1Identification of Salmonella and Listeria monocytogenes by Near-infrared Spectroscopy and PCA显示文摘Near-infrared spectra of pathogenic bacteria (salmonella and Listeria monocytogenes ) were determined, and the spectral data were analyzed by the projection discriminant analysis based on principal component analysis (PCA). The expected results were obtained. 2Modulating effects of Astragalus polysaccharide on immune disorders via gut microbiota and the TLR4/NF-κB pathway in rats with syndrome of dampness stagnancy due to spleen deficiency显示文摘The syndrome of dampness stagnancy due to spleen deficiency(DSSD)is relatively common globally.Although the pathogenesis of DSSD remains unclear,evidence has suggested that the gut microbiota might play a significant role.Radix Astragali,used as both medicine and food,exerts the effects of tonifying spleen and qi.Astragalus polysaccharide(APS)comprises a macromolecule substance extracted from the dried root of Radix Astragali,which has many pharmacological functions.However,whether APS mitigates the immune disorders underlying the DSSD syndrome via regulating gut microbiota and the relevant mechanism remains unknown.Here,we used DSSD rats induced by high-fat and low-protein(HFLP)diet plus exhaustive swimming,and found that APS of moderate molecular weight increased the body weight gain and immune organ indexes,decreased the levels of interleukin-1β(IL-1β),IL-6,and endotoxin,and suppressed the Toll-like receptor 4/nuclear factor-κB(TLR4/NF-κB)pathway.Moreover,a total of 27 critical genera were significantly enriched according to the linear discriminant analysis effect size(LEfSe).APS increased the diversity of the gut microbiota and changed its composition,such as reducing the relative abundance of Pseudoflavonifractor and Paraprevotella,and increasing that of Parasutterella,Parabacteroides,Clostridium XIVb,Oscillibacter,Butyricicoccus,and Dorea.APS also elevated the contents of short-chain fatty acids(SCFAs).Furthermore,the correlation analysis indicated that 12 critical bacteria were related to the body weight gain and immune organ indexes.In general,our study demonstrated that APS ameliorated the immune disorders in DSSD rats via modulating their gut microbiota,especially for some bacteria involving immune and inflammatory response and SCFA production,as well as the TLR4/NF-κB pathway.This study provides an insight into the function of APS as a unique potential prebiotic through exerting systemic activities in treating DSSD.Wenxiao ZHAO Chenchen DUAN Yanli LIU Guangying LU Qin LYU Xiumei LIU Jun ZHENG Xuelian ZHAO Shijun WANG Haijun ZHAO 2023Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,7:1

5The genome-wide meiotic recombination landscape in ciliates and its implications for crossover regulation and genome evolution显示文摘Meiotic recombination is essential for sexual reproduction and its regulation has been extensively studied in many taxa.However,genome-wide recombination landscape has not been reported in ciliates and it remains unknown how it is affected by the unique features of ciliates:the synaptonemal complex(SC)-independent meiosis and the nuclear dimorphism.Here,we show the recombination landscape in the model ciliate Tetrahymena thermophila by analyzing single-nucleotide polymorphism datasets from 38 hybrid progeny.We detect 1021 crossover(CO)events(35.8 per meiosis),corresponding to an overall CO rate of 9.9 cM/Mb.However,gene conversion by non-crossover is rare(1.03 per meiosis)and not biased towards G or C alleles.Consistent with the reported roles of SC in CO interference,we find no obvious sign of CO interference.CO tends to occur within germ-soma common genomic regions and many of the 44 identified CO hotspots localize at the centromeric or subtelomeric regions.Gene ontology analyses show that CO hotspots are strongly associated with genes responding to environmental changes.We discuss these results with respect to how nuclear dimorphism has potentially driven the formation of the observed recombination landscape to facilitate environmental adaptation and the sharing of machinery among meiotic and somatic recombination.Lu Fu Chen Gu Kazufumi Mochizuki Jie Xiong Wei Mia Guangying Wang 2024Journal of Genetics and Genomics2024,51,3:0

9Zeolitic germanosilicate analogue to pharmacosiderite crystallized in an acidic medium显示文摘Zeolites are typically synthesized in alkaline or fluoride-containing near-neutral media.Sophisticated organic structure-directing agents have been investigated for such systems with the aim of discovering materials with unprecedented structures and properties for novel technical applications.In contrast,zeolite crystallization in strongly acidic media has yet to be explored.This study demonstrates that a zeolitic silicate phase crystallizes from acidic gels using trimethylamine as an organic additive with the composition 1 SiO_(2):0.3 TMA:0.3 HCl:0.15 HF:55 H_(2)O:(0.1-0.4)GeO_(2).This phase has an interrupted four-connected framework analog to the octahedron/tetrahedron-mixed framework of the mineral family pharmacosiderite.In comparison to the pharmacosiderite-type HK_(3)(Ge_(7)O_(16))(H_(2)O)_(4),the four GeO_(6)-octahedra forming the central[HGe_(4)O_(4)O_(12)]-cluster are replaced by four SiO_(4)-tetrahedra in a[Si_(4)O_(6)(OH)2.89]-unit in the new phase.However,the structure is distorted and may contain connectivity and point defects;thus,healing by the occasional incorporation of GeO_(6)-units is necessary.The refined unit cell has a cubic symmetry,space group P-43m(#215),with a=7.7005(1)Å.Acidic-medium synthesis is a useful way to find new zeolites that move in a fundamentally different direction from sophisticated organic structure-directing agents.Guangying Fu Qiaolin Lang Xiaolong Liu Haonuan Zhao Yiqing Sun Lei Zhao Ahmed Omran Peng Lu Xiaobo Yang Bing Yu Valentin Valtchev 2023Green Carbon2023,1,2:0
10Regulation and management of the biosecurity for synthetic biology显示文摘Synthetic biology(SynBio)is a high-profile interdiscipline combining engineering with science.As a dual-purpose discipline,SynBio is bringing large changes to many fields and providing great benefits to humans.However,due to its characteristic of complexity and uncertainty,SynBio also presents potential biosafety and biosecurity risks.Biosecurity risks refer to unauthorized access,loss,theft,misuse,diversion or intentional release.If a biosecurity accident happens,it would pose a huge threat to humans and nature.Therefore,it is crucial to establish a set of regulations and management practices for the biosecurity risks of SynBio.In this paper,we summarized the sources of the biosecurity risks of SynBio,from its research materials,products,technologies,information to Do-it-yourself synthetic biology.We reviewed and analyzed the current situation of regulation and management of biosecurity for SynBio in the international community and in China.We found that in most countries and regions,SynBio risks commonly follow the regulation and management of Genetically Modified Organisms which has loopholes if applied to the regulation for SynBio without any amendments.Here,we proposed suggestions for the Chinese-featured regulation and management of biosecurity for SynBio,including a top-to-bottom governing framework,a think-tank implementation mechanism,a Synthetic Biology Laboratory Biosecurity Manual safeguarding system,and strengthening biosecurity education on synthetic biology and self-regulation awareness among relevant personnel.Through this work,we aim to improve the standardized process of biosecurity regulation and management for SynBio in China and thereby map out a peaceful,profitable,and practical development path for synthetic biology.Xiaomei Zeng Hailun Jiang Guangying Yang Yakun Ou Shan Lu Jia Jiang Ruipeng Lei Li Su 2022Synthetic and Systems Biotechnology2022,7,2:0
