3Effect of bax,bcl-2 and bcl-xL on regulating apoptosis in tissues of normal liver and hepatocellular carcinoma显示文摘AIM: To investigate the expression of bax, bcl-2 and bcl-xL mRNA in the tissues of normal liver and hepatocellular carcinoma (HCC), and analyze the relationship between the expression of bax, bcl-2 and bcl-xL mRNA and clinical parameters of HCC patients.METHODS: The expression of bax, bcl-2 and bcl-xL mRNA of normal liver and HCC was measured by Northern blot. Statistical analyses were made by t test and correlation analysis.RESULTS: A very low mRNA level was indicated at bax,bcl-2 and bcl-xL in the HCC tissues in contrast to the tissues of normal liver by Northern blot analysis. The analyses of mRNA level revealed that HCC tissues exhibited a mean 7.6-fold decrease in bax, 4.2-fold in bcl-2 and 3.5-fold in bcl-xL in comparison with normal control tissues, respectively. Positive correlation was found between bax and bcl-xL (r=0.7061,P<0.01). There was no significance between the mRNA expression of these three genes and age, gender, tumor differentiation and tumor stage of HCC patients. 4High-Strength and High-Plasticity TWIP Steel for Modern Vehicle显示文摘In this paper new high-strength and high-plasticity twinning induced plasticity (TWIP) steel for modern automobile body was investigated. Some basic experimental results were given. The results indicate the TWlP steel has excellent properties. It exhibits high ultimate tensile strength (600~1100 MPa) and extremely large elongation of 60% to 90%. 5Future Changes and Uncertainties in Temperature and Precipitation over China Based on CMIP5 Models显示文摘Climate changes in future 21 st century China and their uncertainties are evaluated based on 22 climate models from the Coupled Model Intercomparison Project Phase 5(CMIP5). By 2081–2100, the annual mean surface air temperature(SAT) is predicted to increase by 1.3℃± 0.7℃, 2.6℃± 0.8℃ and 5.2℃± 1.2℃ under the Representative Concentration Pathway(RCP) scenarios RCP2.6, RCP4.5 and RCP8.5, relative to 1986–2005, respectively. The future change in SAT averaged over China increases the most in autumn/winter and the least in spring, while the uncertainty shows little seasonal variation.Spatially, the annual and seasonal mean SAT both show a homogeneous warming pattern across China, with a warming rate increasing from southeastern China to the Tibetan Plateau and northern China, invariant with time and emissions scenario.The associated uncertainty in SAT decreases from northern to southern China. Meanwhile, by 2081–2100, the annual mean precipitation increases by 5% ± 5%, 8% ± 6% and 12% ± 8% under RCP2.6, RCP4.5 and RCP8.5, respectively. The national average precipitation anomaly percentage, largest in spring and smallest in winter, and its uncertainty, largest in winter and smallest in autumn, show visible seasonal variations. Although at a low confidence level, a homogeneous wetting pattern is projected across China on the annual mean scale, with a larger increasing percentage in northern China and a weak drying in southern China in the early 21 st century. The associated uncertainty is also generally larger in northern China and smaller in southwestern China. 6Stretching characteristics and its dynamic significance of the northern continental margin of South China Sea显示文摘To investigate the thinning of the whole crust, and the contribution of the upper versus lower crust to the stretching since Cenozoic, we calculated the stretching factor of the northern margin of South China Sea with data such as whole crustal thickness, depth of Cenozoic sedimentary basement and the horizontal displacement of faults. An isometric line drawing on whole crustal stretching factor is then obtained. Along the seismic Line 1530 in Baiyun sag, we also calculated the stretching factors of the upper and lower crust. The results suggest that the whole crustal thickness decreases seaward while the whole crustal stretching factor increases from shelf to slope. The Moho upwells highest beneath where the crust is thinned most. The value of the whole crustal stretching factor ranges from 1.5 to 6. Two areas were thinned intensely: the center of Yinggehai Basin, and the Baiyun sag in the Pearl River Mouth Basin. The calculation of the upper and lower crustal stretching factors from DSP1530 in Baiyun sag shows that the original crust of Baiyun sag should be thinned before deformation. 