cells increased, while expression of AMPA receptors decreased, whilst motor functions were recovered. At postnatal day 120, the number of AMPA receptors was negatively correlated with the number of endogenous neural stem cells in model mice and FGF-2-treated mice. 8Geologic Features of Wangjiatun Deep Gas Reservoirs of Volcanic Rock in Songliao Basin显示文摘Wangjiatun gas pool is located at the north part of Xujiaweizi in Songliao basin. Commercial gas flow has been found in the intermediate and acid volcanic rock of upper Jurassic-lower Cretaceous, which makes a breakthrough in deep nature gas prospecting in Songliao basin. The deep natural gas entrapment regularity is discussed in the paper by the study of deep strata, structure and reservoir. Andesite, rhyolite and little pyroclastic rock are the main reservoirs. There are two types of volcanic reservoir space assemblage in this area: the pore and fissure and the pure fissure. Changes had taken place for volcanic reservoir space during long geologic time, which was controlled by tectonic movement and geologic environment. The developed degree of reservoir space was controlled by tectonic movement, weathering and filtering, corrosion and Filling. There are three types of source-reservoir-caprock assemblage in this area: lower source- upper reservoir model, upper source-lower reservoir model and lateral change model. Mudstone in Dengluoku formation and the compacted volcanic rock of upper Jurassic-lower Cretaceous are the caprock for deep gas reservoirs. Dark mudstone of deep lacustrine facies in Shahezi formation and lower part of Dengluoku formation are the source rock of deep gas.
