| 1 | Characteristics of the microbial communities in the integrated vertical-flow constructed wetlands显示文摘Microorganisms play an important role in removing pollutants from constructed wetlands.We investigated the microbial characteristics in a novel integrated vertical-flow constructed wetland(IVCW) ,which has been in operation in Wuhan,China since 1998.We used phospholipid fatty acid(PLFA) and amoA gene to analyze the structure and diversity of the microbial community within the IVCW.PLFA results suggested that the amount of bacterial PLFA was significantly higher than that of fungal PLFA,but the total microbial biomass represented by PLFA index was low in the system.Microbial spatial distribution showed significantly higher bacterial(both G+and G-) and fungal biomass in the surface than in the subsurface layers.The ratios of monounsaturated to branched PLFA demonstrated that an anaerobic layer sandwiched by two aerobic layers existed in the IVCW,consistent with the redox potential results.Analysis of the amoA revealed the presence of Nitrosomonas-like sequences in the surface substrate of the downflow chamber and apparent diversities of ammonia-oxidizing bacteria in the system.These results suggest that microorganisms,despite their relatively low biomass,have inhabited the IVCW,and the results will offer some valuable information on microbe to system designers and managers. | ZHOU Qiaohong,HE Feng,Zhang Liping,WANG Yanfen,WU Zhenbin State Key Laboratory of Freshwater Ecology and Biotechnology,Institute of Hydrobiology,Chinese Academy of Sciences,Wuhan 430072,China Department of Biology and Chemistry,City University of Hong Kong | 2009 | Journal of Environmental Sciences2009,21,9: | 14 |
| 2 | SHP2 promotes proliferation of breast cancer cells through regulating Cyclin D1 stabilityviathe PI3K/AKT/GSK3β signaling pathway显示文摘Objective:The tyrosine phosphatase SHP2 has a dual role in cancer initiation and progression in a tissue type-dependent manner.Several studies have linked SHP2 to the aggressive behavior of breast cancer cells and poorer outcomes in people with cancer.Nevertheless,the mechanistic details of how SHP2 promotes breast cancer progression remain largely undefined.Methods:The relationship between SHP2 expression and the prognosis of patients with breast cancer was investigated by using the TCGA and GEO databases.The expression of SHP2 in breast cancer tissues was analyzed by immunohistochemistry.CRISPR/Cas9 technology was used to generate SHP2-knockout breast cancer cells.Cell-counting kit-8,colony formation,cell cycle,and EdU incorporation assays,as well as a tumor xenograft model were used to examine the function of SHP2 in breast cancer proliferation.Quantitative RT-PCR,western blotting,immunofluorescence staining,and ubiquitination assays were used to explore the molecular mechanism through which SHP2 regulates breast cancer proliferation.Results:High SHP2 expression is correlated with poor prognosis in patients with breast cancer.SHP2 is required for the proliferation of breast cancer cellsin vitro and tumor growthin vivo through regulation of Cyclin D1 abundance,thereby accelerating cell cycle progression.Notably,SHP2 modulates the ubiquitin–proteasome-dependent degradation of Cyclin D1viathe PI3K/AKT/GSK3βsignaling pathway.SHP2 knockout attenuates the activation of PI3K/AKT signaling and causes the dephosphorylation and resultant activation of GSK3β.GSK3βthen mediates phosphorylation of Cyclin D1 at threonine 286,thereby promoting the translocation of Cyclin D1 from the nucleus to the cytoplasm and facilitating Cyclin D1 degradation through the ubiquitin–proteasome system.Conclusions:Our study uncovered the mechanism through which SHP2 regulates breast cancer proliferation.SHP2 may therefore potentially serve as a therapeutic target for breast cancer. | Yue Yuan Yanling Fan Zicong Gao Xuan Sun He Zhang Zhiyong Wang Yanfen Cui Weijie Song Zhaosong Wang Fei Zhang Ruifang Niu | 2020 | Cancer Biology & Medicine2020,17,3: | 11 |

