1Characterization of the essential role of bone morphogenetic protein 9 (BMP9) in osteogenic differentiation of mesenchymal stem cells (MSCs) through RNA interference显示文摘Mesenchymal stem cells(MSCs)are multipotent stem cells and capable of differentiating into multiple cell types including osteoblastic,chondrogenic and adipogenic lineages.We previously identified BMP9 as one of the most potent BMPs that induce osteoblastic differentiation of MSCs although exact molecular mechanism through which BMP9 regulates osteogenic differentiation remains to be fully understood.Here,we seek to develop a recombinant adenovirus system to optimally silence mouse BMP9 and then characterize the important role of BMP9 in osteogenic differentiation of MSCs.Using two different siRNA bioinformatic prediction programs,we design five siRNAs targeting mouse BMP9(or simB9),which are expressed under the control of the converging H1 and U6 promoters in recombinant adenovirus vectors.We demonstrate that two of the five siRNAs,simB9-4 and simB9-7,exhibit the highest efficiency on silencing exogenous mouse BMP9 in MSCs.Furthermore,simB9-4 and simB9-7 act synergistically in inhibiting BMP9-induced expression of osteogenic markers,matrix mineralization and ectopic bone formation from MSCs.Thus,our findings demonstrate the important role of BMP9 in osteogenic differentiation of MSCs.The characterized simB9 siRNAs may be used as an important tool to investigate the molecular mechanism behind BMP9 osteogenic signaling.Our results also indicate that recombinant adenovirus-mediated expression of siRNAs is efficient and sustained,and thus may be used as an effective delivery vehicle of siRNA therapeutics.Shujuan Yan Ruyi Zhang Ke Wu Jing Cui Shifeng Huang Xiaojuan Ji Liping An Chengfu Yuan Cheng Gong Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Bo Liu Rex C.Haydon Michael J.Lee Russell R.Reid Jennifer Moriatis Wolf Qiong Shi Hue H.Luu Tong-Chuan He Yaguang Weng 2018Genes & Diseases2018,5,2:8
2Mean water level setup/setdown in the inlet-lagoon system induced by tidal action—a case study of Xincun Inlet,Hainan Island in China显示文摘With the tides propagating from the open sea to the lagoon,the mean water level (MWL) in the inlet and lagoon becomes different from that at the open sea,and a setup/setdown is generated. The change of MWL (setup/setdown) in the system imposes a great impact on regulating the development of tidal marshes,on determining the long-term water level for harbor maintenance,on the planning for the water front development with the flood control for the possible inundation,and on the interpretation of the historical sea level change when using tidal marsh peat deposits in the lagoon as the indicator for open sea's sea level. In this case study on the mechanisms which control the setup/setdown in Xincun Inlet,Hainan in China,the 2-D barotropic mode of Eulerian-Lagrangian CIRCulation (ELCIRC) model was utilized. After model calibration and verification,a series of numerical experiments were conducted to examine the effects of bottom friction and advection terms,wetting and drying of intertidal areas,bathymetry and boundary conditions on the setup/setdown in the system. The modeling results show that setup occurs over the inlet and lagoon areas with an order of one tenth of the tide range at the entrance. The larger the bottom friction is,a larger setup is generated. Without the advection term,the setup is reduced due to a decrease of water level gradient to compensate for the disappearance of the advection term. Even without overtides,a setup can still be developed in the system. Sea level rise and dredging in the inlet and tidal channel can cause a decrease of setup in the system,whereas shoaling of the system can increase the setup. 