| 4 | Effect of Gubi prescription on caveolin-1 expression and phosphoinositide 3 kinase/protein kinase B and Fas signal pathways in rats with knee osteoarthritis显示文摘OBJECTIVE: To investigate the effects of Gubi prescription on the expression of caveolin-1, and the phosphoinositide 3 kinase/protein kinase B(PI3 K/Akt) and Fas signal pathways in rats with knee osteoarthritis(KOA).METHODS: Forty KOA model rats were established using a modification of Hulth's method. Rats were divided into five groups by the random number method: model, positive drug(Vicolli group), and high-, medium-, and low-dose Gubi prescription groups(n = 8/group). In the sham surgery group(n = 8), only anterior and posterior cruciate ligaments of rats were exposed during surgery. A normal group(n = 8) consisted of rats with no treatment. Rats were intragastrically administered corresponding drugs once every day for eight consecutive weeks. Then, rat synovial membranes were extracted and histomorphological changes were recorded. m RNA expression was measured by q-PCR.Serum superoxide dismutase(SOD), malondialdehyde(MDA), nitric oxide(NO), and interleukin 1β(IL-1β) levels were measured. Western blotting determined the effects of Gubi prescription on protein expressions of caveolin-1, Bax, Bcl-2, Fas, and caspase-3 in chondrocytes from KOA rats. The knee cartilage of rats was excised and cultured under aseptic conditions. After coincubation of chondrocytes with Gubi prescription-containing serum, IL-1β, and si RNA, Western blotting was used to determine the protein expressions of caveolin-1, Bax, Bcl-2, Fas, and caspase-3.RESULTS: The morphological score of the articular synovium in the model group was significantly higher than in the normal group(P < 0.01). The morphological score in the high-and mediumdose Gubi prescription groups was lower than in the model group(P < 0.05). Chondrocytes from the decoction-containing serum group had a lower expression of Bax(P < 0.05), and higher expressions of Bcl-2(P < 0.05) and caspase-3(P < 0.05) compared with the model group. Chondrocytes in the decoction-containing serum group had higher expressions of Bax and Bcl-2(P < 0.01) and lower expressions of caveolin-1 and Fas(P < 0.05) compared with the model group. Compared with the model group, Bax and caspase-3 expressions were reduced in the chondrocytes of all three Gubi prescription groups(P < 0.05) whereas Bcl-2 expression was increased(P < 0.05). Compared with the model group, the expressions of caveolin-1 and Fas(P < 0.05) were reduced in groups that received high-and medium-doses of Gubi prescription. | 5 | The C-terminal 17 amino acids of the photoreceptor UVR8 is involved in the fine-tuning of UV-B signaling^FA显示文摘Plant UV-B responses are mediated by the photoreceptor UV RESISTANCE LOCUS 8 (UVR8). In re-sponse to UV-B irradiation, UVR8 homodimers dissociate into monomers that bind to the E3 ubiquitin ligase CONSTITUTIVE PHOTOMORPHOGENIC1 (COP1). The in-teraction of the C27 domain in the C-terminal tail of UVR8 with the WD40 domain of COP1 is critical for UV-B sig-naling. However, the function of the last 17 amino acids (C17) of the C-terminus of UVR8, which are adjacent to C27, is unknown, although they are largely conserved in land plants. In this study, we established that Arabidopsis thaliana UVR8 C17 binds to full-length UVR8, but not to COP1, and reduces COP1 binding to the remaining portion of UVR8, including C27. We hypothesized that overexpression of C17 in a wild-type background would have a dominant negative effect on UVR8 activity;however, C17 overexpression caused strong silencing of endogenous UVR8, precluding a detailed analysis. We therefore generated YFP-UVR8N423 transgenic lines, in which C17 was deleted, to examine C17 function in-directly. YFP-UVR8N423 was more active than YFP-UVR8, suggesting that C17 inhibits UV-B signaling by attenuating binding between C27 and COP1. | 6 | Using MiddRAD-seq data to develop polymorphic microsatellite markers for an endangered yew species显示文摘Microsatellites are highly polymorphic markers which have been used in a wide range of genetic studies.In recent years, various sources of next-generation sequencing data have been used to develop new microsatellite loci, but compared with the more common shotgun genomic sequencing or transcriptome data, the potential utility of RAD-seq data for microsatellite ascertainment is comparatively under-used.In this study, we employed MiddRAD-seq data to develop polymorphic microsatellite loci for the endangered yew species Taxus florinii. Of 8,823,053 clean reads generated for ten individuals of a population, 94,851(~1%) contained microsatellite motifs. These corresponded to 2993 unique loci, of which 526(~18%) exhibited polymorphism. Of which, 237 were suitable for designing microsatellite primer pairs, and 128 loci were randomly selected for PCR validation and microsatellite screening. Out of the 128 primer pairs, 16 loci gave clear, reproducible patterns, and were then screened and characterized in 24 individuals from two populations. The total number of alleles per locus ranged from two to ten(mean=4.875), and within-population expected heterozygosity from zero to 0.789(mean = 0.530),indicating that these microsatellite loci will be useful for population genetics and speciation studies of T. florinii. | 7 | Improvement of machining consistency during through-mask electrochemical large-area machining显示文摘Electrochemical machining(ECM) is an important machining technique for the aeronautical manufacturing industry. Through-mask ECM is a form of ECM for machining metal parts with a hole array. In order to extend the machining area, a serpentine flow channel with multiple curves was used for through-mask ECM. With the extension of the flow channel, ensuring a machining consistency along the flow channel has been a challenge. The electrolyte conductivity is the main factor affecting the machining consistency. To analyze the change rules of the electrolyte conductivity, variations in the bubble rate and the temperature of the electrolyte in the electrolyte flow were explored under different power sources. Results indicate that pulse-power machining can reduce variations in the bubble rate and the temperature in the serpentine flow channel, and then the electrolyte conductivity can be stabilized within a very small range. Experiments using through-mask ECM were conducted in two types of power sources. Experimental results support the importance of pulse-power machining. A 14×28 hole array with a 2.5 mm diameter was fabricated by a pulsed power source. | 8 | Expression of nerve growth factor precursor, mature nerve growth factor and their receptors during cerebral ischemia-reperfusion injury显示文摘We investigated nerve growth factor precursor (proNGF) and mature NGF expression in ischemic and non-ischemic cortices after cerebral ischemia-reperfusion injury. In both ischemic and non-ischemic cortices, proNGF was found to be present in the extracellular space and cytoplasm. In addition, mature NGF was expressed in extracellular space, but with a very low signal. In ischemic cortex only, proNGF was significantly decreased, reaching a minimal level at 1 day. Mature NGF was increased at 4 hours, then reached a minimal level at 3 days. The p75 neurotrophin receptor (p75NTR) was significantly decreased after ischemia, and increased at 3 days after ischemia. These results confirmed that proNGF was the predominant form of NGF during the pathological process of cerebral ischemia-reperfusion injury.
