1Ketogenic diet poses a significant effect on imbalanced gut microbiota in infants with refractory epilepsy显示文摘AIM To investigate whether patients with refractory epilepsy and healthy infants differ in gut microbiota(GM),and how ketogenic diet(KD) alters GM.METHODS A total of 14 epileptic and 30 healthy infants were recruited and seizure frequencies were recorded. Stool samples were collected for 16 S r DNA sequencing using the Illumina Miseq platform. The composition of GM in each sample was analyzed with MOTHUR,and intergroup comparison was conducted by R software.RESULTS After being on KD treatment for a week,64% of epileptic infants showed an obvious improvement,with a 50% decrease in seizure frequency. GM structure in epileptic infants(P1 group) differed dramatically from that in healthy infants(Health group). Proteobacteria,which had accumulated significantly in the P1 group,decreased dramatically after KD treatment(P2 group). Cronobacter predominated in the P1 group and remained at a low level both in the Health and P2 groups. Bacteroides increased significantly in the P2 group,in which Prevotella and Bifidobacterium also grew in numbers and kept increasing.CONCLUSION GM pattern in healthy infants differed dramatically from that of the epileptic group. 5Biofilm Microenvironment-Responsive Nanotheranostics for Dual-Mode Imaging and Hypoxia-Relief-Enhanced Photodynamic Therapy of Bacterial Infections显示文摘The formation of bacterial biofilms closely associates with infectious diseases.Until now,precise diagnosis and effective treatment of bacterial biofilm infections are still in great need.Herein,a novel multifunctional theranostic nanoplatform based on MnO_(2)nanosheets(MnO_(2)NSs)has been designed to achieve pH-responsive dual-mode imaging and hypoxia-relief-enhanced antimicrobial photodynamic therapy(aPDT)of bacterial biofilm infections.In this study,MnO_(2)NSs were modified with bovine serum albumin(BSA)and polyethylene glycol(PEG)and then loaded with chlorin e_(6)(Ce_(6))as photosensitizer to form MnO_(2)-BSA/PEG-Ce_(6)nanosheets(MBP-Ce_(6)NSs).After being delivered into the bacterial biofilm-infected tissues,the MBP-Ce_(6)NSs could be decomposed in acidic biofilm microenvironment and release Ce_(6)with Mn^(2+),which subsequently activate both fluorescence(FL)and magnetic resonance(MR)signals for effective dual-mode FL/MR imaging of bacterial biofilm infections.Meanwhile,MnO_(2)could catalyze the decomposing of H2O_(2)in biofilm-infected tissues into O_(2)and relieve the hypoxic condition of biofilm,which significantly enhances the efficacy of aPDT.An in vitro study showed that MBP-Ce_(6)NSs could significantly reduce the number of methicillin-resistant Staphylococcus aureus(MRSA)in biofilms after 635nm laser irradiation.Guided by FL/MR imaging,MRSA biofilm-infected mice can be efficiently treated by MBP-Ce_(6)NSs-based aPDT.Overall,MBP-Ce_(6)NSs not only possess biofilm microenvironment-responsive dual-mode FL/MR imaging ability but also have significantly enhanced aPDT efficacy by relieving the hypoxia habitat of biofilm,which provides a promising theranostic nanoplatform for bacterial biofilm infections.Weijun Xiu Siyu Gan Qirui Wen Qiu Qiu Sulai Dai Heng Dong Qiang Li Lihui Yuwen Lixing Weng Zhaogang Teng Yongbin Mou Lianhui Wang 2020Research2020,,1:7
6Modeling and simulation of urea-water-solution droplet evaporation and thermolysis processes for SCR systems显示文摘A reliable mathematical model of urea-water-solution(UWS) droplet evaporation and thermolysis is developed.The well known Abramzon–Sirignano evaporation model is corrected by introducing an adjustment coefficient considering the different evaporation behaviors of UWS droplet at different ambient temperatures. A semidetailed kinetic scheme of urea thermolysis is developed based on Ebrahimian's work. Sequentially, the evaporation characteristics, decomposition efficiency of a single UWS droplet and deposit