2Cystathionine-γ-lyase ameliorates the histone demethylase JMJD3-mediated autoimmune response in rheumatoid arthritis显示文摘Cystathionine-γ-lyase(CSE),an enzyme associated with hydrogen sulfide(H2S)production,is an important endogenous regulator of inflammation.Jumonji domain-containing protein 3(JMJD3)is implicated in the immune response and inflammation.Here,we investigated the potential contribution of JMJD3 to endogenous CSE-mediated inflammation in rheumatoid arthritis(RA).Upregulated CSE and JMJD3 were identified in synovial fibroblasts(SFs)from RA patients as well as in the joints of arthritic mice.Knocking down CSE augmented inflammation in IL-1β-induced SFs by increasing JMJD3 expression.In addition,CSE−/−mice with collagen-induced arthritis(CIA)developed severe joint inflammation and bone erosion.Conversely,overexpressing CSE inhibited JMJD3 expression by the transcription factor Sp-1 and was accompanied by reduced inflammation in IL-1β-treated SFs.Furthermore,JMJD3 silencing or the administration of the JMJD3 inhibitor GSK-J4 significantly decreased the inflammatory response in IL-1β-treated SFs,mainly by controlling the methylation status of H3K27me3 at the promoter of its target genes.GSK-J4 markedly attenuated the severity of arthritis in CIA mice.In conclusion,suppressing JMJD3 expression by the transcription factor Sp-1 is likely responsible for the ability of CSE to negatively modulate the inflammatory response and reduce the progression of RA.Weijun Wu Ming Qin Wanwan Jia Zheng Huang Zhongzheng Li Di Yang Mengwei Huang Chenxi Xiao Fen Long Jianchun Mao Philip K.Moore Xinhua Liu Yi Zhun Zhu 2019Cellular & Molecular Immunology2019,16,8:9

4Quality metrics and outcomes among critically ill patients in China:results of the national clinical quality control indicators for critical care medicine survey 2015-2019显示文摘Background:It is crucial to improve the quality of care provided to ICU patient,therefore a national survey of the medical quality of intensive care units(ICUs)was conducted to analyze adherence to quality metrics and outcomes among critically ill patients in China from 2015 to 2019.Methods:This was an ICU-level study based on a 15-indicator online survey conducted in China.Considering that ICU care quality may vary between secondary and tertiary hospitals,direct standardization was adopted to compare the rates of ICU quality indicators among provinces/regions.Multivariate analysis was performed to identify potential factors for in-hospital mortality and factors related to ventilator-associated pneumonia(VAP),catheter-related bloodstream infections(CRBSIs),and catheter-associated urinary tract infections(CAUTIs).Results:From the survey,the proportions of structural indicators were 1.83%for the number of ICU inpatients relative to the total number of inpatients,1.44%for ICU bed occupancy relative to the total inpatient bed occupancy,and 51.08%for inpatients with Acute Physiology and Chronic Health Evaluation II scores≥15.The proportions of procedural indicators were 74.37%and 76.60%for 3-hour and 6-hour surviving sepsis campaign bundle compliance,respectively,62.93%for microbiology detection,58.24%for deep vein thrombosis prophylaxis,1.49%for unplanned endotracheal extubations,1.99%for extubated inpatients reintubated within 48 hours,6.38%for unplanned transfer to the ICU,and 1.20%for 48-hour ICU readmission.The proportions of outcome indicators were 1.28‰for VAP,3.06‰for CRBSI,3.65‰for CAUTI,and 10.19%for in-hospital mortality.Although the indicators varied greatly across provinces and regions,the treatment level of ICUs in China has been stable and improved based on various quality control indicators in the past 5 years.The overall mortality rate has dropped from 10.19%to approximately 8%.Conclusions:The quality indicators of medical care in China’s ICUs are heterogeneous,which is reflected in geographic disparities and grades of hospitals.This study is of great significance for improving the homogeneity of ICUs in China.Xi Rui Fen Dong Xudong Ma Longxiang Su Guangliang Shan Yanhong Guo Yun Long Dawei Liu Xiang Zhou 2022Chinese Medical Journal2022,,9:4

6Hollow Carbon Microspheres/MnO_2 Nanosheets Composites:Hydrothermal Synthesis and Electrochemical Behaviors显示文摘This article reported the electrochemical behaviors of a novel hollow carbon microspheres/manganese dioxide nanosheets(micro-HC/nano-MnO2) composite prepared by an in situ self-limiting deposition method under hydrothermal condition. The results of scanning electron microscopy reveal that MnO2 nanosheets homogeneously grow onto the surface of micro-HC to form a loose-packed microstructure. The quantity of MnO2 required in the electrode layer has thereby been reduced significantly, and higher specific capacitances have been achieved. 7A bird nest-like manganese dioxide and its application as electrode in supercapacitors显示文摘A novel bird nest-like nanostructured MnO2(BNNS-MnO2) was prepared by a facile and cost-effective strategy. Their structures and morphologies were characterized by field emission scanning electron microscopy, transmission electron microscopy and powder X-ray diffraction. Capacitive behaviors were investigated by cyclic voltammetry and galvanostatic charge-discharge. The obtained nano-MnO2 possesses a well designed loose-assembled hierarchical nanoarchitecture with an appropriate crystallinity which gives rise to excellent performances as an electrode material for supercapacitors. A maximum specific capacitance of 917 F/g has been obtained at a current density of 5 mA/cm2 in 6 mol/L KOH aqueous solution, and a specific capacitance of 210 F/g has been maintained for 500 cycles. 