| 1 | Effect of cDNA fragments in different length derived from Potato Virus Y coat protein gene on the induction of RNA-mediated virus resistance显示文摘We have reported that cDNA derived from entire coat protein (CP) gene of potato virus Y (PVY) could induce resistance to PVY infection in transgenic tobacco plants, and the resistance was further demonstrated to be RNA-mediated rather than coat protein-mediated. In this study, we cloned cDNA fragments of 202 bp, 417 bp, and 603 bp in length derived from the 3′ end of the PVY CP gene, and the cDNA fragments were introduced into tobacco (var. NC89) plants via Agrobacterium-mediated transformation system. The results of resistance assay showed that the CP cDNA fragments of 417 bp and 603 bp could confer resistance of the trans-genic plants to PVY infection, but the fragment of 202 bp in length could not. Molecular analysis revealed that the resistance was RNA-mediated, which is believed to be a result of post-transcriptional gene silencing. | 2 | Overexpression of phospholipase Dα gene enhances drought and salt tolerance of Populus tomentosa显示文摘The cDNA of AtPLDα (Arabidopsis thaliana Phospholipase Dα) gene was introduced into P. tomentosa (Populus tomentosa) under the control of the Cauliflower mosaic virus 35S promoter. Southern and Northern blot analyses suggested that the AtPLDα gene has been transferred into the P. tomentosa genome. No obvious morphological or developmental difference was observed between the transgenic and wild-type (WT) plants. Drought and salt tolerance and gene expression of seedlings of several transgenic lines and WT plants (control) were studied. The results showed that the rhizogenesis rate and the average root-length of transgenic lines were significantly higher than WT plants after mannitol and NaCl treatment under the same growth conditions. Northern blot analysis indicated that the higher the PLDα expression in the transgenic plants, the more tolerant the transgenic plants are to drought and salt treatment. Meanwhile, another group of these transgenic lines and WT plants (control) were treated with PEG6000 and NaCl separately. The contents of chlorophylls and the activities of some anti-oxidant enzymes (superoxide dismutase, guaiacol peroxidase and catalase) as well as malondialde-hyde and relative electrical conductivity were analyzed. | 4 | Isolation of a strong matrix attachment region (MAR) and identification of its function in vitro and in vivo显示文摘Inclusion of MARs in transgene cassettes enhances their expression and reduces position-effect variations in the transgenic host. Four new MARs (TM2, TM3, AM1 and AM2) were isolated from tobacco and Arabidopsis by PCR method. The nuclei isolated from suspension-cultured cells of rice were used to prepare nuclear matrix. With a characterized MAR (TM1) as a positive control, the Matrix-MAR interactions were tested by an in vitro binding assay to identify the DNA sequences as MARs and their binding strength to nuclear matrix in vitro was compared. The results showed that TM2 and TM3 had stronger binding strength than TM1. To determine the functions of the four new MARs in vivo, binary vectors pBI121 carrying a uidA GUS reporter gene were modified with direct repeat MARs inserted on both sides of the reporter gene cassette and were transferred into tobaccos via Agrobacterium-mediated transformation procedure. | 9 | Towards sustainable protection against insect-borne plant viral diseases:Phytohormones and beyond显示文摘Due to the immobility of plants,around 75% of 1100 plant virus species are piercing-sucking insect transmitted.Host plant-mediated interactions between viruses and insects play vital roles in the population dynamics of vectors and the epidemiology of plant diseases.A successful viral pathogen has to evolve multiple strategies to manipulate host immune responses and also the ecological environment to facilitate effective transmission by insect vectors.Among these strategies,reprogramming the phytohormone signaling pathways is critical to the establishment of an effective virus transmission among plants and disease pandemic.Here,we review recent studies on the plant-virus inter-relationships with a focus on molecular and biochemical mechanisms that drive vector-borne viral diseases.Defense-related phytohormones such as Jasmonic acid(JA),Salicylic acid(SA)and Ethylene(ET)have been reported to be directly regulated by viral proteins.This knowledge is essential for the further design and/or development of effective and sustainable strategies to protect viral damages so as to increase crop yield and food security.Future efforts in this area should be focused on integrating and meta-analysis of big data generating from dynamics and multiple dimensional pathogen-vector-crop interactions under real agricultural conditions to achieve sustainable protection against plant diseases. | WANG Ning YAO XiangMei ZHU ChangXiang WEN FuJiang YE Jian | 2016 | Science Foundation in China2016,24,2: | 0 |