formation are simulated. As a result, the relation of evaporation time, relative velocity, exhaust temperature and droplet initial diameter is presented. Synchronously, it indicates that temperature is the decisive factor for urea thermolysis. Different temperatures result in different deposit components, and deposit yield is significantly influenced by temperature and decomposition time. 7Effect of a transverse plasma jet on a shock wave induced by a ramp显示文摘We conducted experiments in a wind tunnel with Mach number 2 to explore the evolution of a transverse plasma jet and its modification effect on a shock wave induced by a ramp with an angle of 24°. The transverse plasma jet was created by arc discharge in a small cylindrical cavity with a 2 mm diameter orifice. Three group tests with different actuator arrangements in the spanwise or streamwise direction upstream from the ramp were respectively studied to compare their disturbances to the shock wave. As shown by a time-resolved schlieren system, an unsteady motion of the shock wave by actuation was found: the shock wave was significantly modified by the plasma jet with an upstream motion and a reduced angle. Compared to spanwise actuation, a more intensive impact was obtained with two or three streamwise actuators working together. From shock wave structures, the control effect of the plasma jet on the shock motion based on a thermal effect, a potential cause of shock modification, was discussed. Furthermore, we performed a numerical simulation by using the Improved Delayed Detached Eddy Simulation(IDDES) method to simulate the evolution of the transverse plasma jet plume produced by two streamwise actuators. The results show that flow structures are similar to those identified in schlieren images. 9Design and preliminary experimental investigation of a 4K Stirling-type pulse tube cryocooler with precooling显示文摘A Stirling-type pulse tube cryocooler (PTC) with precooling was designed and manufactured to investigate its performance at 4 K. Numerical simulation was carried out based on the well-known regenerator model REGEN with an emphasis on the performance of a 4 K stage regenerator of the Stirling-type PTC as influenced by the warm end temperature, pressure ratio, frequency and average pressure with helium-4 and helium-3 as the working fluid respectively. This study demonstrates that the use of a cold inertance tube can significantly improve the efficiency of a 4 K Stirling-type PTC. A preliminary experimental investigation was carried out with helium-4 as the working fluid and a refrigeration temperature of 4.23 K was achieved. 10Effect of neoadjuvant chemotherapy on the expression of hormone receptors and Ki67 in Chinese breast cancer patients:A retrospective study of 525 patients显示文摘This study was designed to investigate the effect of neoadjuvant chemotherapy on the expression of hormone receptors and Ki67 in Chinese female breast cancer patients. The expression of estrogen receptor(ER), progesterone receptor(PR) and Ki67 among 525 neoadjuvant chemotherapy cases was studied by immunohistochemistry.Differences between specimens made through preoperative core needle biopsy and excised tissue biopsy were observed. The positive rates of ER, PR and Ki67 in core needle biopsy and excised tissue biopsy were 65.3% and 63.2%, 51.0% and 42.6%, 65.6% and 43.4%, respectively. The expression of ER, PR and Ki67 in core needle biopsy and excised tissue biopsy had no statistically significant difference. However, after neoadjuvant chemotherapy, the discordance rates of ER, PR and Ki67 were 15.2%(79/521), 26.9%(140/520) and 44.8%(225/502), respectively. The ER, PR and Ki67 status changed from positive to negative in 7.5%(39/521), 13.3%(69/520) and 21.1%(106/502) of the patients, whereas ER, PR and Ki67 status changed from negative to positive in 7.7%(40/521), 13.6%(71/520)and 23.7%(119/502) of the patients, respectively. 