7Prefrontal cortical circuits in anxiety and fear:an overview显示文摘Pathological anxiety is among the most difficult neuropsychiatric diseases to treat pharmacologically,and it represents a major societal problem.Studies have implicated structural changes within the prefrontal cortex(PFC)and functional changes in the communication of the PFC with distal brain structures in anxiety disorders.Treatments that affect the activity of the PFC,including cognitive therapies and transcranial magnetic stimulation,reverse anxiety-and fear-associated circuit abnormalities through mechanisms that remain largely unclear.While the subjective experience of a rodent cannot be precisely determined,rodent models hold great promise in dissecting well-conserved circuits.Newly developed genetic and viral tools and optogenetic and chemogenetic techniques have revealed the intricacies of neural circuits underlying anxiety and fear by allowing direct examination of hypotheses drawn from existing psychological concepts.This review focuses on studies that have used these circuit-based approaches to gain a more detailed,more comprehensive,and more integrated view on how the PFC governs anxiety and fear and orchestrates adaptive defensive behaviors to hopefully provide a roadmap for the future development of therapies for pathological anxiety.Yihua Chen Nengyuan Hu Jianming Yang Tianming Gao 2022Frontiers of Medicine2022,16,4:1

13Prediction of cognitive scores by joint use of movie-watching fMRI connectivity and eye tracking via Attention-CensNet显示文摘Background:Brain functional connectivity under the naturalistic paradigm has been shown to be better at predicting individual behaviors than other brain states,such as rest and doing tasks.Nevertheless,the state-of-the-art methods have found it difcult to achieve desirable results from movie watching paradigm functional magnetic resonance imaging(mfMRI)-induced brain functional connectivity,especially when there are fewer datasets.Incorporating other physical measurements into the prediction method may enhance accuracy.Eye tracking,becoming popular due to its portability and lower expense,can provide abundant behavioral features related to the output of human's cognition,and thus might supplement the mfMRI in observing participants'subconscious behaviors.However,there are very few studies on how to effectively integrate the multimodal information to strengthen the performance by a unified framework.objective:A fusion approach with mfMRI and eye tracking,based on convolution with edge-node switching in graph neural networks(CensNet),is proposed in this article.Methods:In this graph model,participants are designated as nodes,mfMRI derived functional connectivity as node features,and different eye-tracking features are used to compute similarity between participants to construct heterogeneous graph edges.By taking multiple graphs as different channels,we introduce squeeze-and-excitation attention module to CensNet(A-CensNet)to integrate graph embeddings from multiple channels into one.Results:The proposed model outperforms those using a single modality and single channel,and state-of-the-art methods.Conclusions:The results indicate that brain functional activities and eye behaviors might complement each other in interpreting trait-like phenotypes.Jiaxing Gao Lin Zhao Tianyang Zhong Changhe Li Zhibin He Yaonei Wei Shu Zhang Lei Guo Tianming Liu Junwei Han Xi Jiang Tuo Zhang 2023Psychoradiology2023,3,1:0
14Targeting cAMP in D1-MSNs in the nucleus accumbens, a new rapid antidepressant strategy显示文摘Studies have suggested that the nucleus accumbens(NAc)is implicated in the pathophysiology of major depression;however,the regulatory strategy that targets the NAc to achieve an exclusive and outstanding anti-depression benefit has not been elucidated.Here,we identified a specific reduction of cyclic adenosine monophosphate(cAMP)in the subset of dopamine D1 receptor medium spiny neurons(D1-MSNs)in the NAc that promoted stress susceptibility,while the stimulation of cAMP production in NAc D1-MSNs efficiently rescued depression-like behaviors.Ketamine treatment enhanced cAMP both in D1-MSNs and dopamine D2 receptor medium spiny neurons(D2-MSNs)of depressed mice,however,the rapid antidepressant effect of ketamine solely depended on elevating cAMP in NAc D1-MSNs.We discovered that a higher dose of crocin markedly increased cAMP in the NAc and consistently relieved depression 24 h after oral administration,but not a lower dose.The fast onset property of crocin was verified through multicenter studies.Moreover,crocin specifically targeted at D1-MSN cAMP signaling in the NAc to relieve depression and had no effect on D2-MSN.These findings characterize a new strategy to achieve an exclusive and outstanding anti-depression benefit by elevating cAMP in D1-MSNs in the NAc,and provide a potential rapid antidepressant drug candidate,crocin.Yue Zhang Jingwen Gao Na Li Peng Xu Shimeng Qu Jinqian Cheng Mingrui Wang Xueru Li Yaheng Song Fan Xiao Xinyu Yang Jihong Liu Hao Hong Ronghao Mu Xiaotian Li Youmei Wang Hui Xu Yuan Xie Tianming Gao Guangji Wang Jiye Aa 2024Acta Pharmaceutica Sinica B2024,14,2:0
