2Detection and characterization of diverse alpha-and betacoronaviruses from bats in China显示文摘Bats have been implicated as important reservoir hosts of alpha- and betacoronaviruses. In this study, diverse coronaviruses(CoVs) were detected in 50 of 951(positive rate 5.3%) intestinal specimens of eight bat species collected in four provinces and the Tibet Autonomous Region of China by pan-coronavirus RT-PCR screening. Based on 400-nt RNA-dependent RNA polymerase(Rd RP) sequence analysis, eight belonged to genus Alphacoronavirus and 42 to Betacoronavirus.Among the 50 positive specimens, thirteen gave rise to CoV full-length Rd RP gene amplification for further sequence comparison, of which three divergent sequences(two from a unreported province) were subjected to full genome sequencing. Two complete genomes of betacoronaviruses(JTMC15 and JPDB144) and one nearly-complete genome of alphacoronavirus(JTAC2) were sequenced and their genomic organization predicted. The present study has identified additional numbers of genetically diverse bat-borne coronaviruses with a wide distribution in China. 3miR-30a-5p promotes glomerular podocyte apoptosis via DNMT1-mediated hypermethylation under hyperhomocysteinemia显示文摘Abnormal elevation of homocysteine(Hcy)level is closely related to the development and progression of chronic kidney disease(CKD),with the molecular mech2anisms that are not fully elucidated.Given the demonstration that miR-30a-5p is specifically expressed in glomerular podocytes,in the present study we aimed to investigate the role and potential underlying mechanism of miR-30a-5p in glomerular podocyte apoptosis induced by Hcy.We found that elevated Hcy downregulates miR-30a-5p expression in the Cbs^(+/-)-mice and Hcy-treated podocytes,and miR-30a-5p directly targets the 3′-untranslated region(3′-UTR)of the forkhead box A1(FOXA1)and overexpression of miR-30a-5p inhibits FOXA1 expression.By nMS-PCR and MassARRAY quantitative methylation analysis,we showed the increased DNA methylation level of miR-30a-5p promoter both in vivo and in vitro.Meanwhile,dual-luciferase reporter assay showed that the region between--1400 and--921 bp of miR-30a-5p promoter is a possible regulatory element for its transcription.Mechanistic studies indicated that DNA methyltransferase enzyme 1(DNMT1)is the key regulator of miR-30a-5p,which in turn enhances miR-30a-5p promoter methylation level and thereby inhibits its expression.Taken together,our results revealed that epigenetic modification of miR-30a-5p is involved in glomerular podocyte injury induced by Hcy,providing a diagnostic marker candidate and novel therapeutic target in CKD induced by Hcy.Ning Ding Lin Xie Fei Ma Shengchao Ma Jiantuan Xiong Guanjun Lu Huiping Zhang Yideng Jiang 2022Acta Biochimica et Biophysica Sinica2022,54,1:4
4Synthesis and properties of novel soluble poly(aryl amine ketone)s as high-performance polymers显示文摘A series of poly(aryl amine ketone)s have been obtained by the condensation polymerization of different aromatic dibromides with different primary aromatic diamines via palladium-catalyzed aryl amination reaction. The structures of polymers are characterized by means of FT-IR, 1H NMR spectroscopy, and elemental analysis. The results show an agreement with the proposed structures. DSC and TG measurements show that polymers possess high glass transition temperature (Tg>170℃) and good thermal stability with high decomposition temperatures (TD>450℃). 6Extracellular-superoxide dismutase DNA methylation promotes oxidative stress in homocysteine-induced atherosclerosis显示文摘In the present study,we investigate the effect of homocysteine(Hcy)on extracellular-superoxide dismutase(ECSOD)DNA methylation in the aorta of mice,and explore the underlying mechanism in macrophages,trying to identify the key targets of Hcy-induced EC-SOD methylation changes.ApoE--mice are fed different diets for 15 weeks,EC-SODand DNAmethyltransferase1(DNMT1)expression levels aredetected byRT-PCRandwestern blot analysis.EC-SOD methylation levels are assessed by ntMS-PCR.After EC-SOD overexpression or knockdown in macrophages,following the transfection of macrophages with pEGFP-N1-DNMT1,the methylation levels of ECSOD are detected.Our data show that the concentrations of Hcy and the area of atherogenic lesions are significantly