11Immobilization of acetylcholinesterase on one-dimensional gold nanoparticles for detection of organophosphorous insecticides显示文摘This paper reports a simple method for immobilization of acetylcholinesterase (AChE) on one-dimensional (1D) gold (Au) nanoparticles for detection of organophosphorous (OP) insecticides. 1D Au nanoparticles were prepared by electrodeposition in the pores of an alumina template which was subsequently removed by 2.0 M NaOH solution. They were characterized by XRD and FESEM. The immobilized AChE retained its biological activity and catalyzed the hydrolysis of acetylthiocholine to form thiocholine, which was subsequently oxidized to produce detectable signals. Based on the inhibition toward the enzymatic activity of AChE by OP insecticides, sensitive detection of methamidophos (an OP insecticide) was performed. Under optimal conditions, the sensors could be used for the determination of methamidophos ranging from 0.004 to 24 μg/mL with the detection limit of 0.001 μg/mL. The developed OP insecticide biosensors exhibited satisfactory stability and reproducibility. 12Cholesterol-tuned liposomal membrane rigidity directs tumor penetration and anti-tumor effect显示文摘Recently, liposomes have been widely used in cancer therapeutics, but their anti-tumor effects are suboptimal due to limited tumor penetration. To solve this problem, researchers have made significant efforts to optimize liposomal diameters and potentials, but little attention has been paid to liposomal membrane rigidity. Herein, we sought to demonstrate the effects of cholesterol-tuned liposomal membrane rigidity on tumor penetration and anti-tumor effects. In this study, liposomes composed of hydrogenated soybean phospholipids(HSPC), 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy(polyethylene glycol)-2000](DSPE-PEG2000) and different concentrations of cholesterol were prepared. It was revealed that liposomal membrane rigidity decreased with the addition of cholesterol. Moderate cholesterol content conferred excellent diffusivity to liposomes in simulated diffusion medium, while excessive cholesterol limited the diffusion process. We concluded that the differences of the diffusion rates likely stemmed from the alterations in liposomal membrane rigidity, with moderate rigidity leading to improved diffusion. Next, the in vitro tumor penetration and the in vivo anti-tumor effects were analyzed. The results showed that liposomes with moderate rigidity gained excellent tumor penetration and enhanced anti-tumor effects. 14泌尿生殖系统小细胞癌:11年临床治疗经验显示文摘当前对于泌尿生殖系统的小细胞癌治疗及判断预后的影响因素尚存在较大争议。本研究回顾性分析了在2002年至2013年在复旦大学附属肿瘤医院诊治的18例泌尿生殖系统小细胞癌患者的临床资料,其中有10例来源于前列腺,7例来源于膀胱,1例来源于肾上腺。平均随访时间为15.5个月,平均无疾病进展时间为9.3个月。对其中的13例患者施行原发病灶切除术,在诊断为局限性泌尿生殖系统小细胞癌的患者中6例接受了根治性手术治疗,13例患者接受了以顺铂为基础的化疗。经统计学分析发现与乳酸脱氢酶较高的患者相比,乳酸脱氢酶处于正常范围的患者无疾病进展时间及总生存时间都明显延长(P=0.030,P=0.010),对于局限性小细胞癌患者,接受根治性手术治疗的患者与未接受根治手术的患者相比在无疾病进展时间上无统计学差异,但是在总生存时间上有明显差别。因此血液中乳酸脱氢酶高于正常值往往预示着患者的预后较差,对于诊断为局限性泌尿生殖系统小细胞肿瘤的患者,强烈推荐及早对患者施行根治性手术并联合以顺铂为基础的化疗。Kun Chang 2014Asian Journal of Andrology2014,16,5:5
15Cloning, expression and protective immunity evalua-tion of the full-length cDNA encoding succinate dehydrogenase iron-sulfur protein of Schistosoma japonicum显示文摘1071-bp fragment was obtained from the Schistosoma japonicum (Chinese strain) adult cDNA library after the 3′ and 5′ ends of the incomplete expression sequence tag (EST) of succinate dehydrogenase iron-sulfur protein of Schistosoma japonicum (SjSDISP) were amplified by the anchored PCR with 2 pairs of primers designed according to the EST of SjSDISP and the sequence of multiclone sites of the library vector. Sequence analysis indicated that the fragment was a full-length cDNA with a complete open reading frame (ORF), encoding 278 amino acid residues. The fragment was cloned into prokary- otic expression vector pQE30, and subsequently sequenced and expressed in Escherichia coli. SDS-PAGE and Western-blot analyses showed that the recombinant protein was about 32 kD and could be recognized by the polyclonal antisera from rabbits immunized with Schistosoma japonicum adult worm antigen. Compared with the FCA controls, mice vaccinated with rSjSDISP (test) or rSjGST (posi- tive control) all revealed high levels of specific antibody and significant reduction in worm burden, liver eggs per gram (LEPG), fecal eggs per gram (FEPG) and intrauterine eggs. These results suggest that SjSDISP may be a novel and partially protective vaccine candidate against schistosomiasis. 