Identification of mecciRNAs and their roles in the mitochondrial entry of proteins显示文摘Mammalian mitochondria have small genomes encoding very limited numbers of proteins.Over one thousand proteins and noncoding RNAs encoded by the nuclear genome must be imported from the cytosol into the mitochondria. Here,we report the identification of hundreds of circular RNAs(mecciRNAs)encoded by the mitochondrial genome. We provide both in vitro and in vivo evidence to show that mecciRNAs facilitate the mitochondrial entry of nuclear-encoded proteins by serving as molecular chaperones in the folding of imported proteins.Known components involved in mitochondrial protein and RNA importation, such as TOM40 and PNPASE,interact with mecciRNAs and regulate protein entry. The expression of mecciRNAs is regulated, and these transcripts are critical for the adaption of mitochondria to physiological conditions and diseases such as stresses and cancers by modulating mitochondrial protein importation.mecciRNAs and their associated physiological roles add categories and functions to the known eukaryotic circular RNAs and shed novel light on the communication between mitochondria and the nucleus. | Xu Liu Xiaolin Wang Jingxin Li Shanshan Hu Yuqi Deng Hao Yin Xichen Bao Qiangfeng Cliff Zhang Geng Wang Baolong Wang Qinghua Shi Ge Shan | 2020 | Science China(Life Sciences)2020,63,10: | 15
Magnetic performance and corrosion resistance of electroless plating CoWP film显示文摘The CoWP film with good magnetic performance and corrosion resistance was electrolessly plated from alkaline solution. The technical parameters of the electroless plating system were optimized. When the pH value of electroless plating solution was 11.0 and the reducing agent (NaH2PO2) content was 0.4 mol L 1, the target chemical reactions proceeded in the electroless plating solution smoothly with negligible interference and side effects. CoWP film prepared under optimal deposition condition contained more hexagonal close-packed (hcp) cobalt (ε-Co) of [110] and crystallographic orientation of Co [002]. VSM analysis reveals that the saturation magnetization of the CoWP film is 100~220 Am2 kg 1 and coercivity is (2.87~4.38)×104 A·cm 1. The corrosion behavior of CoWP film in 3.5% NaCl aqueous solution was studied by electrochemical experiments. The results prove that the corrosion resistance of the CoWP film deposited under the optimal depo-sition condition exhibites relatively lower corrosion current of (3.054~3.162)×10 6 A·cm 2. The surface morphology analysis indicates that the film is smooth and composed of regular-shaped crystallites. | Jiang, Li Pan, Shanshan Lu, Jianbiao Ke, Xin He, Wenhao Yu, Yundan Wei, Guoying Ge, Hongliang | 2012 | Rare Metals2012,31,3: | 6

Analysis of the binding sites with NL-101 to amino acids and peptides by HPLC/MS/MS显示文摘The binding between NL-101, a novel nitrogen mustard anti-cancer drug, with amino acids and peptides has been investigated by high performance liquid chromatography electrospray tandem mass spectrometry(HPLC/ESI-MS/MS). This study offers supporting data of the interaction among drug and amino acids and peptides, which could potentially explain the cytotoxic and mutagenic effects of the drug. Collision-induced dissociation(CID) experiment demonstrated that under the same collision energy, the amino group combined with NL-101 adducts are sensitive and often produce more fragment ions; the carboxyl group combined with NL-101 adducts are hard to break and display fewer fragment ions. In addition, when other group(like sulfhydryl group) of amino acids binds to NL-101, CID spectra show different fragmentation pattern. These differences could display structural information about the drug adducts and be utilized as location of the authentic binding sites. | Lingzi Dai Nian Guo Yaqin Liu Shanshan Shen Qiufu Ge Yuanjiang Pan | 2019 | Chinese Chemical Letters2019,30,1: | 4

