| 3 | Methylation Modifications in Eukaryotic Messenger RNA显示文摘RNA methylation modifications have been found for decades of years, which occur at different RNA types of numerous species, and their distribution is species-specific. However, people rarely know their biological functions. There are several identified methylation modifications in eukaryotic messenger RNA(mRNA), such as N7-methylguanosine(m7G) at the cap, N6-methyl-20-O-methyladenosine(m6Am), 20-O-methylation(Nm) within the cap and the internal positions, and internal N6-methyladenosine(m6A) and 5-methylcytosine(m5C). Among them, m7G cap was studied more clearly and found to have vital roles in several important mRNA processes like mRNA translation, stability and nuclear export. m6A as the most abundant modification in mRNA was found in the 1970s and has been proposed to function in mRNA splicing, translation, stability, transport and so on. m6A has been discovered as the first RNA reversible modification which is demethylated directly by human fat mass and obesity associated protein(FTO) and its homolog protein, alkylation repair homolog 5(ALKBH5). FTO has a special demethylation mechanism that demethylases m6A to A through two over-oxidative intermediate states: N6-hydroxymethyladenosine(hm6A) and N6-formyladenosine(f6A). The two newly discovered m6A demethylases, FTO and ALKBH5, significantly control energy homeostasis and spermatogenesis, respectively, indicating that the dynamic and reversible m6A,analogous to DNA and histone modifications, plays broad roles in biological kingdoms and brings us an emerging field 'RNA Epigenetics'. 5-methylcytosine(5mC) as an epigenetic mark in DNA has been studied widely, but m5C in mRNA is seldom explored. | 5 | Reversible RNA Modification N^1-methyladenosine(m^1A) in mRNA and tRNA显示文摘More than 100 modifications have been found in RNA. Analogous to epigenetic DNA methylation, epitranscriptomic modifications can be written, read, and erased by a complex network of proteins. Apart from N^6-methyladenosine(m^6A), N^1-methyladenosine(m^1A) has been found as a reversible modification in tRNA and mRNA. m^1A occurs at positions 9, 14, and 58 of tRNA, with m^1A58 being critical for t RNA stability. Other than the hundreds of m^1A sites in mRNA and long non-coding RNA transcripts, transcriptome-wide mapping of m^1A also identifies>20 m^1A sites in mitochondrial genes. m^1A in the coding region of mitochondrial transcripts can inhibit the translation of the corresponding proteins. Iron is the major coloring element. Mossbauer spectroscopy has shown that the iron of the glaze exists in the form of structural iron (Fe2+, Fe3+). The quantitative relationship between the main wavelength of every color glaze and the relative content (Fe2+/Fe3+) of structural iron has been determined. | 10 | The RNA N^(6)-methyladenosine demethylase ALKBH9B modulates ABA responses in Arabidopsis显示文摘The mRNA modification N^(6)-methyladenosine(m^(6)A)plays vital roles in plant development and biotic and abiotic stress responses.The RNA m^(6)A demethylase ALKBH9 B can remove m^(6)A in alfalfa mosaic virus RNA and plays roles in alfalfa mosaic virus infection in Arabidopsis.However,it is unknown whether ALKBH9 B also exhibits demethylation activity and has a biological role in endogenous plant mRNA.We demonstrated here that mRNA m^(6)A modification is induced by the phytohormone abscisic acid(ABA)and that ALKBH9 B has m^(6)A demethylation activity on endogenous mRNA.Knocking out ALKBH9 B led to hypersensitivity to ABA treatment during seed germination and early seedling development.We further showed that ALKBH9 B removes the m^(6)A modification in the ABA INSENSITIVE 1(ABI1)and BRI1-EMS-SUPPRESSOR 1(BES1)transcripts following ABA treatment,affecting the stability of these mRNAs.Furthermore,we determined that ALKBH9 B acts genetically upstream of the transcription factors ABI3 and ABI5,and its regulatory function in ABA responses depended on ABI3 and ABI5.Our findings reveal the important roles of the m^(6)A modification in ABA responses and highlight the role of ALKBH9 Bmediated m^(6)A demethylation in regulating ABA responses post-transcriptionally. | Jun Tang Junbo Yang Qiang Lu Qian Tang Shuyan Chen Guifang Jia | 2022 | Journal of Integrative Plant Biology2022,64,12: | 1 |

| 16 | Response of Chinese fir seedlings to low phosphorus stress and analysis of gene expression differences显示文摘Chinese fir(Cunninghamia lanceolata) is an excellent fast-growing timber species occurring in southern China and has significant value in the forestry industry. In order to enhance the phosphorus utilization efficiency in Chinese fir, four clones named X6, S3, S39 and FK were used, and low phosphorus(LP) stress experiments were performed to analyze the response of different clones to phosphorus deficiency. According to the results on seedling height, maximum root length, leaf blade aspect ratio, root ratio, malondialdehyde content, acid phosphates activity,proline content, soluble protein level, and chlorophyll a and b levels of the tested clones, compared to the control groups(CK), the phosphorus high efficiency clone X6 was screen out for transcriptome sequencing experiments. De novo RNA-seq was then used to sequence the root transcriptomes of X6 under LP stress and CK, and we then compared the gene expression differences under the two conditions. A total of 3416 SDEGs were obtained by comparing the LP and CK groups, among which 1742 were up-regulated and 1682 were down-regulated. All SDEGs obtained from the LP and CK treated samples were subjected to KEGG annotation and classification. Through classification statistical analysis using WEGO software,607 SDEGs obtained KEGG pathway annotations, which were related to 206 metabolic pathways. In Chinese fir subjected to LP stress, 53 SDEGs related with phosphorus metabolism, and phosphate uptake and transport were obtained from our transcriptome data. The present study analyzed the neu-roprotective effects of gastrodine by measuring EphB2 expression in rat neural cells cultured in vitro and injured by kainic acid. Gastrodine (12.5, 25, and 50 mg/L) improved morphology in kainic acid-injured neurons, reduced lactate dehydrogenase leakage, decreased neuronal apoptosis, and increased EphB2 expression in neuronal cells. A moderate dose of 25 mg/L gastrodine resulted in the most significant effects.
