| 1 | Baicalin induces ferroptosis in bladder cancer cells by downregulating FTH1显示文摘Ferroptosis is a non-apoptotic regulated cell death caused by iron accumulation and subsequent lipid peroxidation.Currently,the therapeutic role of ferroptosis on cancer is gaining increasing interest.Baicalin an active component in Scutellaria baicalensis Georgi with anticancer potential various cancer types;however,the effects of baicalein on bladder cancer and the underlying molecular mechanisms remain largely unknown.In the study,we investigated the effect of baicalin on bladder cancer cells5637 and KU-19-19.As a result,we show baicalin exerted its anticancer activity by inducing apoptosis and cell death in bladder cancer cells.Subsequently,we for the first time demonstrate baicalin-induced ferroptotic cell death in vitro and in vivo,accompanied by reactive oxygen species(ROS) accumulation and intracellular chelate iron enrichment.The ferroptosis inhibitor deferoxamine but not necrostatin-1,chloroquine(CQ),N-acetyl-L-cysteine,L-glutathione reduced,or carbobenzoxy-valyl-alanyl-aspartyl-[O-methyl]-fluoromethylketone(Z-VAD-FMK) rescued baicalin-induced cell death,indicating ferroptosis contributed to baicalin-induced cell death.Mechanistically,we show that ferritin heavy chain1(FTH1) was a key determinant for baicalin-induced ferroptosis.Overexpression of FTH1 abrogated the anticancer effects of baicalin in both 5637 and KU19-19 cells.Taken together,our data for the first time suggest that the natural product baicalin exerts its anticancer activity by inducing FTH1-dependent ferroptosis,which will hopefully provide a prospective compound for bladder cancer treatment. | Na Kong Xiaying Chen Jiao Feng Ting Duan Shuiping Liu Xueni Sun Peng Chen Ting Pan Lili Yan Ting Jin Yu Xiang Quan Gao Chengyong Wen Weirui Ma Wencheng Liu Mingming Zhang Zuyi Yang Wengang Wang Ruonan Zhang Bi Chen Tian Xie Xinbing Sui Wei Tao | 2021 | Acta Pharmaceutica Sinica B2021,11,12: | 23 |

| 4 | Stability influence factors analysis and construction of a deep beam anchorage structure in roadway roof显示文摘Deep beam anchorage structures based on spatial distribution analysis of the cable prestressed field have been proposed for roadway roof support. Stability and other factors that influence deep beam structures are studied in this paper using mechanical calculations, numerical analysis and field measurements. A mechanical model of deep beam structure subjected to multiple loading is established, including analysis of roof support in the return airway of S1203 working face in the Yuwu coal mine, China. The expression of maximum shear stress in the deep beam structure is deduced according to the stress superposition criterion. It is found that the primary factors affecting deep beam structure stability are deep beam thickness, cable pre-tension and cable spacing. The variation of maximum shear stress distribution and prestressed field diffusion effects according to various factors are analyzed using Matlabòand FLAC3D^(TM) software, and practical support parameters of the S1203 return airway roof are determined.According to the observations of rock pressure, there is no evidence of roof separation, and the maximum values of roof subsidence and convergence of wall rock are 72 and 48 mm, respectively. | 6 | Characteristics of Late-Quaternary Activity and Seismic Risk of the Northeastern Section of the Longmenshan Fault Zone显示文摘Following the 2008 Wenchuan M8 earthquake,the seismic risk of the northeastern section of the Longmenshan fault zone and the adjacent Hanzhong basin has become an issue that receives much concern.It is facing,however,the problem of a lack of sufficient data because of little previous work in these regions.The northeastern section of the Longmenshan fault zone includes three major faults:the Qingchuan fault,Chaba-Lin’ansi fault,and Liangshan south margin fault,with the Hanzhong basin at the northern end.This paper presents investigations of the geometry,motion nature,and activity ages of these three faults,and reveals that they are strike slip with normal faulting,with latest activity in the Late Pleistocene.It implies that this section of the Longmenshan fault zone has been in an extensional setting,probably associated with the influence of the Hanzhong basin.Through analysis of the tectonic relationship between the Longmenshan fault zone and the Hanzhong basin,this work verifies that the Qingchuan fault played an important role in the evolution of the Hanzhong basin,and further studies the evolution model of this basin.Finally,with consideration of the tectonic setting of the Longmenshan fault zone and the Hanzhong basin as well as seismicity of surrounding areas,this work suggests that this region has no tectonic conditions for great earthquakes and only potential strong events in the future. | WANG Mingming ZHOU Bengang YANG Xiaoping XIE Chao GAO Xianglin | 2013 | Acta Geologica Sinica(English Edition)2013,87,6: | 6 |

| 8 | Lactobacillus reuteri-derived extracellular vesicles maintain intestinal immune homeostasis against lipopolysaccharideinduced inflammatory responses in broilers显示文摘Background:Lactobacillus reuteri strains are widely used as probiotics to prevent and treat inflammatory bowel disease by modulating the host’s immune system.However,the underlying mechanisms by which they communicate with the host have not been clearly understood.Bacterial extracellular vesicles(EVs)have been considered as important mediators of host-pathogen interactions,but their potential role in commensals-host crosstalk has not been widely studied.Here,we investigated the regulatory actions of EVs produced by L.reuteri BBC3,a gut-associated commensal bacterium of Black-Bone chicken,in the development of lipopolysaccharide(LPS)-induced intestinal inflammation in a chicken model using both in vivo and in vitro experiments.Results:L.reuteri BBC3 produced nano-scale membrane vesicles with the size range of 60-250 nm.Biochemical and proteomic analyses showed that L.reuteri BBC3-derived EVs(LrEVs)carried DNA,RNA and several bioactive proteins previously described as mediators of other probiotics’beneficial