| 1 | Paleozoic and Mesozoic Basement Magmatisms of Eastern Qaidam Basin,Northern Qinghai-Tibet Plateau:LA-ICP-MS Zircon U-Pb Geochronology and its Geological Significance显示文摘The eastern margin of the Qaidam Basin lies in the key tectonic location connecting the Qinling,Qilian and East Kunlun orogens.The paper presents an investigation and analysis of the geologic structures of the area and LA-ICP MS zircon U-Pb dating of Paleozoic and Mesozoic magmatisms of granitoids in the basement of the eastern Qaidam Basin on the basis of 16 granitoid samples collected from the South Qilian Mountains,the Qaidam Basin basement and the East Kunlun Mountains.According to the results in this paper,the basement of the basin,from the northern margin of the Qaidam Basin to the East Kunlun Mountains,has experienced at least three periods of intrusive activities of granitoids since the Early Paleozoic,i.e.the magmatisms occurring in the Late Cambrian(493.1±4.9 Ma),the Silurian(422.9±8.0 Ma-420.4±4.6 Ma) and the Late Permian-Middle Triassic(257.8±4.0 Ma-228.8±1.5 Ma),respectively.Among them,the Late Permian-Middle Triassic granitoids form the main components of the basement of the basin.The statistics of dated zircons in this paper shows the intrusive magmatic activities in the basement of the basin have three peak ages of 244 Ma(main),418 Ma,and 493 Ma respectively.The dating results reveal that the Early Paleozoic magmatism of granitoids mainly occurred on the northern margin of the Qaidam Basin and the southern margin of the Qilian Mountains,with only weak indications in the East Kunlun Mountains.However,the distribution of Permo-Triassic(P-T) granitoids occupied across the whole basement of the eastern Qaidam Basin from the southern margin of the Qilian Mountains to the East Kunlun Mountains.An integrated analysis of the age distribution of P-T granitoids in the Qaidam Basin and its surrounding mountains shows that the earliest P-T magmatism(293.6-270 Ma) occurred in the northwestern part of the basin and expanded eastwards and southwards,resulting in the P-T intrusive magmatism that ran through the whole basin basement.As the Cenozoic basement thrust system developed in the eastern Qaidam Basin,the nearly N-S-trending shortening and deformation in the basement of the basin tended to intensify from west to east,which went contrary to the distribution trend of N-S-trending shortening and deformation in the Cenozoic cover of the basin,reflecting that there was a transformation of shortening and thickening of Cenozoic crust between the eastern and western parts of the Qaidam Basin,i.e.,the crustal shortening of eastern Qaidam was dominated by the basement deformation(triggered at the middle and lower crust),whereas that of western Qaidam was mainly by folding and thrusting of the sedimentary cover(the upper crust). | CHEN Xuanhua George GEHRELS YIN An LI Li JIANG Rongbao | 2012 | Acta Geologica Sinica(English Edition)2012,86,2: | 32 |
| 2 | Community analysis of ammonia-oxidizing bacteria in activated sludge of eight wastewater treatment systems显示文摘We investigated the communities of ammonia-oxidizing bacteria(AOB) in activated sludge collected from eight wastewater treatment systems using polymerase chain reaction(PCR) followed by terminal restriction fragment length polymorphism(T-RFLP),cloning,and sequencing of the α-subunit of the ammonia monooxygenase gene(amoA).The T-RFLP fingerprint analyses showed that different wastewater treatment systems harbored distinct AOB communities.However,there was no remarkable difference among the AOB TRFLP profiles from different parts of the same system.The T-RFLP fingerprints showed that a full-scale wastewater treatment plant(WWTP) contained a larger number of dominant AOB species than a pilot-scale reactor.The source of influent affected the AOB community,and the WWTPs treating domestic wastewater contained a higher AOB diversity than those receiving mixed domestic and industrial wastewater.However,the AOB community structure was little affected by the treatment process in this study.Phylogenetic analysis of the cloned amoA genes clearly indicated that all the dominant AOB in the systems was closely related to Nitrosomonas spp.not to Nitrosospira spp.Members of the Nitrosomonas oligotropha and Nitrosomonas communis clusters were found in all samples,while members of Nitrosomonas europaea cluster occurred in some systems. | Xiaohui Wang Xianghua Wen Craig Criddle George Wells Jie Zhang Yin Zhao | 2010 | Journal of Environmental Sciences2010,22,4: | 17 |
| 3 | Cytotoxic genes from traditional Chinese medicine inhibit tumor growth both in vitro and in vivo显示文摘OBJECTIVE: Little effort has been made to study the protein-encoding genes isolated from traditional Chinese medicine(TCM) drugs, and the delivery of these genes into malignant cells through recombinant adeno-associated virus(r AAV) vectors has not been attempted. METHODS: We synthesized the c DNAs of five known cytotoxic proteins isolated from TCM drugs and the FLAG epitope-tagged c DNAs were subcloned into a r AAV plasmid vector. The protein expression was confi rmed by Western blot assay. Various cancer cell lines were transfected with the above plasmids and cell growth was monitored both in vitro and in vivo. The best cytotoxic gene was further packaged into r AAV vectors, under the control of a liver cancer-specifi c promoter. The liver tumor growth was then monitored following intratumor administration of the r AAV vectors.RESULTS: The expression plasmids, encoding individual potential cytotoxic genes tagged with FLAG epitope, were successfully generated and sequenced. Among these genes, trichosanthin(TCS) gene yielded the most promising results for the inhibition of cancer cell growth in vitro. The over-expressed TCS functioned as a type I ribosome-inactivating protein, followed by inducing apoptosis that is associated with the Bcl-PARP signaling pathway. | 4 | Pristimerin enhances recombinant adeno-associated virus vector-mediated transgene expression