1Structural changes of gut microbiota in Parkinson's disease and its correlation with clinical features显示文摘The aim of this study was to compare the structure of gut microbiota in Parkinson's disease(PD) patients and healthy controls;and to explore correlations between gut microbiota and PD clinical features. We analyzed fecal bacterial composition of 24 PD patients and 14 healthy volunteers by using 16 S rRNA sequencing. There were significant differences between PD and healthy controls, as well as among different PD stages. The putative cellulose degrading bacteria from the genera Blautia(P=0.018),Faecalibacterium(P=0.048) and Ruminococcus(P=0.019) were significantly decreased in PD compared to healthy controls.The putative pathobionts from the genera Escherichia-Shigella(P=0.038), Streptococcus(P=0.01), Proteus(P=0.022), and Enterococcus(P=0.006) were significantly increased in PD subjects. Correlation analysis indicated that disease severity and PD duration negatively correlated with the putative cellulose degraders, and positively correlated with the putative pathobionts. The results suggest that structural changes of gut microbiota in PD are characterized by the decreases of putative cellulose degraders and the increases of putative pathobionts, which may potentially reduce the production of short chain fatty acids, and produce more endotoxins and neurotoxins; and these changes is potentially associated with the development of PD pathology.Wei Li Xiaoli Wu Xu Hu Tao Wang Shan Liang Yunfeng Duan Feng Jin Bin Qin 2017Science China(Life Sciences)2017,60,11:26
2Fluid inclusion evidence for petroleum accumulation in northern Qaidam Basin显示文摘Based on the results of fluid inclusion study(microscopic characteristics,homogenization temperatures and grains with oil inclusions),the pool-forming time of the Nanbaxian and Mabei 1 oilfields in northern Qaidam Basin was discussed in this paper.Fluid inclusions in northern Qaidam Basin are relatively abundant,including aqueous inclusions and hydrocarbon inclusions.These inclusions are small(5-10 μm),and mainly distributed in quartz fissures,overgrowths and cementations.The abundances of grains with oil inclusions(GOI) are commonly low.The GOI values of most samples are between 2.0%-10.5%,and those of about 40 percent of total samples exceed 5%.The homogenization temperatures of fluid inclusions in different samples are markedly dissimilar,indicating that these inclusions were formed in different periods.According to the measured homogenization temperatures of fluid inclusions and in combination with burial history and thermal evolution history,the Nanbaxian and Mabei 1 oilfields are deduced,which have experienced two hydrocarbon charging periods,with the former of N1-N21 and N22-N23,and the latter of N1 and N1 end-N2,respectively.YANG Yunfeng LI Xianqing FENG Songbao WANG Kangdong KONG Longxi DONG Peng XU Feng PENG Dehua CHEN Yan 2010Chinese Journal Of Geochemistry2010,29,4:5
3Analysis and prediction of loudspeaker large-signal symptoms显示文摘Nonlinear lumped-parameter force factor Bl(x), stiffness Kms(x) and inductance Le(x) of electrodynamic loudspeakers change frequency responses and generate some nonlinear effects for large stimulus: harmonic and intermodulation distortion, DC component in diaphragm displacement, instability of vibration and jumping effects. By modeling the nonlinear system under large-signal conditions, relationship between the nonlinear parameters and large-signal behavior can be revealed and help to provide guidance to diagnose loudspeakers. 4Regulation of U6 Promoter Activity by Transcriptional Interference in Viral Vector-Based RNAi显示文摘The direct negative impact of the transcriptional activity of one component on the second one in cis is referred to as transcriptional interference (TI). U6 is a type III RNA polymerase III promoter commonly used for driving small hairpin RNA (shRNA) expression in vector-based RNAi. In the design and construction of viral vectors, multiple transcription units may be arranged in close proximity in a space-limited vector. Determining if U6 promoter activity can be affected by TI is critical for the expression of target shRNA in gene therapy or loss-of-function studies. In this research, we designed and implemented a modified retroviral system where shRNA and exogenous gene expressions were driven by two independent transcriptional units. We arranged U6 promoter driving shRNA expression and UbiC promoter in two promoter arrangements. In primary macrophages, we found U6 promoter activity was inhibited by UbiC promoter when in the divergent arrangement but not in tandem. In contrast, PKG promoter had no such negative impact. Instead of enhancing U6 promoter activity, CMV enhancer had significant negative impact on U6 promoter activity in the presence of UbiC promoter. 5A Multifunctional Lentiviral-Based Gene Knockdown with Concurrent Rescue that Controls for Off-Target Effects of RNAi显示文摘The efficient, stable delivery of siRNA into cells, and the appropriate controls for non-specific off-target effects of siRNA are major limitations to functional studies using siRNA technology. To overcome these drawbacks, we have developed a single lentiviral vector that can concurrently deplete endogenous gene expression while expressing an epitope-tagged siRNA-resistant target gene in the same cell. To demonstrate the functional utility of this system, we performed RNAi-depleted α-actinin-1 (α-ACTN1) expression in human T cells. α-ACTN1 RNAi resulted in inhibited chemotaxis to SDF-1α, but it can be completely rescued by concurrent expression of RNAi-resistant α-ACTN1 (rr-α-ACTN1) in the same cell. The presence of a GFP tag on rr-α-ACTN1 allowed for detection of appropriate subcellular localization of rr-α-ACTN1. 6Nonlinear behavior of electrodynamic loudspeaker suspension at low frequencies显示文摘The suspension of electrodynamic loudspeakers includes a surround of the cone and a spider, and it is characterized by the mechanic stiffness in the lumped-parameter model. By solving the nonlinear differential equation of motion which considers the nonlinearity of suspension at low frequencies numerically and measuring different kinds of surrounds and spiders, the nonlinear behavior of suspension is theoretically and experimentally studied. Since the nonlinear stiffness of spiders and surrounds can be measured and fitted respectively before assembled into loudspeakers, which spider works best with which surround is studied. 10Tetra-armed conjugated microporous polymers for gas adsorption and photocatalytic hydrogen evolution显示文摘Two novel tetra-armed conjugated microporous polymers with different geometries have been designed and synthesized via Suzuki-Miyaura cross coupling polycondensation. Both polymers are stable in various organic solvents tested and are thermally stable. The pyrene-containing polymer of PrPy with the rigid pyrene unit shows a higher Brunauer-Emmet-Teller specific surface area of 1219 m^2 g^(-1) than the tetraphenylethylene-containing polymer of PrTPE(770 m^2 g^(-1)), which leads to a high CO_2 uptake ability of 3.89 mmol g^(-1) at 1.13 bar/273 K and a H_2 uptake ability of 1.69 wt% at 1.13 bar/77 K. 19Dependence of femtosecond time-resolved magneto-optical Kerr rotation on the direction of polarization of the probe beam显示文摘We demonstrate that the femtosecond time-resolved magneto-optical Kerr rotation oscillates with the direction of polarization of the probe beam when a sample of Al0.25Ga0.75As/GaAs multi-quantum wells is excited by a circularly polarized pump and detected by a linearly polarized probe at wavelengths from 800 to 830 nm. Analytical expressions are derived to explain the mechanism, which is in good agreement with the numerical computation and the experimental data.
