3Diagnostic performance of different specimens in detecting enterovirus A71 in children with hand,foot and mouth disease显示文摘Hand,foot and mouth disease(HFMD)is a major public health problem among children in the Asia-Pacific region.The optimal specimen for HFMD virological diagnosis remains unclear.Enterovirus A71(EV-A71)neutralizing antibody titres detected in paired sera were considered the reference standard for calculating the sensitivity,specificity,positive and negative predictive value of throat swabs,rectal swabs,stool,blood samples and cerebrospinal fluid(CSF)by RT-PCR or ELISA assay.In this study,clinical samples from 276 HFMD patients were collected for analysing the sensitivity of different kind of specimens.Our results showed that stool had the highest sensitivity(88%,95%CI:74%–96%)and agreement with the reference standard(91%).The order of diagnostic yield for EV-A71 infection was stool samplerectal swab>throat swab>blood sample>CSF sample,and using a combination of clinical samples improved sensitivity for enterovirus detection.The sensitivity of ELISA for IgM antibody detection in sterile-site specimens was significantly higher than that of RT-PCR(serum/plasma:62%vs.2%,CSF:47%vs.0%)(P<0.002).In conclusion,our results suggest that stool has the highest diagnostic yield for EV-A71-infected HFMD.If stool is unavailable,rectal swabs can be collected to achieve a similar diagnostic yield.Otherwise,throat swabs may be useful in detecting positive samples.Although IgM in blood or CSF is diagnostically accurate,it lacks sensitivity,missing 40%–50%of cases.The higher proportion of severe cases and shorter interval between onset and sampling contributed to the increase in congruency between clinical testing and the serological reference standard.Yonghong Zhou Chongchen Zhou Kai Wang Qi Qiu Yibing Cheng Yu Li Peng Cui Lu Liang Peng Li Xiaowei Deng Lili Wang Wen Zheng Hui Gong Fang Wang Meng Xu Justin Jang Hann Chu Lance Turtle Hongjie Yu 2023Virologica Sinica2023,38,2:1

6Combination of Click Chemistry and Enzymatic Ligation for Stable and Efficient Protein Immobilization for Single-Molecule Force Spectroscopy显示文摘Protein immobilization is an essential method for both basic and applied research for protein,and covalent,site-specific attachment is the most desirable strategy.Classic methods typically rely on a heterobifunctional cross-linker,such as N-hydroxysuccinimide(NHS)-linker-maleimide,or a similar two-step process.It utilizes the amino-reactive NHS and the thiolreactive maleimide to conjugate protein to the solid support.However,NHS as a chemical is susceptible to hydrolysis during storage and handling,and maleimide reacts nonspecifically with all cysteines available in the protein,leading to an inconsistent result.To solve these problems,we have developed a method by combining a strain-promoted azide–alkyne cycloaddition(SPAAC)click reaction and an OaAEP1(C247A)-based enzymatic ligation.The method was demonstrated by the successful immobilization of enhanced green fluorescent protein(eGFP),which was visualized by fluorescent imaging.Moreover,the correct folding and stability of the immobilized protein were verified by atomic force microscopy-based single-molecule force spectroscopy(AFM-SMFS)measurement with a high success rate(89%).Finally,the strength of the 1,2,3-triazole linkage from the azide-dibenzocyclooctyne(DBCO)-based SPAAC reaction was quantified with an ultrahigh rupture force>1.7 nN.Thus,this stable,efficient,and site-specific immobilization method can be used for many challenging systems,especially SMFS studies.Shengchao Shi Ziyi Wang Yibing Deng Fang Tian Qingsong Wu Peng Zheng 2022CCS Chemistry2022,4,2:0
7Modeling for critical state line of granular soil with evolution of grain size distribution due to particle breakage显示文摘Determination of the critical state line(CSL)is important to characterize engineering properties of granular soils.Grain size distribution(GSD)has a significant influence on the location of CSL.The influence of particle breakage on the CSL is mainly attributed to the change in GSD due to particle breakage.However,GSD has not been properly considered in modeling the CSL with influence of particle breakage.This study aims to propose a quantitative model to determine the CSL considering the effect of GSD.We hypothesize that the change of critical state void ratio with respect to GSD is caused by the same mechanism that influences of the change of minimum void ratio with respect to GSD.Consequently,the particle packing model for minimum void ratio proposed by Chang et al.