2Targeting BMI-1-mediated epitheliale-mesenchymal transition to inhibit colorectal cancer liver metastasis显示文摘Liver is the most common metastatic site for colorectal cancer(CRC),there is no satisfied approach to treat CRC liver metastasis(CRCLM).Here,we investigated the role of a polycomb protein BMI-1 in CRCLM.Immunohistochemical analysis showed that BMI-1 expression in liver metastases was upregulated and associated with T4 stage,invasion depth and right-sided primary tumor.Knockdown BMI-1 in high metastatic HCT116 and LOVO cells repressed the migratory/invasive phenotype and reversed epithelialemesenchymal transition(EMT),while BMI-1 overexpression in low metastatic Ls174 T and DLD1 cells enhanced invasiveness and EMT.The effects of BMI-1 in CRC cells were related to upregulating snail via AKT/GSK-3βpathway.Furthermore,knockdown BMI-1 in HCT116 and LOVO cells reduced CRCLM using experimental liver metastasis mice model.Meanwhile,BMI-1 overexpression in Ls174 T and DLD1 significantly increased CRCLM.Moreover,sodium butyrate,a histone deacetylase and BMI-1 inhibitor,reduced HCT116 and LOVO liver metastasis in immunodeficient mice.Our results suggest that BMI-1 is a major regulator of CRCLM and provide a potent molecular target for CRCLM treatment.Zhiyao Xu Zhuha Zhou Jing Zhang Feichao Xuan Mengjing Fan Difan Zhou Zhenyu Liuyang Ximei Ma Yiyang Hong Yihong Wang Sherven Sharma Qinghua Dong Guanyu Wang 2021Acta Pharmaceutica Sinica B2021,11,5:8
3Iso-UniK:lightweight multi-process unikernel through memory protection keys显示文摘Unikernel,specializing a minimalistic libOS with an application,is an attractive design for cloud computing.However,the Achilles’heel of unikernel is the lack of multi-process support,which makes it less flexible and applicable.Many applications rely on the process abstraction to isolate different components.For example,Apache with the multi-processing module isolates a request handler in a process to guarantee security.Prior art tackles the problem by simulating multi-process with multiple unikernels,which is incompatible with existing cloud providers and also introduces high overhead.This paper proposes Iso-UniK,a new unikernel design enabling multi-task applications with the support of both functionality and isolation.Iso-UniK leverages a recent hardware feature,named Intel Memory Protection Key(Intel MPK),to provide lightweight and efficient isolation for multi-process in unikernel.Our design has three benefits compared with previous approaches.First,Iso-UniK does not need hypervisor support and is thus compatible with existing cloud computing platforms;second,Iso-UniK promises fast system calls with only 45 cycles;last,a process can be isolated with a flexible configuration.We have implemented a prototype based on OSv,a unikernel system supporting unmodified applications.Iso-UniK can achieve fast fork operation with only 66μs for multi-process applications.Our evaluation shows that the isolation and multi-process support in Iso-UniK will not damage the applications’performance.Guanyu Li Dong Du Yubin Xia 2018Cybersecurity2018,1,1:0
4Emerging drug design strategies in anti-influenza drug discovery显示文摘Influenza is an acute respiratory infection caused by influenza viruses(IFV),According to the World Health Organization(WHO),seasonal IFV epidemics result in approximately 3-5 million cases of severe illness,leading to about half a million deaths worldwide,along with severe economic losses and social burdens.Unfortunately,frequent mutations in IFV lead to a certain lag in vaccine development as well as resistance to existing antiviral drugs.Therefore,it is of great importance to develop anti-IFV drugs with high efficiency against wild-type and resistant strains,needed in the fight against current and future outbreaks caused by different IFV strains.In this review,we summarize general strategies used for the discovery and development of antiviral agents targeting multiple IFV strains(including those resistant to available drugs).Structure-based drug design,mechanism-based drug design,multivalent interaction-based drug design and drug repurposing are amongst the most relevant strategies that provide a framework for the development of antiviral drugs targeting IFV.Chuanfeng Liu Lide Hu Guanyu Dong Ying Zhang Edeildo Ferreira da Silva-Jinior Xinyong Liu Luis Menendez-Arias Peng Zhan 2023Acta Pharmaceutica Sinica B2023,13,12:0
