| 1 | Meteorological control on CO_2 flux above broad-leaved Korean pine mixed forest in Changbai Mountains显示文摘The impacts of temperature, photosynthetic active radiation (PAR) and vapor pressure deficit (VPD) on CO2 flux above broad-leaved Korean pine mixed forest in the Changbai Mountains were studied based on eddy covariance and meteorological factors measurements.The results showed that, daytime CO2 flux was mainly controlled by PAR and they fit Michaelis-Menten equation. Meanwhile VPD also had an influence on the daytime flux. Drier air reduced the CO2 assimilation of the ecosystem, the drier the air, the more the reduction of the assimilation. And the forest was more sensitive to VPD in June than that in July and August. The respiration of the ecosystem was mainly controlled by soil temperature and they fit exponential equation. It was found that this relationship was also correlated with seasons; respiration from April to July was higher than that from August to November under the same temperature. Daily net carbon exchange of the ecosystem and the daily mean air temperature fit exponential equation. It was also found that seasonal trend of net carbon exchange was the result of comprehensive impacts of temperature and PAR and so on. These resulted in the biggest CO2 uptake in June and those in July and August were next. | 2 | Seasonal patterns and environmental control of ecosystem respiration in subtropical and temperate forests in China显示文摘Continuous measurement of carbon dioxide exchange using the eddy covariance (EC) technique was made at two ChinaFLUX forest sites including the young subtropical Pinus plantation (Qianyanzhou) and old temperate broad-leaved Korean pine mixed forest (Changbai Mountains) as part of the ChinaFLUX network. Seasonal patterns and environmental control of ecosystem respiration in the subtropical and temperate forests were evaluated by the often-used multiplicative model and Q10 model as a function of temperature and soil water content. The resuits suggested that ( i ) temperature was found to be a dominant factor in the ecosystem respiration, and most of the temporal variability of ecosystem respiration was explained by temperature. However, in the drought-stressed ecosystem, soil water content controlled the temporal variability of ecosystem respiration other than temperature effects, and soil water content became a dominat factor when severe drought affected the ecosystem respiration; (ii) the regression models analysis revealed that in the drier soil, ecosystem respiration was more sensitive to soil moisture than was expressed by the often-used multiplicative model. | 3 | Eddy flux corrections for CO_2 exchange in broad-leaved Korean pine mixed forest of Changbai Mountains显示文摘Based on analysis of mechanisms causing energy no-closure and nocturnal low fluxes issues for CO2 exchange studies by eddy covariance method, corrections were done with the raw data sets obtained from Changbai Mountains forest flux site, to evaluate the impacts of sonic anemometer tilt, frequency response limitations and advection on estimation of CO2 exchange, respectively. The results show that the planar fit coordinate transforming method is superior to the streamline coordinate transforming method in tilt correction. The latter could cause a systematical underestimation of eddy fluxes relating with the angle of sensor and terrain tilt. The underestimation of CO2 and energy fluxes for frequency response limitations average 3.0% and 2.0% during daytime, respectively, which increase by 9.0% and 5.5% during nighttime, respectively. The corrections of frequency response limitations are closely related to atmospheric stability. The advection loss of CO2 fluxes is dominated by nocturnal vertical advection, which is at least 18% when the horizontal advection is neglected. | 4 | Photosynthesis-transpiration coupling model at canopy scale in terrestrial ecosystem显示文摘At the hypothesis of big leaf, an ecosystem photosynthesis-transpiration coupling cycle model was established by the scaled SMPT-SB model from single leaf to canopy, and model parameterization methods were discussed. Through simulating the canopy light distribution, canopy internal conductance to CO2 can be scaled from single leaf to canopy by integrating to canopy using the relationship between single internal conductance and photosynthetic photon flux density. Using the data observed by eddy covariance method from the Changbai Mountains site of ChinaFLUX, the application of the model at the canopy scale was examined. Under no water stress, the simulated net ecosystem photosynthesis rate fitted with the observed data very well, the slope and R2 of the line regression equation of the observed and simulated values were 0.7977 and 0.8892, respectively (n = 752), and average absolute error was 3.78 μmol CO2 m-2s-1; the slope, R2 and average absolute error of transpiration rate were 0.7314, 0.4355 and 1.60mmol H2O m-2 s-1, respectively (n = 752). The relationship between canopy photosynthesis,transpiration and external environmental conditions was discussed by treating the canopy as a whole and neglecting the comprehensive feedback mechanism within canopy, and it was noted that the precipitation course affected the transpiration rate simulation badly. Compared to the models based on eco-physiological processes, the SMPT-SB model was simple and easy to be used. | 5 | The Effects of Groundwater Depth on the Soil Evaporation in Horqin Sandy Land, China显示文摘The interactions between groundwater depth and soil hydrological processes, play an important role in both arid and semi-arid ecosystems. The effect of groundwater depth on soil water variations were neglected or not explicitly treated. In this paper, we combine a simulation experiment and a water flow module of HYDRUS-1D model to study the variation in soil evaporation under different groundwater depth conditions and the relationship between groundwater depth and evaporation efficiency in Horqin Sandy Land, China.The results showed that with