1Sodium oligomannate therapeutically remodels gut microbiota and suppresses gut bacterial amino acids-shaped neuroinflammation to inhibit Alzheimer's disease progression显示文摘Recently,increasing evidence has suggested the association between gut dysbiosis and Alzheimer's disease(AD)progression,yet the role of gut microbiota in AD pathogenesis remains obscure.Herein,we provide a potential mechanistic link between gut microbiota dysbiosis and neuroinflammation in AD progression.Using AD mouse models,we discovered that,during AD progression,the alteration of gut microbiota composition leads to the peripheral accumulation of phenylalanine and isoleucine,which stimulates the differentiation and proliferation of pro-inflammatory T helper 1(Thl)cells.The brain-infiltrated peripheral Th1 immune cells are associated with the Ml microglia aaivation,contributing to AD-associated neuroinflammation.Importantly,the elevation of phenylalanine and isoleucine concentrations and the increase of Th1 cell frequency in the blood were also observed in two small independent cohorts of patients with mild cognitive impairment(MCI)due to AD.Furthermore,GV-971,a sodium oligomannate that has demonstrated solid and consistent cognition improvement in a phase 3 clinical trial in China,suppresses gut dysbiosis and the associated phenylalanine/isoleucine accumulation,harnesses neuroinflammation and reverses the cognition impairment.Together,our findings highlight the role of gut dysbiosis-promoted neuroinflammation in AD progression and suggest a novel strategy for AD therapy by remodelling the gut microbiota.Xinyi Wang Guangqiang Sun Teng Feng Jing Zhang Xun Huang Tao Wang Zuoquan Xie Xingkun Chu Jun Yang Huan Wang Shuaishuai Chang Yanxue Gong Lingfei Ruan Guanqun Zhang Siyuan Yan Wen Lian Chen Du Dabing Yang Qingli Zhang Feifei Lin Jia Liu Haiyan Zhang Changrong Ge Shifu Xiao Jian Ding Meiyu Geng 2019Cell Research2019,29,10:192
2A mouse model of severe acute pancreatitis induced with caerulein and lipopolysaccharide显示文摘AIM: To establish a non-traumatic, easy to induce and reproducible mouse model of severe acute pancreatitis (SAP)induced with caerulein and lipopolyasccharide (LPS).METHODS: Thirty-two healthy mature NIH female mice were selected and divided at random into four groups (each of 8 mice), i.e., the control group (NS group), the caerulein group (Ch group), the lipopolysaccharide group (LPS group),and the caerulein+LPS group (Cn+LPS group). Mice were injected intraperitoneally with caerulein only, or LPS only,and caerulein and LPS in combination. All the animals were then killed by neck dislocation three hours after the last intraperitoneal injection. The pancreas and exo-pancreatic organs were then carefully removed for microscopic examination. And the pancreatic acinus was further observed under transmission electron microscope (TEM). Pancreatic weight, serum amylase, serum nitric oxide (NO)concentration, superoxide dismutase (SOD) and malondialdehyde (MDA) concentration of the pancreas were assayed respectively.RESULTS: (1) NS animals displayed normal pancreatic structure both in the exocrine and endocrine. In the LPS group, the pancreas was slightly edematous, with the infiltration of a few inflammatory cells and the necrosis of the adjacent fat tissues. All the animals of the Cn group showed distinct signs of a mild edematous pancreatitis characterized by interstitial edema, infiltration of neutrophil and mononuclear cells, but without obvious parenchyma necrosis and hemorrhage. In contrast, the Cn+LPS groupshowed more diffuse focal areas of nonviable pancreatic and hemorrhage as well as systemic organ dysfunction.According to Schmidt's criteria, the pancreatic histologic score showed that there existed significant difference in the Cn+LPS group in the interstitial edema, inflammatory infiltration,parenchyma necrosis and parenchyma homorrhage in comparison with those of the Cn group, LPS group and NS group (P<0.01 or P<0.05). (2) The ultrasturcture of acinar cells was seriously damaged in the Cn+LPS group. Chromatin margination of nuclei was present, the number and volume of vacuoles greatly increased. Zymogen granules (ZGs) were greatly decreased in number and endoplasmic reticulum exhibited whorls. The swollen mitochondria appeared, the crista of which was decreased in number or disappeared.