1Protective immunity against Rickettsia heilongjiangensis in a C3H/HeN mouse model mediated by outer membrane protein B-pulsed dendritic cells显示文摘Rickettsia heilongjiangensis is an obligate intracellular bacterium that causes Far-Eastern tick-borne spotted fever. Outer membrane protein B(Omp B) is an important surface protein antigen of rickettsiae. In the present study, the omp B gene of R. heilongjiangensis was divided into four fragments, resulting in four recombinant proteins(OmpB-p1, Omp B-p2, Omp B-p3, and Omp B-p4). Each Omp B was used in vitro to stimulate murine bone marrow-derived dendritic cells(BMDCs) of C3H/He N mice, and the Omp B-pulsed BMDCs were transferred to naive C3H/He N mice. On day 14 post-transfer of BMDCs, the mice were challenged with R. heilongjiangensis and the rickettsial loads in the mice were quantitatively determined on day 7 post-challenge. Mice receiving BMDCs pulsed with Omp B-p2, Omp B-p3, or Omp B-p4 exhibited significantly lower bacterial load compared with mice receiving Omp B-p1-pulsed BMDCs. CD4+ and CD8+ T cells isolated from the spleen of C3H/He N mice receiving BMDCs pulsed with each OmpB were co-cultured with BMDCs pulsed with the respective cognate protein. In flow cytometric analysis, the expression level of CD69 on CD4+ or CD8+ T cells from mice receiving BMDCs pulsed with Omp B-p2, OmpB-p3, or Omp B-p4 was higher than that on cells from mice receiving Omp B-p1-pulsed BMDCs, while the expression level of tumor necrosis factor(TNF)-α on CD8+ T cells and interferon(IFN)-γ on the CD4+ and CD8+ T cells from mice receiving Omp B-p2,-p3, or-p4 was significantly higher than on cells from mice receiving Omp B-p1-pulsed BMDCs. 3Concentrations and distribution of novel brominated flame retardants in the atmosphere and soil of Ny-Alesund and London Island,Svalbard,Arctic显示文摘Novel brominated flame retardants(NBFRs)were investigated in Arctic air and soil samples collected from Ny-?lesund and London Island,Svalbard,during Chinese scientific research expeditions to the Arctic during 2014–2015.The concentrations of 9 NBFRs in the Arctic air and soil were 4.9–8.7 pg/m3(average 6.8 pg/m3)and 101–201 pg/g dw(average 150 pg/g dw),respectively.The atmospheric concentration of hexabromobenzene(HBB)was significantly correlated with that of pentabromotoluene(PBT)and pentabromobenzene(PBBz),suggesting similar source and environmental fate in the Arctic air.No significant spatial difference was observed among the different sampling sites,both for air and soil samples,indicating that the effects of the scientific research stations on the occurrence of NBFRs in the Arctic were minor.The fugacities from soil to air of pentabromoethylbenzene(PBEB),2,3-dibromopropyl 2,4,6-tribromophenyl ether(DPTE),and decabromodiphenylethane 1,2-bis(pentabromophenyl)ethane(DBDPE)were lower than the equilibrium value,indicating a nonequilibrium state of these compounds between air and soil,the dominant impact of deposition and the net transport from air to soil.The correlation analysis between the measured and predicted soil-atmosphere coefficients based on the absorption model showed that the impact of the soil organic matter on the distribution of NBFRs in the Arctic region was minor.To the best of our knowledge,this work is one of the limited reports on atmospheric NBFRs in the Arctic and the first study to investigate the occurrence and fate of NBFRs in the Arctic soil.Yanfen Hao Wenying Meng Yingming Li Xu Han Huili Lu Pu Wang Ruiqiang Yang Qinghua Zhang Guibin Jiang 2020Journal of Environmental Sciences2020,32,11:1

5Synthesis of carbon dots with a tunable photoluminescence and their applications for the detection of acetone and hydrogen peroxide显示文摘Carbon dots(CDs) with multi-color emissive properties and a high photoluminescent quantum yield(PLQY) have attracted great attention recently due to their potential applications in chemical,environmental,biological and photo-electronic fields.Solvent-dependent effect in photoluminescence provides a facial and effective approach to tune the emission of CDs.In this study,green emissive nitrogen-doped carbon dots(N-CDs) are synthesized from p-hydroquinone and ethylenediamine through a simple hydrothermal method.The as-prepared N-CDs possess a robust excitation-independent green luminescence and a high PLQY of up to 15.9%.Further spectroscopic characterization indicates that the high PLQY is achieved by the balance of nitrogen doping states and the surface passivation extent in CDs.The N-CDs also exhibit solvent-dependent multi-color emissive property and distinct PLQY in different solvents(the maximum can reach up to 25.3%).Furthermore,the as-prepared N-CDs are applied as fluorescence probes to detect acetone and H2O2 in water.This method has exhibited a low detection limit of acetone(less than 0.1 %) and a quick and linear response to the H_2O_2 with the concentration from 0 to 120 μmol/L.This work broadens the knowledge of applying CDs as probes in the bio and chemical sensing fields.Shitong Cui Yanfen Wu Yao Liu Qingwen Guan Yida Zhang Yilin Zhang Song Luo Meng Xu Juncheng Wang 2020Chinese Chemical Letters2020,31,2:1
