1Establishment and application of distant hybridization technology in fish显示文摘Hybridization is widely used. However, for a long time, systematic theories and technologies related to hybridization in fish have been lacking. In this study, through long-term systematic research, we investigated and obtained the main rules regarding inheritance and reproduction related to fish distant hybridization. Furthermore, we established one-step and multistep breeding technologies that were suitable for interspecific hybridization and intraspecific hybridization. Simultaneously, we used these two breeding technologies to produce a batch of diploid fish lineages and tetraploid fish lineages and improved fishes. In addition, we widely discuss the methods, technologies and results of hybridization breeding, referring to the domestic and foreign literature on fish hybridization. 2Large chromosomal segment deletions by CRISPR/LbCpf1-mediated multiplex gene editing in soybean显示文摘The creation of new soybean varieties has been limited by genomic duplication and redundancy.Efficient multiplex gene editing and large chromosomal segment deletion through clustered regularly interspaced palindromic repeats(CRISPR)/CRISPR-associated protein(Cas)systems are promising strategies for overcoming these obstacles.CRISPR/Cpf1 is a robust tool for multiplex gene editing.However,large chromosomal excision mediated by CRISPR/Cpf1 has been reported in only a few non-plant species.Here,we report on CRISPR/LbCpf1-induced large chromosomal segment deletions in soybean using multiplex gene targeting.The CRISPR/LbCpf1 system was optimized for direct repeat and guide RNA lengths in crispr RNA(crRNA)array.The editing efficiency was evaluated using LbCpf1 driven by the CaMV35S and soybean ubiquitin promoter.The optimized system exhibited editing efficiencies of up to 91.7%.Our results showed eight gene targets could be edited simultaneously in one step when a single eight-gRNA-target crRNA array was employed,with an efficiency of up to 17.1%.We successfully employed CRISPR/LbCpf1 to produce small fragments(<1 Kb)and large chromosomal segment deletions(10 Kb-1 Mb)involving four different gene clusters in soybean.Together,these data demonstrate the power of the CRISPR/LbCpf1 platform for multiplex gene editing and chromosomal segment deletion in soybean,supporting the use of this technology in both basic research and agricultural applications.Kaixuan Duan Yuanyuan Cheng Jing Ji Chenchen Wang Yongshu Wei Yuanchao Wang 2021Journal of Integrative Plant Biology2021,63,9:5
3Parathyroid hormone increases alveolar bone homoeostasis during orthodontic tooth movement in rats with periodontitis via crosstalk between STAT3 and β-catenin显示文摘Periodontitis patients are at risk of alveolar bone loss during orthodontic treatment.The aim of this study was to investigate whether intermittent parathyroid hormone(1–34)treatment(iPTH)could reduce alveolar bone loss during orthodontic tooth movement(OTM)in individuals with periodontitis and the underlying mechanism.A rat model of OTM in the context of periodontitis was established and alveolar bone loss was observed.The control,iPTH and iPTH+stattic groups received injections of vehicle,PTH and vehicle,or PTH and the signal transducer and activator of transcription 3(STAT3)inhibitor stattic,respectively.iPTH prevented alveolar bone loss by enhancing osteogenesis and suppressing bone resorption in the alveolar bone during OTM in rats with periodontitis.This effect of iPTH was along with STAT3 activation and reduced by a local injection of stattic.iPTH promoted osteoblastic differentiation and might further regulate the Wnt/β-catenin pathway in a STAT3-dependent manner.The findings of this study suggest that iPTH might reduce alveolar bone loss during OTM in rats with periodontitis through STAT3/β-catenin crosstalk.Cheng Zhang Tiancheng Li Chenchen Zhou Li Huang Yuyu Li Han Wang Peipei Duan Shujuan Zou Li Mei 2020International Journal of Oral Science2020,12,4:4
4Diphenyl cyclopropenone-centered polymers for site-specific CO-releasing and chain dissociation显示文摘The synthesis and stimuli-responsiveness of a diphenyl cyclopropenone(DPCP)-centered poly(methyl acrylate)(PMA)are presented.DPCP-centered PMA could release carbon monoxide(CO)upon UV light in a switched on-and-off manner.The CO-releasing process can be reported by the variations in photoluminescence spectra.In addition,DPCP moiety covalently embedded in the crosslinked polyurethane could also release CO under UV light.Of special,DPCP-centered PMA in solution was selectively dissociated at the phenol ester bond under the ultrasound,and a force-induced hydrolyzation reaction was revealed by D20 exchanging^1 H NMR spectra.The kinetic study reveals that small quantity of water could enhance the chain scission rate.This work provides a DPCP-centered polymer for sitespecific CO-releasing and chain dissociation.Chenchen Shao Hanyi Duan Yuqin Min Xinghong Zhang 2020Chinese Chemical Letters2020,31,1:2