increased in ApoE--mice fed with a high-methionine diet,and have a positive correlation with the levels of superoxide anions,which indicates that Hcy-activated superoxide anions enhance the development of atherogenic lesions.EC-SOD expression is suppressed by Hcy,and the content of superoxide anion is increased when EC-SOD is silenced by RNAi in macrophages,suggesting that EC-SOD plays a major part in oxidative stress induced by Hcy.Furthermore,the promoter activity of EC-SOD is increased following transfection with the-1/-110o fragment,and EC-SOD methylation level is significantly suppressed by Hcy,and more significantly decreased upon DNMT1 overexpression.In conclusion,Hcy may alter the DNA methylation status and DNMT1 acts as the essential enzyme in the methyl transfer process to disturb the status of EC-SOD DNA methylation,leading to decreased expression of Ec-sODandincreasedoxidativestressandatherosclerosis.Shengchao Ma Guanjun Lu Qing Zhang Ning Ding Yuzhen Jie Hui Zhang Lingbo Xu Lin Xie Xiaoling Yang Huiping Zhang Yideng Jiang 2022Acta Biochimica et Biophysica Sinica2022,54,9:1

10Sensor Network Structure Recognition Based on P-law显示文摘A sensor graph network is a sensor network model organized according to graph network structure.Structural unit and signal propagation of core nodes are the basic characteristics of sensor graph networks.In sensor networks,network structure recognition is the basis for accurate identification and effective prediction and control of node states.Aiming at the problems of difficult global structure identification and poor interpretability in complex sensor graph networks,based on the characteristics of sensor networks,a method is proposed to firstly unitize the graph network structure and then expand the unit based on the signal transmission path of the core node.This method which builds on unit patulousness and core node signal propagation(called p-law)can rapidly and effectively achieve the global structure identification of a sensor graph network.Different from the traditional graph network structure recognition algorithms such as modularity maximization and spectral clustering,the proposed method reveals the natural evolution process and law of graph network subgroup generation.Experimental results confirm the effectiveness,accuracy and rationality of the proposed method and suggest that our method can be a new approach for graph network global structure recognition.Chuiju You Guanjun Lin Jinming Qiu Ning Cao Yundong Sun Russell Higgs 2023Computer Systems Science & Engineering2023,46,8:0
11Synthesis and properties of novel heat-resistant fluorescent conjugated polymers with bisindolylmaleimide显示文摘Conjugated polymers with bisindolylmaleimide(BIM) backbone are obtained by the condensation polymerization of methyl and octanyl N-substituted BIMs with 4,4'-difluoro-diphenylsulfone and4,4'-difluoro-diphenylketone. The structures of polymers are confirmed by FTIR and NMR spectroscopy.The polymers exhibit both high glass transition temperatures(T_g>175 ℃) and high decomposition temperatures(T_5>395 ℃). 12Multidisciplinary Treatment for Severe Secondary Raynaud’s Phenomenon:A Case Report显示文摘Raynaud’s phenomenon is a symptom complex manifested as intermittent fingertip ischemia caused by cold or other sympathetic drivers.Secondary Raynaud’s phenomenon is often more severe and could even lead to finger ulceration,making it particularly complicated to treat.We describe a case of severe Raynaud’s phenomenon secondary to subclinical hypothyroidism lasting for more than 6 hours in a 65-year-old woman.The patient was also diagnosed with hypothyroidism,epilepsy,and secondary soft tissue infection of the right middle and ring fingers.After careful multidisciplinary consultation and discussion,the patient received vasodilation,anticoagulation,thyroxine supplementation,stellate ganglion block,hyperbaric oxygen therapy and debridement.The patient responded well to the medication,avoiding amputation or obviously dysfunction.Multidisciplinary team gathering the doctors from different departments proposes appropriate strategies for patients with severe Raynaud’s phenomenon and could improve the prognosis and satisfaction of patient effectively.Cuibo Leng Guanjun Lin Hong Cao Zijia Liu 2022Chinese Medical Sciences Journal2022,37,4:0
