| 2 | Ginsenoside Rh2 suppresses colon cancer growth by targeting the miR-150-3p/SRCIN1/Wnt axis显示文摘Ginsenoside Rh2,which is extracted from ginseng,exerts antitumor activity.Recent studies suggest that Rh2 may suppress the growth of colon cancer(CC)in vitro.However,the underlying mechanism remains unclear.In this study,we identified the relative levels of miR-150-3p in CC tissues and cells by a comprehensive strategy of data mining,computational biology,and real-time reverse transcription PCR(qRT-PCR)experiments.The regulatory effects of miR-150-3p/SRCIN1 on the proliferative and invasive abilities of CC cells are evaluated by CCK-8,EdU,wound healing,and transwell assays.Cell cycle-and apoptosis-related protein levels are assessed by western blot analysis.An in vivo tumor formation assay was conducted to explore the effects of miR-150-3p on tumor growth.Furthermore,bioinformatics and dual luciferase reporter assays are applied to determine the functional binding of miRNA to mRNA of the target gene.Finally,the relationship between Rh2 and miR-150-3p was further verified in SW620 and HCT-116 cells.miR-150-3p is downregulated in CC tissues and cell lines.Functional assays indicate that the upregulation of miR-150-3p inhibits tumor growth both in vivo and in vitro.In addition,SRCIN1 is upregulated in CC and predicts a poor prognosis,and it is the direct target for miR-150-3p.Moreover,the miR-150-3p mimic decreases Topflash/Fopflash-dependent luciferase activity,resulting in the inhibition of Wnt pathway activity.Rh2 can suppress the growth of CC by increasing miR-150-3p expression.Rh2 alleviates the accelerating effect on Wnt pathway activity,cell proliferation/migration,and colony formation caused by miR-150-3p inhibition.Rh2 inhibits the miR-150-3p/SRCIN1/Wnt axis to suppress colon cancer growth. | Shipeng Li Wenfeng Han Qichen He Yang Wang Gang Jin Youcheng Zhang | 2023 | Acta Biochimica et Biophysica Sinica2023,55,4: | 2 |
| 3 | Therapeutic effect of Qinghuayin(清化饮)against chronic atrophic gastritis through the inhibition of toll or interleukin-1 receptor domain-containing adaptor inducing interferon-β signaling pathway显示文摘OBJECTIVE:To examine the efficacy of Qinghuayin(清化饮,QHY)in rat chronic atrophic gastritis(CAG)models and explored the molecular mechanism of QHY in treating CAG.METHODS:In total,65 Wistar rats were randomly divided into the control(n=10)and CAG groups(n=55).CAG model rats were further divided into five groups:model(n=10),vitacoenzyme(n=10),low-dose QHY(n=10),medium-dose QHY(n=10),and high-dose QHY groups(n=10).We analyzed histopathological changes using hematoxylin and eosin staining and measured interleukin(IL)-6 and IL-8 levels in serum using enzyme-linked immunosorbent assay(ELISA)(Boster Bio,Pleasanton,USA).In addition,gastrin(GAS),pepsinogen I(PGI),and PGII expressions were evaluated using ELISA.The protein and m RNA expression of toll-like receptor 4(TLR4)and toll or interleukin-1 receptor domaincontaining adaptor inducing interferon-β(TRIF)was detected by Western blotting and quantitative reverse transcription-polymerase chain reaction,respectively.RESULTS:Our results revealed that histopathological changes in CAG model rates could be restored by low-,medium-,and high-dose QHY.The changes in GAS and PGI/II expression demonstrated that QHY improved CAG.Serum IL-6 and IL-levels were decreased by QHY administration.TLR4 and TRIF were upregulated at the m RNA and protein levels in the model group but downregulated by QHY administration.CONCLUSION:We concluded that QHY could effectively improve the histopathological changes of the gastric mucosa induced by CAG in rats.The therapeutic mechanism of QHY may be related to inhibition of the inflammatory factors IL-6 and IL-8 and suppression of TLR4/TRIF m RNA and protein expression. | HE Youcheng YAO Xiaoling LI Sihan ZHOU Hongjian XU Ruoying ZHENG Rong XIAO Wendi SU Ning LIN Ping HUANG Minghan | 2022 | Journal of Traditional Chinese Medicine2022,42,2: | 1 |
| 4 | Variability of size-fractionated chlorophyll a in the high-latitude Arctic Ocean in summer 2020显示文摘The size structure of phytoplankton has considerable effects on the energy flow and nutrient cycling in themarine ecosystem,and thus is important to marine food web and biological pump.However,its dynamics in the high-latitude Arctic Ocean,particularly ice-covered areas,remain poorly understood.We investigated size-fractionated chlorophyll a(Chl a)and related environmental parameters in the highly ice-covered Arctic Ocean during the summer of 2020,and analyzed the relationship between Chl adistribution and water mass through cluster analysis.Results showed that inorganic nutrients were typically depleted in the upper layer of the Canada Basin region,and that phytoplankton biomass was extremely low(mean=0.05±0.18 mg·m^(−3))in the near-surface layer(upper 25 m).More than 80%of Chl a values were<0.1 mg·m^(−3) in the water column(0-200 m),but high values appeared at the ice edge or in corresponding ice areas on the shelf.Additionally,the mean contribution of both nanoplankton(2-20μm)(41%)and picoplankton(<2μm)(40%)was significantly higher than that of microplankton(20-200μm)(19%).Notably,the typical subsurface chlorophyll maximum(0.1 mg·m^(−3))was found north of 80°N,where the concentration of sea ice reached approximately 100%.The Chl aprofile results showed that the deep chlorophyll maximum of total-,micro-,nano-,and picoplankton was located at depth of 40,39,41,and 38 m,respectively,indicating that nutrients are the primary factor limiting phytoplankton growth in the ice-covered Arctic Ocean during summer.These phenomena suggest that,despite the previous literatures pointing to significant light limitation under the Arctic ice,the primary limiting factor for phytoplankton in summer is still nutrient. | CAI Ting HAO Qiang BAI Youcheng LAN Musheng HE Jianfeng CHEN Jianfang | 2022 | Advances in Polar Science2022,33,3: | 0 |
