| 1 | Hydrodynamic Gene Delivery of Interleukin-22 Protects the Mouse Liver from Concanavalin A-, Carbon Tetrachloride-,and Fas Ligand-Induced Injury via Activation of STAT3显示文摘Interleukin-22 (IL-22) is a recently identified T cell-derived cytokine whose biological significance remains obscure. Previously, we have shown that IL-22 plays a protective role in T cell-mediated hepatitis induced by Concanavalin A (Con A), acting as a survival factor for hepatocytes. In the present paper, we demonstrate that hydrodynamic gene delivery of IL-22 cDNA driven either by a liver-specific albumin promoter or a human cytomegalovirus (CMV) promoter results in IL-22 protein expression, STAT3 activation, and expression of several anti-apoptotic proteins, including Bcl-xL, Bcl-2, and Mcl-1 in the liver. Immunohistochemical analysis reveals that IL-22 protein expression is mainly detected in the cytoplasm of hepatocytes. Overexpression of IL-22 by hydrodynamic gene delivery significantly protects against liver injury, necrosis, and apoptosis induced by administration of Con A, carbon tetrachloride (CCl4), or the Fas agonist Jo-2 mAb. Western blot analyses show that overexpression of IL-22 significantly enhances activation of STAT3 and expression of Bcl-xL, Bcl-2,and Mcl-1 proteins in liver injury induced by Con A. In conclusion, hydrodynamic gene delivery of IL-22 protects against liver injury induced by a variety of toxins, suggesting the therapeutic potential of IL-22 in treating human liver disease. | 2 | Hepatitis B Virus X Protein Sensitizes Primary Mouse Hepatocytes to Ethanol-and TNF-α-Induced Apoptosis by a Caspase-3-Dependent Mechanism显示文摘It is well-documented that alcohol drinking together with hepatitis viral infection accelerates liver injury; however the underlying mechanisms remain unknown. In this paper, we demonstrated that primary hepatocytes from transgenic mice overexpressing hepatitis B virus X protein (HBX) were more susceptible to ethanol- and TNF-α-induced apoptotic killing. Compared to normal control mouse hepatocytes, ethanol and/or TNF-α treatment led to a significant increase in reactive oxygen species, mitochondrial permeability transition, cytochrome C release,caspase-3 activity, and poly (ADP-ribose) polymerase degradation in hepatocytes from HBX transgenic mice.Blocking caspase-3 activity antagonized ethanol- and TNF-α-induced apoptosis in primary hepatocytes from HBX transgenic mice. | 5 | Biodiesel Production from Waste Edible Oils and Grease Containing Free Fatty Acids显示文摘Till now, most part of the biodiesel is produced from the refined vegetable oils using methanol as feedstock in the presence of an alkali catalyst. However, large amount of waste edible oils and grease are available. The difficulty with alkali-catalyzed esterification of these oils is that they often contain large amount of free fatty acids (FFA), polymers and decomposition products. These free fatty acids can quickly react with the alkali catalyst to produce soaps that inhibit the separation of the ester and glycerine. An esterification and transesterification process is developed to convert the high FFA oil to its monoesters. The first step, the acidcatalyzed esterification with glycerine and these FFA reduces the FFA content of the oil and grease to less than3%, and then an azeotropic distillation solvent is used to remove the water. The major factors affecting the conversion efficiency of the process such as glycerol to free fatty acid molar ratio, catalyst amount, reaction temperature and reaction duration are analyzed. The second step, alkali-catalyzed transesterification process converts the products of the first step to its monoesters and glycerol, and then the glycerol is recycled for utilization in the first step. | 8 | Free fatty acids profiling in cold-pressed rapeseed oil pretreated by microwave显示文摘This study proposed a strategy based on simple chemical derivatization coupled with neutral loss scan tandem to electrospray ionization source of mass spectrometer (NLS-ESI-MS/MS) for rapid and sensitive detection of free fatty acids (FFAs) in coldpressed rapeseed oils without sample purification and enrichment. The method was applied to investigate contents of FFAs and their changes in cold-pressed rapeseed oil pretreated by microwave. Results showed that main FFAs in rapeseed oil control samples were myristic acid (C14:0), palmitic acid (C16:0), stearic acid (C18:0), oleic acid (C18:1), linoleic acid (C18:2) and linolenic acid (C18:3). The highest content of C18:1 was 7163.69 nmol/g and C18:2 was 5221.18 nmol/g among 8 oil samples. FFAs generally increased after microwave pretreatment except C14:0 in oil sample of ‘Xinyou 17’ and ‘Suyou 6’. C18:1 and C18:2 significantly changed (P<0.05) after microwave pretreatment. With this established simple,practical and reliable analysis approach, changes of FFAs would be monitored to ensure safety and quality of edible oils. | 10 | Microencapsulation of nutmeg essential oil by spray-drying with different wall materials显示文摘Nutmeg essential oil with volatile components is sensitive to oxygen, light, water and high temperature. Spray-drying is widely used as an encapsulation technique for improving compound stability during processing and storage. In this study, influence of encapsulated matrix on spray-dried nutmeg essential oil microcapsule was investigated using blends of maltodextrin and either gum Arabic(GA-MD), or octenyl succinic acid modified starch(OSA-MD) as wall materials. Characteristics of spray-dried powders from these 2 wall materials were evaluated and compared. Powder property analysis indicated that GA-MD microcapsules presented larger size, higher moisture and poorer flowability compared with OSA-MD microcapsules. Fourier transforms infrared spectroscopy and thermogravimetric analysis showed that both GA-MD and OSA-MD microcapsules improved the stability of nutmeg essential oil. However, GA-MD microcapsules exhibited higher storage stability than OSA-MD microcapsules. | 12 | Physicochemical properties and volatile components of hempseed oils in Bama region显示文摘A comparison study has