| 1 | Synthesis of Ag-SiO_2 composite nanospheres and their catalytic activity显示文摘A simple,mild,and time-saving method is employed to synthesize Ag-SiO2 composite nanospheres with Ag nanoparticles uniformly distributed on the surface of SiO2 nanoparticles.The chemical elements and the morphology of Ag-SiO2 composite nanospheres were analyzed with transmission electron microscopy(TEM),X-ray power diffraction(XRD),and X-ray photoelectron spectroscopy(XPS).On the surface of Ag-SiO2 composite nanospheres,silane coupling agent(KH-550)is introduced as an intermediary to connect the surfaces of SiO2 nanospheres and Ag nanoparticles,which is also helpful for avoiding the aggregation of Ag nanoparticles.It is found that Ag-SiO2 composite nanospheres have very good catalytic properties for the reduction of organic dyes,which may have potential application in wastewater treatment. | HAN QingYan LI GuiAn WANG DeXuan HE EnJie DONG Jun GAO Wei LI JunNa LIU TingZhuo ZHANG ZhongYue ZHENG HaiRong | 2014 | Science China Chemistry2014,57,6: | 2 |
| 2 | Effect of long-term low dose of aspirin on severity of disease following onset of acute cerebral infarction显示文摘BACKGROUND: Aspirin can decrease the incidence risk of high-risk crowdgroup of cerebral infarction, but there are still controversy if it might decrease the degree of disease in degree of patients with acute cerebral infarction. OBJECTIVE: To observe the effect of lower dose of aspirin during taking for a long time on disease degree of disease following onset of acute cerebral infarction. DESIGN: Grouping according to the admission time and 1∶1 paired observation. SETTING: Department of Neurology, Qilu Hospital of Shandong University. PARTICIPANTS: The participants in present study were 321 patients with acute cerebral infarction who received treatments in the Department of Neurology, Qilu Hospital of Shandong University from January 1999 to June 2000. There were 190 male and 131 female ,with mean (65±11)years of age. Inclusive criteria: ① A focal neurological disturbance occurred suddenly and had lasted for more than 24 hours, patients were admitted within 3 days after onset of disease; ② A computed tomography of the brain was performed and excluded hemorrhage in all patients; ③ The patients were proved internal carotid occlusions by clinical features and image findings; ④ The functions of limbs were normal (before the first stroke) or almost normal (before the second stroke). Exclusive criteria: ①The patients who had have cardiogenic cerebral embolism; ②The patients who had taken warfarin orally and other platelet agglutination drugs. METHODS: ①All the patients were divided into 2 groups according to whether they had taken aspirin before: aspirin-treated group (n=110) and blank control group(n=211). there were 70 male and 40 female in aspirin-treated group, with average(65±10) years of age.All patients had taken 50-100 mg/d aspirin for 6 months to 10 years before onset. There were 120 male and 91 female in blank control group, with average (65±13)years of age . Patients received a clinical scoring within 3 days and similar therapeutic measures (such as anti-platelet agglutination, improving cerebral circulation and metabolism-promoting reagent). Two groups of patients had the same basic conditions except for taking aspirin or not before. ②The matched pairs were made between 50 cases selected from aspirin-treated group and 50 cases from non-aspirin-treated groups according to age, gender, and other stroke risk factors. ③ Evaluation : Degree of disease after onset was evaluated by means of Acute Cerebral Infarction Clinical Neurologic Impairment Degree Scoring Standard of Carotid Artery System. MAIN OUTCOME MEASURES: Acute Cerebral Infarction Clinical Neurologic Impairment Degree Scoring Standard of Carotid Artery System. RESULTS: All 321 patients entered the stage of analysis with no loss in the midway. ① The symptom following onset of acute cerebral infarction was evaluated with clinical neurologic impairment scoring criteria, there were no significant differences between aspirin group and blank control group [(17.39 ± 9.90) vs(16.22 ± 9.98) (t=1.025, P > 0.05)]. ② No significant differences were found in 1:1 matched pairs of 100 cases from aspirin group and blank control group (t=1.74, P > 0.05). | 3 | Inhibition of beta-site amyloid precursor protein-cleaving enzyme and beta-amyloid precursor protein genes in SK-N-SH cells显示文摘BACKGROUND:Previous