| 1 | Oral administration of dibutyryl adenosine cyclophosphate improved growth performance in weaning piglets by enhancing lipid fatty acids metabolism显示文摘Dibutyryl adenosine cyclophosphate(dbc AMP-Ca), an analog of cyclic adenosine monophosphate(c AMP), plays greater roles in regulating physiological activities and energy metabolism than c AMP. The aim of this study was to investigate the effect of oral administration of dbc AMP-Ca on growth performance and fatty acids metabolism in weaning piglets. A total of 14 early weaning piglets(7 ± 1 d of age,3.31 ± 0.09 kg, Landrace ? Large White ? Duroc) were randomly divided into 2 groups: control group and dbc AMP-Ca group, and the piglets received 7 m L of 0.9% Na Cl or 1.5 mg dbc AMP-Ca dissolved in 7 m L of 0.9% Na Cl per day for 10 d, respectively. The results showed that the average daily gain(ADG)increased by 109.17%(P < 0.05) in the dbc AMP-Ca group compared with the control group. Besides,dbc AMP-Ca significantly decreased blood high density lipoprotein cholesterol(HDLC) concentration(P < 0.05) and significantly increased blood low density lipoprotein cholesterol(LDLC) concentration(P < 0.05) compared with the control group. Further, liver C18:2 n6 t content significantly increased in dbc AMP-Ca group(P < 0.05) compared with the control group. With the increase of C18:2 n6 t content,the m RNA expression levels of peroxisome proliferator-activated receptor a(PPARa) and hormone sensitive glycerol three lipase(HSL), of which genes are related to lipid metabolism, were also significantly increased(P < 0.05 or P < 0.01). | 2 | Paternal Zn-deficiency abolishes metabolic effects in offspring induced by diet type显示文摘Accumulating evidence implicates that offspring are susceptible to paternal alterations in numerous fetal disorders,such as growth and metabolic defects.However,less study has been conducted to define the relationship between paternal zinc deficiency(ZnD)and energy metabolism of offspring.In the present study,we used a paternal ZnD exposure(Zn at 0.3 mg/g)model to test energy metabolism of male and female offspring with the intervention of diet type(high-fat diet and low-fat diet).Our results demonstrated that paternal ZnD decreased body weight(BW)gain per week(P<0.01)and ME intake per week(P<0.05)at 11 weeks in male offspring with high-fat diet intervention but not in female offspring.Further,anabolism and catabolism of hepatic energy products also exhibited alterations.ZnD attenuated liver glucose but increased lipids content accompanied with elevated adiponectin and reduction in leptin level in serum,which exhibited lipid metabolic disturbance and smaller ratio of liver weight to BW in male but not female offspring.The qRT-PCR and liver energy metabolites analysis revealed that paternal ZnD mainly induced reduction in glucose tolerance and lowered glucose uptaking ability in male offspring and thereby alleviated glycolysis and the tricarboxylic acid cycle(TCA)cycle,which displayed a male gender-dependency.Therefore,we propose that paternal ZnD abolishes metabolic effects in male offspring induced by diet type intervention.Our findings reveal a novel link between paternal Zn-D and offspring energy metabolism. | Guanya Li Zhenglin Dong Shusheng Yue Dan Wan Yulong Yin | 2022 | Animal Nutrition2022,,1: | 0 |
| 3 | Bariatric surgery induces alterations in effective connectivity between the orbitofrontal cortex and limbic regions in obese patients显示文摘Obese subjects show enhanced brain responses in motivation and reward neurocircuitry encompassing sensory and somatic integration-interception,motivation-reward(striatal),emotion,and memory processes,which attenuate frontal region activation during food cues.Bariatric surgery(BS) is the only reliable treatment for morbid obesity.Unfortunately,it is unknown how BS affects neurocircuitry after weight loss.We aimed to examine effects of BS on the basal activity of brain areas involved in reward and motivation processing,emotion,memory,and gut-brain interaction.We combined resting-state fMRI with amplitude of low-frequency fluctuation(ALFF) and Granger causality analysis(GCA) to assess interactions between regions within the frontal-mesolimbic circuitry in 16 obese subjects(OB) and 22 normal-weight(NW) subjects.The OB group was studied at baseline and 1 month post BS.Comparisons between OB and NW,and preand post BS showed significant differences in ALFF in areas involved in drive(caudate,orbitofrontal cortex(OFC)),arousal(thalamus),and conditioning/memory(amygdala,hippocampus)(P < 0.05,FDR correction).GCA revealed that in the OB group,the