in human cell lines in vitro and murine hepatocytes in vivo显示文摘OBJECTIVE:In the present study,we systemically evaluated the ability of two bioactive compounds from traditional Chinese medicine,celastrol and pristimerin,to enhance recombinant adeno-associated virus(rAAV) serotype vector-mediated transgene expression both in human cell lines in vitro,and in murine hepatocytes in vivo.METHODS:Human cell lines were infected with rAAV vectors with either mock treatment or treatment with celastrol or pristimerin.The transgene expression,percentage of nuclear translocated viral genomes and the ubiquitination of intracellular proteins were investigated post-treatment.In addition,nonobese diabetic/severe combined immunodeficient gamma(NSG) mice were tail vain-injected with rAAV vectors and co-administered with either dimethyl sulfoxide,celastrol,pristimerin or a positive control,bortezomib.The transgene expression in liver was detected and compared over time.RESULTS:We observed that treatment with pristimerin,at as low as 1 umol/L concentration,significantly enhanced rAAV2 vector-mediated transgene expression in vitro,and intraperitoneal coadministration with pristimerin at 4 mg/(kgd) for 3 d dramatically facilitated viral transduction in murine hepatocytes in vivo.The transduction efficiency of the tyrosine-mutant rAAV2 vectors as well as that of rAAV8 vectors carrying oversized transgene cassette was also augmented significantly by pristimerin.The underlying molecular mechanisms by which pristimerin mediated the observed increase in the transduction efficiency of rAAV vectors include both inhibition of proteasomal degradation of the intracellular proteins and enhanced nuclear translocation of the vector genomes.CONCLUSION:These studies suggest the potential beneficial use of pristimerin and pristimerincontaining herb extract in future liver-targeted gene therapy with rAAV vectors. | Li-na Wang Yuan Wang Yuan Lu Zi-fei Yin Yuan-hui Zhang George V.Aslanidi Arun Srivastava Chang-quan Ling Chen Ling | 2014 | Journal of Integrative Medicine2014,12,1: | 8 |

| 10 | Genomic Surveillance for SARS-CoV-2 Variants of Concern from Imported COVID-19 Cases-the Mainland of China,2021显示文摘Introduction:After the epidemic in Wuhan City was brought under control in 2020,local outbreaks of coronavirus disease 2019(COVID-19)in the mainland of China were mainly due to imported COVID-19 cases.The ongoing evolution of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has continued to generate new variants.Some have been designated as variants of concern(VOCs)by the World Health Organization(WHO).To better assess the role of imported SARS-CoV-2 surveillance and the prevalence of VOCs in 2021,the genomic surveillance data of SARS-CoV-2 from imported COVID-19 cases of 2021 in the mainland of China were analyzed.Methods:The analyses included the number of sequence submissions,time of sequence deposition,and time of detection of the VOCs in order to determine the timeliness and sensitivity of the surveillance.The proportions of VOCs were analyzed and compared with data from the Global Initiative of Sharing All Influenza Data(GISAID).Results:A total of 3,355 sequences of imported cases were submitted from 29 provincial-level administrative divisions,with differences in the number of sequence submissions and median time of sequence deposition.A total of 2,388 sequences with more than 90%genomic coverage were used for lineage analysis.The epidemic trend from Alpha to Delta to Omicron in imported cases was consistent with that in the GISAID.In addition,VOCs from imported cases were usually identified after WHO designation and before causing local outbreaks.Conclusions:The global distribution of SARSCoV-2 VOCs changed rapidly in 2021.Robust genomic surveillance of the imported SARS-CoV-2 in the mainland of China is of great significance.Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),the etiological agent of coronavirus disease 2019(COVID-19),is constantly mutating under the different circumstances of global transmission(1).The emerging SARS-CoV-2 variants may have potential adverse impacts on epidemic traits and severity.To some extent,it is also capable of escaping natural and vaccine-induced immunity(2–3).Some of them were designated as variants of concern(VOCs)by the World Health Organization(WHO)(4).Therefore,robust surveillance is essential to assess the evolution of viruses in real time.After the epidemic in Wuhan City was brought under control in 2020,several COVID-19 outbreaks in the mainland of China have been proven to relate to SARS-CoV-2 contaminated cold-chain products(5–7),while most were caused by transmission through imported cases on flights,at isolation facilities,or in designated hospitals(8–9).Therefore,genomic surveillance for SARS-CoV-2 from imported cases is of great significance for monitoring the risk of different variants that were imported into the mainland of China,assessing the risk of importation-associated domestic spread,and helping guide public health interventions.On March 17,2020,the China CDC released a notice and launched genomic surveillance for SARS-CoV-2 from imported COVID-19 cases nationwide.The laboratories of provincial CDCs were required to conduct SARS-CoV-2 whole-genome sequencing for samples from imported cases and submit the genomic sequences to the China CDC in time.This study includes the analysis of genomic surveillance data of imported SARS-CoV-2 cases of 2021 from the mainland of China. | Yenan Feng Xiang Zhao Zhixiao Chen Kai Nie Zeyuan Yin Ying Xia Ji Wang Peihua Niu Ruhan A Lili Li Dayan Wang Wenjie Tan Xuejun Ma Shiwen Wang Huanyu Wang George F.Gao Cao Chen Wenbo Xu | 2022 | China CDC weekly2022,4,31: | 1 |

Permanent magnet motors are used as a benchmark type for concrete algorithm development and evaluation.Algorithms are presented, their properties are established, and their accuracy and robustness are evaluated by simulation case studies under both normal operations and inter-turn winding faults.
