2Development of 'Multi-Resistance Rice' by Pyramiding of Insect (Cry1C) and Blast Resistance (Pi1 and Pi2) Genes显示文摘[Objective] The paper was to improve the blast resistance of insect-resistant transgenic rice. [Method] The Japonica rice variety Jikang10, a new transgenic variety with exogenous insect-resistant gene Cry1C, was used as the receptor, and Kongyu 131, a traditional breeding variety with broad-spectrum high blast resistance genes Pi1 and Pi2, was used as the donor to breeding new rice varieties. The genes were polymerized by hybridization and multi-generation backcrossing, and the offspring of each generation was screened by molecular marker assisted selection, field identification of multi-resistance against insect pests and diseases and agronomic trait selection. [Result] Four lines SK01, SK02, SK03 and SK04 with better resistances to insect pests and blast and outstanding agronomic traits in field were selected. 3Single‑Atom Cobalt‑Based Electrochemical Biomimetic Uric Acid Sensor with Wide Linear Range and Ultralow Detection Limit显示文摘Uric acid(UA)detection is essential in diagnosis of arthritis,preeclampsia,renal disorder,and cardiovascular diseases,but it is very challenging to realize the required wide detection range and low detection limit.We present here a single-atom catalyst consisting of Co(Ⅱ)atoms coordinated by an average of 3.4 N atoms on an N-doped graphene matrix(A-Co-NG)to build an electrochemical biomimetic sensor for UA detection.The A-Co-NG sensor achieves a wide detection range over 0.4-41,950μM and an extremely low detection limit of 33.3±0.024 nM,which are much better than previously reported sensors based on various nanostructured materials.Besides,the A-Co-NG sensor also demonstrates its accurate serum diagnosis for UA for its practical application.Combination of experimental and theoretical calculation discovers that the catalytic process of the A-Co-NG toward UA starts from the oxidation of Co species to form a Co^3+-OH-UA*,followed by the generation of Co^3+-OH+^*UA_H,eventually leading to N-H bond dissociation for the formation of oxidized UA molecule and reduction of oxidized Co^3+to Co^2+for the regenerated A-Co-NG.This work provides a promising material to realize UA detection with wide detection range and low detection limit to meet the practical diagnosis requirements,and the proposed sensing mechanism sheds light on fundamental insights for guiding exploration of other biosensing processes.Fang Xin Hu Tao Hu Shihong Chen Dongping Wang Qianghai Rao Yuhang Liu Fangyin Dai Chunxian Guo Hong Bin Yang Chang Ming Li 2021Nano-Micro Letters2021,13,1:2

5Propofol inhibits cells migration and invasion via HOTAIR/miR-93/HIF-1α-mediated lactate secretion in colon cancer显示文摘Propofol,a common intravenous anesthetic used in clinical,has been shown to regulate cells proliferation,inflammation,angiogenesis and metastasis as well as exerting anti-cancer effect in several different types of cancer.However,the functions and mechanisms of propofol in lactate secretion and invasion of cancer cells remain unknown.The primary aim of this study was to investigate the role of propofol in cells migration and invasion of colon cancer(CRC).Scratches assay,Transwell assay were used to detect colon cells migration and invasion.Realtime Quantitative Polymerase Chain Reaction(RT-qPCR),Western blotting were utilized to detect the expression of related molecules.Dual-luciferase assay was performed to identify transcriptional activity of target genes,RNA pulldown assay was used to confirm the interactions of RNAs.Results indicated that propofol inhibited cell migration and invasion through suppressing lactic acid production in CRC.Moreover,propofol reduced lactic acid production by down-regulating the expression of hypoxia inducible factor-1α(HIF-1α)and monocarboxylate transporter 4(MCT4).In addition,propofol inhibited the expression of HIF-1αand cells migration/invasion via suppressing the interaction of HOX transcript antisense RNA(HOTAIR)with miR-93.The present study indicated that propofol inhibited colon cancer cell migration and invasion by reducing lactic acid production and the expression of HIF-1αand MCT4 via HOTAIR/miR-93 axis.These data may provide potential metabolic targets for CRC treatment.RUONAN GU WENJING GUO WENYANG WANG GUIHUAN LI XIAOJU LAI ZHIBIN HUANG WANLU ZHAO ZHICONGWU HUA CHEN WENYANG LUO FANGYIN ZENG FAN DENG 2021BIOCELL2021,45,6:0
6Application of AHP and DTOPSIS in Sugarcane Breeding显示文摘The weights of ten important traits of sugarcane,including the number of hair groups,plant height,defoliation capability,stalk diameter,ratooning property,stalk number of disease resistance,cane yield,sucrose content and sugar yield,were distributed with the Analytic Hierarchy Process( AHP) method. Ten Yuetang series of sugarcane lines were evaluated by Dynamic Technique for Order Preference by Similarity to Ideal Solution( DTOPSIS). AHP results indicated that the weights of ten traits were 0. 013 4,0. 022 7,0. 030 2,0. 040 8,0. 065 3,0. 088 1,0. 116 2,0. 160 6,0. 198 4 and 0. 264 5,respectively,with random consistency ratio C. R. = 0. 098 1 < 0. 1,suggesting that the distribution of the weights of evaluation indicators was reasonable. Comprehensive evaluation of 10 sugarcane lines by DTOPSIS method combining with the conclusion of AHP showed that the comprehensive performances of Yuetang 03-393 and Yuetang 03-373 were much better than other sugarcane lines,with relative approach degree C i ranking the first and second,respectively,which was basically consistent with the practical survey,suggesting that Yuetang 03-393 and Yuetang 03-373 were suitable for large-scale popularization. 7Simulating androgen receptor selection in designer yeast显示文摘Androgen receptor(AR)mutation is closely associated with prostate cancer(PCa)and is one of the mechanisms of resistance to PCa therapies such as AR antagonists.Although sequencing technologies like next-generation sequencing(NGS)contributes to the high-throughput and precise detection of AR mutations carried by PCa patients,the lack of interpretations of these clinical genetic variants has still been a roadblock for PCa-targeted precision medicine.Here,we established a designer yeast reporter assay to simulate natural androgen receptor(AR)selection using AR antagonists.Yeast HIS3 gene transactivation was associated with the ligand-induced recruitment of steroid receptor coactivator-1(SRC-1)by AR mutants,where yeast growth in histidine-free medium was determined as the outcome.This assay is applicable to determine a wide range of clinical AR mutants including those with loss of function relating to androgen insensitivity syndrome(AIS),and those associated with PCa conferring resistance to AR antagonists such as enzalutamide(ENZ),bicalutamide(BIC),and cyproterone acetate(CPA).One clinical AR mutant previously reported to confer ENZ-resistance,F877L,was found to confer partial resistance to CPA as well using designer yeast.Our simple and efficient assay can enable precise one-pot screening of AR mutants,providing a reference for tailored medicine.Haoran Zhang Lu Zhang Yipeng Xu Shaoyong Chen Zhenyi Ma Mingdong Yao Fangyin Li Bo Li Yingjin Yuan 2022Synthetic and Systems Biotechnology2022,7,4:0