(3) Pancreatic weight and serum amylase levels in the Cn +LPS was significantly higher than those of the NS group and the LPS group respectively (P<0.01 or P<0.05).However, the pancreatic wet weight and serum amylase concentration showed no significant difference between the Cn+LPS group and the Cn group. 3Upper mantle anisotropy in the Ordos Block and its margins显示文摘Based on the polarization analysis of teleseismic data,SKS (SKKS) fast-wave directions and delay times between fast and slow shear waves were determined for each of the 111 seismic stations from both permanent and temporary broadband seismograph networks deployed in the Ordos Block and its margins.Both the Silver and Chan and stacking analysis methods were used.In this way,an image of upper mantle anisotropy in the Ordos Block and its margins was acquired.In the western and northern margins of the Ordos Block,the fast-wave directions are consistently NW-SE.The fast-wave directions are mainly NWW-SEE and EW in the southern margin of the Ordos Block.In the eastern margin of the Ordos Block,the fast-wave directions are generally EW,although some run NEE-SWW or NWW-SEE.In the Ordos Block,the fast-wave directions trend near N-S in the north,but switch to near EW in the south.The delay time between fast and slow waves falls into the interval 0.48-1.50 s,and the average delay time at the stations in the Ordos Block is less than that in its margins.We suggest that the anisotropy of the stable Ordos Block is mainly caused by 'fossil' anisotropy frozen in the ancient North China Craton.The NE-trending push of the northeastern margin of the Tibetan Plateau has caused NW-SE-trending lithospheric extension in the western and northern margins of the Ordos Block,and made the upper mantle flow southeastwards.This in turn has resulted in the alignment of the upper mantle peridotite lattice with the direction of material deformation.In the southern margin of the Ordos Block,the collision between the North China and Yangtze blocks resulted in the fast-wave direction running parallel to the collision boundary and the Qinling Orogen.Combining this with the APM and velocity structure of the Qinling Orogen,we propose that eastward-directed asthenospheric-mantle channel flow may have occurred beneath the Qinling Orogen.In the eastern margin of the Ordos Block,the complex anisotropic characteristics of the Fenhe Graben and Taihang Orogen may be caused by the interaction of western Pacific Plate subduction,regional extensional tectonics,and the orogeny.For station YCI,the apparent splitting parameters (the fast-wave directions range from 45° to 106° and the delay times range from 0.6 to 1.5 s) exhibit systematic variations as a function of incoming polarization with a periodicity of π/2.This variation can be best explained by a two-layer anisotropic model (φlower=132°,δtlower=0.8 s,φupper=83°,δtupper=0.5 s).The upper layer anisotropy beneath station YCI can again be attributed to 'fossil' anisotropy frozen in the ancient North China Craton.The lower layer anisotropy is affected by the tectonic activity of the western Ordos Block.The NW-SE trending extension caused by the NE trending push of the northeastern margin of the Tibetan Plateau affected the deformation of the lower anisotropic layer beneath station YCI.By comparing the fast-wave directions with GPS velocity directions,we see that the crust and upper mantle possibly have vertically coherent deformation in the margins of the Ordos Block,whereas the internal deformation characteristics of the Ordos Block are complex and require further study.CHANG LiJun WANG ChunYong DING ZhiFeng 2011Science China Earth Sciences2011,54,6:25