studies have demonstrated that Piper futokadsura stem selectively inhibits expression of amyloid precursor protein(APP) at the mRNA level.In addition,the piperlonguminine(A) and dihydropiperlonguminine(B) components(1:0.8),which can be separated from Futokadsura stem,selectively inhibit expression of the APP at mRNA and protein levels. OBJECTIVE:Based on previous findings,the present study investigated the effects ofβ-site amyloid precursor protein cleaving enzyme(BACE1) and APP genes on the production ofβ-amyloid peptide 42(Aβ42) in human neuroblastoma cells(SK-N-SH cells) using small interfering RNAs (siRNAs) and A/B components separated from Futokadsura stem,respectively. DESIGN,TIME AND SETTING:A gene interference-based randomized,controlled,in vitro experiment was performed at the Key Laboratory of Cardiovascular Remodeling and Function Research,Ministries of Education and Public Health,and Institute of Pharmacologic Research, School of Pharmaceutical Science & Department of Biochemistry,School of Medicine,Shandong University between July 2006 and December 2007. MATERIALS:SK-N-SH cells were provided by Shanghai Institutes of Biological Sciences,Chinese Academy of Sciences,Shanghai,China;mouse anti-human BACE1 monoclonal antibody was purchased from R&D Systems,USA;mouse anti-human APP monoclonal antibody was purchased from Cell Signaling Technology,USA;and horseradish peroxidase(HRP)-conjugated goat anti-mouse IgG was provided by Sigma,USA. METHODS:The human BACE1 cDNA sequence was obtained from NCBI website (www.ncbi.nlm.nih.gov/sites/entrez).Three pairs of siRNAs,specific to human BACE1 gene,were synthesized through the use of Silencer^(?) pre-designed siRNA specification,and were transfected into SK-N-SH cells with siPORT NeoFX transfection agent to compare the effects of different concentrations of siRNAs(10-50 nmol/L) on SK-N-SH cells.Futokadsura stem was separated and purified with chemical methods,and the crystal was composed of A/B components,with an A to B ratio of 1:0.8.The A/B(1:0.8) components were added to the SK-N-SH cells at different concentrations(13.13,6.56,and 3.28 mg/mL). MAIN OUTCOME MEASURES:Using RT-PCR and Western blot methods,BACE1 and APP expression at mRNA and protein levels was detected in SK-N-SH cells following treatment with different siRNAs and concentrations of Futokadsura stem-separated A/B components,respectively. Altered Aβ42 secretion by SK-N-SH cells was determined by ELISA. RESULTS:BACE1 mRNA and protein levels were significantly suppressed by 40 and 50 nmol/L siRNAs at 48 hours post-transfection.A/B components(1:0.8),which were separated from Futokadsura stem,selectively inhibited mRNA and protein expression of APP in SK-N-SH cells. Aβ42 secretion by SK-N-SH cells was significantly decreased following treatment with siRNAs or A/B components. | 4 | Etiological factors of Alzheimer disease and recent advances of its treatment显示文摘OBJECTIVE: To investigate the etiological factors of Alzheimer disease (AD) and the advance in drug treatment. DATA SOURCES: Using the terms 'Alzheimer disease, etiology, drug treatment', we searched Medline database for AD treatment-related English literatures which were published between January 1993 and September 2006. Other literatures were obtained by searching concrete magazines and papers by hand. STUDY SELECTION: The data were selected primarily. Epidemiologic study and randomized controlled clinical trials were selected, and those studies with repetitive or similar contents were excluded. DATA EXTRACTION: Totally 1 537 AD and its treatment-related literatures were collected, and 32 of them were involved. Altogether 1 505 non-randomized controlled clinical trials, repetitive studies and reviews were excluded. DATA SYNTHESIS: The restriction of curative effect is a progressive neural degenerative disease. Although the etiological hypothesis of this disease has been introduced, the etiological factors of this disease are still unclear. The current treatments mainly involve: preventing against Aβ formation and clearing Aβ, application of antioxidant and free radical scavengers, application of anti-inflammatory preparation, application of cholinergic preparation, hormonal therapy, application of metabolic enhancer, application of neurotrophic factor and nerve protectant, gene therapy and so on. CONCLUSION: The etiological factors of AD are still unclear, and symptomatic treatment is much taken in clinical therapy.