15Functional diversity of subgroup 5 R2R3-MYBs promoting proanthocyanidin biosynthesis and their key residues and motifs in tea plant显示文摘The tea plant(Camellia sinensis)is rich in polyphenolic compounds.Particularly,flavan-3-ols and proanthocyanidins(PAs)are essential for the flavor and disease-resistance property of tea leaves.The fifth subgroup of R2R3-MYB transcription factors comprises the primary activators of PA biosynthesis.This study showed that subgroup 5 R2R3-MYBs in tea plants contained at least nine genes belonging to the TT2,MYB5,and MYBPA types.Tannin-rich plants showed an expansion in the number of subgroup 5 R2R3-MYB genes compared with other dicotyledonous and monocot plants.The MYBPA-type genes of tea plant were slightly expanded.qRT–PCR analysis and GUS staining analysis of promoter activity under a series of treatments revealed the differential responses of CsMYB5s to biotic and abiotic stresses.In particular,CsMYB5a,CsMYB5b,and CsMYB5e responded to high-intensity light,high temperature,MeJA,and mechanical wounding,whereas CsMYB5f and CsMYB5g were only induced by wounding.Three genetic transformation systems(C.sinensis,Nicotiana tabacum,and Arabidopsis thaliana)were used to verify the biological function of CsMYB5s.The results show that CsMYB5a,CsMYB5b,and CsMYB5e could promote the gene expression of CsLAR and CsANR.However,CsMYB5f and CsMYB5g could only upregulate the gene expression of CsLAR but not CsANR.A series of site-directed mutation and domain-swapping experiments were used to verify functional domains and key amino acids of CsMYB5s responsible for the regulation of PA biosynthesis.This study aimed to provide insight into the induced expression and functional diversity model of PA biosynthesis regulation in tea plants.Tianming Jiao Yipeng Huang Ying-Ling Wu Ting Jiang Tongtong Li Yanzhuo Liu Yvchen Liu Yunyun Han Yajun Liu Xiaolan Jiang Liping Gao Tao Xia 2023Horticulture Research2023,10,8:0
16Amplitude Ratio of Regional Seismic Phases: An Efficient Indicator of Thermal State of the Lithosphere显示文摘By analyzing seismograms of short period records at the Beijing SeismoJogicaJ Observatory, the present paper investigates the amplitude ratio of seismic phases. The results indicate that the amplitude ratio of Sn/Lg is correlated with the lithosphere structure, the thermal state, and strong earthquake occurrence in the region the seismic rays pass through. 17Distinct and shared endothermic strategies in the heat producing tissues of tuna and other teleosts显示文摘Although most fishes are ectothermic,some,including tuna and billfish,achieve endothermy through specialized heat producing tissues that are modified muscles.How these heat producing tissues evolved,and whether they share convergent molecular mechanisms,remain unresolved.Here,we generated a high-quality genome from the mackerel tuna(Euthynnus affinis)and investigated the heat producing tissues of this fish by single-nucleus and bulk RNA sequencing.Compared with other teleosts,tuna-specific genetic variation is strongly associated with muscle differentiation.Single-nucleus RNA-seq revealed a high proportion of specific slow skeletal muscle cell subtypes in the heat producing tissues of tuna.Marker genes of this cell subtype are associated with the relative sliding of actin and myosin,suggesting that tuna endothermy is mainly based on shivering thermogenesis.In contrast,cross-species transcriptome analysis indicated that endothermy in billfish relies mainly on nonshivering thermogenesis.Nevertheless,the heat producing tissues of the different species do share some tissue-specific genes,including vascular-related and mitochondrial genes.Overall,although tunas and billfishes differ in their thermogenic strategies,they share similar expression patterns in some respects,highlighting the complexity of convergent evolution.Baosheng Wu Xueli Gao Mingling Hu Jing Hu Tianming Lan Tingfeng Xue Wenjie Xu Chenglong Zhu Yuan Yuan Jiangmin Zheng Tao Qin Peidong Xin Ye Li Li Gong Chenguang Feng Shunping He Huan Liu Haimeng Li Qing Wang Zhenhua Ma Qiang Qiu Kun Wang 2023Science China(Life Sciences)2023,66,11:0
18Two memory associated genes regulated by amyloid precursor protein intracellular domain Novel insights into the pathogenesis of learning and memory impairment in Alzheimer's disease显示文摘In this study,we employed chromatin immunoprecipitation,a useful method for studying the locations of transcription factors bound to specific DNA regions in specific cells,to investigate amyloid precursor protein intracellular domain binding sites in chromatin DNA from hippocampal neurons of rats,and to screen out five putative genes associated with the learning and memory functions.The promoter regions of the calcium/calmodulin-dependent protein kinase II alpha and glutamate receptor-2 genes were amplified by PCR from DNA products immunoprecipitated by amyloid precursor protein intracellular domain.An electrophoretic mobility shift assay and western blot analysis suggested that the promoter regions of these two genes associated with learning and memory were bound by amyloid precursor protein intracellular domain(in complex form).Our experimental findings indicate that the amyloid precursor protein intracellular domain is involved in the transcriptional regulation of learning-and memory-associated genes in hippocampal neurons.These data may provide new insights into the molecular mechanism underlying the symptoms of progressive memory loss in Alzheimer's disease.Chuandong Zheng Xi Gu Zhimei Zhong Rui Zhu Tianming Gao Fang Wang 2012Neural Regeneration Research2012,7,5:0