5Microwave-assisted continuous flow phytosynthesis of silver nanoparticle/reduced graphene oxide composites and related visible light catalytic performance显示文摘The creation of an environmentally friendly synthesis method for silver nanomaterials(Ag-NPs)is an urgent concern for sustainable nanotechnology development.In the present study,a novel straightforward and green method for the preparation of silver nanoparti-cle/reduced graphene oxide(AgNP/rGO)composites was successfully developed through the combination of phytosynthesis,continuous flow synthesis and microwave-assistance.Oriental persimmon(Diospyros kaki Thunb.)extracts were used as both plant reducing and capping agents for fast online synthesis of AgNP/rGO composites.The experimental param-eters were optimized and the morphologies of the prepared materials were investigated.The characterization results reveal that spherical AgNPs were quickly synthesized and uni-formly dispersed on rGO sheets using the proposed online system.Fourier transform in-frared spectroscopy analysis confirmed that phenols,flavonoids,and other substances in the plant extracts played a decisive role in the synthesis of AgNP/rGO composites.Using sodium borohydride(NaBH4)degradation of p-nitrophenol(4-NP)as a model,the catalytic activity of the prepared AgNP/rGO materials was evaluated.The complete degradation of 4-NP was achieved within 12 min through the use of AgNP/rGO materials,and the compos-ite had a much better catalytic activity than the bare AgNPs and rGO had.Compared with the conventional chemical method,our online method is facile,fast,cost-efficient,and en-vironmentally friendly.Houyu Wang Chun-Gang Yuan Chenchen Liu Xuelei Duan Qi Guo Yiwen Shen Jingfu Liu Yongsheng Chen 2022Journal of Environmental Sciences2022,34,5:2
6Modulating effects of Astragalus polysaccharide on immune disorders via gut microbiota and the TLR4/NF-κB pathway in rats with syndrome of dampness stagnancy due to spleen deficiency显示文摘The syndrome of dampness stagnancy due to spleen deficiency(DSSD)is relatively common globally.Although the pathogenesis of DSSD remains unclear,evidence has suggested that the gut microbiota might play a significant role.Radix Astragali,used as both medicine and food,exerts the effects of tonifying spleen and qi.Astragalus polysaccharide(APS)comprises a macromolecule substance extracted from the dried root of Radix Astragali,which has many pharmacological functions.However,whether APS mitigates the immune disorders underlying the DSSD syndrome via regulating gut microbiota and the relevant mechanism remains unknown.Here,we used DSSD rats induced by high-fat and low-protein(HFLP)diet plus exhaustive swimming,and found that APS of moderate molecular weight increased the body weight gain and immune organ indexes,decreased the levels of interleukin-1β(IL-1β),IL-6,and endotoxin,and suppressed the Toll-like receptor 4/nuclear factor-κB(TLR4/NF-κB)pathway.Moreover,a total of 27 critical genera were significantly enriched according to the linear discriminant analysis effect size(LEfSe).APS increased the diversity of the gut microbiota and changed its composition,such as reducing the relative abundance of Pseudoflavonifractor and Paraprevotella,and increasing that of Parasutterella,Parabacteroides,Clostridium XIVb,Oscillibacter,Butyricicoccus,and Dorea.APS also elevated the contents of short-chain fatty acids(SCFAs).Furthermore,the correlation analysis indicated that 12 critical bacteria were related to the body weight gain and immune organ indexes.In general,our study demonstrated that APS ameliorated the immune disorders in DSSD rats via modulating their gut microbiota,especially for some bacteria involving immune and inflammatory response and SCFA production,as well as the TLR4/NF-κB pathway.This study provides an insight into the function of APS as a unique potential prebiotic through exerting systemic activities in treating DSSD.Wenxiao ZHAO Chenchen DUAN Yanli LIU Guangying LU Qin LYU Xiumei LIU Jun ZHENG Xuelian ZHAO Shijun WANG Haijun ZHAO 2023Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,7:1
