1Protocatechuic aldehyde protects cardiomycoytes against ischemic injury via regulation of nuclear pyruvate kinase M2显示文摘Rescuing cells from stress damage emerges a potential therapeutic strategy to combat myocardial infarction.Protocatechuic aldehyde(PCA)is a major phenolic acid in Chinese herb Danshen(Salvia miltiorrhiza root).This study investigated whether PCA regulated nuclear pyruvate kinase isoform M2(PKM2)function to protect cardiomyocytes.In rats subjected to isoprenaline,PCA attenuated heart injury and protected cardiomyocytes from apoptosis.Through DARTS and CETSA assays,we identified that PCA bound and promoted PKM2 nuclear translocation in cardiomyocytes exposed to oxygen/glucose deprivation(OGD).In the nucleus,PCA increased the binding of PKM2 to β-Catenin via preserving PKM2 acetylation,and the complex,in cooperation with T-cell factor 4(TCF4),was required for transcriptional induction of genes encoding anti-apoptotic proteins,contributing to rescuing cardiomyocyte survival.In addition,PCA ameliorated mitochondrial dysfunction and prevented mitochondrial apoptosis dependent on PKM2.Consistently,PCA increased the binding of PKM2 to β-Catenin,improved heart contractive function,normalized heart structure and attenuated oxidative damage in mice subjected to artery ligation,but the protective effects were lost in Pkm2-deficient heart.Together,we showed that PCA regulated nuclear PKM2 function to rescue cardiomyocyte survival via β-Catenin/TCF4 signaling cascade,suggesting the potential of pharmacological intervention of PKM2 shuttle to protect the heart.Xunxun Wu Lian Liu Qiuling Zheng Haiping Hao Hui Ye Ping Li Hua Yang 2021Acta Pharmaceutica Sinica B2021,11,11:4

6Fabrication and photoluminescence of Er-doped ZnO thin films on SiO_2/Si substrate by pulsed laser deposition显示文摘Er doped ZnO thin films were grown on Si substrates using SiO2 buffer layer by pulsed laser deposition (PLD) method. The obtained films crystallize well and show high c-axis orientation. The Er content was evidently detected by the energy dispersive X-ray spectroscopy (EDS). Upon annealing in O2 ambience at different temperatures, the films show different photoluminescence properties at 1.54 μm. The samples annealed at 700 and 850 ℃ show intense photoluminescence peaks which enhance with the annealing temperature, while no obvious luminescence peaks are observed for the as-grown samples or annealed at 500 ℃. 10Dihydrotanshinone I preconditions myocardium against ischemic injury via PKM2 glutathionylation sensitive to ROS显示文摘Ischemic preconditioning(IPC)is a potential intervention known to protect the heart against ischemia/reperfusion injury,but its role in the no-reflow phenomenon that follows reperfusion is unclear.Dihydrotanshinone I(DT)is a natural compound and this study illustrates its role in cardiac ischemic injury from the aspect of IPC.Pretreatment with DT induced modest ROS production and protected cardiomyocytes against oxygen and glucose deprivation(OGD),but the protection was prevented by a ROS scavenger.In addition,DT administration protected the heart against isoprenaline challenge.Mechanistically,PKM2 reacted to transient ROS via oxidization at Cys423/Cys424,leading to glutathionylation and nuclear translocation in dimer form.In the nucleus,PKM2 served as a co-factor to promote HIF-1a-dependent gene induction,contributing to adaptive responses.In mice subjected to permanent coronary ligation,cardiac-specific knockdown of Pkm2 blocked DT-mediated preconditioning protection,which was rescued by overexpression of wild-type Pkm2,rather than Cys423/424-mutated Pkm2.In conclusion,PKM2 is sensitive to oxidation,and subsequent glutathionylation promotes its nuclear translocation.Although IPC has been viewed as a protective means against reperfusion injury,our study reveals its potential role in protection of the heart from no-reflow ischemia.Xunxun Wu Lian Liu Qiuling Zheng Hui Ye Hua Yang Haiping Hao Ping Li 2023Acta Pharmaceutica Sinica B2023,13,1:1

19Integrating Conjugate Gradients Into Evolutionary Algorithms for Large-Scale Continuous Multi-Objective Optimization显示文摘Large-scale multi-objective optimization problems(LSMOPs)pose challenges to existing optimizers since a set of well-converged and diverse solutions should be found in huge search spaces.While evolutionary algorithms are good at solving small-scale multi-objective optimization problems,they are criticized for low efficiency in converging to the optimums of LSMOPs.By contrast,mathematical programming methods offer fast convergence speed on large-scale single-objective optimization problems,but they have difficulties in finding diverse solutions for LSMOPs.Currently,how to integrate evolutionary algorithms with mathematical programming methods to solve LSMOPs remains unexplored.In this paper,a hybrid algorithm is tailored for LSMOPs by coupling differential evolution and a conjugate gradient method.On the one hand,conjugate gradients and differential evolution are used to update different decision variables of a set of solutions,where the former drives the solutions to quickly converge towards the Pareto front and the latter promotes the diversity of the solutions to cover the whole Pareto front.On the other hand,objective decomposition strategy of evolutionary multi-objective optimization is used to differentiate the conjugate gradients of solutions,and the line search strategy of mathematical programming is used to ensure the higher quality of each offspring than its parent.In comparison with state-of-the-art evolutionary algorithms,mathematical programming methods,and hybrid algorithms,the proposed algorithm exhibits better convergence and diversity performance on a variety of benchmark and real-world LSMOPs.Ye Tian Haowen Chen Haiping Ma Xingyi Zhang Kay Chen Tan Yaochu Jin 2022IEEE/CAA Journal of Automatica Sinica2022,9,10:1
204×4 MIMO fiber-wireless transmission based on an integrated four-channel directly modulated optical transceiver显示文摘In this paper, an integrated compact four-channel directly modulated analog optical transceiver is proposed and fabricated. The 3 d B bandwidth of this optical transceiver exceeds 20 GHz, and the measured spurious-free dynamic range is up to 91.2 d B · Hz^2∕3. The optical coupling efficiency(CE) is improved by using a precise submicron alignment technique for lens coupling in a transmitter optical subassembly, and the highest CE is achieved when the oblique angle of the arrayed waveguide grating using a silica-based planar lightwave circuit(AWG-PLC) in receiver optical sub assembly is set to 42°. Based on the proposed optical transceiver, we have experimentally demonstrated a 6.624 Gbit/s 4 × 4 multi-input multioutput(MIMO) 16-quadrature amplitude modulation orthogonal frequency division multiplexing(16 QAM-OFDM) radio signal over 15.5 km standard single mode fiber, together with 1.2 m wireless transmission in both an uplink and a downlink. To cope with the channel interference and noise of the fiber-wireless transmission system, a low-complexity MIMO demodulation algorithm based on lattice reduction zero-forcing(LR-ZF) is designed. In our experiment, 1.6 d B power penalty is achieved by using the proposed LR-ZF algorithm, compared to the commonly used zero-forcing algorithm.Moreover, this LR-ZF algorithm has much less complexity than the optimal maximum-likelihood sequence estimation(MLSE) at a given transmission performance.