| 4 | A pH-sensitive supramolecular nanosystem with chlorin e6 and triptolide co-delivery for chemo-photodynamic combination therapy显示文摘The combination of Ce6,an acknowledged photosensitizer,and TPL,a natural anticancer agent,has been demonstrated as a useful strategy to reinforce the tumor growth suppression,as well as decrease the systemic side effects compared with their monotherapy.However,in view of the optimal chemo-photodynamic combination efficiency,there is still short of the feasible nanovehicle to steadily co-deliver Ce6 and TPL,and stimuli-responsively burst release drugs in tumor site.Herein,we described the synergistic antitumor performance of a pH-sensitive supramolecular nanosystem,mediated by the host–guest complexing betweenβ-CD and acid pH-responsive amphiphilic co-polymer mPEG-PBAE-mPEG,showing the shell–core structural micelles with the tightβ-CD layer coating.Both Ce6 and TPLwere facilely co-loaded into the spherical supramolecular NPs(TPL+Ce6/NPs)by one-step nanoprecipitation method,with an ideal particle size(156.0 nm),acid pH-responsive drug release profile,and enhanced cellular internalization capacity.In view of the combination benefit of photodynamic therapy and chemotherapy,as well as co-encapsulation in the fabricated pH-sensitive supramolecular NPs,TPL+Ce6/NPs exhibited significant efficacy to suppress cellular proliferation,boost ROS level,lower MMP,and promote cellular apoptosis in vitro.Particularly,fluorescence imaging revealed that TPL+Ce6/NPs preferentially accumulated in the tumor tissue area,with higher intensity than that of free Ce6.As expected,upon 650-nm laser irradiation,TPL+Ce6/NPs exhibited a cascade of amplified synergistic chemo-photodynamic therapeutic benefits to suppress tumor progression in both hepatoma H22 tumor-bearingmice and B16 tumor-bearingmice.More importantly,lower systemic toxicitywas found in the tumor-bearingmice treated with TPL+Ce6/NPs.Overall,the designed supramolecular TPL+Ce6/NPs provided a promising alternative approach for chemo-photodynamic therapy in tumor treatment. | Yihan Wu Jingjing Li Xuemei Zhong Jinfeng Shi Yanfen Cheng Chenglin He Jiaxin Li Liang Zou Chaomei Fu Meiwan Chen Jinming Zhang Huile Gao | 2022 | Asian Journal of Pharmaceutical Sciences2022,17,2: | 4 |
| 5 | The HDAC inhibitor GCJ-490A suppresses c-Met expression through IKKα and overcomes gefitinib resistance in non-small cell lung cancer显示文摘Objective:The novel compound GCJ-490A has been discovered as a pan-histone deacetylase(HDAC)inhibitor that exerts potent inhibitory activity against HDAC1,HDAC3,and HDAC6.Because of the important roles of HDACs in lung cancer development and the high distribution of GCJ-490A in lung tissue,we explored the anti-tumor potency of GCJ-490A against non-small cell lung cancer(NSCLC)in vitro and in vivo in this study.Methods:The in vitro effects of GCJ-490A alone or combined with the EGFR inhibitor gefitinib against NSCLC were measured with proliferation,apoptosis,and colony formation assays.NSCLC xenograft models were used to investigate the efficacy of GCJ-490A combined with gefitinib for the treatment of NSCLC in vivo.Western blot assays,luciferase reporter assays,chromatin immunoprecipitation assays,quantitative real time-PCR,immunohistochemistry,and transcription factor activity assays were used to elucidate possible mechanisms.Results:GCJ-490A effectively inhibited NSCLC cell proliferation and induced apoptosis in vitro and in vivo.Interestingly,inhibition of HDAC1 and HDAC6 by GCJ-490A increased histone acetylation at the IKKαpromoter and enhanced IKKαtranscription,thus decreasing c-Met.Moreover,this c-Met downregulation was found to be essential for the synergistic anti-tumor activity of GCJ-490A and gefitinib.Conclusions:These findings highlight the promising potential of HDAC inhibitors in NSCLC treatment and provide a rational basis for the application of HDAC inhibitors in combination with EGFR inhibitors in clinical trials. | Ting He Yinglei Gao Yanfen Fang Yangming Zhang Shuwei Zhang Fajun Nan Jian Ding Yi Chen | 2022 | Cancer Biology & Medicine2022,19,8: | 3 |