an increase in groundwater depth, the evaporation of soil and the recharge of groundwater decrease. In this study, the groundwater recharge did not account for more than 21% of the soil evaporation for the depths of groundwater examined. The soil water content at 60 cm was less affected by the evaporation efficiency when the mean groundwater depth was 61 cm during the experimental period. In addition, the evaporation efficiency(the ratio of actual evaporation to potential evaporation) decreases with the increase in groundwater depth during the experiment. | 6 | Nasal delivery of broadly neutralizing antibodies protects mice from lethal challenge with SARS-CoV-2 delta and omicron variants显示文摘Multiple new variants of severe acute respiratory syndrome coronavirus 2(SARS-Co V-2)have constantly emerged,as the delta and omicron variants,which have developed resistance to currently gained neutralizing antibodies.This highlights a critical need to discover new therapeutic agents to overcome the variants mutations.Despite the availability of vaccines against coronavirus disease 2019(COVID-19),the use of broadly neutralizing antibodies has been considered as an alternative way for the prevention or treatment of SARS-Co V-2 variants infection.Here,we show that the nasal delivery of two previously characterized broadly neutralizing antibodies(F61 and H121)protected K18-h ACE2 mice against lethal challenge with SARS-Co V-2 variants.The broadly protective efficacy of the F61 or F61/F121 cocktail antibodies was evaluated by lethal challenge with the wild strain(WIV04)and multiple variants,including beta(B.1.351),delta(B.1.617.2),and omicron(B.1.1.529)at 200or 1000 TCID_(50),and the minimum antibody administration doses(5-1.25 mg/kg body weight)were also evaluated with delta and omicron challenge.Fully prophylactic protections were found in all challenged groups with both F61 and F61/H121 combination at the administration dose of 20 mg/kg body weight,and corresponding mice lung viral RNA showed negative,with almost all alveolar septa and cavities remaining normal.Furthermore,low-dose antibody treatment induced significant prophylactic protection against lethal challenge with delta and omicron variants,whereas the F61/H121 combination showed excellent results against omicron infection.Our findings indicated the potential use of broadly neutralizing monoclonal antibodies as prophylactic and therapeutic agent for protection of current emerged SARS-Co V-2 variants infection. | Jia Lu Qiangling Yin Rongjuan Pei Qiu Zhang Yuanyuan Qu Yongbing Pan Lina Sun Ding Gao Cuiqin Liang Jingwen Yang Wei Wu Jiandong Li Zongqiang Cui Zejun Wang Xinguo Li Dexin Li Shiwen Wang Kai Duan Wuxiang Guan Mifang Liang Xiaoming Yang | 2022 | Virologica Sinica2022,37,2: | 4 |
| 7 | Antibody Cocktail Exhibits Broad Neutralization Activity Against SARS-CoV-2 and SARS-CoV-2 Variants显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-Co V-2)has precipitated multiple variants resistant to therapeutic antibodies.In this study,12 high-affinity antibodies were generated from convalescent donors in early outbreaks using immune antibody phage display libraries.Of them,two RBD-binding antibodies(F61 and H121)showed high-affinity neutralization against SARS-Co V-2,whereas three S2-target antibodies failed to neutralize SARS-Co V-2.Following structure analysis,F61 identified a linear epitope located in residues G446–S494,which overlapped with angiotensinconverting enzyme 2(ACE2)binding sites,while H121 recognized a conformational epitope located on the side face of RBD,outside from ACE2 binding domain.Hence the cocktail of the two antibodies achieved better performance of neutralization to SARS-Co V-2.Importantly,these two antibodies also showed efficient neutralizing activities to the variants including B.1.1.7 and B.1.351,and reacted with mutations of N501 Y,E484 K,and L452 R,indicated that it may also neutralize the recent India endemic strain B.1.617.The unchanged binding activity of F61 and H121 to RBD with multiple mutations revealed a broad neutralizing activity against variants,which mitigated the risk of viral escape.Our findings revealed the therapeutic basis of cocktail antibodies against constantly emerging SARS-Co V-2 variants and provided promising candidate antibodies to clinical treatment of COVID-19 patients infected with broad SARS-Co V-2 variants. | Yuanyuan Qu Xueyan Zhang Meiyu Wang Lina Sun Yongzhong Jiang Cheng Li Wei Wu Zhen Chen Qiangling Yin Xiaolin Jiang Yang Liu Chuan Li Jiandong Li Tianlei Ying Dexin Li Faxian Zhan Youchun Wang Wuxiang Guan Shiwen Wang Mifang Liang | 2021 | Virologica Sinica2021,36,5: | 3 |

| 12 | Photosynthate supply drives soil respiration of Fraxinus mandshurica seedlings in northeastern China: evidences from a shading and nitrogen addition experiment显示文摘Improved understanding of the link between photosynthesis and below-ground processes is needed to better understand ecosystem carbon(C) cycling and its feedback to climate change. We conducted a short-term shading and nitrogen(N) addition experiment from June to September 2013 to investigate the effect of photosynthate supply by Manchurian Ash(Fraxinus mandshurica) seedlings on soil respiration(SR). Shading significantly reduced SR in early and middle growing season, but not in late growing season, leading to a decrease in mean SR by 24 % in N-unfertilized treatments. N addition increased mean SR by 42 % in un-shaded treatment. The stimulation of SR was largely attributed to accelerated autotrophic respiration by increasing photosynthesis, leaf area index and belowground biomass. Shading reduced mean SR by 32 % in N addition treatment. The strengthened shading effect on SR resulted from N addition was because of more photosynthates supply at low soil temperature. The results showed that the nitrogen application rates at 45 kg·hm-2 with the ratio of 75% NO3-N to 25% NH4-N resulted in the highest potassium and reducing sugar contents in the flue-cured tobacco leaving with the highest quality grade and value.
