2Cloning, expression and protective immunity evalua-tion of the full-length cDNA encoding succinate dehydrogenase iron-sulfur protein of Schistosoma japonicum显示文摘1071-bp fragment was obtained from the Schistosoma japonicum (Chinese strain) adult cDNA library after the 3′ and 5′ ends of the incomplete expression sequence tag (EST) of succinate dehydrogenase iron-sulfur protein of Schistosoma japonicum (SjSDISP) were amplified by the anchored PCR with 2 pairs of primers designed according to the EST of SjSDISP and the sequence of multiclone sites of the library vector. Sequence analysis indicated that the fragment was a full-length cDNA with a complete open reading frame (ORF), encoding 278 amino acid residues. The fragment was cloned into prokary- otic expression vector pQE30, and subsequently sequenced and expressed in Escherichia coli. SDS-PAGE and Western-blot analyses showed that the recombinant protein was about 32 kD and could be recognized by the polyclonal antisera from rabbits immunized with Schistosoma japonicum adult worm antigen. Compared with the FCA controls, mice vaccinated with rSjSDISP (test) or rSjGST (posi- tive control) all revealed high levels of specific antibody and significant reduction in worm burden, liver eggs per gram (LEPG), fecal eggs per gram (FEPG) and intrauterine eggs. These results suggest that SjSDISP may be a novel and partially protective vaccine candidate against schistosomiasis. 3Marsdenia tenacissima injection induces the apoptosis of prostate cancer by regulating the AKT/GSK3β/STAT3 signaling axis显示文摘Marsdenia tenacissima injection,a standard Marsdenia tenacissima extract(MTE),has been approved as an adjuvant therapeutic agent for various cancers.Our previous study showed that MTE inhibited the proliferation and metastasis of prostate cancer(PCa)cells.However,the underlying mechanisms and active ingredients of MTE against PCa were not completely understood.This study revealed that MTE induced significant decreases in cell viability and clonal growth in PCa cells.In addition,MTE induced the apoptosis of DU145 cells by reducing the mitochondrial membrane potential and increasing the expression of Cleaved Caspase 3/7,Cyt c,and Bax.In vivo,DU145 xenografted NOD-SCID mice treated with MTE showed significantly decreased tumor size.TUNEL staining and Western blot confirmed the pro-apoptotic effects of MTE.Network pharmacology analysis collected 196 ingredients of MTE linked to 655 potential targets,and 709 PCa-associated targets were retrieved,from which 149 overlapped targets were screened out.Pathway enrichment analysis showed that the HIF-1,PI3K-AKT,and ErbB signaling pathways were closely related to tumor apoptosis.Western blot results confirmed that MTE increased the expression of p-AKT^(Ser473) and p-GSK3β^(Ser9),and decreased the expression of p-STAT3^(Tyr705) in vitro and in vivo.A total of 13 compounds in MTE were identified by HPLC-CAD-QTOF-MS/MS and UPLCQTOF-MS/MS.Molecular docking analysis indicated that six compounds may interact with AKT,GSK3β,and STAT3.In conclusion,MTE induces the endogenous mitochondrial apoptosis of PCa by regulating the AKT/GSK3β/STAT3 signaling axis,resulting in inhibition of PCa growth in vitro and in vivo.LI Xiaolan HE Songhua LIANG Wei ZHANG Weiquan CHEN Xin LI Qiaofeng YANG Xin LIU Yanying ZHU Dan LI Li LIU Buming SU Zhiheng CHEN Jie GUO Hongwei 2023Chinese Journal of Natural Medicines2023,21,2:2
4Intercalation and hybrid heterostructure integration of two-dimensional atomic crystals with functional organic semiconductor molecules显示文摘Van der Waals(vdW)integration affords semiconductor heterostructures without constrains of lattice matching and opens up a new realm of functional devices by design.A particularly interesting approach is the electrochemical intercalation of two-dimensional(2D)atomic crystal and formation of superlattices,which can provide scalable production of novel vdW heterostructures.However,this approach has been limited to the use of organic cations with non-functional aliphatic chains,therefore failed to take the advantage of the vast potentials in molecular functionalities(electronic,photonic,magnetic,etc.).Here we report the integration of 2D crystal(MoS_(2),WS_(2),highly oriented pyrolytic graphite(HOPG),WSe_(2) as model systems)with electrochemically inert organic molecules that possess semiconducting characteristics(including perylene-3,4,9,10-tetracarboxylic dianhydride(PTCDA),pentacene and fullerene),through on-chip electrochemical intercalation.An unprecedented long-range spatial feature of intercalation has been achieved,which allowed facile assembly of a vertical MoS_(2)-PTCDA-Si junction.The intercalated heterostructure shows significant modulation of the lateral transport,and leads to a molecular tunneling characteristic at the vertical direction.The general intercalation of charge neutral and functional molecules defines a versatile platform of inorganic/organic hybrid vdW heterostructures with significantly extended molecular functional building blocks,holding great promise in future design of nano/quantum devices.Wen He Han Zang Songhua Cai Zhangyan Mu Cheng Liu Mengning Ding Peng Wang Xinran Wang 2020Nano Research2020,13,11:2

7Determination of Chlorogenic Acid in Stems of Mussaenda pubescens Ait.f.by High Performance Liquid Chromatography显示文摘[Objectives] This study was conducted to establish a method for determination of chlorogenic acid in Mussaenda pubescens Ait.f.[Methods]HPLC used SHISEIDO C_(18) MG Ⅱ column( 5 μm,4.6 mml.D.× 250 mm) as chromatographic column and acetonitrile-0.4% phosphoric acid solution( 9∶91) as mobile phase.The separation was performed at a flow rate of 1.0 ml/min and a column temperature of 30 ℃,and the detection wavelength was set at 327 nm.[Results]Chlorogenic acid had a good linear relation in the range of 0.254 7-2.547 5 μg( R2= 0.999 9).The recovery rate was 97.8%,RSD = 1.82%( n = 9).[Conclusions]The content of chlorogenic acid in M.pubescens was determined by ultrasonic extraction and HPLC.The method was simple,stable and reliable and could be used for the quality control of M.pubescens.Qianhua ZHANG Dongping TU Lichun ZHAO Yi LUO Songhua HE Tao ZHU 2018Agricultural Biotechnology2018,7,6:1

9Flexible piezoelectric PVDF nanofiber film sensor by blow spinning显示文摘Flexible sensors can be widely applied in the area of lightweight,wearable and intelligent electronic devices.In aerospace,for example,flexible sensors are suitable for measuring pressure in complex aircraft structures.In recent years,flexible sensors have been well developed;however,most of them lack circuit parts,meaning that the sensors have to be connected to an external electrical source.Additionally,the current traditional design limits their flexibility.Thus,this article proposes a self-powered flexible sensor based on PVDF nanofiber film,which is created through an efficient blow spinning method.The piezoelectric PVDF nanofiber films were prepared,and the flexible pressure sensor was assembled.The PVDF nanofiber film sensor showed good sensitivity and stability,and it can be applied for the detection of human physiological signals.HE Wei DAI ZhiJun ZOU KangKang LI XueYing HAO SongHua WANG HaoLun 2023Science China(Technological Sciences)2023,66,3:0