effects such as glucosyltransferase,serine protease and elongation factor Tu.In vivo broiler experiments showed that administration of LrEVs exerted similar effects as L.reuteri BBC3 in attenuating LPS-induced inflammation by improving growth performance,reducing mortality and decreasing intestinal injury.LrEVs suppressed the LPS-induced expression of pro-inflammatory genes(TNF-α,IL-1β,IL-6,IL-17 and IL-8),and improved the expression of anti-inflammatory genes(IL-10 and TGF-β)in the jejunum.LrEVs could be internalized by chicken macrophages.In vitro pretreatment with LrEVs reduced the gene expression of TNF-α,IL-1βand IL-6 by suppressing the NF-κB activity,and enhanced the gene expression of IL-10 and TGF-βin LPS-activated chicken macrophages.Additionally,LrEVs could inhibit Th1-and Th17-mediated inflammatory responses and enhance the immunoregulatory cells-mediated immunosuppression in splenic lymphocytes of LPS-challenged chickens through the activation of macrophages.Finally,we revealed that the reduced content of both vesicular proteins and nucleic acids attenuated the suppression of LrEVs on LPS-induced inflammatory responses in ex vivo experiments,suggesting that they are essential for the LrEVs-mediated immunoregulation.Conclusions:We revealed that LrEVs participated in maintaining intestinal immune homeostasis against LPSinduced inflammatory responses in a chicken model.Our findings provide mechanistic insight into how commensal and probiotic Lactobacillus species modulate the host’s immune system in pathogens-induced inflammation. | Rujiu Hu Hua Lin Mimi Wang Yuezhen Zhao Haojing Liu Yuna Min Xiaojun Yang Yupeng Gao Mingming Yang | 2021 | Journal of Animal Science and Biotechnology2021,12,3: | 5 |
| 9 | A Volumetric Model for Evaluating Tight Sandstone Gas Reserves in the Permian Sulige Gas Field,Ordos Basin,Central China显示文摘To accurately measure and evaluate reserves is critical for ensuring successful production of unconventional oil and gas. This work proposes a volumetric model to evaluate the tight sandstone gas reserves of the Permian Sulige gas field in the Ordos Basin. The reserves can be determined by four major parameters of reservoir cutoffs, net pay, gas-bearing area and compression factor Z, which are controlled by reservoir characteristics and sedimentation. Well logging, seismic analysis, core analysis and gas testing, as well as thin section identification and SEM analysis were used to analyze the pore evolution and pore-throat structure. The porosity and permeability cutoffs are determined by distribution function curve,empirical statistics and intersection plot. Net pay and gas-bearing area are determined based on the cutoffs, gas testing and sand body distribution, and the compression factor Z is obtained by gas component. The results demonstrate that the reservoir in the Sulige gas field is characterized by ultralow porosity and permeability, and the cutoffs of porosity and permeability are 5% and 0.15×10^(–3) μm^2, respectively. The net pay and gas-bearing area are mainly affected by the sedimentary facies, sand body types and distribution. The gas component is dominated by methane which accounts for more than 90%, and the compression factor Z of H_8(P_2h_8) and S_1(P_1s_1) are 0.98 and 0.985, respectively. The distributary channels stacked and overlapped, forming a wide and thick sand body with good developed intergranular pores and intercrystalline pores. The upper part of channel sand with good porosity and permeability can be sweet spot for gas exploration. The complete set of calculation systems proposed for tight gas reserve calculation has proved to be effective based on application and feedback. | 10 | Qualitatively and quantitatively assessing the aggregation ability of sludge during aerobic granulation process combined XDLVO theory with physicochemical properties显示文摘Inexact mechanism of aerobic granulation still impedes optimization and application of aerobic granules. In this study, the extended Derjaguin, Landau, Verwey, and Overbeek(XDLVO) theory and physicochemical properties were combined to assess the aggregation ability of sludge during aerobic granulation process qualitatively and quantitatively. Results show that relative hydrophobicity of sludge and polysaccharide content of extracellular polymeric substances(EPS) increased, while electronegativity of sludge decreased during acclimation phase. After 20 days' acclimation, small granules began to form due to high aggregation ability of sludge. Since then, coexisted flocs and granules possessed distinct physicochemical properties during granulation and maturation phase. The relative hydrophobicity decreased while electronegativity increased for flocs, whereas that for granules presented reverse trend. Through analyzing the interaction energy using the XDLVO theory, small granules tended to self-grow rather than self-aggregate or attach of flocs due to poor aggregation ability between flocs and granules during the granulation phase. | 17 | A facile design for multifunctional AIEgen based on tetraaniline derivatives显示文摘Aniline oligomers have been widely used in many fields due to their excellent physicochemical properties. Owing to strong intermolecular interactions, their emission is always weakened or quenched when they are in high concentration or aggregated state, which greatly limits their fluorescent applications. Inspired by the concept of aggregation-induced emission(AIE), herein we introduced large steric groups onto the aniline oligomer to prevent the formation of packing structure. In particular, diphenyl vinyl group was bonded with oligomeric tetraaniline by a facile synthetic procedure with high yield. The obtained aniline oligomer derivative exhibited typical AIE features, which was also confirmed by density functional theoretical calculation. More importantly, this AIE oligomer was able to detect Fe^(3+) ions selectively and quantitatively. The fluorescence intensity decreased linearly along with the increment of Fe^(3+) concentration. Moreover, we demonstrated that this AIE oligomer could stain live bacteria, such as E. coli and S. aureus efficiently.