7Microenvironmental stiffness mediates cytoskeleton re-organization in chondrocytes through laminin-FAK mechanotransduction显示文摘Microenvironmental biophysical factors play a fundamental role in controlling cell behaviors including cell morphology,proliferation,adhesion and differentiation,and even determining the cell fate.Cells are able to actively sense the surrounding mechanical microenvironment and change their cellular morphology to adapt to it.Although cell morphological changes have been considered to be the first and most important step in the interaction between cells and their mechanical microenvironment,their regulatory network is not completely clear.In the current study,we generated silicon-based elastomer polydimethylsiloxane(PDMS)substrates with stiff(15:1,PDMS elastomer vs.curing agent)and soft(45:1)stiffnesses,which showed the Young’s moduli of~450 k Pa and 46 kPa,respectively,and elucidated a new path in cytoskeleton re-organization in chondrocytes in response to changed substrate stiffnesses by characterizing the axis shift from the secreted extracellular protein lamininβ1,focal adhesion complex protein FAK to microfilament bundling.We first showed the cellular cytoskeleton changes in chondrocytes by characterizing the cell spreading area and cellular synapses.We then found the changes of secreted extracellular linkage protein,lamininβ1,and focal adhesion complex protein,FAK,in chondrocytes in response to different substrate stiffnesses.These two proteins were shown to be directly interacted by Co-IP and colocalization.We next showed that impact of FAK on the cytoskeleton organization by showing the changes of microfilament bundles and found the potential intermediate regulators.Taking together,this modulation axis of lamininβ1-FAK-microfilament could enlarge our understanding about the interdependence among mechanosensing,mechanotransduction,and cytoskeleton re-organization.Chenchen Zhou Mengmeng Duan Daimo Guo Xinmei Du Demao Zhang Jing Xie 2022International Journal of Oral Science2022,14,2:0
8Annexin A5 derived from matrix vesicles protects against osteoporotic bone loss via mineralization显示文摘Matrix vesicles(MVs)have shown strong effects in diseases such as vascular ectopic calcification and pathological calcified osteoarthritis and in wound repair of the skeletal system due to their membranous vesicle characteristics and abundant calcium and phosphorus content.However,the role of MVs in the progression of osteoporosis is poorly understood.Here,we report that annexin A5,an important component of the matrix vesicle membrane,plays a vital role in bone matrix homeostasis in the deterioration of osteoporosis.We first identified annexin A5 from adherent MVs but not dissociative MVs of osteoblasts and found that it could be sharply decreased in the bone matrix during the occurrence of osteoporosis based on ovariectomized mice.We then confirmed its potential in mediating the mineralization of the precursor osteoblast lineage via its initial binding with collagen type I to achieve MV adhesion and the subsequent activation of cellular autophagy.Finally,we proved its protective role in resisting bone loss by applying it to osteoporotic mice.Taken together,these data revealed the importance of annexin A5,originating from adherent MVs of osteoblasts,in bone matrix remodeling of osteoporosis and provided a new strategy for the treatment and intervention of bone loss.Guanyue Su Demao Zhang Tiantian Li Tong Pei Jie Yang Shasha Tu Sijun Liu Jie Ren Yaojia Zhang Mengmeng Duan Xinrui Yang Yang Shen Chenchen Zhou Jing Xie Xiaoheng Liu 2023Bone Research2023,11,4:0
9Local administration of liposomal-based Plekhf1 gene therapy attenuates pulmonary fibrosis by modulating macrophage polarization显示文摘Idiopathic pulmonary fibrosis(IPF)is a fatal interstitial lung disease with limited therapeutic options.Macrophages,particularly alternatively activated macrophages(M2),have been recognized to contribute to the pathogenesis of pulmonary fibrosis.Therefore,targeting macrophages might be a viable therapeutic strategy for IPF.Herein,we report a potential nanomedicinebased gene therapy for IPF by modulating macrophage M2 activation.In this study,we illustrated that the levels of pleckstrin homology and FYVE domain containing 1(Plekhf1)were increased in the lungs originating from IPF patients and PF mice.Further functionality studies identified the pivotal role of Plekhf1 in macrophage M2 activation.Mechanistically,Plekhf1 was upregulated by IL-4/IL-13 stimulation,after which Plekhf1 enhanced PI3K/Akt signaling to promote the macrophage M2 program and exacerbate pulmonary fibrosis.Therefore,intratracheal administration of Plekhf1 siRNA-loaded liposomes could effectively suppress the expression of Plekhf1 in the lungs and notably protect mice against BLM-induced lung injury and fibrosis,concomitant with a significant reduction in M2 macrophage accumulation in the lungs.In conclusion,Plekhf1 may play a crucial role in the pathogenesis of pulmonary fibrosis,and Plekhf1 siRNA-loaded liposomes might be a promising therapeutic approach against pulmonary fibrosis.Lifeng Yan Chenchen Hou Juan Liu Yi Wang Chenxi Zeng Jun Yu Tianyu Zhou Qing Zhou Shengzhong Duan Weining Xiong 2023Science China(Life Sciences)2023,66,11:0