8Experimental Study on Deep Desulfurizer in LF Process显示文摘CaO-Al2O3-SiO2-CaF2-MgO was selected as the slag system for desulfurization in LF process.The reaction between steel and slag during desulfurization has been simulated by using Factsage software to study the influence of component on the sulfur distribution ratio.In order to research the influence of CaO content,aluminum powder content and its granularity on desulfurization,laboratory experiments have been carried out in a 200 kg inductive furnace.Results showed that the optimal composition of deep desulfurizer is wCaO=64% and aluminium powder 10% with a granularity of 30 μm.Industrial trials showed that the main composition range of final slag in LF process is wCaO=53.0%-57.0%,wAl2O3=23.4%-25.1%,wSiO2=8.1%-10.0%,and wCaF2=3.2%-4.7%.The sulfur mass percent in steel is lower than 0.0008% with a desulfurization rate above 89%.According to the result of industrial production,this desulfurizer could meet the production requirement for ultra-low sulfur steel,of which sulfur mass percent is under 0.0015%.WU Long PEI Fen CHEN Yut LI Shi-qi 2012Journal of Iron and Steel Research(International)2012,19,4:2
9Enhanced uptake and anti-maturation effect of celastrol-loaded mannosylated liposomes on dendritic cells for psoriasis treatment显示文摘Psoriasis is an autoimmune skin disease in which dendritic cells(DCs) trigger the progression of psoriasis by complex interactions with keratinocytes and other immune cells. In the present study,we aimed to load celastrol, an anti-inflammatory ingredient isolated from Chinese herbs, on mannosylated liposomes to enhance DC uptake as well as to induce DC tolerance in an imiquimod-induced psoriasis-like mouse model. Mannose was grafted onto liposomes to target mannose receptors on DCs. The results demonstrated that compared with unmodified liposomes, DCs preferred to take up more fluorescence-labeled mannosylated liposomes. After loading celastrol into mannose-modified liposomes,they effectively inhibited the expression of maturation markers, including CD80, CD86 and MHC-II, on DCs both in vitro and in vivo. Additionally, after intradermal injection with a microneedle, celastrolloaded mannose-modified liposomes(CEL-MAN-LPs) achieved a superior therapeutic effect compared with free drug and celastrol-loaded unmodified liposomes in the psoriasis mouse model in terms of the psoriasis area and severity index, histology evaluation, spleen weight, and expression of inflammatory cytokines. 13A dandelion-like carbon microsphere/MnO_2 nanosheets composite for supercapacitors显示文摘This article reported the electrochemical performance of a novel cabon microsphere/MnO2nanosheets(CMS/MnO2) composite prepared by a in situ self-limiting deposition method under hydrothermal condition. The results of scanning electron microscopy(SEM) and transmission electron microscopy(TEM) revealed that MnO2nanosheets homogeneously grew onto the surface of CMS to form a loose-packed and dandelion-like core/shell microstructure. The unique microstructure plays a basic role in electrochemical accessibility of electrolyte to MnO2active material and a fast diffusion rate within the redox phase. 14Preparation of nano-PANI@MnO_2 by surface initiated polymerization method using as a nano-tubular electrode material:The amount effect of aniline on the microstructure and electrochemical performance显示文摘In this study,nano-polyanline and manganese oxide nanometer tubular composites(nano-PANI@MnO2)were prepared by a surface initiated polymerization method and used as electrochemical capacitor electrode materials; and the effect of aniline amount on the microstructure and electrochemical performance was investigated. The microstructures and surface morphologies of nano-PANI@MnO2 were characterized by X-ray diffraction,scanning electron microscopy and fourier transformation infrared spectroscope. The electrochemical performance of these composite materials was performed with cyclic voltammetry,charge–discharge test and electrochemical impedance spectroscopy,respectively. The results demonstrate that the feed ratio of aniline to MnO2 played a very important role in constructing the hierarchically nano-structure,which would,hence,determine the electrochemical performance of the materials. Using the templateassisted strategy and controlling the feed ratio of aniline to MnO2,the nanometer tubular structure of nanoPANI@MnO2 was obtained. 