4Seismic anisotropy of upper mantle in eastern China显示文摘Based on the polarization analysis of teleseismic SKS waveform data recorded at 65 seismic stations which respectively involved in the permanent and temporary broadband seismograph networks deployed in eastern China, the SKS fast-wave direction and the delay time between the fast and slow shear waves at each station were determined by use of SC method and the stacking analysis method, and then the image of upper mantle anisotropy in eastern China was acquired. In the study region, from south to north, the fast-wave polarization directions are basically EW in South China, gradually clockwise rotate to NWW-SEE in North China, then to NW-SE in Northeast China. The delay time falls into the interval [0.41 s, 1.52 s]. Anisotropic characteristics in eastern China indicate that the upper mantle anisotropy is possibly caused by both the collision between the Indian and Eurasian Plates and the subduction from the Pacific and Philippine Sea Plates to the Eurasian Plate. The collision between two plates made the crust of western China thickening and uplifting and the material eastwards extruding, and then caused the upper mantle flow eastwards and southeastwards. The subduction of Pacific Plate and Philippine Sea Plate has resulted in the lithosphere and the asthenosphere deformation in eastern China, and made the alignment of upper mantle peridotite lattice parallel to the deformation direction. 5Comparison of Undernutrition Prevalence of Children under 5 Years in China between 2002 and 2013显示文摘Objective To describe the undernutrition status of children under 5-year in China, and study the trend between 2002 and 2013. Methods The study was based on two national surveys. Undernutrition was determined against WHO's 2006 growth standards. The prevalence in 2013 and 2002 was weighted by China sixth National Population Census(2010). The relationship between undernutrition and gender/age groups/different areas use weighted logistic regression. Results The results indicated the overall prevalence of stunting, underweight, and wasting of Chinese children under 5-year was 8.1%, 2.4%, and 1.9% in 2013, respectively. The prevalence of stunting was higher for children aged 12-47 month, while underweight was higher for children aged 48-59 month. The prevalence of undernutrition was higher in rural areas than in urban areas, especially in poor rural areas. There was a decline of stunting, underweight, and wasting between 2002 and 2013 among the children, with greater reduction in rural areas than in urban areas. Conclusion The prevalence of undernutrition of children under 5-year remains high in rural areas especially in poor rural areas in China. 6Upper mantle anisotropy and crust-mantle deformation pattern beneath the Chinese mainland显示文摘Over the past 10 years,the number of broadband seismic stations in China has increased significantly.The broadband seismic records contain information about shear-wave splitting which plays an important role in revealing the upper mantle anisotropy in the Chinese mainland.Based on teleseismic SKS and SKKS phases recorded in the seismic stations,we used the analytical method of minimum transverse energy to determine the fast wave polarization direction and delay time of shear-wave splitting.We also collected results of shear-wave splitting in China and the surrounding regions from previously published papers.From the combined dataset we formed a shear-wave splitting dataset containing 1020 parameter pairs.These splitting parameters reveal the complexity of the upper mantle anisotropy image.Our statistical analysis indicates stronger upper mantle anisotropy in the Chinese mainland,with an average shear-wave time delay of 0.95 s;the anisotropy in the western region is slightly larger(1.01 s)than in the eastern region(0.92 s).On a larger scale,the SKS splitting and surface deformation data in the Tibetan Plateau and the Tianshan region jointly support the lithospheric deformation mode,i.e.the crust-lithospheric mantle coherent deformation.In eastern China,the average fast-wave direction is approximately parallel to the direction of the absolute plate motion;thus,the upper mantle anisotropy can be attributed to the asthenospheric flow.The area from the Ordos block to the Sichuan Basin in central China is the transition zone of deformation modes between the east and the west regions,where the anisotropy images are more complicated,exhibiting'fossil'anisotropy and/or two-layer anisotropy.The collision between the Indian Plate and the Eurasian Plate is the main factor of upper mantle anisotropy in the western region of the Chinese mainland,while the upper mantle anisotropy in the eastern region is related to the subduction of the Pacific Plate and the Philippine Sea Plate beneath the Eurasian Plate.WANG ChunYong CHANG LiJun DING ZhiFeng LIU QiongLin LIAO WuLin Lucy M FLESCH 2014Science China Earth Sciences2014,57,1:18