17Tissue expression and developmental regulation of chicken cathelicidin antimicrobial peptides显示文摘Cathelicidins are a major family of antimicrobial peptides present in vertebrate animals with potent microbicidal and immunomodulatory activities. Four cathelicidins, namely fowlicidins 1 to 3 and cathelicidin B1, have been identified in chickens. As a first step to understand their role in early innate host defense of chickens, we examined the tissue and developmental expression patterns of all four cathelicidins. Real-time PCR revealed an abundant expression of four cathelicidins throughout the gastrointestinal, respiratory, and urogenital tracts as well as in all primary and secondary immune organs of chickens. Fowlicidins 1 to 3 exhibited a similar tissue expression pattern with the highest expression in the bone marrow and lung, while cathelicidin B1 was synthesized most abundantly in the bursa of Fabricius. Additionally, a tissue-specific regulatory pattern was evident for all four cathelicidins during the first 28 days after hatching. The expression of fowlicidins 1 to 3 showed an age-dependent increase both in the cecal tonsil and lung, whereas all four cathelicidins were peaked in the bursa on day 4 after hatching, with a gradual decline by day 28. An abrupt augmentation in the expression of fowlicidins 1 to 3 was also observed in the cecum on day 28, while the highest expression of cathelicidin B1 was seen in both the lung and cecal tonsil on day 14. 18Efficacy of Rebixiao Chinese herbal tablets and Chinese formula granules in acute gout arthritis patients:a randomized,multicenter,double-blind,controlled trial显示文摘OBJECTIVE:To evaluate the clinical efficacy and safety of Rebixiao(RBX)Chinese herbal tablets(CHT)and Chinese formula granules(CFG)in the treatment of acute gout arthritis(AGA).METHODS:This randomized,multicenter,double-blind,controlled trial included 165 AGA patients with the damp-heat symptom pattern who were randomly divided into an RBX CHT group and an RBX CFG group and treated for 7 d at three centers.The total effective rates of the joint symptom score,Traditional Chinese Medicine(TCM)symptoms score,erythrocyte sedimentation rate(ESR),and C-reactive protein(CRP)were used to evaluate the clinical efficacy.Safety assessments were also performed.RESULTS:Of the 165 enrolled patients,147 completed the clinical observation.There was no difference in baseline between the two groups.The total effective rates of the joint symptom score were 94.36%and 97.36%,and the total effective rates of the TCM symptoms score were 95.77%and 97.36%in the CFG group and CHT group,respectively.No statistical difference was found between the two groups(P>0.05).Additionally,ESR and CRP were similar in both groups(P>0.05).Furthermore,treatment efficacy regarding TCM and joint symptoms,the ESR,and CRP were consistent within each center and among the different centers(P>0.05).In addition,the incidence of adverse events was 4.22%and 2.63%in the CFG group and CHT group,respectively,and no difference was observed between the two groups(P>0.05).CONCLUSION:RBX CFG and CHT have significant and similar efficacy in the treatment of AGA,and CFG did not increase adverse side effects.Wang Shasha Zhang Ying Dai Min Yang Min Leng Wenfei Peng Xiufeng Li Guojun Yang Jinbing Gan Jianping Zhu Fenglin Li Yanping Wu Bin 2020Journal of Traditional Chinese Medicine2020,40,4:3