Beneficial Effects of Celastrol on Immune Balance by Modulating Gut Microbiota in Experimental Ulcerative Colitis Mice显示文摘Ulcerative colitis(UC)is a chronic inflammatory bowel disease caused by many factors including colonic inflammation and microbiota dysbiosis.Previous studies have indicated that celastrol(CSR)has strong anti-inflammatory and immune-inhibitory effects. Here,we investigated the effects of CSR on colonic inflammation and mucosal immunity in an experimental colitis model, and addressed the mechanism by which CSR exerts the protective effects. We characterized the therapeutic effects and the potential mechanism of CSR on treating UC using histological staining, intestinal permeability assay, cytokine assay, flow cytometry, fecal microbiota transplantation(FMT),16S rRNA sequencing, untargeted metabolomics, and cell differentiation.CSR administration significantly ameliorated the dextran sodium sulfate(DSS)-induced colitis in mice, which was evidenced by the recovered body weight and colon length as well as the decreased disease activity index(DAI)score and intestinal permeability.Meanwhile,CSR down-regulated the production of pro-inflammatory cytokines and up-regulated the amount of anti-inflammatory mediators at both mRNA and protein levels, and improved the balances of Treg/Th1 and Treg/Th17 to maintain the colonic immune homeostasis.Notably, all the therapeutic effects were exerted in a gut microbiota-dependent manner.Furthermore,CSR treatment increased the gut microbiota diversity and changed the compositions of the gut microbiota and metabolites, which is probably associated with the gut microbiota-mediated protective effects. In conclusion, this study provides the strong evidence that CSR may be a promising therapeutic drug for UC. | Mingyue Li Weina Guo Yalan Dong Wenzhu Wang Chunxia Tian Zili Zhang Ting Yu Haifeng Zhou Yang Gui Kaming Xue Junyi Li Feng Jiang Alexey Sarapultsev Huafang Wang Ge Zhang Shanshan Luo Heng Fan Desheng Hu | 2022 | Genomics, Proteomics & Bioinformatics2022,20,2: | 1
Immune regulation of the gut-brain axis and lung-brain axis involved in ischemic stroke显示文摘Local ischemia often causes a series of inflammatory reactions when both brain immune cells and the peripheral immune response are activated. In the human body, the gut and lung are regarded as the key reactional targets that are initiated by brain ischemic attacks.Mucosal microorganisms play an important role in immune regulation and metabolism and affect blood-brain barrier permeability. In addition to the relationship between peripheral organs and central areas and the intestine and lung also interact among each other. Here,we review the molecular and cellular immune mechanisms involved in the pathways of inflammation across the gut-brain axis and lung-brain axis.We found that abnormal intestinal flora, the intestinal microenvironment, lung infection, chronic diseases, and mechanical ventilation can worsen the outcome of ischemic stroke. This review also introduces the influence of the brain on the gut and lungs after stroke, highlighting the bidirectional feedback effect among the gut, lungs, and brain. | Xiaodi Xie Lei Wang Shanshan Dong ShanChun Ge Ting Zhu | 2024 | Neural Regeneration Research2024,19,3: | 1

Novel photoelectric material of perovskite-like(CH_(3))_(3)SPbI_(3) nanorod arrays with high stability显示文摘Organometallic halide perovskite materials make great achievements in optoelectronic fields, especially in solar cells,in which the organic cations contain amine components. However, the amine with NàH bonds is easily hydrolyzed with moisture in the air, weakening the perovskite materials stability. It is desirable to develop other non-amine stable perovskite materials. In this work,sulfur-based perovskite-like(CH_(3))_(3)SPbI_(3) nanorod arrays were fabricated by a solution-processed method, which can be indexed hexagonal crystal structure in the space group P63 mc.The binding force is exceptionally strong between the non-amine(CH_(3))_(3) S+and[PbI_(6)]_(4)-octahedral,leading to high stability of(CH_(3))_(3)SPbI_(3).The(CH_(3))_(3)SPbI_(3) nanorod arrays can keep the morphology and crystal structure in an ambient atmosphere over 60 days.In addition,the(CH_(3))_(3)SPbI_(3) nanorod arrays can offer direct charge transfer channels, which show excellent optoelectronic properties.The(CH_(3))_(3)SPbI_(3) nanorod arrays-based solar cells with VOx hole transfer layers achieved a power conversion efficiency of 2.07%with negligible hysteresis. And the(CH_(3))_(3)SPbI_(3) nanorod arrays were also effectively applied in photodetectors with interdigitated gold electrodes. This work demonstrates that sulfur-based perovskite-like(CH_(3))_(3)SPbI_(3) is a novel promising stable compound with great potential for practical optoelectronic applications. | Ruiyuan Hu Chuangye Ge Liang Chu Yifei Feng Shanshan Xiao Yuhui Ma Wei Liu Xing'ao Li Mohammad Khaja Nazeeruddin | 2021 | Journal of Energy Chemistry2021,30,8: | 0
Observation and analysis of morphology abnormalities in development of Oryzias melastigma embryos显示文摘Fish embryos are widely used as models in toxicology, drug development, and human disease research because of their high sensitivity, observability, and operability, providing the basis for an in-depth understanding of the embryogenesis.Increasing studies have indicated that birth defects are hereditary. In this study,we used Oryzias melastigma as a model to conduct a study of 185-day embryogenesis and observed self-induced non-pathological abnormal embryogenesis.O.melastigma experienced pre-puberty stage, adolescence stage, and senescence stage, and individuals produced up to 102 eggs per day. However, the fecundity was markedly reduced during the senescent stage.During the active egg and blastodisc stages,pseudo-fertilization and pseudo-blastocysts were observed.During cleavage at the 4-to 32-cell stages,we observed blastomeres separation or dislocation.Excessively separated blastomeres formed double blastoderms, eventually resulting in conjoined twins.During the blastula stage,we observed abnormally increased cell volume, narrowed and elongated blastocysts, and abnormally coated blastoderms. At the organogenesis stage,we observed abnormal numbers of Kupff er’s vesicles and conjoined twins.Abnormality in the location and number of oil droplets were observed in various development stages.Abnormal development was more commonly observed in fertilized eggs produced by broodstock in pre-puberty or senescence stages, which is probably related to the age of fish and the egg quality. This study can provide the materials for comparative analysis in toxicological and molecular studies of O.melastigma, and may provide evidence for other economic fish that produce sticky eggs. | Jingjing SONG Haiyi WU Kaikai LIU Wendan CHI Shanshan GE Zhipeng ZHANG Mingqi WANG Yi YU Daode YU | 2021 | Journal of Oceanology and Limnology2021,39,4: | 0