7High Potassium Aggravates the Oxidative Stress Inducedy by Magnesium Deficiency in Rice Leaves显示文摘Magnesium(Mg) deficiency in plant affects photosynthesis and many other metabolic processes.Rice(Oryza sativa L.cv.'Wuyunjing 7') plants were grown in hydroponics culture at three Mg and two potassium(K) levels under greenhouse conditions to examine the induction of oxidative stress and consequent antioxidant responses in rice leaves due to Mg deficiency.At low Mg(0.2 mmol L-1 Mg supply for two weeks after transplanting) and high K(6 mmol L-1) for 21 days,the rice plants showed severe Mg deficiency and a significant decreases in the dry matter production.The Mg deficiency in leaves decreased chlorophyll concentrations,photosynthetic activity,and soluble protein,but significantly increased the concentrations of soluble sugars and malondialdehyde(MDA) and the activities of superoxide dismutase(SOD,EC 1.15.1.1),catalase(CAT,EC 1.11.1.6) and peroxidase(POD,EC 1.11.1.7).In addition,Mg concentrations in the leaves and in the shoot biomass were negatively related to the activities of the three antioxidative enzymes and the concentration of MDA in leaves.There were very significant interactive effects between Mg and K supplied in the culture solution on shoot biomass yield,chlorophyll content,photosynthesis rate,the activities of SOD,CAT and POD,and MDA content in the leaves of rice.It is suggested that the high K level in the nutrient solution aggravated the effect of low Mg supply-induced Mg deficiency and created the oxidative damage in rice plants.DING Yu-Chuan CHANG Chun-Rong LUO Wen WU Yan-Shou REN Xiao-Li WANG Ping XU Guo-Hua 2008Pedosphere2008,18,3:17
8Seismic anisotropy of upper mantle in Sichuan and adjacent regions显示文摘Based on the polarization analysis of teleseismic SKS waveform data recorded at 94 broadband seis-mic stations in Sichuan and adjacent regions, the SKS fast-wave direction and the delay time between the fast and slow shear waves were determined at each station using the grid searching method of minimum transverse energy and the stacking analysis method, and the image of upper mantle anisot-ropy was acquired. The fast-wave polarization directions are mainly NW-SE in the study area, NWW-SEE to its northeast and NS to its west. The delay time falls into the interval [0.47 s, 1.68 s]. The spatial variation of the fast-wave directions is similar to the variation of GPS velocity directions. The anisotropic image indicates that the regional tectonic stress field has resulted in deformation and flow of upper mantle material, and made the alignment of upper mantle peridotite lattice parallel to the di-rection of material deformation. The crust-upper mantle deformation in Sichuan and adjacent regions accords with the mode of vertically coherent deformation. In the eastern Tibetan Plateau, the crustal material was extruded to east or southeast due to SE traction force of the upper mantle material. The extrusion might be obstructed by a rigid block under the Sichuan Basin and the crust has been de-formed. 9Upper mantle anisotropy of the eastern Himalayan syntaxis and surrounding regions from shear wave splitting analysis显示文摘Polarization analysis of teleseismic data has been used to determine the XKS(SKS,SKKS,and PKS)fast polarization directions and delay times between fast and slow shear waves for 59 seismic stations of both temporary and permanent broadband seismograph networks deployed in the eastern Himalayan syntaxis(EHS)and surrounding regions.The analysis employed both the grid searching method of the minimum tangential energy and stacking analysis methods to develop an image of upper mantle anisotropy in the EHS and surrounding regions using the newly obtained shear wave splitting parameters and previously published results.The fast polarization directions are oriented along a NE-SW azimuth in the EHS.However,within the surrounding regions,the fast directions show a clockwise rotation pattern around the EHS from NE-SW,to E-W,to NW-SE,and then to N-S.In the EHS