(2017)is extended to predict critical state void ratio.The developed model is validated by experimental results of CSLs for several types of granular materials.Then the evolution of GSD due to particle breakage is incorporated into the model.The model is further evaluated using the experimental results on rockfill material,which illustrates the applicability of the model in predicting CSL for granular material with particle breakage.Ching S.Chang Yibing Deng 2020Geoscience Frontiers2020,11,2:0

9Detection of weak non-covalent cation-π interactions in NGAL by single-molecule force spectroscopy显示文摘Cation-π interaction is an electrostatic interaction between a cation and an electron-rich arene.It plays an essential role in many biological systems as a vital driving force for protein folding,stability,and receptor-ligand interaction/recognition.To date,the discovery of most cation-π interactions in proteins relies on the statistical analyses of available three-dimensional(3D)protein structures and corresponding computational calculations.However,their experimental verification and quantification remain sparse at the molecular level,mainly due to the limited methods to dynamically measure such a weak non-covalent interaction in proteins.Here,we use atomic force microscopy-based single-molecule force spectroscopy(AFM-SMFS)to measure the stability of protein neutrophil gelatinase-associated lipocalin(also known as NGAL,siderocalin,lipocalin 2)that can bind iron through the cation-π interactions between its three cationic residues and the iron-binding tri-catechols.Based on a site-specific cysteine engineering and anchoring method,we first characterized the stability and unfolding pathways of apo-NGAL.Then,the same NGAL but bound with the iron-catechol complexes through the cation-π interactions as a holo-form was characterized.AFM measurements demonstrated stronger stabilities and kinetics of the holo-NGAL from two pulling sites,F122 and F133.Here,NGAL is stretched from the designed cysteine close to the cationic residues for a maximum unfolding effect.Thus,our work demonstrates high-precision detection of the weak cation-π interaction in NGAL.Jingyuan Nie Yibing Deng Fang Tian Shengchao Shi Peng Zheng 2022Nano Research2022,15,5:0
10Highly Dynamic Polynuclear Metal Cluster Revealed in a Single Metallothionein Molecule显示文摘Human metallothionein(MT)is a small-size yet efficient metal-binding protein,playing an essential role in metal homeostasis and heavy metal detoxification.MT contains two domains,each forming a polynuclear metal cluster with an exquisite hexatomic ring structure.The apoprotein is intrinsically disordered,which may strongly influence the clusters and the metal-thiolate(M-S)bonds,leading to a highly dynamic structure.However,these features are challenging to identify due to the transient nature of these species.The individual signal from dynamic conformations with different states of the cluster and M-S bond will be averaged and blurred in classic ensemble measurement.To circumvent these problems,we combined a single-molecule approach and multiscale molecular simulations to investigate the rupture mechanism and chemical stability of the metal cluster by a single MT molecule,focusing on the Zn4S11 cluster in theαdomain upon unfolding.Unusual multiple unfolding pathways and intermediates are observed for both domains,corresponding to different combinations of M-S bond rupture.None of the pathways is clearly preferred suggesting that unfolding proceeds from the distribution of protein conformational substates with similar M-S bond strengths.Simulations indicate that the metal cluster may rearrange,forming and breaking metal-thiolate bonds even when MT is folded independently of large protein backbone reconfiguration.Thus,a highly dynamic polynuclear metal cluster with multiple conformational states is revealed in MT,responsible for the binding promiscuity and diverse cellular functions of this metal-carrier protein.Guodong Yuan Felipe Curtolo Yibing Deng Tao Wu Fang Tian Qun Ma Yutong Liu Jinglin Zuo Guilherme Menegon Arantes Peng Zheng 2021Research2021,,1:0