7Biofunctional magnesium coated Ti6Al4V scaffold enhances osteogenesis and angiogenesis in vitro and in vivo for orthopedic application显示文摘The insufficient osteogenesis and osseointegration of porous titanium based scaffold limit its further application.Early angiogenesis is important for scaffold survival.It is necessary to develop a multifunctional surface on titanium scaffold with both osteogenic and angiogenic properties.In this study,a biofunctional magnesium coating is deposited on porous Ti6Al4V scaffold.For osseointegration and osteogenesis analysis,in vitro studies reveal that magnesium-coated Ti6Al4V co-culture with MC3T3-E1 cells can improve cell proliferation,adhesion,extracellular matrix(ECM)mineralization and ALP activity compared with bare Ti6Al4V cocultivation.Additionally,MC3T3-E1 cells cultured with magnesium-coated Ti6Al4V show significantly higher osteogenesisrelated genes expression.In vivo studies including fluorochrome labeling,micro-computerized tomography and histological examination of magnesium-coated Ti6Al4V scaffold reveal that new bone regeneration is significantly increased in rabbits after implantation.For angiogenesis studies,magnesium-coated Ti6Al4V improve HUVECs proliferation,adhesion,tube formation,wound-healing and Transwell abilities.HUVECs cultured with magnesium-coated Ti6Al4V display significantly higher angiogenesis-related genes(HIF-1αand VEGF)expression.Microangiography analysis reveal that magnesium-coated Ti6Al4V scaffold can significantly enhance the blood vessel formation.This study enlarges the application scope of magnesium and provides an optional choice to the conventional porous Ti6Al4V scaffold with enhanced osteogenesis and angiogenesis for further orthopedic applications.Peng Gao Bo Fan Xiaoming Yu Wenwen Liu Jie Wu Lei Shi Di Yang Lili Tan Peng Wan Yulin Hao Shujun Li Wentao Hou Ke Yang Xiaokang Li Zheng Guo 2020Bioactive Materials2020,5,3:15
8Vascular endothelial growth factor concentration in vitreous humor of patients with severe proliferative diabetic retinopathy after intravitreal injection of conbercept as an adjunctive therapy for vitrectomy显示文摘Background:Proliferative diabetic retinopathy(PDR)is a progressive stage of diabetic retinopathy featured by the formation of neovascular and proliferative membrane.Vascular endothelial growth factor(VEGF)acts as a pivot factor in the development of neovascularization.This study was to investigate the changes of intravitreal VEGF concentrations of severe PDR after intravitreal injection of conbercept(IVC)and its potential advantages to the following vitrectomy.Methods:This was a prospective,interventional,randomized controlled study.Sixty eyes(60 patients)with severe PDR and 20 eyes from 20 patients with rhegmatogenous retinal detachment complicated with proliferative vitreoretinopathy were enrolled in this study.PDR eyes were randomly assigned to three groups by sortation randomization method with 20 eyes in each based on the interval of preoperative IVC(group A:7 days,group B:14 days,group C:non-IVC).Another 20 eyes without diabetes were enrolled as the non-diabetic control group(group D),receiving PPV directly.Vitreous specimens of all 80 patients were collected and evaluated afterwards.The intravitreal VEGF concentration of the four groups,and the total surgical time and the intraoperative bleeding rate of the PDR groups were recorded.Results:The mean intravitreal VEGF concentrations of groups A-D were 66.6±43.3,93.1±52.3,161.4±106.1 and 1.8±1.2 pg/mL,respectively.It increased significantly in PDR patients(groups A,B and C)(P=0.002,<0.001,and<0.001,respectively).PDR patients with preoperative IVC(groups A and B)presented significantly lower VEGF concentrations(P<0.001 and 0.001),intraoperative bleeding rates(P=0.004)and total surgical time(P<0.001,P=0.003)compared with group C.No statistical differences were presented between groups A and B on the three parameters.Conclusion:Seven days and 14 days of preoperative IVC are equally efficient and safe for the vitrectomy of severe PDR patients through decreasing vitreous VEGF concentrations,intraoperative bleeding rate and total surgical times.Bing Li Meng-Da Li Jun-Jie Ye Zhe Chen Zi-Jian Guo Yu Di 2020Chinese Medical Journal2020,,6:14