3Multiobjective optimization using an immunodominance and clonal selection inspired algorithm显示文摘Based on the mechanisms of immunodominance and clonal selection theory,we propose a new multiobjective optimization algorithm,immune dominance clonal multiobjective algorithm(IDCMA).IDCMA is unique in that its fitness values of current dominated individuals are assigned as the values of a custom distance measure,termed as Ab-Ab affinity,between the dominated individuals and one of the nondominated individuals found so far.According to the values of Ab-Ab affin-ity,all dominated individuals(antibodies) are divided into two kinds,subdominant antibodies and cryptic antibodies.Moreover,local search only applies to the sub-dominant antibodies,while the cryptic antibodies are redundant and have no func-tion during local search,but they can become subdominant(active) antibodies during the subsequent evolution.Furthermore,a new immune operation,clonal proliferation is provided to enhance local search.Using the clonal proliferation operation,IDCMA reproduces individuals and selects their improved maturated progenies after local search,so single individuals can exploit their surrounding space effectively and the newcomers yield a broader exploration of the search space.The performance comparison of IDCMA with MISA,NSGA-Ⅱ,SPEA,PAES,NSGA,VEGA,NPGA,and HLGA in solving six well-known multiobjective function optimization problems and nine multiobjective 0/1 knapsack problems shows that IDCMA has a good performance in converging to approximate Pareto-optimal fronts with a good distribution.GONG MaoGuo JIAO LiCheng MA WenPing DU HaiFeng 2008Science in China(Series F)2008,51,8:6
4Modeling the circulation and sediment transport in the Beibu Gulf显示文摘Water circulation and sediment transport in the Beibu Gulf are important for its environmental protection and resource exploitation.By employing the Regional Ocean Modeling System(ROMS),we studied the seasonal variation of circulation,sediment transport and long-term morphological evolution in the Beibu Gulf.The simulation results show that the circulation induced by tide and wind is cyclonic both in winter and summer in the gulf and that the wind-driven circulation is stronger in winter than that in summer.The sediment concentration is higher in the Qiongzhou Strait,west of the Hainan Island and the coast of Vietnam and the Leizhou Peninsula.The sediment is transported westwards in winter and eastwards in summer in the Qiongzhou Strait.The west entrance of the Qiongzhou Strait is dominated by westward transport all the year round.The sediment discharged by rivers is deposited near the river mouths.The simulated result demonstrates that the sediment transport is mainly controlled by tidal induced bottom resuspension in the Beibu Gulf.Four characteristics are summarized for the distribution patterns of erosion and deposition.(1) The erosion and deposition are insignificant in most area of the gulf.(2) Sediment deposition is more significant in the mouths of Qiongzhou Strait.(3) The erosion is observed in the seabed of Qiongzhou Strait.(4) Erosion and deposition occur alternatively in the west of Hainan Island.CHENG Gaolei GONG Wenping WANG Yaping XIA Zhen 2017Acta Oceanologica Sinica2017,36,4:5
5Intelligent multi-user detection using an artificial immune system显示文摘Artificial immune systems (AIS) are a kind of new computational intelligence methods which draw inspiration from the human immune system. In this study, we introduce an AIS-based optimization algorithm, called clonal selection algorithm, to solve the multi-user detection problem in code-division multipleaccess communications system based on the maximum-likelihood decision rule. Through proportional cloning, hypermutation, clonal selection and clonal death, the new method performs a greedy search which reproduces individuals and selects their improved maturated progenies after the affinity maturation process. Theoretical analysis indicates that the clonal selection algorithm is suitable for solving the multi-user detection problem. 7Immunodominance and clonal selection inspired multiobjective clustering显示文摘The biological immune system is a highly parallel and distributed adaptive system. The information processing abilities of the immune system provide important insights into the field of computation. Based on immunodominance in the biological immune system and the clonal selection mechanism, a novel data mining method, Immune Dominance Clonal Multiobjective Clustering algorithm (IDCMC), is presented. The algorithm divides an individual population into three sub-populations according to three different measurements, and adopts different evolution and selection strategies for each sub-population. The update of each sub-population, however, is not carried out in isolation. The periodic combination operation of the analysis of the three sub-populations represents considerable advantages