| 3 | The interferon inducing pathways and the hepatitis C virus显示文摘The innate immune response is triggered by a variety of pathogens, including viruses, and requires rapid induction of typeⅠ?interferons (IFN), such as IFNβ and IFNα. IFN induction occurs when specific pathogen motifs bind to specific cellular receptors. In non-professional immune, virally-infected cells, IFN induction is essentially initiated after the binding of dsRNA structures to TLR3 receptors or to intracytosolic RNA helicases, such as RIG-Ⅰ/MDA5. This leads to the recruitment of specific adaptors, such as TRIF for TLR3 and the mitochondrial-associated IPS-1/VISA/MAVS/CARDIF adapter protein for the RNA helicases, and the ultimate recruitment of kinases, such as MAPKs, the canonical IKK complex and the TBK1/IKKε kinases, which activate the transcription factors ATF-2/ c-jun, NF-κB and IRF3, respectively. The coordinated action of these transcription factors leads to induction of IFN and of pro-inflammatory cytokines and to the establishment of the innate immune response. HCV can cleave both the adapters TRIF and IPS-1/VISA/MAVS/ CARDIF through the action of its NS3/4A protease. Helicobacter pylori genes in gastric diseases显示文摘Helicobacter pylori(H. pylori) infection is present in more than half the world's population and has been associated with several gastric disorders, such as gastritis, peptic ulceration, and gastric adenocarcinoma.The clinical outcome of this infection depends on host and bacterial factors where H. pylori virulence genes seem to play a relevant role. Studies of cag A and vac A genes established that they were determining factors in gastric pathogenesis. However, there are gastric cancer cases that are cag A-negative. Several other virulence genes have been searched for, but these genes remain less well known that cag A and vac A. Thus, this review aimed to establish which genes have been suggested as potentially relevant virulence factors for H. pylori-associated gastrointestinal diseases. | 5 | Dietary supplementation with free methionine or methionine dipeptide mitigates intestinal oxidative stress induced by Eimeria spp. challenge in broiler chickens显示文摘Background: This study evaluated the effects of Eimeria spp. challenge and dietary supplementation with free methionine or methionine dipeptide on animal performance;expression of genes associated with the immune system, antioxidant system, and amino acid transport in the jejunum;and redox status of the jejunum of broiler chickens.Methods: A randomized, 2 × 3 factorial design was used, in which Eimeria spp. challenge was the first factor(Eimeria-challenged, EC, or unchallenged, UC, broilers) and methionine supplementation was the second factor(non-supplemented, NS;free DL-methionine, DL-Met;and methionine dipeptide, DL-methionyl-DL-methionine, DLMMet). At 14 days of age, chickens were inoculated orally with sporulated oocysts of Eimeria acervulina, Eimeria praecox, Eimeria maxima, and Eimeria mitis. Birds were killed by cervical dislocation 144 h post-inoculation(PI), and the jejunum was collected for biochemical and molecular analyses.Results: EC broilers had a 13% lower feed intake(FI), 37% lower body weight gain(BWG), and 39% higher feed conversion ratio(FCR) than UC broilers. Chickens fed the DL-Met diet had higher BWG(about 12% higher) and better FCR(about 12% lower) than chickens fed the NS diet. EC chickens had lower relative weight of the bursa of Fabricius(51.8%) and higher relative weights of the spleen and whole intestine(53.6% and 26.3%, respectively) than UC chickens. Eimeria spp. challenge led to an increase in the levels of oxidative substances, such as nitrite and thiobarbituric acid reactive substances(TBARS), in the jejunum of chickens 144 h PI. Among UC chickens, those fed the DL-Met diet had higher total antioxidant capacity(TAC) and lower catalase(CAT) and superoxide dismutase(SOD) activities. EC chickens that received the NS diet had higher carbonylated protein content(CP). This result was associated with their lower TAC and catalase activity. The lower TAC in EC chickens might have been due to reduced expression of catalase(CAT) and superoxide dismutase 1(SOD1) genes. Chickens fed the DL-Met and DLMMet diets had lower nitrite content. Eimeria spp. challenge suppressed neutral amino acid transporter 1(B0 AT1),peptide transporter 1(PEPT1), toll-like receptor 5(TLR5), interleukin 2(IL2), and occludin(OCLN) gene expression and enhanced cationic amino acid transporter 1(CAT-1) and interferon gamma(IFNG) gene expression. | 6 | Biomarkers in precision therapy in colorectal cancer显示文摘Colorectal cancer(CRC)is the most commonly diagnosed cancer in Europe.Because CRC is also a major cause of cancerrelated deaths worldwide,a lot of research has been focused on the