9The impact of climate change on forest fire danger rating in China’s boreal forest显示文摘The Great Xing'an Mountains boreal forests were focused on in the northeastern China.The simulated future climate scenarios of IPCC SRES A2a and B2a for both the baseline period of 1961-1990 and the future scenario periods were downscaled by the Delta Method and the Weather Generator to produce daily weather data.After the verification with local weather and fire data,the Canadian Forest Fire Weather Index System was used to assess the forest fire weather situation under climate change in the study region.An increasing trend of fire weather severity was found over the 21st century in the study region under the both future climate change scenarios,compared to the 1961-1990 baseline period.The annual mean/maximum fire weather index was predicted to rise continuously during 2010-2099,and by the end of the 21st century it is predicted to rise by 22%-52% across much of China's boreal forest.The significant increases were predicted in the spring from of April to June and in the summer from July to August.In the summer,the fire weather index was predicted to be higher than the current index by as much as 148% by the end of the 21st century.Under the scenarios of SRES A2a and B2a,both the chance of extremely high fire danger occurrence and the number of days of extremely high fire danger occurrence was predicted to increase in the study region.It is anticipated that the number of extremely high fire danger days would increase from 44 days in 1980s to 53-75 days by the end of the 21st century.YANG Guang DI Xue-ying GUO Qing-xi SHU Zhan ZENG Tao YU Hong-zhou WANG Chao 2011Journal of Forestry Research2011,22,2:12
10A CFD/CSD model for aeroelastic calculations of large-scale wind turbines显示文摘In this paper,a CFD/CSD model coupling N-S equations and structural equations of motion in the time domain is described for aeroelastic analysis of large wind turbines.The structural modes of blades are analyzed with one-dimensional beam models.By combining point matched sliding grid for wind turbine rotation and deforming grid for structural vibrations,a hybrid dynamic grid strategy is designed for the multi-block structured grid system of a wind turbine.The dual time-stepping approach and finite volume scheme are applied to the three-dimensional unsteady preconditioned N-S equations,and DES approach is employed to simulate the unsteady massively separated flows.A modal approach is adopted to calculate the structural response,and a predictor-corrector scheme is used to solve the structural equations of motion.CFD and CSD solvers are tightly coupled via successive iterations within each physical time step.As a result,a time-domain CFD/CSD model for aeroelastic analysis of a large wind turbine is achieved.The presented method is applied to the NH1500 large wind turbine under the rated condition,and the calculated aeroelastic characteristics agree well with those of the prescribed vortex wake method.GUO TongQing LU ZhiLiang TANG Di WANG TongGuang DONG Lu 2013Science China(Technological Sciences)2013,56,1:11

12Improved corrosion resistance and thermal stability of sintered Nd-Fe-B magnets with holmium substitution显示文摘The effects of Ho substitution for Nd on the microstructure, corrosion resistance and thermal stability of the Nd-Fe-B magnets were investigated. The(Nd,Ho)-O phase was formed with increasing Ho substitution. The results of potentiodynamic polarization and highly accelerated stress test show improved corrosion resistance with increasing Ho substitution. The optimum mass loss 0.29 mg/cm^2 is achieved.Moreover, the average temperature coefficients for remanence and coercivity in the range of 25-150℃are both closer to zero, indicating improved thermal stability. 13Future changes in coverage of 1.5℃ and 2℃warming thresholds显示文摘The areas covered by 1.5 ℃ and 2.0 ℃ warming thresholds under RCP2.6, RCP4.5, and RCP8.5 were analyzed based on 22 CMIP5 models. More than 90% of the model runs are in agreement that by the end of the 21 st century, near-surface air temperature changes over ~5%(~2%), ~40%(~18%), and ~92%(~86%)of the globe will cross the 1.5 ℃(2.0 ℃) threshold under RCP2.6, RCP4.5, and RCP8.5, respectively. Under RCP8.5, nearly the whole of North America, Europe + Russia, Africa, and Asia–Russia will cross the 1.5 ℃(2.0 ℃) threshold in ~2050(~2060), while the coverage rates over South America and Oceania are ~80%(~75%) and ~50%(~30%), respectively. The threshold-onset time(TOT) for 2 ℃ warming is earliest over Europe + Russia and North America, followed by Africa, Asia–Russia, South America, and finally Oceania under the RCP4.5 and RCP8.5 scenarios. 