15AIM2 deficiency in B cells ameliorates systemic lupus erythematosus by regulating Blimp-1-Bcl-6 axis-mediated B-cell differentiation显示文摘Absent in melanoma 2(AIM2)has been reported to be a component of inflammasomes in innate immune cells.Surprisingly,AIM2 is expressed by B cells,and higher AIM2 expression is observed in the B cells from lupus patients.To date,the inflammasome-indepe ndent functi on of AIM2 in B cells remai ns un clear.Here,we report in creased expressi on of AIM2 in human tonsil memory and germinal center(GC)B cells and in memory B cells and plasma cells from the circulation and skin lesions of lupus patients.Conditional knockout of AIM2 in B cells reduces the CD19^(+)B-cell frequency in lymph nodes and spleens,and dampens KLH-induced lgG1-antibody production.In a pristane-induced mouse model of lupus,AIM2 deficiency in B cells attenuates lupus symptoms and reduces the frequency of GC B cells,T follicular helper(Tfh)cells,plasmablast cells,and plasma cells.Furthermore,the loss of AIM2 in human B cells leads to the increased expression of Blimp-1 and reduces the expression of Bcl-6.However,the silendng of Blimp-1 and Bcl-6 has no significant effect on AIM2 expression,indicating that AIM2 might be the upstream regulator for Blimp-1 and Bcl-6.In addition,IL-10 is found to upregulate AIM2 expression via DNA demethylation.Together,our findings reveal that AIM2 is highly expressed in the B cells of lupus patients and promotes B-cell differentiation by modulating the Bcl-6-Blimp-1 axis,providing a novel target for SLE treatment.Ming Yang Di Long Longyuan Hu Zhidan Zhao Qianwen Li Yunkai Guo Zhenghao He Ming Zhao Liwei Lu Fen Li Hai Long Haijing Wu Qianjin Lu 2021Signal Transduction and Targeted Therapy2021,6,10:1
16A hierarchical porous carbon membrane from polyacrylonitrile/polyvinylpyrrolidone blending membranes: Preparation,characterization and electrochemical capacitive performance显示文摘Novel hierarchical porous carbon membranes were fabricated through a simple carbonization procedure of well-defined blending polymer membrane precursors containing the source of carbon polyacrylonitrile(PAN) and an additive of polyvinylpyrrolidone(PVP), which was prepared using phase inversion method. The as-fabricated materials were further used as the active electrode materials for supercapacitors.The effects of PVP concentration in the casting solution on structure feature and electrochemical capacitive performance of the as-prepared carbon membranes were also studied in detail. As the electrode material for supercapacitor, a high specific capacitance of 278.0 F/g could be attained at a current of 5 m A/cm2 and about 92.90% capacity retention could be maintained after 2000 charge/discharge cycles in 2 mol/L KOH solution with a PVP concentration of 0.3 wt% in the casting solution. 17Energy Storage Mechanism of Vanadium Nitride via Intercalating Different Atomic Radius for Expanding Interplanar Spacing显示文摘As a promising anode material in supercapacitors,vanadium nitride has been widely concerned due to its ultra-high theoretical specific capacitance.However,its routine test capacitance value is still far from the theoretical value and its energy storage mechanism is controversial.In order to solve these two key problems,here we prepare interplanar spacing expanded vanadium nitride materials with different impurity atoms intercalation from two anionic precursors of vanadium-based metal organic frameworks with different functional groups.The obtained vanadium nitride reaches a higher specific capacitance;and further,through ex situ X-Ray diffraction and in situ Raman,the charge storage of vanadium nitride is contributed by two processes:the first benefit is from the K^(+) de/intercalation in the interplanar spacing,and the other one is derived from the redox reaction with OH−by adsorption on surface.Furthermore,both of the first principle calculation and extended experiments support this idea.We believe that such detailed research on the energy storage mechanism can provide a clear idea for the application of metal nitrides in supercapacitors and other energy storage devices.Ying Liu Lingyang Liu Long Kang Fen Ran 2022Energy & Environmental Materials2022,5,2:1

20SMYD3-PARP16 axis accelerates unfolded protein response and mediates neointima formation显示文摘Neointimal hyperplasia after vascular injury is a representative complication of restenosis.Endoplasmic reticulum(ER)stress-induced unfolded protein response(UPR)is involved in the pathogenesis of vascular intimal hyperplasia.PARP16,a member of the poly(ADP-ribose)polymerases family,is correlated with the nuclear envelope and the ER.Here,we found that PERK and IRE1 a are ADPribosylated by PARP16,and this might promote proliferation and migration of smooth muscle cells(SMCs)during the platelet-derived growth factor(PDGF)-BB stimulating.Using chromatin immunoprecipitation coupled with deep sequencing(ChIP-seq)analysis,PARP16 was identified as a novel target gene for histone H3 lysine 4(H3 K4)methyltransferase SMYD3,and SMYD3 could bind to the promoter of Parp16 and increased H3 K4 me3 level to activate its host gene’s transcription,which causes UPR activation and SMC proliferation.Moreover,knockdown either of PARP16 or SMYD3 impeded the ER stress and SMC proliferation.On the contrary,overexpression of PARP16 induced ER stress and SMC proliferation and migration.In vivo depletion of PARP16 attenuated injury-induced neointimal hyperplasia by mediating UPR activation and neointimal SMC proliferation.This study identified SMYD3-PARP16 is a novel signal axis in regulating UPR and neointimal hyperplasia,and targeting this axis has implications in preventing neointimal hyperplasia related diseases.Fen Long Di Yang Jinghua Wang Qing Wang Ting Ni Gang Wei Yizhun Zhu Xinhua Liu 2021Acta Pharmaceutica Sinica B2021,11,5:1