and surrounding regions,the fast directions of seismic anisotropy determined using shear wave splitting analysis correlate with surficial geological features including major sutures and faults and with the surface deformation fields derived from global positioning system(GPS)data.The coincidence between structural features in the crust,surface deformation fields and mantle anisotropy suggests that the deformation in the crust and lithospheric mantle is mechanically coupled.In the EHS,the coherence between the fast directions and the NE direction of the subduction of the Indian Plate beneath the Tibetan Plateau suggests that the lithospheric deformation is caused mainly by subduction.In the regions surrounding the EHS,we speculate that a westward retreat of the Burma slab could contribute to the curved anisotropy pattern.The Tibetan Plateau is acted upon by a NE-trending force due to the subduction of the Indian Plate,and also affected by a westward drag force due to the westward retreat produced by the eastward subduction of the Burma slab.The two forces contribute to a curved lithospheric deformation that results in the alignment of the upper mantle peridotite lattice parallel to the deformation direction,and thus generates a curved pattern of fast directions around the EHS.CHANG LiJun WANG ChunYong DING ZhiFeng YOU HuiChuan LOU Hai SHAO CuiRu 2015Science China Earth Sciences2015,58,10:16

11Effect of Saccharomyces cerevisiae on alfalfa nutrient degradation characteristics and rumen microbial populations of steers fed diets with different concentrate-to-forage ratios显示文摘Live yeast(Saccharomyces cerevisiae) constitutes an effective additive for animal production;its probiotic effect may be related to the concentrate-to-forage ratio(CTFR).The objective of this study was to assess the effects of S.cerevisiae(SC) on fiber degradation and rumen microbial populations in steers fed diets with different levels of dietary concentrate.Ten Simmental × Local crossbred steers(450 ± 50 kg BW) were assigned to a control group or an SC group.Both groups were fed the same basal diet but the SC group received SC supplementation(8 × 10~9 cfu/h/d through the ruminal fistula)following a two-period crossover design.Each period consisted of four phases,each of which lasted 17 d:10 d for dietary adaptation,6 d for degradation study,and 1 d for rumen sample collection.From the 1^(st) to the 4^(th) phase,steers were fed in a stepwise fashion with increasing CTFRs,i.e.,30:70,50:50,70:30,and 90:10.The kinetics of dry matter and fiber degradation of alfalfa pellets were evaluated;the rumen microbial populations were detected using real-time PCR.The results revealed no significant(P> 0.05) interactions between dietary CTFR and SC for most parameters.Dietary CTFR had a significant effect(P< 0.01) on degradation characteristics of alfalfa pellets and the copies of rumen microorganism;the increasing concentrate level resulted in linear,quadratic or cubic variation trend for these parameters.SC supplementation significantly(P< 0.05) affected dry matter(DM) and neutral detergent fiber(NDF)degradation rates(c_(DM),c_(NDF)) and NDF effective degradability(ED_(NDF)).Compared with the control group,there was an increasing trend of rumen fungi and protozoa in SC group(P < 0.1);copies of total bacteria in SC group were significantly higher(P< 0.05).Additionally,percentage of Ruminobacter amylophilus was significantly lower(P< 0.05)but percentage of Selenomonas ruminantium was significantly higher(P< 0.05) in the SC group.In a word,dietary CTFR had a significant effect on degradation characteristics of forage and rumen microbial population.S.cerevisiae had positive effects on DM and NDF degradation rate or effective degradability of forage;S.cerevisiae increased rumen total bacteria,fungi,protozoa,and lactate-utilizing bacteria but reduced starch-degrading and lactate-producing bacteria.Gengzhi Ding Ying Chang Liping Zhao Zhenming Zhou Liping Ren Qingxiang Meng 2014Journal of Animal Science and Biotechnology2014,5,4:15