14Simulated Microgravity Conditions and Carbon Ion Irradiation Induce Spermatogenic Cell Apoptosis and Sperm DNA Damage显示文摘Objective To investigate the effect of simulated microgravity and carbon ion irradiation(CIR) on spermatogenic cell apoptosis and sperm DNA damage to the testis of male Swiss Webster mice,and assess the risk associated with space environment.Methods Sperm DNA damage indicated by DNA fragmentation index(DFI) and high DNA stainability(HDS) was measured by sperm chromatin structure assay(SCSA).Apoptosis of spermatogenic cells was detected by annexin V-propidium iodide assay.Bax(the expression levels of p53) and proliferating cell nuclear antigen(PCNA) were measured by immunoblotting;p53 and PCNA were located by immunohistology.Results HDS,DFI,apoptosis index,and the expression levels of p53 and Bax were detected to be significantly higher in the experimental groups(P<0.05) compared with those in the control group;however,the PCNA expression varied to a certain degree.p53-and PCNA-positive expression were detected in each group,mainly in relation to the spermatogonic cells and spermatocytes.Conclusion The findings of the present study demonstrated that simulated microgravity and CIR can induce spermatogenic cell apoptosis and sperm DNA damage.Sperm DNA damage may be one of the underlying mechanisms behind male fertility decline under space environment.These findings may provide a scientific basis for protecting astronauts and space traveler's health and safety.LI Hong Yan ZHANG Hong MIAO Guo Ying XIE Yi SUN Chao DI Cui Xia LIU Yang LIU Yuan Yuan ZHANG Xin MA Xiao Fei XU Shuai GAN Lu ZHOU Xin 2013Biomedical and Environmental Sciences2013,26,9:10

16On the Kinematic Characteristics and Dynamic Process of Boundary faults of the Nansha Ultra-crust Layer-Block显示文摘The Nansha ultra-crust layer-block is confined by ultra-crustal boundary faults of distinctive features, bordering the Kangtai-Shuangzi-Xiongnan extensional faulted zone on the north, the Baxian-Baram-Yoca-Cuyo nappe faulted zone on the south, the Wan’an-Natuna strike-slip tensional faulted zone on the west and the Mondoro-Panay strike-slip compressive faulted zone on the east. These faults take the top of the Nansha asthenosphere as their common detachmental surface. 17Coordinated control for medium voltage DC distribution centers with flexibly interlinked multiple microgrids显示文摘From flexible interconnection among feeders to hybrid alternating current(AC) and direct current(DC)distribution structures of future smart distribution systems medium-voltage DC distribution centers with flexibly interlinked multiple microgrids(MGs) will have wide applications on the demand side. A generic coordinated control framework based on a distributed cooperation scheme is proposed for such DC centers, as opposed to centralized control structures. A novel unified control only using local measurements is proposed for these interlinking converters. During normal power disturbances, automatic coordinated power control and mutual support among sub-systems can be realized, thereby improving DC voltage and AC frequency stability to enable multiple MGs to be treated as a real unified cluster. Moreover, with this method, interlinking converters can realize seamless transition in power dispatching mode, common DC bus voltage control mode, and MG support mode without communication and control system switching. A simplified dynamic model has been developed to verify the proposed control strategy. 19Fluorescence regional integration and differential fluorescence spectroscopy for analysis of structural characteristics and proton binding properties of fulvic acid sub-fractions显示文摘Structural characteristics and proton binding properties of sub-fractions(FA3–FA13) of fulvic acid(FA), eluted stepwise by pyrophosphate buffer were examined by use of fluorescence titration combined with fluorescence regional integration(FRI) and differential fluorescence spectroscopy(DFS). Humic-like(H-L) and fulvic-like(F-L) materials, which accounted for more than 80% of fluorescence response, were dominant in five sub-fractions of FA. Based on FRI analysis, except the response of F-L materials in FA9 and FA13, maximum changes in percent fluorescence response were less than 10% as pH was increased from 2.5 to 11.5.Contents of carboxylic and phenolic groups were compared for fluorescence peaks of FA sub-fractions based on pH-dependent fluorescence derived from DFS. Static quenching was the dominant mechanism for binding of protons by FA sub-fractions. Dissociation constants(p Ka) were calculated by use of results of DFS and the modified Stern-Volmer relationship. The p Kaof H-L, F-L, tryptophan-like and tyrosine-like materials of FA subfractions exhibited ranges of 3.17–4.06, 3.12–3.97, 4.14–4.45 and 4.25–4.76, respectively, for acidic pHs. At basic pHs, values of p Ka for corresponding materials were in ranges of 9.71–10.24, 9.62–10.99, 9.67–10.31 and 9.33–10.28, respectively. At acidic pH, protein-like(P-L)materials had greater affinities for protons than did either H-L or F-L materials. The dicarboxylic and phenolic groups were likely predominant sites of protonation for both H-L and F-L materials at both acidic and basic pHs.