in its global search ability. The clustering task is a multiobjective optimization problem, which is more robust with respect to the variety of cluster structures of different datasets than a single-objective clustering algorithm. In addition, the new algorithm can determine the number of clusters automatically, which should identify the most promising clustering solutions in the candidate set. 8Protective immunity against Rickettsia heilongjiangensis in a C3H/HeN mouse model mediated by outer membrane protein B-pulsed dendritic cells显示文摘Rickettsia heilongjiangensis is an obligate intracellular bacterium that causes Far-Eastern tick-borne spotted fever. Outer membrane protein B(Omp B) is an important surface protein antigen of rickettsiae. In the present study, the omp B gene of R. heilongjiangensis was divided into four fragments, resulting in four recombinant proteins(OmpB-p1, Omp B-p2, Omp B-p3, and Omp B-p4). Each Omp B was used in vitro to stimulate murine bone marrow-derived dendritic cells(BMDCs) of C3H/He N mice, and the Omp B-pulsed BMDCs were transferred to naive C3H/He N mice. On day 14 post-transfer of BMDCs, the mice were challenged with R. heilongjiangensis and the rickettsial loads in the mice were quantitatively determined on day 7 post-challenge. Mice receiving BMDCs pulsed with Omp B-p2, Omp B-p3, or Omp B-p4 exhibited significantly lower bacterial load compared with mice receiving Omp B-p1-pulsed BMDCs. CD4+ and CD8+ T cells isolated from the spleen of C3H/He N mice receiving BMDCs pulsed with each OmpB were co-cultured with BMDCs pulsed with the respective cognate protein. In flow cytometric analysis, the expression level of CD69 on CD4+ or CD8+ T cells from mice receiving BMDCs pulsed with Omp B-p2, OmpB-p3, or Omp B-p4 was higher than that on cells from mice receiving Omp B-p1-pulsed BMDCs, while the expression level of tumor necrosis factor(TNF)-α on CD8+ T cells and interferon(IFN)-γ on the CD4+ and CD8+ T cells from mice receiving Omp B-p2,-p3, or-p4 was significantly higher than on cells from mice receiving Omp B-p1-pulsed BMDCs. 9Feedback between tidal hydrodynamics and morphological changes induced by natural process and human interventions in a wave-dominated tidal inlet: Xiaohai, Hainan, China显示文摘The feedback between morphological evolution and tidal hydrodynamics in a wave-dominated tidal inlet, Xiaohai, China is investigated through data analysis and numerical model experiments. Historically, Xiaohai Inlet had two openings, located at the north and south of Neizhi Island (a rocky outcrop), respectively. The evolution of Xiaohai Inlet was dominated by the natural process before 1972. In addition to the natural process, human interventions, including the closure of the north opening, 50% of freshwater reduction, and increase of land reclamation, have altered tidal hydrodynamics and morphological evolution since 1972. A series of numerical model simulations were conducted to investigate the influence of morphological changes on the hydrodynamics and the influence of human activities on the inlet evolution. The natural process has caused narrowing and shoaling of the inlet throat, development of the flood-tidal delta, and shoaling of the tidal channel inside the lagoon. Human interventions have accelerated these changes. Consequently, the tidal propagation from the offshore into the lagoon has been impeded and the tidal energy has been dissipated substantially. Tidal current has changed from ebb-dominant to flood-dominant in most parts of the inlet system whereas the inlet throat has remained as ebb-dominant, the tidal prism has decreased consistently, and sediment has continued to deposit inside the inlet. As a result, the changes of morphology, hydrodynamics, and sediment transport show a positive feedback. The human interventions have had both advantageous and adverse influences on the stability of the inlet. The closure of the North Opening has decreased the longshore sediment input to the inlet, and increased the tidal prism, ebb velocity, and sediment transport in the south opening, thus enhancing the inlet's stability. However, reducing the river discharge and landfill of the tidal flats has resulted in a decrease of the tidal prism, the ebb velocity, and the ability to export sediment, thus having the tendency to deteriorate the inlet's stability.