6Characterization of a novel bifunctional enzyme from buffalo rumen metagenome and its effect on in vitro ruminal fermentation and microbial community composition显示文摘To efficiently use lignocellulosic materials in ruminants,it is crucial to explore effective enzymes,especially bifunctional enzymes.In this study,a novel stable bifunctional cellulase-xylanase protein from buffalo rumen metagenome was expressed and characterized,CelXyn2.The enzyme displayed optimal activity at pH 6.0 and 45℃.The residual endoglucanase and xylanase activities were 90.6%and 86.4%after a 60-min pre-incubation at 55℃.Hydrolysis of rice straw,wheat straw,sheepgrass and sugar beet pulp by CelXyn2 showed its ability to degrade both cellulose and hemicellulose polymers.Treatment with CelXyn2 improved the hydrolysis of agricultural residues with an evident increase in production of total gas,lactate and volatile fatty acids.The results of 16S rRNA and real-time PCR showed that the effect on in vitro ruminal microbial community depended on fermenta-tion substrates.This study demonstrated that CelXyn2 could strengthen lignocellulose hydrolysis and in vitro ruminal fermentation.These characteristics of CelXyn2 distinguish it as a promising candidate for agricultural application.Zhenxiang Meng Jing Ma Zhanying Sun Chengjian Yang Jing Leng Weiyun Zhu Yanfen Cheng 2023Animal Nutrition2023,,2:0
7Comprehensive Evaluation of New Maize Varieties for Grain and Fodder Based on Membership Function Method显示文摘To screen new maize(Zea mays L.)varieties suitable for food and fodder dual-purpose in Du'an Yao Autonomous County of Guangxi,the agronomic characters,yield and quality indexes of 12 new maize varieties were measured,and the correlation between various indexes were analyzed,and the comprehensive performance of tested varieties was evaluated by membership function method.The results showed that Guidan 671 had the highest grain yield and whole-plant biomass at 10908 and 49965 kg/hm^(2),respectively,and the second was Zhaoyu 215 with a grain yield and whole-plant biomass of 10086 and 47175 kg/hm^(2),respectively.Grain yield was highly significantly positively correlated with ear diameter and 100-grain weight(P<0.01),and significantly correlated with whole-plant biomass,starch content,ear length and grain number per row(P<0.05);and the whole-plant biomass was highly significantly correlated with the number of grains per row(P<0.01),and significantly correlated with starch content,panicle length,plant height and panicle height(P<0.05).The comprehensive performance scores of the tested varieties from high to low were Guidan 671,Zhaoyu 215,Guidan 669,Guidan 6208,Guidan 666,Guidan 6205,Guidan 660,Guidan 6203,Guidan 6206,Guidan 162,Guidan 668 and Guidan 673.According to the values of membership function and combined with various indexes,Guidan 671 and Zhaoyu 215 had good comprehensive performance,and could be used as the first choice for food and fodder dual-purpose maize varieties in Du'an Yao Autonomous County.Chenglin ZOU Qiongxiang LIN Kaijian HUANG Ruining ZHAI Meng YANG Aihua HUANG Runxiu MO Xinxing WEI Yanfen HUANG 2023Agricultural Biotechnology2023,12,2:0
8Production,purific tion,characterization and application of two novel endoglucanases from buffalo rumen metagenome显示文摘Background Lignocellulose biomass is the most abundant and renewable material in nature.The objectives of this study were to characterize two endoglucanases Trep Cel3 and Trep Cel4,and determine the effect of the combination of them(1.2 mg Trep Cel3,0.8 mg Trep Cel4)on in vitro rumen fermentation characteristics.In this study,three nature lignocellulosic substrates(rice straw,RS;wheat straw,WS;leymus chinensis,LC)were evaluated for their in vitro digestibility,gas,NH3-N and volatile fatty acid(VFA)production,and microbial protein(MCP)synthesis by adding enzymatic combination.Methods Two endoglucanases’genes were successfully expressed in Escherichia coli(E.coli)BL21(DE3),and enzymatic characteristics were further characterized.The combination of Trep Cel3 and Trep Cel4 was incubated with lignocellulosic substrates to evaluate its hydrolysis ability.Results The maximum enzymatic activity of Trep Cel3 was determined at p H 5.0 and 40℃,while Trep Cel4 was at p H 6.0 and 50℃.They were stable over the temperature range of 30 to 60℃,and active within the p H range of 4.0 to 9.0.The Trep Cel3 and Trep Cel4 had the highest activity in lichenan 436.9±8.30 and 377.6±6.80 U/mg,respectively.The combination of Trep Cel3 and Trep Cel4 exhibited the highest efficiency at the ratio of 60:40.Compared to maximum hydrolysis of Trep Cel3 or Trep Cel4 separately,this combination was shown to have a superior ability to maximize the saccharification yield from lignocellulosic substrates up to 188.4%for RS,236.7%for wheat straw WS,222.4%for LC and 131.1%for sugar beet pulp(SBP).Supplemental this combination enhanced the dry matter digestion(DMD),gas,NH3-N and VFA production,and MCP synthesis during in vitro rumen fermentation.Conclusions The Trep Cel3 and Trep Cel4 exhibited the synergistic relationship(60:40)and significantly increased the saccharification yield of lignocellulosic substrates.The combination of them stimulated in vitro rumen fermentation of lignocellulosic substrates.This combination has the potential to be a feed additive to improve agricultural residues utilization in ruminants.If possible,in the future,experiments in vivo should be carried out to fully evaluate its effect.Zhenxiang Meng Chengjian Yang Jing Leng Weiyun Zhu Yanfen Cheng 2023Journal of Animal Science and Biotechnology2023,14,3:0