13Baicalein attenuates cardiac hypertrophy in mice via suppressing oxidative stress and activating autophagy in cardiomyocytes显示文摘Baicalein is a natural flavonoid extracted from the root of Scutellaria baicalensis that exhibits a variety of pharmacological activities.In this study,we investigated the molecular mechanisms underlying the protective effect of baicalein against cardiac hypertrophy in vivo and in vitro.Cardiac hypertrophy was induced in mice by injection of isoproterenol(ISO,30 mg·kg^(−1)·d^(−1))for 15 days.The mice received caudal vein injection of baicalein(25 mg/kg)on 3rd,6th,9th,12th,and 15th days.We showed that baicalein administration significantly attenuated ISO-induced cardiac hypertrophy and restored cardiac function.The protective effect of baicalein against cardiac hypertrophy was also observed in neonatal rat cardiomyocytes treated with ISO(10μM).In cardiomyocytes,ISO treatment markedly increased reactive oxygen species(ROS)and inhibited autophagy,which were greatly alleviated by pretreatment with baicalein(30μM).We found that baicalein pretreatment increased the expression of catalase and the mitophagy receptor FUN14 domain containing 1(FUNDC1)to clear ROS and promote autophagy,thus attenuated ISO-induced cardiac hypertrophy.Furthermore,we revealed that baicalein bound to the transcription factor FOXO3a directly,promoting its transcription activity,and transactivated catalase and FUNDC1.In summary,our data provide new evidence for baicalein and FOXO3a in the regulation of ISO-induced cardiac hypertrophy.Baicalein has great potential for the treatment of cardiac hypertrophy.Bing-yan Liu Ling Li Gao-li Liu Wei Ding Wen-guang Chang Tao Xu Xiao-yu Ji Xian-xin Zheng Jing Zhang Jian-xun Wang 2021Acta Pharmacologica Sinica2021,42,5:13
14Variations of shear wave splitting in the 2013 Lushan Ms7.0 earthquake region显示文摘In this paper, variations of shear wave splitting in the 2013 Lushan Ms7.0 earthquake sequence were studied. By analyzing shear wave particle motion of local events in the shear wave window, the fast polarization directions and the delay time between fast and slow shear waves were derived from seismic recordings at eight stations on the southern segment of the Longmenshan fault zone. In the study region, the fast polarization directions show partition characteristics from south to north. And the systematic changes of the time delays between two split shear waves were also observed. As for spatial distribution, the NE fast polarization directions are consistent with the Longmenshan fault strike in the south of focal region, whereas the NW fast direction is parallel to the direction of regional principal compressive stress in the north of focal region. Stations BAX and TQU are respectively located on the Central and Front-range faults, and because of the direct influence of these faults, the fast directions at both stations show particularity. In time domain, after the main shock, the delay times at stations increased rapidly, and decreased after a period of time. Shear-wave splitting was caused mostly by stress-aligned microcracks in rock below the stations. The results demonstrate changes of local stress field during the main shock and the aftershocks. The stress on the Lushan Ms7.0 earthquake region increased after the main shock, with the stress release caused by the aftershocks and the stress reduced in the late stage.CHANG LiJun DING ZhiFeng WANG ChunYong 2014Science China Earth Sciences2014,57,9:12

17Role of macrophages in peripheral nerve injury and repair显示文摘Resident and inflammatory macrophages are essential effectors of the innate immune system. These cells provide innate immune defenses and regulate tissue and organ homeostasis. In addition to their roles in diseases such as cancer, obesity and osteoarthritis, they play vital roles in tissue repair and disease rehabilitation. Macrophages and other inflammatory cells are recruited to tissue injury sites where they promote changes in the microenvironment. Among the inflammatory cell types, only macrophages have both pro-inflammatory (Ml) and anti-inflammatory (M2) actions, and M2 macrophages have four subtypes. The co-action of Ml and M2 subtypes can create a favorable microenvironment, releasing cytokines for damaged tissue repair. In this review, we discuss the activation of macrophages and their roles in severe peripheral nerve injury. 