discovery and development of biomarkers to improve the diagnostic process and to predict treatment outcomes.Up till now only a few biomarkers are recommended by expert panels.Current TNM criteria,however,cause substantial under-and overtreatment of CRC patients.Consequently,there is a growing need for new and efficient biomarkers to ensure optimal treatment allocation.An ideal biomarker should be easily translated into clinical practice,to identify patients who can be spared from treatment or benefit from therapy,ultimately resulting in precision medicine in the future.In this review we aim to provide an overview of a number of frequently studied biomarkers in CRC and,at the same time,we will emphasize the challenges and controversies that withhold the clinical introduction of these biomarkers.We will discuss both prognostic and predictive markers of chemotherapy,aspirin therapy as well as overall therapy toxicity.Currently,only mutant KRAS,mutant BRAF,MSI and the Oncotype DXColon Cancer Assay are used in clinical practice.Other biomarker studies showed insufficient evidence to be introduced into clinical practice.Divergent patient selection criteria,absence of validation studies and a large number of single biomarker studies are possibly responsible.We therefore recommend that future studies focus on combining key markers,rather than analysing single markers,standardizing study protocols,and validate the results in independent study cohorts,followed by prospective clinical trials. | Marlies S.Reimers Eliane C.M.Zeestraten Peter J.K.Kuppen Gerrit Jan Liefers Cornelis J.H.van de Velde | 2013 | Gastroenterology Report2013,1,3: | 3 |

| 9 | Architecting Fault Tolerance with Exception Handling: Verification and Validation显示文摘When building dependable systems by integrating untrusted software components that were not originally designed to interact with each other, it is likely the occurrence of architectural mismatches related to assumptions in their failure behaviour. These mismatches, if not prevented during system design, have to be tolerated during runtime. This paper presents an architectural abstraction based on exception handling for structuring fault-tolerant software systems. This abstraction comprises several components and connectors that promote an existing untrusted software element into an idealised fault-tolerant architectural element. Moreover, it is considered in the context of a rigorous software development approach based on formal methods for representing the structure and behaviour of the software architecture. The proposed approach relies on a formal specification and verification for analysing exception propagation, and verifying important dependability properties, such as deadlock freedom, and scenarios of architectural reconfiguration. The formal models are automatically generated using model transformation from UML diagrams: component diagram representing the system structure, and sequence diagrams representing the system behaviour. Finally, the formal models are also used for generating unit and integration test cases that are used for assessing the correctness of the source code. | 11 | Biliary tract reconstruction using jejunal tube:an experimental study in dogs显示文摘BACKGROUND:To physiologically reconstruct the biliary tract,Crema et al suggested the application of the Monti principle to the biliary tract,already used in humans for the urinary tract.With this technique,a jejunal segment is transversely retubularized.This study aimed to evaluate the efficacy of jejunal tube interposition between the common bile duct and duodenum in dogs.METHODS:Thirteen dogs underwent a laparoscopic common bile duct ligature,followed by a biliodigestive connection by jejunal tube interposition after one week.The levels of glutamic-pyruvic and glutamic-oxalacetic transaminases,total bilirubins,alkaline phosphatase and gamma-glutamyltransferase were assessed before surgery and thereafter weekly until euthanasia,which was performed 6 weeks after biliodigestive connection.RESULTS:Data on 9 dogs were analyzed statistically.The dogs presented with obstructive jaundice after common bile duct ligature,as confirmed by biochemical examination.They showed a statistically significant reduction in cholestasis after biliodigestive connection by jejunal tube interposition and were healthy until the end of the experiment.CONCLUSION:A statistically significant reduction was seen in total bilirubin and canalicular enzymes(alkaline phosphatase and gamma-glutamyltransferase)in the 9 dogs 6 weeks after biliodigestive connetion by jejunal tube interposition. | Eliane Anrain Trentini Eduardo Crema Juan Carlos Llanos Mauro Masson Lerco Luiz Alberto Magna Luiz Sergio Leonardi | 2009 | Hepatobiliary & Pancreatic Diseases International2009,8,2: | 2 |