OFC had greater connectivity to limbic regions(amygdala,hippocampus,and medial thalamus) and the caudate.Post BS,the connectivity of the OFC to limbic regions decreased,whereas the connectivity from the amygdala and hippocampus to the caudate and thalamus was enhanced,particularly in subjects with lower body mass index(BMI).OFC activation in the OB group was associated with BMI prior to surgery,and changes in OFC post surgery were associated with alterations in BMI.Overall,the functional connectivity of the OFC was significantly decreased.As it is important for salience attribution and connected to limbic brain regions involved with emotional reactivity and conditioning after BS,its significant association with BMI changes indicates the contribution of OFC changes to the improved control of eating behavior after surgery. | Shijun DUAN Gang JI Guanya LI Yang HU Wenchao ZHANG Jia WANG Dardo TOMASI Nora DVOLKOW Yongzhan NIE Guangbin CUI Gene-Jack WANG Yi ZHANG | 2020 | Science China(Information Sciences)2020,63,7: | 0 |

| 5 | Role of iron in host-microbiota interaction and its effects on intestinal mucosal growth and immune plasticity in a piglet model显示文摘Iron is an essential trace element for both the host and resident microbes in the gut. In this study, iron was administered orally and parenterally to anemic piglets to investigate the role of iron in host-microbiota interaction and its effects on intestinal mucosal growth and immune plasticity. We found that oral iron administration easily increased the abundance of Proteobacteria and Escherichia-Shigella, and decreased the abundance of Lactobacillus in the ileum. Furthermore, similar bacterial changes, namely an increase in Proteobacteria, Escherichia-Shigella, and Fusobacterium and a reduction in the Christensenellaceae_R-7_group,were observed in the colon of both iron-supplemented groups. Spearman's correlation analysis indicated that the changed Fusobacterium, Fusobacteria and Proteobacteria in the colon were positively correlated with hemoglobin, colon and spleen iron levels. Nevertheless, it was found that activated m TOR1 signaling, improved villous height and crypt depth in the ileum,enhanced immune communication, and increased protein expression of IL-22 and IL-10 in the colon of both iron-supplemented groups. | 6 | The effect of nano-calcium carbonate onβ-glucosidase immobilized by alginate and chitosan显示文摘Nanotechnology has become the most promising domain to boost the efficiency of enzymes.Enzymes are vital as a green catalyst in many industries,food,pharmaceutical and biomedical,etc.The immobilization process of the enzyme increases its catalytic properties.In this research,a novel method is presented to describe the effect of nano-calcium carbonate on the characteristics of immobilizedβ-glucosidase,which was extracted from the Agrocybe aegirit.The nano-CaCO_(3)was produced using the eco-friendly natural deep eutectic solvent.The pure nano-CaCO_(3)was observed as vaterite,with a size of about 300 nm.The nano-calcium carbonate was coated by a natural polymer sodium alginate compound and then adsorbed chitosan.Further,this obtained composite is cross-linked by the bioactive genipin to immobilize theβ-glucosidase.The enzyme/protein loading ratio to the supports was 1:4,respectively.The recovery efficiency of immobilizedβ-glucosidase was 89.3%,and immobilization yield was 96.452%.Chitosan-coated nano-CaCO_(3)was used as a carrier for immobilization ofβ-glucosidase to improve its stability and reusability.In addition to stability and reusability,pH tolerance,temperature tolerance,and enzyme kinetics are the significant parameters that illustrate the proficiency of an immobilized enzyme.The measured optimal enzymatic reaction conditions for the immobilizedβ-glucosidase were 50℃and pH 6.Furthermore,it has shown noticeable improvements in thermo-stability and pH tolerance.Temperature tolerance was observed 50%to the initial activity of the immobilized enzyme even after the 3 h of incubation at 50℃,while pH tolerance was noticed more than 50%and 40%at pH 7 and 8,respectively.The K_(m)and V_(max)values of free and immobilizedβ-glucosidase to 4-nitrophenylβ-D-glucopyranoside were 1.549μmol/L/min,0.346 mmol/L and 0.532μmol/L/min,0.080 mmol/L,respectively.The immobilizedβ-glucosidase retains its storability 80%even after 30 days of storage at 4℃and maintains 93.1%of its residual activity by reusing up to 10 cycles. | Mariam Iqbal Yuefeng Deng Qiaoyue Chen Chengli Yang Yihui Zhu Ziqian Chen Jianjun Wang Kaishun Hu Guanya He Dali Li | 2022 | Green Synthesis and Catalysis2022,3,3: | 0 |