18Polymorphisms of alcohol dehydrogenase-2 and aldehyde dehydrogenase-2 and esophageal cancer risk in Southeast Chinese males显示文摘AIM:To evaluate the impact of alcohol dehydrogenase-2(ADH2) and aldehyde dehydrogenase-2(ALDH2) polymorphisms on esophageal cancer susceptibility in Southeast Chinese males.METHODS:Two hundred and twenty-one esophageal cancer patients and 191 healthy controls from Taixing city in Jiangsu Province were enrolled in this study.ADH2 and ALDH2 genotypes were examined by polymerase chain reaction and denaturing high-performance liquid chromatography.Unconditional logistic regression was used to calculate the odds ratios(OR) and 95% confi dence interval(CI) .RESULTS:The ADH G allele carriers were more susceptible to esophageal cancer,but no association was found between ADH2 genotypes and risk of esophageal cancer when disregarding alcohol drinking status.Regardless of ADH2 genotype,ALDH2G/A or A/A carriers had significantly increased risk of developing esophageal cancer,with homozygous individuals showing higher esophageal cancer risk than those who were heterozygous.A signif icant interaction between ALDH2 and drinking was detected regarding esophageal cancer risk;the OR was 3.05(95% CI:1.49-6.25) .Compared with non-drinkers carrying both ALDH2 G/G and ADH2 A/A,drinkers carrying both ALDH2 A allele and ADH2 G allele showed a signif icantly higher risk of developing esophageal cancer(OR = 8.36,95% CI:2.98-23.46) .CONCLUSION:Both ADH2 G allele and ALDH2 A allele significantly increase the risk of esophageal cancer development in Southeast Chinese males.ALDH2 A allele significantly increases the risk of esophageal cancer development especially in alcohol drinkers.Alcohol drinkers carrying both ADH2 G allele and ALDH2 A allele have a higher risk of developing esophageal cancer.Jian-Hua Ding Su-Ping Li Hai-Xia Cao Jian-Zhong Wu Chang-Ming Gao Ping Su Yan-Ting Liu Jian-Nong Zhou Jun Chang Gen-Hong Yao 2009World Journal of Gastroenterology2009,15,19:10
19Neoadjuvant oxaliplatin and capecitabine combined with bevacizumab plus radiotherapy for locally advanced rectal cancer: results of a single-institute phase II study显示文摘Background:Neoadjuvant chemoradiotherapy followed by surgery is recommended as the standard of care for locally advanced rectal cancer,reducing local recurrence but not distant metastasis.Intensified systemic therapy is warranted to reduce the risk of distant metastasis.The present study aimed to evaluate the safety and efficacy of neo-adjuvant oxaliplatin and capecitabine(XELOX)combined with bevacizumab plus radiotherapy for locally advanced rectal cancer.Methods:Patients with stages II to III rectal cancer received one cycle of induction chemotherapy and concurrent chemoradiotherapy with XELOX plus bevacizumab.Surgery was performed 6-8 weeks after completion of radiotherapy,and postoperative chemotherapy with three cycles of XELOX and two cycles of capecitabine were given.The primary endpoints were pathologic complete response(pCR)rate and safety,and the secondary endpoints were 3-year overall survival and progression-free survival.Results:Forty-five patients were enrolled between February 2013 and April 2015.All completed the neoadjuvant therapy.Seven patients(15.6%)refused subsequent surgical therapy for personal reasons,and the other 38 patients received radical resection,with a sphincter preservation rate of 84.2%and a pCR rate of 39.5%.Toxicity was acceptable,with grades 3-4 hematological toxicity and diarrhea observed in six and two patients,respectively.Incidence of anastomotic leak that required surgical intervention was 13.3%.After a median follow-up period of 37 months,five patients developed disease progression and two died of cancer.The 3-year overall survival rate and 3-year progres-sion-free survival rate were 95.3%and 88.6%,respectively.Conclusions:The addition of bevacizumab to neoadjuvant chemoradiotherapy resulted in a satisfying pCR rate and 3-year survival,but also may increase the risk of anastomotic leak,thus this regimen is not suitable to be considered for regular recommendation for locally advanced rectal cancer.Xin Yu Qiao-xuan Wang Wei-wei Xiao Hui Chang Zhi-fan Zeng Zhen-hai Lu Xiao-jun Wu Gong Chen Zhi-zhong Pan De-sen Wan Pei-rong Ding Yuan-hong Gao 2018Cancer Communications2018,38,1:10