| 6 | Spatiotemporal Trends and Ecological Determinants in Population by Elevation in China Since 1990显示文摘It is very important to understand the ecological and socio-economic factors in population distribution and their changes over time for the compilation of regional development planning and the guidance of rational population flow.Using surface-based population data for China from 1990 to 2015,the national distribution and dynamics of the human population by elevation are quantified based on 1-km cell-size gridded distribution datasets and 1-km cell-size DEM(digital elevation model).A geographical detector model is used to quantitatively analyze the dominant role of natural geographical factors,such as topography and climate,on the spatial distribution of population.Results show that:1)the population size and density decrease rapidly with elevation below 1000 m above the sea level,and the gap in population density between low-altitude areas and high-altitude areas increases with time because of the continuous growth of population density in low-altitude areas;2)the distribution of the population can be divided into five steps according to integrated population density(IPD),in proportions of 43∶35∶21∶1∶0,and that these proportions have remained stable over the last 25 yr;3)the basic pattern of population spatial distribution is determined by natural geographical environment factors,such as topography,climate,geomorphology,and their interactions;and 4)the development of society and the economy are the driving forces for the dynamic change in the population distribution during the study period,with the distribution pattern and dynamics of population by altitude in China providing a comprehensive reflection of various geographical elements on different spatial scales. | MA Chaoqun HE Yanfen | 2021 | Chinese Geographical Science2021,31,2: | 2 |

| 8 | The effect of increased atmospheric temperature and CO_2 concentration during crop growth on the chemical composition and in vitro rumen fermentation characteristics of wheat straw显示文摘This experiment was c onducted to investigate the effects of increased atmospheric temperature and CO_2concentration during crop growth on the chemical composition and in vitro rumen fermentation characteristics of wheat straw.The field experiment was carried out from November 2012 to June 2013 at Changshu(31°32'93 'N,120°41'88' E) agro-ecological experimental station.A total of three treatments were set.The concentration of CO_2was increased to 500 umol/mol in the first treatment(CO_2 group).The temperature was increased by 2 ℃ in the second treatment(TEM group) and the concentration of CO_2 and temperature were both increased in the third treatment(CO_2 + TEM group).The mean temperature and concentration of CO_2 in control group were 10.5 ℃ and413 umol/mol.At harvesting,the wheat straws were collected and analyzed for chemical composition and in vitro digestibility.Results showed that dry matter was significantly increased in all three treatments.Ether extracts and neutral detergent fiber were significantly increased in TEM and CO_2 + TEM groups.Crude protein was significantly decreased in CO_2 + TEM group.In vitro digestibility analysis of wheat straw revealed that gas production was significantly decreased in CO_2 and CO_2 + TEM groups.Methane production was significantly decreased in TEM and CO_2 + TEM groups.Ammonia nitrogen and microbial crude protein were significantly decreased in all three treatments.Total volatile fatty acids were significantly decreased in CO_2 and CO_2 + TEM groups.In conclusion,the chemical composition of the wheat straw was affected by temperature and CO_2 and the in vitro digestibility of wheat straw was reduced,especially in the combined treatment of temperature and CO_2. | Xiangyu He Yanping Wu Min Cai Chunlong Mu Weihong Luo Yanfen Cheng Weiyun Zhu | 2015 | Journal of Animal Science and Biotechnology2015,6,4: | 1 |

Methods: Human ovarian cancer HO-8910 cells were cultured in vitro. The inhibitory effect of SGI-1776 combination with DDP in sub-toxic concentration on induction on viability of human ovarian cancer HO-8910 cells was evaluated by the MTT assay. Cell apoptosis rate was analyzed by flow cytometry. The proteins expression level related to apoptosis were analyzed by Western blot. Results: SGI-1776 combination with DDP in sub-toxic concentration significantly inhibited the proliferation of human ovarian cancer HO-8910 cells, and proliferation inhibition rate was increased drastically compared with normal saline(NS) group or DDP group in sub-toxic concentration or SGI-1776 group in sub-toxic concentration(P﹤0.01). Apoptosis rate markedly increased after the treatment of SGI-1776 combination with DDP in sub-toxic concentration for 48 h. Western blot showed that the expression of bcl-2 protein was down-regulated and protein level of Bax and Cyto-c were depressed by SGI-1776 combination with DDP in sub-toxic concentration.