been performed in hempseed oils (HSO) extracted from different cultivars in Bama region. Squeezing properties, chemical compositions, thermal stability and flavor profile of pressed HSO were determined in this study. Results indicated that there were no significant differences in crude fat, protein, fiber of different hempseeds and fatty acid composition in HSOs. Hempseeds with small size and dark-colored peel have lower squeezing oil yield than big ones. Average content of total phytosterol, tocopherol and phenol in HSO from small seeds were 872.0 mg/100 g, 124.1 mg/100 g and 15.5 mg SA/100 g respectively. Thermal oxidation by differential scanning calorimetry (DSC) revealed three step oxidation of HSO with mean onset and oxidation temperature at 124oC and 135-315oC. Oxidation temperature of HSO from small seeds was higher than that from big seeds (big size and light-colored peel). Main volatile ingredients in HSO contained 11 kinds of monoterpene and 7 kinds of sesquiterpene. Volatile terpene content in HSO from small seeds was nearly 30% higher than that of big seeds. Quality of HSO with different peel feature has significant difference in squeezing yield, thermal stability and volatile flavors. | 13 | Epidemiology and prevention of hepatitis B virus in China显示文摘Hepatitis B virus (HBV) infection is prevalent in China. Approximately 600 million people have ever been infected by HBV. About 130 million are HBV chronic carriers and 30 million HB patients. Among them, 50% of HBV carriers are caused by carrier mothers to born infants. Around 300 000 people died of liver disease including liver cirrhosis and primary hepatocellular carcinoma each year and 50% of them died of primary hepatocellular carcinoma. HBV infection is not only the health problem but also becoming a social problem. HBV chronic carriers and patients have endured the great pressure from disease burden and social discrimination. According to the report of the national screening program of HBV released by the ministry of health in 2008, China has taken many effective measures to control the HBV infection, including vaccine immunization program, strengthening the management of blood sources and blood productions, prevention of nosocomial HBV infection, strengthening health education on HBV infection and safe injection techniques. The implementation of HB vaccine immunization program, which China officially introduced into the national immunization program since 1992, has dramatically reduced the incidence of HBV infection among infants and children. Integrated with other interventions, the rate of HBV infection decreased gradually. According to the survey of the national screening program of HBV in 2006, compared with the incidence of HBV in 1992, the incidence rate of HBsAg positive has decreased 26.36% , the number of children who have ever been infected by HBV decreased 80 million since 1992. However some problems are still existing. | 14 | Study on the protection of Er-doped phosphate glass waveguide surface in ion-exchange processing显示文摘A novel method, sputtering K9 glass film, is proposed to solve the surface corrosion of Er-doped phosphate glass during ion-exchange processing for optical waveguide fabrication. The corrosion causes are analyzed to be the intrinsically weak stabilization of phosphate glass structure, hydrophile and weakly acidic property of phosphate radical. Experimental results show that the K9 glass film could not only protect the Er-doped phosphate glass surface from being corroded but also give no influence on the waveguide fabrication. The effect of thickness of K9 glass film on the optical property of waveguide is also investigated and the op- timal thickness is found to be 60―80 nm. | 15 | Arsenic exposure and glutamate-induced gliotransmitter release from astrocytes显示文摘The present study used cultures of primary astrocytes, isolated from neonatal rats, to verify the hypothesis that arsenite-induced neurotoxicity can influence neuronal function by altering glutamate-induced gliotransmitter release. Primary astrocytes were exposed to 0, 2.5, 5, 10, 20 or 30 μM arsenite for 24 hours. Cell viability and morphological observations revealed that 5 μM arsenic exposure could induce cytotoxicity. Cells were then cultured in the presence of 0, 2.5, 5, or 10 μM arsenite for 24 hours and stimulated with 25 μM glutamate for 10 minutes. Results showed that [Ca2+]i in astrocytes exposed to 5 and 10 μM arsenite was significantly increased and levels of D-serine, γ-aminobutyric acid and glycine in cultures exposed to 2.5-10 μM arsenite were also increased. However, glutamate levels in the media were significantly increased only after treatment with 10 μM arsenite. Based on the methods of finite element analysis,the comparative analysis of these two cable-stayed bridge schemes about static performances,dynamic performances,stability and wind resistance behavior were carried out. | 20 | Cellular antioxidant activity and cytotoxicity assay of canolol显示文摘To better understand canolol as an efficient intracellular antioxidant agent in food related fields, potential cellular antioxidant activity and cytotoxicity were evaluated using multiple assays, including cellular antioxidant activity(CAA), 3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-di-phenytetrazoliumromide(MTT) and methylene blue assay. CAA assay was used to identify the half maximal effective concentration of canolol and other bioactive components, such as sinapic acid, resveratrol and sesamol in oil crops. Results showed that antioxidant capacity(without PBS wash) varies between different compounds. Capacity variation followed the order: sinapic acid > canolol > resveratrol > sesamol. While with PBS wash, the order changed to sinapic acid > resveratrol > canolol > sesamol. Half maximal inhibitory concentration of canolol measured by MTT was 1887.08±1.22 μmol/L, implying that cytotoxicity of canolol was greatly lower than that of resveratrol, sesamol and sinapic acid. Methylene blue assay showed that cell viability was still up to 90% exposed to canolol at a high concentration of 4444 μmol/L.
